Objective To study the effects of atorvastatin on expression of lysyl oxidase(LOX) in myocardial tissue of rats with diabetic cardiomyopathy (DCM) and its mechanism.Methods Thirty DCM SD rats were randomly divided into 3 groups:DCM group,treatment group (atorvastain 2 mg · kg-1 · d-1 by gastric gavage) and β-aminopropionitrile group(β-aminopropionitrile 80 mg · kg-1 · d-1 by gastric gavage),10 cases in each group.Other 10 SD rats were selected as the control group.At the end of week 8,the rats were killed for extracting the myocardial tissue RNA and protein.Expression levels of LOX,MMP-2 and NF-κB mRNAs and proteins in myocardial tissue of DCM rats were measured by RT-PCR and Western blot.Results The expression levels of LOX,MMP-2 and NF-κB mRNAs and proteins in the DCM group were significantly higher than those in the control group (P＜0.01),and compared with the DCM group,the expression of BAPN LOX,MMP-2 and NF-κB mRNA and proteins in the treatment group were significantly deceased (P＜0.01).Conclusion Atorvastatin can reverse the expression of LOX in myocardial tissue of DCM rat,and then may regulate the expression of MMP-2 and NF-κB.

The growth-propagation and decolorization-degradation systems for the model dy e Biebrich Scarlet by Trametes hirsuta were established preliminar ily. It was showed that 30℃ and the static culture were better; the effects of compo nents in culture media on the efficiencies of decolorization and degradation wer e not significant; considering the convenience of observing the changes of dye c o lors and shortenig the culture period, the potato liquid medium had the advantag e of others as a preferable medium for reaction systems of Trametes hirsuta . The decolorization and degradation of all dyes Biebrich Scarlet and Direct Deep Blu e L-3RB, Reactive Blue X-BR, Basic Violet 5BN, and Methylene Blue by Tramete s hirsuta were better.

Objective To explore the relationship between severity of symptom and immune function in left-to-right shunt congenital heart disease (CHD) infants combined with pneumonia. Methods The clinical data of 45 left-to-right shunt CHD combined with pneumonia infants were retrospectively analyzed. The infants were divided into 2 groups according to the modified Ross score:mild group (0-6 scores, 28 cases) and severe group (7-12 scores, 17 cases);then, the infants were divided into 2 groups according to deformities:single deformity group (the infants had a kind of deformity such as ventricular septal defect, atrial septal defect and patent ductus arteriosus, 21 cases) and composite/complex deformity group (the infants combined 2 or more deformities or other malformations, 24 cases). Twelve cases of healthy check-up infants were selected as control group. The levels of lymphocyte subsets (including CD3+, CD4+, CD8+, CD4+/CD8+, NK cell and B lymphocyte) and IgG, IgA, IgM were detected in every group. Results There were no statistical differences in IgG, IgA, IgM, CD8 +, CD4 +/CD8 +, NK cell and B lymphocyte among mild group, severe group and control group or single deformity group, composite/ complex deformity group and control groups (P>0.05). The CD3+ in severe group was significantly lower than that in mild group and control group (0.59 ± 0.10 vs. 0.69 ± 0.13 and 0.69 ± 0.12), the CD4+was significantly lower than that in control group (0.34 ± 0.07 vs. 0.45 ± 0.09), and there were statistical differences (P<0.05). For CD4+, though there was statistically difference among single deformity group, composite/complex deformity group and control groups (P<0.05), no statistically significant difference was in multiple comparison among the 3 groups (P>0.05). The CD4 + in the composite/ complex deformity group was significantly lower than that in the single deformity group and the control group (0.34 ± 0.07 vs. 0.45 ± 0.15 and 0.45 ± 0.09), and there was statistical difference (P<0.05). Conclusions In left-to-right shunt CHD infants combined with pneumonia, the reduction of CD3+and CD4+may be one of the causes of serious illness.

The progressive injury of retinal ganglion cells ( RGCs) is a common occurrence in several eye diseases, which ultimately may lead to irreversible blindness. Currently, there are still no effective or ideal treatments for it in practice, however some recent studies show that stem cell transplantation may provide a promising new idea for neuroprotection and replacement of retinal ganglion cells. This paper will review the research progress of stem cell transplantation-based treatment.

