1.Evaluation on efficacy-toxicity correlation of aqueous extracts from Aconiti Lateralis Radix Praeparata on diarrheal model rats based on "warmly invigorating spleen Yang" efficacy.

The diarrheal rat model was established by orally administering senna. The preventive experiment was concurrent for 6 days. The treatment experiment modeling had lasted for 12 days. The administration started at the 7th day, and lasted for 6 days. During the experiment, efforts were made in symptom score and weighing. After the experiment, hearts, livers, spleens, kidneys, brains, adrenals and thymuses were collected and weighed, lactic dehydrogenase (LDH) and cardiac troponin-I (cTn-I) in serum were detected. The efficacy of aqueousextracts from Aconiti Lateralis Radix Praeparata in preventing and treating rats with diarrheal and its accompanying toxicity were respectively studied. The result shows that aqueous extracts from Aconiti Lateralis Radix Praeparata could improve syndromes of rats with diarrheal. The 50% effective doses (ED50) of preventive and treatment administrations were 1.420 4 g · kg(-1) and 1.048 9 g · kg(-1), respectively. Aqueous extracts from Aconiti Lateralis Radix Praeparata could decrease the ratio of heart to body weight, and increase serum LDH and cTn-I. It was concluded that aqueous extracts from Aconiti Lateralis Radix Praeparata had a specific preventive and treatment effect on rats with diarrheal caused by senna, but with specific toxicity on heart.

3.Erythromycin for improving enteral nutrition tolerance in adult critical patients: a systematic review and Meta-analysis

Objective To systematically review the efficacy and safety of erythromycin on enteral nutrition (EN) tolerance in adult critical care patients.Methods Databases including PubMed,EMbase,the Cochrane Library,CNKI and Wangfang data were retrieved up to June,2013 to collect the randomized controlled trial (RCT) concerning erythromycin in improving EN tolerance or increasing the successful rate of postpyloric EN tube as compared with other treatments.Two reviewers independently screened the literature,extracted the data,and assessed the quality of methodology.Then Meta-analysis was performed using RevMan 5.2 software.Results A total of 16 RCTs were included.Ten RCTs involving 668 patients were included for evaluating erythromycin in improving EN tolerance.Six RCTs involving 353 patients were included for evaluating erythromycin to increase the successful rate of postpyloric EN tube.The result of Meta-analysis showed that compared with placebo,erythromycin could significantly improve the successful rate of postpyloric EN tube placement [relative risk (RR) =1.82,95% confidence interval (95%CI) 1.40-2.37,P<0.000 01],while there was no significant difference between erythromycin and metoclopramide (RR=1.04,95% CI 0.79-1.36,P=0.799).In patients who needed early EN,compared with placebo or blank control,erythromycin had higher successful gastric feeding rate over 5 days (RR =1.89,95% CI 1.19-3.00,P=0.007).In patients who failed EN,compared with metoclopramide,erythromycin could significantly increase the successful gastric EN rate for 24 hours (RR=1.30,95%CI 1.02-1.66,P=0.03),72 hours (RR=1.57,95%CI 1.15-2.14,P=0.005) and 144 hours (RR=2.04,95%CI 1.23-3.37,P=0.006).The median time of EN intolerance was postponed in erythromycin group than that in metoclopramide group.Adverse reactions were reported in 5 studies.There was no statistic difference except for the higher diarrhea rate in the combination treatment group compared with erythromycin group.Conclnsions Postpyloric EN tube placement rate can be improved by erythromycin,which could be a choice of substitute for bedside intubation without fluoroscopy or endoscopic assistance.Based on the evidence,we recommended that intravenous erythromycin in a small dose of approximately 3 mg/kg weight as an option for EN intolerance in critical patients.
4.An experimental study on permeability of basic fibroblast growth factor through placental barrier in rats

Objective: To investigate if bFGF can penetrate placental barrier. Methods: Sixteen day pregnant Wistar rats were selected. bFGF labeled with 125 I was injected peritoneally into the rats. The radioactivity of bFGF in different organs were determined in 30 min. Results: (1) 125 I bFGF was detected in the brain, heart, liver,lung and spleen. (2)With the same dose of 125 I bFGF, the concentration of it in the brain was at lowest level of all other organs.(3) In the range of safe dose, the permeability of bFGF through placental barrier was increased obviously. Conclusion: bFGF may penetrate placental barrier into rat's brain, which makes possible for the therapeutic intervention of bFGF in feotus. [
5.Study on anti-inflammatory efficacy accompanied by side effects of different components of Sophorae Tonkinensis Radix et Rhizoma.

