1.Progress in epigenetic regulation of regulatory T cell

Regulatory T (Treg) cells exert significant influence on the control of autoimmunity and maintenance of homeostasis,while the progressive roles of Treg cells in solid tumor and hematological malignance have also attracted much attention recently.Except for the specific expression of transcription factor forkhead box protein 3 (Foxp3),natural Treg (nTreg) cells have Treg-specific hypomethylation pattern in a limited number of key genes,which is indispensable for stable Treg cells and functional nTreg cells to define the T-cell subpopulation possessing Treg-type epigenome more accurately.The expression of Foxp3 and Treg-specific hypomethylation phenotype have close relationship with the epigenetic regulation,which involves DNA methylation,histone methylation,acetylation,microRNA,etc.This review summarizes the different mechanisms of epigenetic regulation according to the two important features of Treg cells.

Post-transcriptional regulation of prokaryotes is of pivotal importance to the quality control and stability main -tenance of multiple proteins .The ubiquitin and ubiquitin-like proteins-mediated modifications have widely participated in the manipulation of cell cycle control , subcellular localization and cellular metabolisms .Typical ubiquitin like proteins (UBLs) are ubiquitin,SUMO,Nedd8 and FAT10.Recently, the dynamic regulation mechanisms between sumoylation and de-sumoylation have become the cutting edge in the era of ubiquitin-like modifications.In this review, we briefly summa-rized the entire SUMO-mediated modification systems and the intricate balance regulation .We also discussed the counter acting star molecule SENP1 and its relationship with tumor initiation and progression ,immunological regulation and develop-mental homeostasis .
3.Current status and prospects of quantitative immunoassay technologies

Quantitative immunoassay technique is the common method of quantitative detection in clinical laboratory. Several important branches of quantitative immunoassay were formed by changing the tracer or the Antigen-antibody complex separation method, including radioimmunoassay, fluorescent immunoassay, enzyme immunoassay, chemiluminescence, colloidal gold, immuno-turbidimetric analysis and homogeneous immunoassay. Different immunoassay techniques have their own characteristics, also apply to different detecting conditions in clinic. This paper reviewed several common kinds of quantitative immunoassay technology, and discussed both their advantages and disadvantages, which provide reference for the application and development of clinical testing technology.
4.Gene mutation in pituitary adenomas of human

The single colonel composition of pituitary adenomas of human attests to the molecular basis of pituitary tumorigenesis. This review offers a specific overview of the involvement of alternation of tumor oncogenes and tumor suppressor genes (TSGs) in the pathogenesis of human pituitary adenomas. Mutations of gsp and H ras oncogenes genes and amplification of PKC have been identified in pituitary adenomas. Especially the mutation of gsp has been detected in 40% human GH secreting pituitary adenomas. The role of TSGs(tumor suppressor genes) including MEN 1, p53, Rb and p16 in pituitary adenoma formation and progression has yet be observed. Providing evidence proves that activation of oncogenes and inactivation of tumor suppressor genes seem to be at least one mechanism by which human pituitary adenomas origin and progress.

Objective:To observe the effect of ischemic preconditioning(IP)on hepatocellular apoptosis caused by ischemia-reperfus ion(IR)injury and expression of apoptosis regulatory genes(Survivin,HIF-1?)protein,to explore the protective mechanisms of IP on IR injury in rat liver.Methods:Ninty SD rats were randomly divided into sham operation(SO)group,ischemia-reperfusion(IR)group,ischemic preconditioning(IP)group.In SO group,the hepatoduodenal ligament of rat was only pulled and separated,and then closed the abdomen;IR group subjected to 30 minutes of ischemia;IP group was achieved by ischemia for 5 minutes,followed by reperfusion for 5 minutes before continuous to ischemia for 30 minutes.Ten rats of each group were killed after reperfusion for 2,6,12,24 hours respectively,then liver tissues were taken out.The rats in SO group were killed after operation for 2 hours.To detect the Apotosis index(A)Iand Survivin,HIF-1? protein levels.Results:AI of hepotocytes significantly increased in IR group and IP group than that in SO group(P
7.The role for Neurotrophin 3 in proprioceptive sensory nerve

