1.Study on the differentiation of human mesenchymal stem cells into vascular endothelial-like cells.

To explore the feasibility of mesenchymal stem cells (MSCs) acting as seed cells in tissue engineering, we isolated human bone marrow MSCs and differentiated them into vascular endothelial-like cells (ELCs) in vitro. Bone marrow mononuclear cells (BMSCs) were isolated by the method of percoll density centrifugation, and seeded in Dulbecco Modified Eagle Medium supplemented with 10% fetal bovine serum. MSCs were purified through multiple adherent cultures, and differentiated into ELCs induced by endothelial cell growth medium-2 (EBM-2) medium containing vascular endothelial growth factor (VEGF), human fibroblast growth factor (hFGF), insulin like growth factors 1 (IGF-1), and human epidermal growth factor (hEGF). The relative biologic characteristics of ELCs including cell morphology and phenotype were studied by inverted microscope and flow cytometry. The induced cells were identified by immunofluorescence with CD31 and Von Willebrand factor (vWF). The results showed that the morphology of MSCs was long-spindle and vortex-like growth. After induction of differentiation, the cells were round, and similar to vascular endothelial cells (ECs). Flow cytometric analysis revealed that ELCs expressed ECs specific surface markers of CD31 and vascular endothelial cadherin (VE-cadherin), but not CD133. Immunofluorescence results also confirmed that ELCs expressed CD31 and vWF. The results suggested that ELCs possed similar cell biological characteristics with ECs. In one word, human MSCs derived from bone marrow have the potential to differentiate into ECs in vitro, and show clinical feasibility acting as ideal donor cells of vascular tissue engineering.

2.Surgical Treatment and Clinical Characteristics of Children′s Thalamic Tumors

Objective To study the clinical characteristics and effects of surgical treatment of children with thalamic tumors.Methods The clinical data of 22 cases were retrospectively studied and followed-up 6 months to 9 years.Results There were 13 boys and 9 girls,their ages ranging from 3 to 13 years.The average duration of symptoms before diagnosis about 2 months.Headache and papilledem were the most symptoms and signs,respectively.Most children′s thalamic tumors were low grade tumors with clear verge.In this group,good results were obtained that total remove 9 cases,subtotal remove 8 cases,partial remove 3 cases,biopsy 2 cases and no surgical death.Conclusions Clinical character of children thalamic tumors is distinct and good surgical results in the nearly future.The long results are determined by type of pathology.

6.Changes in the lymphocyte subgroup after TACE, THPP of primary hepatic carcinoma

Purpose: Observe the varities of lymphocyte subgroup of patients suffering from primary hepatic carcinoma ( PHC) during the treatment by hepatic arterial chemoembolization (TACE) and Hyperthemia Peritoneal Perfusion (THPP). Methods: Twenty seven patients were treated with TACE and THPP a week after TACE. We tested for lymphocyte subgroup three times with the flow cytometry (FCM) for pre-TACE, post-TACE and THPP a month later. 7.EFFECT OF QUERCETIN ON CULTURED HUMAN VASCULARENDOTHELIAL CELLS

Objective To study the effects of quercetin (Que) on the release of endothelin-1(ET-1) and prosta cylin(PGI2) by normal human vascular endothelial cell(VEC). Methods Radioimmunoassay(RIA) was used to assess the amount of ET-1 and PGI2 produced by VEC. VEC proliferation was assessed by tetrazolium(MTT) assay. Results Que increased the normal VEC proliferation at the concentration of 5, 20, 40, 80, 100μmol/L and increased the pro duction of PGI2 and inhibits the release of ET by the normal VEC at the concentration of 5, 20 and 80μmol/L. Que at the concentration of 5, 20 and 80μmol/L had no direct effect on morphology of the normal VEC. Conclusion Que can stimulate the proliferation of VEC and inhibit the release of ET-1 and increase the formation of PGI2. The data sug gest that Que might be beneficial for the prevention and treatment of vascular endothelial injury-related cardiovascu lar diseases, such as atherosclerosis and thromboembolism diseases.
8.Study on invasion-related factors of three human urinary bladder cancer cell lines

