1.Preparation of Metoprolol Succinate Sustained-Release Tablet and Its Content Determination

Objective To establish a method of preparing metoprolol succinate sustained-release tablet and its content determination. Methods The formulation was optimized through the orthogonal design test by using release rate of the drug as an indicator.The different batches of metoprolol succinate sustained-release tablets were determined by HPLC. Results The tablets could release drug steadily and slowly as designed,which was similar to imported tablets. The linear range of metoprolol succinate was 10-70 μg?mL-1( r=0.999 8) . Conclusion The releasing rate of metoprolol succinate sustained-release tablet prepared in optimum condition can meet the requirement. This preparation technology is simple, the assay method is rapid, sensitive and reproducible.
2.Effect of ketamine on plasma adhesion molecules and myocardial injury in patients undergoing cardiac valve replacement

Objective It has been shown that ketamine can effectively suppress expression of cell adhesion molecules in vitro. The purpose of this study was to investigate the effect of ketamine on plasma adhesion molecules in vivo during cardiac valve replacement.Methods Twenty ASA Ⅲ-Ⅳ patients (7 males, 13 females) aged 23-65 yrs, weighing 31-77 kg undergoing cardiac valve replacement were randomly divided into two groups of 10 patients each : ketamine group received ketamine 2 mg?kg-1 i.v. before induction of anesthesia and at the start of CPB and control group received normal saline instead of ketamine. Blood samples were taken from radial artery for determination of plasma P-selectin, L-selectin, ICAM-1, IL-6 and cTnI concentrations before induction of anesthesia (T0 ,baseline) , 20 min after initiation of CPB (T1) , 30 min after aortic declamping (T2) 4 h and 24 h after CPB (T3, T4) . Results There was no significant difference between the two groups in age, weight, operation time, CPB time and aortic cross-clamping time. Objective: To observe the protective effects of Yougui drink(右归饮) on chronic renal failure rats with insufficiency of kidney-Yang(肾阳虚) and investigate its mechanisms.Methods: Chronic(renal) failure model with insufficiency of kidney-Yang was replicated by nephrectomy(5/6 renal cortex) and(propylthiouracil)(PTU) infused into the stomach.The model rats were randomly divided into three(groups:)(model) group,Yougui drink group and sham-operated group.Yougui drink group was given Yougui drink(infused) into stomach daily,while the other two groups were given equivalent water daily for 28 days.The(pathological) study of the kidney tissue was performed and the serum blood urea nitrogen(BUN),creatinine(Cr) and lipid levels were determined after the experiment.Results: Yougui drink significantly decreased serum BUN and Cr levels and increased the reduced triglycerides(TG) in the rats compared to model group(BUN:(13.65?1.37)mmol/L vs.(15.88?1.16) mmol/L,P
4.The role of cell apoptosis in glioma chemotherapy evaluation

Objective To study the role of cell apoptosis during the chemotherapy of human gliocytoma in order to get effective suvilliance on the effect of chemotherapy. Methods Gliocytoma cells were isolated and cultured from 40 human gliocytoma samples. Mitochondrial membrance potential (MMP), cell cycle, the level of Bcl-2 and Bax were detected by flow cytometry (FCM) at 24, 48, 72 hours respectively of incubation with Lomustine (CCNU) and Teniposide (VM-26), and the trends were also analysed. Results MMP decreased greatly, the apoptosis part in the cell cycle ananlysis increase, the expression level of Bcl-2 decreased, and that of Bax increase rapidly, while the Bcl-2/Bax ratio decreased. Conclusions CCNU and VM-26 have singnificant effect in gliocytoma chemotherapy on inducing gliocytoma cell apoptosis. VM-26 has more stronger effect on the cell cycle. MMP is the most sensitive and the fastest index in apoptosis detection.
5.Effects of Rhizoma smilacis glabrae,on cellular and humoral immune responses

A remarkabale inhibitory activity was exhibited by the aqueous extract from Rhizoma smilacisglabrae(RSG) against both picryl chloride(PC)- induced contact dermatitis and sheep red bloodcells (SRBC)- induced footpad reaction.The effect of RSG was displayed more distinctlywheng given after than before the 2nd antigen challenge.RSG also showed a marked anti-in-flammatory activity against xylene- induced ear and egg whiteinduced footpad edema.Addi-tionally,RSG did not show a potable influence on IgM- and IgG-PFC counts against SRBC inmice.An increase but not decrease in serum hemolysin level,however,was observed in both groupsof RSG,peralleled with the finding that hemolytic plaques in PFC test were obviously biggerthan those in control.These results suggest that RSG has a selective activity to suppress the cellularimmune response without inhibiting the humoral immune response.The suppression to cellular im-munity by RSG may be presented mainly through affecting the inflammatory process after lym-phokine release.
6.Correlative factors for organic psychotic symptoms in patients following traumatic brain injury.