OBJECTIVETo investigate the anti-inflammatory efficacy accompanied by side effects of water extract and alcohol extract of Sophorae Tonkinensis Radix et Rhizome (STRR), their molecular mechanism, and interpret the relationship of "toxicity-effect" of toxic medicine.
METHODThe ear swelling by croton oil and granuloma by agar test models were used, water extract and alcohol extract of STRR of different dosages were administrated ig to mice to observe the assident toxicity, at the same time the activities of ALT, AST and the content of SOD, MDA,PEG2, NO, NOS, Cr, BUN, GSH, TG and Gn in serum were tested.
RESULTBoth water extract and alcohol extract of STRR have a strong inhibitory effect on ear swelling by croton oil and granuloma by agar. The activities of ALT, AST in serum were higher than that of normal group. SOD, MDA, PEG2, NO, NOS, GSH, TG and Gn had obvious changes.
CONCLUSIONBoth water extract and alcohol extract of STRR had an anti-inflammatory effect on acute and chronic inflammation. At the same time, side effects and liver toxicity. The anti-inflammatory effect of STRR in probable relation to the reduced inflammatory mediators release. Oxidative damnification might be one of the liver injury mechanism.

6.Improving Effect of Phenylethanoid Glycosides from Tibetan Medicine Phlomis younghusbandii on Rats with Acute High-altitude Cerebral Edema

OBJECTIVE:To investigate the improving effect of phenylethanoid glycosides (PhGCs) from Tibetan medicine Phlomis younghusbandii on rats with acute high-altitude cerebral edema. METHODS:60 Wistar rats were randomly divided into a normoxia control group (isometric sterile water for injection),a hypoxia model group (isometric sterile water for injection),a dexamethasone group(4 mg/kg),and three groups of PhGCs at high(400 mg/kg),middle(200 mg/kg)and low(50 mg/kg)dos-es,with 10 rats in each group. The rats were given drugs,ig,6 d before the establishment of models. On the 4th day of administra-tion,ig,the rats in all groups except the normoxia blank group were placed in a simulated 8 000 m altitude plateau environment for 72 h hypoxic exposure to establish the rat models of high-altitude cerebral edema. Following HE stain,the pathological changes in rats’brain tissues were observed under the light microscope. Dry-wet proportion method was used to determine the water con-tents in rats’brain. The content of MDA and the activities of SOD and GSH in rats’brain tissues were detected. Enzyme-linked im-munosorbent assay was adopted to determine the contents of IL-1β and TNF-α in rats’serum and brain tissues. RESULTS:Com-pared to the rats in the normoxia control group,those in the hypoxia model group showed obvious brain edema,and thickened lacu-nas around cells and vessels and inflammatory cell infiltration, higher water contents and MDA and weaker activities of SOD and GSH in brain,and higher contents of IL-1β and TNF-α in serum and brain tissues. There were statistically significances (P<0.01 or P<0.05). Compared to the rats in the hypoxia model group,those in the groups of PhGCs at high,middleand low dosages demonstrated less inflammatory cell infiltration and lower water contents in brain tissues,in which the groups of PhGCs at high and middle dosages demonstrated lower content of MDA and stronger activities of SOD and GSH in brain tissues, and lower contents of IL-1β and TNF-α in serum and brain tissues. There were statistically significances (P<0.01 or P<0.05). CONCLUSIONS:PhGCs can obviously alleviate the acute cerebral injury in rats which is caused by acute hypoxia and has im-provement effect to some degree on the rats with acute high-altitude cerebral edema.

Objective To study the expression of p53,p16,PCNA protein in esophageal carcinoma and its relationship to sexual distinction,the location of disease,the biological level,the depth of invasion and lymph node metastasis.Methods 118 patients with esophageal carcinoma were included in the study,all of them were treated for the first time.p53,p16 and PCNA protein in the 118 cases of esophageal carcinoma were detected by immunohistochemical assay(SP technique). 8.Study on effect of aqueous extracts from aconite on "dose-time-toxicity" relationships in mice hearts.

