1.Relationship between Organochlorine Pesticides Exposure after Tangshan Earthquake and Breast Cancer:a Casecontrol Study

Objective To explore the relationship between organochlorine pesticides exposure after Tangshan earthquake and the breast cancer. Methods 1∶1 matched case-control study was conducted in 150 newly diagnosed cases of breast cancer with histopathological diagnosis from three city-level hospitals in Tangshan,China and 150 matched controls at the same hospitals,without the related diseases. The levels of derivatives of DDT and HCH in serum were measured by gas chromatography (GC) method and epidemiological survey were conducted by questionnaire for 150 pairs of cases and controls. The relationship between the organochlorine pesticides metabolites and the breast cancer of the exposed population was analyzed by conditional logistic regression model by SAS 8.2 Software. Results Compared with the control,significant higher rates of detection of p,p'-DDD and o,p'-DDT were seen in the case group and the levels of p,p'-DDE,?-HCH and ?-HCH in serum were higher in the case group,and there was a significant difference in serum residues levels of ?-HCH,p,p'-DDE and o,p'-DDT in different exposure areas in Tangshan,the high exposure area showed higher residues levels than the low exposure area. Multivariate conditional logistic regression analysis showed high pesticides exposure after the earthquake,the serum residues of p,p'-DDE and p,p'-DDT were associated with breast cancer risk,ORs were 6.592(95%CI:1.402-31.002),2.405(95%CI:1.033-5.600)and 1.845(95%CI:1.028-3.353) respectively. Conclusion Exposure to environmental organochlorines may be one of the risk factors for breast cancer; Higher pesticides exposure after the earthquake may increases breast cancer risk in Tangshan area.
2.Stereological observation without bias of the effects of exercises and loads on bone morphology in rats

BACKGROUND: Bone morphological stereological observation is limited due to the difficulty in human experimental bone access. Animal studies overcome the limitation of human experiments and can directly evaluate bone quality through bone stereological histomorphometric parameters.OBJECTIVE: To explore the mechanism of the impacts of exercise on long bone development in adolescents through exercise and load experiments in rats.DESIGN: A randomized controlled study by employing experimental animals as subjects.SETTING: Laboratory of Sports of Beijing University of Physical Education and the Key Laboratory of Orthopaedics of Xijing Hospital Affiliated to the Fourth Military Medical University of Chinese PLA.MATERIALS: The study was conducted in the Laboratory of Sports of Beijing University of Physical Education and the Key Laboratory of Orthopaedics of Xijing Hospital Affiliated to the Fourth Military Medical University of Chinese PLA. Totally 120 male 4-week old SD rats were randomly paired into 12 groups with 10 rats each based on their body mass.METHODS: Incremental exercises with the combination of suspension and loading mode of rats were used in the study. The undecalcified slices of the right tibia were observed after double-fluorescence-labeling by tetracycline (TC) and calcein for the analysis of bone histomorphometric parameters including bone mass, structure and metabolism, etc. During the period of growth under different exercise strength and different modes of mechanic load by stereological sampling without bias.static or dynamic parameters of bone trabecula among groups of different modes for mechanical load.RESULTS: The increase or decrease of multiple parameters including bone mass, bone quality, bone structure and bone metabolism in low-loading exercise group were more regular than that of high-loading exercise group and control group. In the suspension group, the bone mineral apposition significantly reduced, and the relative volume of bone trabecula significantly decreased with the significantly increased thickness, decreased numbers and significant of increased separation. The biochemical indicators of bone metabolism and the biomechanical capability of suspension group were significantly lower than that of low-loading exercise group but similar to that of control group. All parameters of loading group were similar to those of low-loading exercise group with the characters of active osteogenesis and osteo-absorption but mainly osteogenesis, and significantly enhanced abilities of anti-destruction and anti-deformity in bone.CONCLUSION: Exercises of different strengths can produce different impacts on long bone development in rats of growth period. The integrated effects of low-loading exercises in bone metabolism, bone mass, bone histomorphology and bone biomechanics are better than those of high-loading exercises. This phenomenon suggests that appropriate increment of gravity load during the exercise processin children and adolescents is good for the improvement of the capability of bone metabolism and bone mechanics, which further improve the bone quality during development.