Objective To observe the effects of T-lymphocyte function subsets,IL-4 and IFN-γ cell factor in different dose and stage after irradiation.Methoda The C57BL/6j mice were divided into shammed irradiation group and model groups.The radiation hurt model was induced by 60Co gamma rays(0.7,1.4,2.8 and 5.6 Gy).The changes of T-lymphocyte subsets CD3,CD4,CD8,IL-4 and IFN-γ in spleen cells were analyzed by flow cytometry in acute injury stage and recovery stage after irradiation.Results The lymphocyte subsets CD3 +,CD4+ and CD8 + decreased after irradiation,which were related to the irradiation dose.At 1 day after irradiation,the decreasing level of IFN-γ was higher than that of IL-4.When irradiation dose was over 2.8 Gy,IL-4 / IFN-γ showed a markedly increased compared with control group.At 25 days after irradiation,CD3 +,CD4+,CD8 +,CD4 +/CD8 +,IL-4 and IFN-γ recovered obviously,but they did not recover to the normal level of shammed irradiation group.Conclusions The depression of mouse immune function induced by γ-irradiation might be caused by changes of CD3,CD4,CD8 and CD4/CD8 ratio,especially the imbalance of IL4 and IFN-γ.

To establish induction and liquid culture system for hairy roots of Danshen (Salvia miltiorrhiza), Agrobacterium rhizogenes A4, LBA9402, 15834 as test bacterium were used to infect aseptic leaves of Danshen. The hairy roots were induced and positive transgenic hairy roots were selected with PCR using rolB and rolC as the target gene. Then hairy roots of S. miltiorrhiza were harvested and salvianolic acids were extracted with 70% methanol containing 1% formic acid. The content of salvianolic acid B (SalB) and rosmarinic acid (RA) were determined by HPLC. According to the above research results, the Danshen hairy roots induced by A. rhizogenes LBA9402 were inoculated into the following group of culture media: MSOH, MS, B5, and 6,7-V liquid media. Then the same methods of extraction and determination for the content of Danshen hairy roots were adopted. Last, the hairy roots of S. miltiorrhiza induced by A. rhizogenes LBA9402 were inoculated into the MSOH liquid media with different pH values. The content of salvianolic acid were extracted with 70% methanol containing 1% formic acid and determined by HPLC. As a result, three kinds of A. rhizogenes A4, LBA9402, 15834 could induce hairy roots and Ri plasmids were integrated into the genome of S. miltiorrhiza by PCR. Danshen hairy roots induced by A. rhizogenes LBA9402 and A4 produced much more salvianolic acid, which were (3.27 ± 0.37)% [including (1.04 ±0.36)% of RA and (2.22 ± 0.29)% of SalB] and (3.17 ± 0.20)% [including (0.92 ± 0.31)% of RA and (2.25 ± 0.26)% of SalB], respectively. Hairy roots induced by A. rhizogenes LBA9402 when they were cultured in MSOH liquid media produced much more salvianolic acid, which was (4.56 ± 0.36)%, including (1.12 ± 0.26)% of RA and (3.44 ± 0.23)% of SalB. Hairy roots induced by A. rhizogenes LBA9402 produced the most salvianolic acid when they were cultured in MSOH liquid media with the pH value 4.81, which was 4.85%, including 1.16% of RA and 3.69% of SalB. So Danshen hairy roots induced by A. rhizogenes LBA9402 and A4 produced much more salvianolic acid when they were cultured in MSOH liquid media with the pH value 4.81. The research had established the foundation on genetic engineering to improve the quality of S. miltiorrhiza.