OBJECTIVETo study the effect of single administration of aqueous extracts from aconite on "dose-toxicity" relationship and "time-toxicity" relationship of mice hearts, through changes in electrocardiogram (ECG) and serum biochemical indexes.
METHODMice were grouped according to different drug doses and time points, and orally administered with water extracts from aconite for once to observe the changes of mice ECG before and after the administration, calculate visceral indexes heart, liver and kidney, and detect levels of CK, LDH, BNP and CTn-I in serum.
RESULTAccording to the "time-toxicity" relationship study, at 5 min after oral administration with aqueous extracts from aconite in mice, the heart rate of mice began rising, reached peak at 60 min and then slowly reduced; QRS, R amplitude, T duration and amplitude and QT interval declined at 5 min, reduced to the bottom at 60 min and then gradually elevated. The levels of CK, LDH, BNP and CTn-I in serum elevated at 5 min and reached the peak at 60 min, with no significant change in ratios of organs to body at different time points. On the basis of the "dose-toxicity" relationship, with the increase in single dose of aqueous extracts from aconite, the heart rate of mice. QRS, T duration and amplitude and QT interval declined gradually, and levels of CK, LDH, BNP and CTn-I in serum slowly elevated, with a certain dose dependence and no significant change in ratios of organs to body in mice.
CONCLUSIONSingle oral administration of different doses of aqueous extracts from aconite could cause different degrees of heart injury at different time points, with a certain dose dependence. Its peak time of toxicity is at 60 min after the administration of aqueous extracts from aconite.

9.A meta-analysis of efficacy and safety of compound glycyrrhizin combined with tripterygium wilfordii polyglycosides in treating psoriasis

Objective:To evaluate the clinical efficacy and safety of compound glycyrrhizin combined with tripterygium wilfordii polyglycosides for psoriasis.Methods:Literature search was conducted in CNKI, Wanfang, VIP databases, Sinomed, PubMed, the Cochrane Library and Embase with the deadline of September 28th, 2020. The randomly controlled trials of compound glycyrrhizin combined with tripterygium wilfordii polyglycosides in treating psoriasis was included. The publication bias and methodology quality of RCTs were evaluated according to the Cochrane 5.0.1 bias risk assessment method. The RevMan 5.3 software was used for meta analysis.Results:Ten studies with 961 patients were included, with the experimental group ( n=486) and the control group ( n=475). The results of meta-analysis showed that the total effective rate [ OR=3.16, 95% CI (2.00-4.99), P<0.001], the cure rate [ OR=2.56, 95% CI (1.94-3.61), P<0.001], the recurrence rate [ OR=0.15, 95% CI (0.04-0.60), P=0.007], the total incidence of adverse reactions [ OR=0.53, 95% CI (0.34-0.82), P=0.004], the incidence of abnormal liver function [ OR=0.17, 95% CI (0.06-0.47), P=0.001] in the experimental group were better than those in the control group. There was no significant difference on the incidence of adverse reactions of digestive tract ( P>0.05) or lower extremity or facial edema ( P>0.05) between both groups. Conclusions:Compound glycyrrhizin combined with tripterygium wilfordii polyglycosides is effective in the treatment of psoriasis than tripterygium wilfordii polyglycosides alone, and can reduce the recurrence rate and the incidence of adverse reactions. However, a large number of large samples and high-quality studies are still needed.
10.Meta-analysis of risk factor on hypertension in China.

OBJECTIVETo evaluate the risk factor of hypertension in China.
METHODSThe results of 24 case-control studies from 1989 to 2001 were analyzed by Meta-analysis method in this study. Random effect model (D-L method) and fix effect model (M-H method) were applied for data processing.
RESULTSThe pooled OR values (95% CI) of drinking, smoking, over-intake of salt, family history of hypertension, quickness to temper and overweight were 1.194 (1.184-1.203), 1.100 (1.096-1.104), 1.165 (1.157-1.173), 2.662 (2.550-2.779), 2.524 (1.902-3.348), 1.616 (1.600-1.633), respectively.
CONCLUSIONDrinking alcohol, smoking, over-intake of salt, family history of hypertension, quickness to temper and overweight were the important risk factors of hypertension in China.