Objective To investigate the various mechanisms o f invasion in urinary bladder cancer. Methods The cloni ng efficiency and activity of adhesion of basement membrane were analysed. The f luidity of the membrane and the expression of E-cadherin, and MMP-2, and CD44 were carried out by fluorescence polarization, immunohistochemical staining and flow cytometry. Results The cloning efficiency of BLX and B LZ was 6.0% and 3.0% in soft agar, respectively. The colony of BLS was n ot formed in soft agar. The fluidity of the membrane was different in the three cell lines. The expressions of E-cadherin and CD44 in BLS and BLZ were greater than that in BLX cell line. Significant difference in MMP-2 was not found in th e three cell lines. Conclusion The research suggests that d ifference in fluidity of the membrane and expression of CD44 may play an importa nt role in invasion of urinary bladder cancer.
9.Video-assisted thoracoscopic surgery and conventional radical operation on stage Ⅰ , Ⅱ esophageal cancer

Objective To compare the results and safety between video-assisted thoracoscopic surgery ( VATS ) and conventional radical operation in patients with stage Ⅰ , Ⅱ esophageal cancer. Methods Retrospectively reviewed 43 patients with stage Ⅰ , Ⅱ esophageal cancer,underwent either VATS radical operation (VATS group,16 cases) or conventional radical operation (control group,27 cases ) from September 2007 to September 2009. Patient's operative characteristics and postoperative courses were compared between two groups. Results In VATS group the operation time was ( 115.6 ± 48.0) min,the peri-operative blood loss was ( 131 ± 71 ) ml,the first postoperative day chest lead quantity was (331 ± 170)ml, the time of postoperative chest tube was (7.25 ± 2.35) d,the postoperative 36 h visual analogue scale (VAS) was (3.4 ± 1.2) scores,the postoperative drainage of chest was ( 1281 ± 534) ml,the 72 h postoperative locomotor activity of right upper extremity was (5.1 ± 1.5) cm. While in control group was ( 145.6 ± 20.6)min, (292 ± 111 ) ml, (494 ± 194) ml, ( 10.00 ± 2.79 )d, (7.3 ± 1.4) scores, ( 1780 ± 731 ) ml, ( 15.6 ± 3.1 )cm respectively (P < 0.01 or < 0.05 ). The lymph node dissection number,the total cost of hospital between were no statistically significant differences in two groups (P >0.05). Conclusion Comparing with conventional radical operation, VATS radical operation for patients with stage Ⅰ , Ⅱ esophageal cancer appears to be as effective but less morbid.
10.Expression and Bioactivites of Recombinant Human Glucagon-like Peptide-1

The synthesized full-length hGLP-1 gene was cloned into pET-32a(+) to get the recombinant plasmid pET32-GLP-1, which could express a fusion protein containing thioredox, hexahistidine, and rhGLP-1 consecutively. The recombinant plasmid containing hGLP-1 was transformed into E.coli BL21 (DE3) and expressed by IPTG induction. The fusion protein was purified from lysates with Ni?IDA His?Bind affinity chromatography. rhGLP-1 with the purity of 90% was achieved after enterokinase digestion, Ni?IDA His?Bind affinity chromatography again, then was concentrated by ultrafiltration. The purified rhGLP-1 showed a single band on IEF gel with an isoelectric point between pH5.2 and pH5.85. ESI mass spectrometry showed that the molecular weight was 3355.0kDa as expected. rhGLP-1 was digested with trypsin followed by mass analysis and the peptide mapping produced two expected fragments with the molecular weights of 2097.7kDa and 1005.5kDa, respectively. The purified rhGLP-1 also showed obvious biological activity for both lowering plasma glucose and stimulating insulin secretion in mice.
