1.Comparison of histopathological staining methods for Cryptococcus neoforman

Objective To compare several histopathological staining methods in demonstrating Cryptococcus neoforman.Methods Hematoxylin-eosin(HE) method,periodic acid-Schiff(PAS) method,Alcian blue method and Grocott-Gomori's methenamine silver method were carried out in the lung and brain tissue samples from 7 autoptical cases identified with Cryptococcus neoforman infection.Results All these methods demonstrated Cryptococcus neoforman infection,HE method showed unclear pink cell body which were difficult to be found.Alcian blue method displayed blue picture,and PAS method showed violet-red body,and Grocott-Gomori's methenamine silver method presented clear dark black picture which was easy to be distinguished from other components.Conclusion Grocott-Gomori's methenamine silver method is the best method in staining Cryptococcus neoforman.
2.Expressions of CD44, CD44V3, CD44V6 and PCNA in human osteosarcoma and the clinical significance

Objective To explore the expressions of CD44, CD44V3, CD44V6 and PCNA and the relationship between the expressions and clinical pathologic changes of human osteosarcoma Methods The expressions of CD44, CD44V3, CD44V6 and PCNA in 15 osteochondromas and 40 osteosarcomas were detected immunohistochemically Results (1) The positive expression rates of CD44, CD44V3 and CD44V6 in osteosarcoma were markedly higher than those in osteochondroma (2) The positive expression rate of CD44V6 was higher than those of CD44 and CD44V3 in osteochondroma (3) The expression of CD44V6 was not involved in the pathological type or the prognosis in osteochondroma (4) The expressions of CD44 and CD44V6, particularly that of CD44V6, were closely involved in the PCNA positive rate Conclusion CD44V6 might act as a marker of malignancy of the bone tumors, but the relationship with the prognosis in malignant bone tumors waits to be further studied
3.Study on the map of plasmid of Y. pestis in the R. opimus natural plague foci in Junggar Basin

Objective To investigate the type of plasmid map of Y. pestis in the R. opimas natural plague loci in Junggar Basin. Methods A total of 39 plasmid DNA of Y. pestis which were isolated from the natural plague loci of Junggar Basin, Tianshan Mountain, Kunlun Mountain, Qinghai-Tibet Plateau and In-ner Mongolia were extracted by the methods of Kado and Liu. The plasmid map was analyzed by the methods of agarose gel eleetrophoretogram. Results Two types of plasmid map were found in 26 Y. pestis which were isolated from Junggar Basin. Of them 23 were 6 × 106, 45 × 106 and 65 × 106 type of plasmid map, and 3 were 6 × 106, 45 × 106 and 72 × 106 type. Conclusion There are two types of plasmid map in the R. opi-mus natural plague loci in Junggar Basin. One type, which is the dominant type in this area, is 6 × 106, 45 × 106 and 65 × 106 type. This type is also similar to the dominant plasmid map type of the nature plague loci of Tianshan Mountain, Kunlun Mountain, Qinghai-Tibet Plateau and Inner Mongolia. The other type is 6 × 106, 45 × 106 and 72 × 106 type, and this type is new plasmid map type of Y. pestis in our country.
4.Expression of survivin and its correlation with cell proliferation and axillary lymph node metastasis in breast cancer

Objective To investigate the expression of survivin and its correlation with tumor cell proliferation and axillary lymph node metastasis in breast cancer. Methods Expressions of survivin and proliferating cell nuclear antigen(PCNA) in 74 cases of breast cancer(34 cases with axillary lymph node metastasis) were measured by immunohistochemistry. Results (1) There was expression of survivin in 64.9% of breast cancer. (2) Labeling idex(LI) of PCNA in breast cancer cells with positive expression of survivin was significantly higher than that with negative survivin. (3) The positive percent of survivin expression in breast cancer with axillary lymph node metastasis was higher than that without axillary lymph node metastasis. Conclusion The expression of survivin in breast cancer is related to PCNA and axillary lymph node metastasis.
5.Malignant transformation of immortalized human fetal osteoblastic cell line hFOB1.19