The present study shows that mushroom juice exhibits a significanteffect on growth of L. acidophilus in reconstituted skim milk. When 5% - 10% mushroom juice was added in skim milk, they shorten generation time and the rate of lactic acid production was accelerated. After 10h incubation at 37℃ cell counts of L. acidophilus in test groups were much more than in control group, were up to 108/mL.
4.The antidiabetic and hepatoprotective effects of magnolol on diabetic rats induced by high-fat diet and streptozotocin.

The effects of magnolol (Mag) on hyperglycemia and hyperlipemia, hepatic oxidative stress and cytochrome P4502E1 (CYP2E1) activity of diabetic rats induced by high-fat diet (HFD) and streptozotocin (STZ) were studied. After oral administration of Mag (25, 50 and 100 mg x kg(-1) x d(-1)) for continuous 10 weeks, the blood glucose and lipids (TC, TG and LDL-C) levels, as well as the hepatic CYP2E1 activity and MDA content of diabetic rats, decreased significantly (P < 0.05 or P < 0.01), whereas the oral glucose tolerance and hepatic antioxidant enzymatic activities (CAT and GSH-Px) of diabetic rats, increased significantly (P < 0.05 or P < 0.01). The results indicated that Mag was effective against the hepatic oxidative damage, hyperglycemia and hyperlipemia of diabetic rats induced by HFD and STZ, and the inhibition of Mag on hepatic CYP2E1 activity could be an important mechanism of Mag against hepatic insulin resistance and oxidative damage.

6.Survey of Relationship between Symptoms and Dietary Factors in 54 Outpatients with Irritable Bowel Syndrome

Background:Irritable bowel syndrome(IBS)is a commonly seen functional intestinal disorder. Dietary factors are important factors affecting the abdominal symptoms of IBS patients. Aims:To investigate the effect of dietary factors on abdominal symptoms of IBS and the application of dietary adjustment strategy in patients lived in Xian,Shaanxi Province and the surrounding areas. Methods:Fifty-four outpatients with abdominal complaints who met the Rome Ⅲ criteria of IBS from Feb. 2015 to Jul. 2015 at the Second Affiliated Hospital of Xian Jiaotong University were enrolled. Twenty healthy volunteers were served as controls. All subjects completed a questionnaire containing issues mainly on IBS symptoms, eating habits,dietary factors causing abdominal symptoms,and the patients’coping styles. Results:Compared with healthy controls,more rice and less spicy food,fatty food,raw/ cold food,acidic beverages and alcohol drinking were observed in diet of IBS patients(P all < 0. 05). Abdominal symptoms of IBS patients could be caused or exacerbated predominantly by cold food(55. 6% ),fatty food(55. 6% ),raw food(48. 1% ),meat(48. 1% )and spicy food (40. 7% ). More than half of the IBS patients(63. 0% ) adopted an elimination diet to minimize their abdominal symptoms. Spicy food(59. 3% ),raw food(48. 1% ),cold food(44. 4% ),fatty food(44. 4% )and meat(37. 0% ) were the most frequently eliminated classes of food. Conclusions:Dietary factors can affect the abdominal symptoms of IBS in patients lived in Xian,Shaanxi Province and the surrounding areas and elimination diet based on foods they perceive as problematic is widely used. The classes of food and eating habits causing abdominal symptoms are quite different between domestic and overseas IBS patients.