OBJECTIVE:
To investigate the correlative factors for organic psychotic symptoms following traumatic brain injury (TBI).
METHODS:
In the current study, 391 subjects who had undergone forensic identification of the organic mental disorders due to TBI were included, both the demographic and post-traumatic information collected. The relevant data were statistically analyzed in those confirmed as organic psychotic syndrome according to Chinese Classification of Mental Disorders 3rd version (CCMD-3).
RESULTS:
Fifty-two subjects (13.3%) were identified as organic psychotic symptoms. The chi-square test showed that the detectable organic psychotic symptoms were associated with the marriage status, damage nature, injury severity and treatment, and the multivariate logistic regression analysis revealed good fitness of treatment and injury severity with the regression model (OR = 0.044, 95% CI: 0.017-0.114; OR = 2.145, 95% CI: 1.201-3.832, respectively).
CONCLUSION
The risks of organic psychotic symptoms following TBI can be involved in the alternative of craniotomy for the cases with trauma and moderate brain injury.

7.Mechanism of liver and lung injury in septic mice

Objective:To investigate the mechanism of liver and lung injury in mouse septic models.Methods:Twenty-four male Kunming mice were subjected to cecal ligation and puncture(CLP)or sham operation.The permeability of microvasculature,water contents,activities of myeloperoxidase(MPO)and the apoptosis of microvascular endothelial cells in lung microvasculature and liver sinus were examined 3 h and 12 h after operation.Results:Both the liver and lung showed a significant increase in microvessel permeability at 12 h in CLP group compared with sham operation group.MPO activity and water content in CLP group were obviously higher than those in the sham operation group.The apoptosis of lung microvascular endothelial cells at 12 h in CLP group(5.03?0.92)% was significantly higher than that of control group(3.48?1.21)%(P

Objective High mobility group box-1 protein ( HMGB1) plays an essential role in regulating energy metabolism of tumor cells via affecting mitochondrial autophagy .The aim of this study was to explore the effect of HMGB 1 on mitochondrial biogene-sis and cell energy metabolism in anoxia environment . Methods HepG2 cells were divided into normoxia control group ( cells were cultured in a culture box containing 5%CO2) , hypoxia control group ( cells were cultured in a culture box containing 1%O2+5%CO2+94%N2 ) , hypoxia HMGB1 siRNA group ( cells were cultured in a culture box containing 1% O2+5% CO2+94% N2 after transfected with HMGB1 siRNA) and hypoxia NC siRNA group ( cells were cultured in a culture box containing 1%O2+5%CO2+94%N2 after trans-fected with negative control siRNA ) .MTS assay was carried out to measure cell proliferation rate .The alterations of mitochondrial bio-genesis associated proteins were detected by RT-PCR and western blot.Mitochondrial density and morphology were determined by transmission electron microscopy (TEM).The ATP content in whole cell extracts was determined with a colorimetric ATP detection kit . Results Compared with the hypoxia control and hypoxia NC siR-NA group, the proliferationof hypoxia HMGB1 siRNA group was significantly inhibited, especially in 48 h and 72 h(P<0.05).Com-pared with hypoxia control group and hypoxia NC siRNA group , the expression of PGC 1α, NRF1and TFAM in hypoxia HMGB1 siRNA group were decreased significantly ( P<0.05) .Western blot results showed that , compared with normoxia control group , the expressions of PGC1α(0.494±0.210 vs 0.090±0.020), NRF1(1.080±0.470 vs 0.581±0.190)and TFAM(1.585±0.340 vs 0.792±0.350) protein in hypoxia 24 h group were increased obviously ( P<0.05) .Compared with hypoxia control group and hypoxia NC siRNA group , the expres-sions of PGC 1α, NRF1 and TFAM protein in hypoxia HMGB1 siRNA group were significantly decreased (P<0.05).Compared with hypoxia control group , the content of ATP in the HMGB 1 siRNA hypoxia group was significantly decreased , and hypoxia 12 h and 24 h were the most obvious ( P<0.05) . Conclusion HMGB1 could maintain cell energy metabolism by regulating mitochondrial biogene-sis so that cells could continue to proliferate in adverse anoxia environment .
10.Periphery analgesic effect of myricetin on a rat model of inflammatory pain

Aim To investigate the periphery analge-sic effect of myricetin on a rat model of inflammatory pain and the mechanism. Methods Rat models of in-flammatory pain were induced by complete Freund ’ s adjuvant ( CFA) injection in left hindlimb plantar cen-ter. The thermal withdrawal latency ( TWL) was meas-ured before and after CFA or myricetin treatment. Elec-trophysiological method was used to identify the effect of myricetin on the action potential frequency and the voltage dependent potassium channel currents in small DRG neurons. Results Rats with CFA injected showed thermal hyperalgesia ( P <0. 05 ) and TWL in-creased significantly after myricetin intraperitoneally in-jected ( P <0. 05 ) . Current clamp recording showed the action potential frequency of small DRG neurons in rats was inhibited by myricetin ( P<0. 01 ) and voltage calmap recording showed the inhibitory effect of myr-icetin was enhanced by calcium depended potassium channel currents ( P<0. 05 ) . Conclusion Myricetin exerts periphery analgesic effect by enhancing calcium depended potassium channel currents and inhibiting excitability of small neurons of dorsal root ganglion.