ObjectiveTo investigate the effects of using transforming growth factor-β3 (TGF-β3) combined with dental pulp stem cells (DPSCs) on the regeneration of rabbit facial nerve.MethodsSixteen New Zealand adult rabbits were selected randomly.These rabbits were divided into three groups (TGF-β3 and DPSCs treated group,TGF-β3 control group and PBS control group.treatment group (silicone guidance channel with collagen protein sponge were filled with TGF-β3 and DPSCs),TGF-β3 control group (silicone guidance channel with collagen protein sponge were filled with TGF-β3) and PBS control group(silicone guidance channel with collagen protein sponge were filled with PBS).After 3 months,a series of examinations were performed,including gross morphology,histological staining,neuroelectrophysiological tset.ResultsIn the experimental group,the diameter of the regenerating nerve and near distal nerve stem were almost the same.There were no formation of neuroma.The adventitial angiogenesis was rich and with tough texture.3 months after operation,in the experimental group,the facial nerve membrane integrity,nerve fibers arranged in neat rows,form a more complete,myelin swelling.The total number of regeneration of nerve fibers in the experimental group were more than of control groups,statistical analysis was significant (P＜0.05).Diameter of regenerating nerve fibers in the experimental group were greater than that of control groups,statistical analysis was significant (P＜0.05).The ultra-thin section showed that the regenerated fibers in the treatment group were mainly myelinated never fiber.The layer structure of myelin sheath was clear,and there were rich organells axoplasma.The neuroelectrophysiological examinations revealed that the latency of nerve and muscle action conduction in the treatment group was shorter than that of the control groups,the treatment group:(1.96±0.32) ms,the TGF-β3 control group:(2.35±0.41) ms,the PBS control group:(3.42±0.55) ms.The wave amplitude of nerve and muscle action conduction in the treatment group was obviously higher than that of control groups,the treatment group:(11.06±3.25) mV,the TGF-β3 control group:(8.40± 1.68) mV,the PBS control group:(4.62±0.77) mV.ConclusionThe combination of TGF-β3 and DPSCs can improve the effects on the repair of facial nerve injury.

Objective To investigate the feasibility of combing the application of transforming growth factor-β3 (TGF-β3) with concentration of 100 ng/μl and dental pulp stem cells (DPSCs) for recovering rabbit facial nerve transverse trauma.Methods Thirty-six healthy adult Zelanian rabbits of clean-grade were selected and randomly divided into group DPSCs+TGF-β3 (experimental group), group TGF-β3 (control group 1) and group PBS (control group 2) with 12 rabbits in each group.The operations for all three groups were applied at rabbit's left cheek.A model of traumatic transection was set on upper buccal branch, then 100 ng/μl TGF-β3 solution and 0.1 ml of 1 ×108/L DPSCs suspension were added into regeneration chamber for the experimental group, while the same amount of 100 ng/μl TGF-β3 solution was added for group TGF-β3 and the same amount of PBS for group PBS.The recovery of facial nerve regeneration with the prepared animal's specimen was evaluated in the 1st, 4th and 12th week after the operation on sacrificed rabbits.Results The effects on nerve regeneration recovery for the experimental group was superior to that of control groups 1 and 2 with all the 36 models included in the result analysis, and that of control group 1 was superior to that of control group 2, which was getting better with the extension of time.Conclusions The combined application of TGF-β3 and DPSCs can effectively promote the facial nerve regeneration, which is better than that of single application of TGF-β3.Meanwhile, the effect with TGFβ3 application is better than that with application of PBS.

Objective To investigate and analyze cognitive similarities and differences of nurses factors that lead to the tense nurse-patient relationship from both attitudes of nurses and patients. Methods By using the method of questionnaire, 284 nurses and 305 patients were investigated respectively in a top'three hospital of a city. Results There was a significant difference in cognitive scores of nurses factors that lead to the tense nurse- patient relationship (P < 0.01), patients' scores were (3.90±0.47) points which were higher than (3.52±0.35) points of the nurses′. In dimensions of prioritization, both nurses and patients believed that technical ability, professional knowledge and service attitude were the main factors leading to the tense relationship between nurses and patients. While nurses held that communication ability was more important, patients held that the nurse′s own personality was more important. Conclusions There are similarities and differences of attitudes to nurs's factors between nurses and patients regarding tension in nurse- patient relationship. The tense relationship between nurses and patients is affected to a great extent by nurses' professional knowledge, technical ability and service attitude. Managers should objectively evaluate and fully pay attention to cognitive similarities and differences of both sides, strengthen nurses′theoretical knowledge and clinical skills training, improve service attitude, and develop good character,in order to provide high quality nursing service for patients.