Objective Explore reflux esophagitis age,symptoms,and the relationship between the degree of esophageal damage.Methods Summing up 198 cases of young and middle-aged patients and 104 cases of elderly pa-tients with the symptom score and Endoscopic mucosal injurs classification.Results Under the old group gastroscopy serious esophagitis more than the young group,old age group was 49.97%,25.25%for the young group,but the clini-cal symptoms of heart burn not obvious.Conclusion Reflux esophagitis damaged esophagus increased with age in-crease,but clinical heartburn symptoms do not necessarily obvious,it should attach importance to the brink throat dis-comfort,and other symptoms.
8.The involvement of outward potassium currents in methamphetamine-induced hippocampal neuron damage in fetal rat

Objective To investigate the effects of methamphetamine (Meth) on the outward K+ currents and elucidate the role of outward K+ channels in Meth induced hippocampal neuron damage.Methods Hippocampal neurons were harvest from 18-day-old embryonic rats and were divided into two groups:the control group and the Meth treated group.Both of 4-AP and TEA sensitive K+ currents were recorded after the treatment of Meth by performing the whole cell patch clamp.Furthermore,the MTT and TUNEL assays were performed to evaluate the effects of K+ channel on hippocampal neuron damage mediated by Meth.For statistical comparison,One-way ANOVA and LSD multiple comparison test or t-test was used.P-value < 0.05 was considered to be statistically significant.Results The density of 4-AP sensitive K+ channel currents in Meth treated group [(120.1 ± 19.6) pA/pF,n =7] were significantly increased when compared with control group [(87.4 ± 12.5) pA/pF,n =10,P <0.01] and the increments of the currents induced by Meth was dose dependent.The MTT data showed that the cell viability was obviously decreased in Meth treated group (48.72 ± 4.38) % relative to the control group (100.07 ± 3.36) %.Moreover,application of K+ channel antagonist,4-AP (61.39 ± 3.15)%,and the high K+ solution (78.25 ± 9.42) % substantially enhanced the cell viability.The TUNEL assay showed there were protective effects of 4-AP and the high K+ solution against neuron damage observed during cells exposed to Meth.Conclusions The increments of 4-AP sensitive K+ channel currents induced by Meth might be involved in hippocampal neuron damage.
9.Preparation and evaluation of quality control serum for antinuclear antibody testing

Objective In order to further enhance the repeatability and comparability for antinuclear antibody testing results in clinical laboratory,explore the reliable preparation method,and develop the quality control serum that is capable of meeting the needs of routine antinuclear antibody testing.Methods According to selection requirements,the serum from patient which is used to test antinuclear antibody was selected,and then we prepared the quality control serum.through evaluation of index in appearance of serum,aseptic level,uniformity and stability in intra-assay and inter-assay etc,the study optimized the method and condition for preparation,also confirmed the performance of prepared quality control serum.Results Prepared quality control serums are able to preserved stable at-20 ℃ 270 d,it can be keep stable more than 8 h after rewarming at room temperature.There are no significant differences between the commercialization of quality control performance and the freez-thawed uniformity and stability in intra-assay and inter-assay,it are keep consistent to result that before freezing testing.Conclusions Suitable methods and experimental conditions are explored for preparation of antinuclear antibody testing quality control substance.Each batch of prepared quality control substance has good uniformity and stability.The prepared quality control serum are able to meet needs of antinuclear antibody testing,and meet quality control requirements.
10.Clinical Significance of Lymphocyte Subsets Change in Breast Cancer Patients before and after Treatment

Objective To investigate the changes of lymphocyte subsets before and after the treatment with operation and chemotherapy in patients with breast cancer.Methods Using cytometer to measure lymphocyte subsets of CD3+,CD4+, CD8+,CD4+/CD8+ and NK of 25 breast cancer patients before and after treatment.The all 25 patients were treated with operation and chemotherapy after pathological diagnosis.The article compared the figures of T cell subsets before and after treatment.Results Compared with before operation and chemotherapy,the CD8+ statistically decreased after 1~3 months of treatment (t=2.57),and NK statistically increased after 3~6 (t=-2.23).After 6 months of treatment,the patients’ CD3+(t=-2.25),CD4+(t=-2.07)were statistically lower and NK (t=-3.56)was statistically higher.There was no significant difference for other items.Conclusion Lymphocyte subsets test against breast cancer patients can reflect immuni-ty level at a certain level during treatment.But how to explain the figures in different stage against treatment effectiveness and prognosis need further research.