Neurotrophin 3(NT-3) is required for proprioceptive neuron survival and physiological function.During embryonic development,NT-3 deficient in developing muscle dues to loss of proprioceptive neurons in dorsal root ganglia or their axons by apoptosis,and then to the absence of muscle spindles.The chemoattractant effect of NT-3 on dorsal root ganglion axons has been improved that NT-3 is a key requirement for the development of proprioceptive projection to motor neurons.Depletion of NT-3 in the period of adult educes to the decline of the function of proprioception.NT-3 has extensive protective effect on proprioceptive neuron and enhances them to restore the normal physiological function under the pathological state.
8.Effects of Dexmedetomidine Combined with Fentanyl on Analgesic Effects and Hemodynamics of Orthopedic Patients after Surgery

OBJECTIVE:To investigate the influence of dexmedetomidine combined with fentanyl on analgesic effects and hemodynamics of orthopedics patients after surgery.METHODS:A total of 80 patients undergoing orthopedics surgery were randomly divided into observation group (40 cases) and control group (40 cases).Observation group was given patient-controlled analgesia (PCA) of Dexmedetomidine hydrochloride injection+Fentanyl citrate injection,maintaining analgesia for 48 h.Control group was given PCA of Fentanyl citrate injection,maintaining analgesia for 48 h.The MAP,SpO2,HR,VAS and the occurrence of ADR were observed in 2 groups from the application of multi-function monitor to connecting to PCA (T0),12 h after connecting (T1) and 48 h after connecting (T2).RESULTS:At T0,there was no statistical significance in VAS score between 2 groups (P>0.05).VAS scores of 2 groups at T1-2 were significantly higher than at T0.Those of 2 groups at T1 were significantly higher than at T2;but the observation group was significantly lower than the control group,with statistical significance (P<0.05).There was no statistical significance in MAP,SpO2 or HR between 2 groups at different time points (P>0.05).The incidence of respiratory depression,sharp pain,nausea and vomiting,hypotension in observation group were significantly lower than control group,with statistical significance (P<0.05).CONCLUSIONS:Dexmedetomidine combined with fentanyl show significant analgesic effect after orthopedics surgery,and have no effect on hemodynamics with good safety.
9.Analysis of brain of sleep disorders in patients with leukoaraiosis and its influencing factors

Objective To investigate the leukoaraiosis(LA)influencing factors of sleep disorder patients. Meth-ods 90 cases of LA patients which were conformed MRI diagnostic criteria were chosen as LA group,90 cases of normal old people which the gender and age were matched with LA group were chosen as NC group;by asking the medical history to inquirying blood vessel hazards;pittsburgh sleep quality index(PSQI)was adopted to evaluate the condition of sleep disorders,according to the performance MRI of skull to part LA group as four grades. Results In LA group,hypertension and diabetes incidence were significantly higher than that in group NC(P < 0. 05);LA group,PSQI score was significantly higher than that of NC group(P < 0. 01). LA group with the LA increasing grade,PSQI score increased(P < 0. 05). Regression analysis showed that age,hypertension,diabetes and MoCA were associated with LA in patients with sleep disorders. Conclusion Patients with LA have a sleep disorder, which is closely related with the degree of cognitive impairment,LA lesions,age,hypertension,diabetes etc.

Autophagy is an important pathway in the body to remove damaged cell organelles and dysfunctional proteins to maintain protein equilibrium and intracellular homeostasis by the lysosomal system .As a degenerative disease of central nervous system , the main feature of Alzheimer ’ s disease is abnormal protein aggregation .However , the role of autophagy in the pathogenesis of Alzheimer ’ s disease is not fully clear yet .The aim of this paper is to take β-amyloid peptide as a starting point to explore the relationship between autophagy and β-amyloid peptide in the pathogenesis of Alzheimer’s disease.