Objective To establish a malignant transformed human fetal osteoblastic human cells from the immortalized cell line hFOB1.19 in order to explore the molecular mechanism in tumorigenesis of osteosarcoma. Methods hFOB1.19 cells were treated sequentially by an initiated factor, N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) and a promoter, 12-O-tetradecanoyl phorbol-13-acetate (TPA). The features of malignancy of transformed cells were identified by cell morphology, DNA content analysis, colony forming frequency on soft agar and tumorigenetic test in nude mice. Results Continuous passaging after the treatments resulted in the formation of a few paramorph foci and exhibited in an extensively random orientation. The poly-ploid of DNA was 81.08% in experiment group, much higher than that in control group (55.03%). Compared with that of negative control cell, colony formation efficiency of transformed cells in semisolid agar showed a a significant increase. The transformed cells formed tumors subcutaneously in the nude mice, which were verified to be poorly differentiated osteosarcoma histopathological examination. Conclusion Mocking the process of malignant transformation of human cells, we establish malignant transformed immortalized human fetal osteoblastic human cells (hFOB1.19) model.
6.Comparison of the Inhibition Effects of Arsenacetylic Acid on Experimental H_(22) Hepatoma-Bearing Mice in Two Different Forms of Administration

OBJECTIVE: To compare inhibition effects of arsenacetylic acid(ASAC) on experimental H22 hepatoma-bearing mice in different forms of administration. METHODS: The mice were divided into 5 groups at random after inoculated with H22 hepatoma into their right axillas hypodermic by intraperitoneal injection(ip) and intravenous injection(iv), and then injected with normal saline,cyclophosphamide and different dosage of ASAC, observe the rate of tumor strain becoming tumor, the inhibition effects of the subjects and the effects of the subjects on mice's viscera. RESULTS: Compared with ip,either high, moderate or low dosage of ASAC by iv did have an obvious inhibitory effect on tumor and the tumor inhibition rates were 46.59%, 46.31% and 32.48% respectively; however, the spleen index in groups that of lower dosage of ASAC by two forms of drug administration were increased; there were death by poisoning in the group that iv with high dosage of ASAC.CONCLUSION: Compared with ip, the administration of ASAC by iv has a better effect on tumor.

Epiretinal membrane (ERM) is a relatively common macular disease that forms along the surface of the internal limiting membrane (ILM) of the retina to some reason.The pathogenesis is not clear.Microincision vitrectomy surgery has been confirmed as a minimally invasive and very safe modality of treatment.Deciding when to perform a vitrectomy can be difficult.There are many factors which can affect the postoperative visual acuity,such as age,the thickness of macular,integrity of photoreceptor inner segment/outer segment (IS/OS) junction.
8.Application of stem cell transplantation in repair of the retinal ganglion cells injury

The progressive injury of retinal ganglion cells ( RGCs) is a common occurrence in several eye diseases, which ultimately may lead to irreversible blindness. Currently, there are still no effective or ideal treatments for it in practice, however some recent studies show that stem cell transplantation may provide a promising new idea for neuroprotection and replacement of retinal ganglion cells. This paper will review the research progress of stem cell transplantation-based treatment.

10.Effect of nordihydroguaiaretic acid on expressions of VEGF and its receptor KDR in vitro

Objective To explore the effect of nordihydroguaiaretic acid (NDGA) on the expressions of vascular endothelial growth factor (VEGF) and its receptor, kinase-inserted domain containing receptor(KDR) and the possible mechanism. Methods The expression of VEGF in human malignant glioma cell line SHG-44 and that of KDR in human umbilical vein endothelial cell (HUVEC) line ECV-304 were observed 1~3 d after NDGA treatment with immunohistochemistry, in situ hybridization and image analysis. Results The expression of VEGF was declined at protein or mRNA levels in SHG-44 cells after treated with 100 μmol/L NDGA for 1 to 3 d. The expression of KDR in endothelial cells with 100 μmol/L NDGA treatment for 1 to 3 d was decreased too, in a more obvious way compared with the decline of VEGF expression in SHG-44 cells. Conclusion The results suggest that NDGA inhibits the expression of VEGF in glioma cells as well as that of VEGF receptor KDR in endothelial cells, which may be the important molecular mechanism of anti-angiogenesis of NDGA.
