1.Measurement of serum and urine TGF?1 level in bladder cancer patients and its clinical significance

Purpose:To study the role of TGF?1 in the occu rrence and development of bladder transitional cell carcinoma. Methods:To quantitatively measure the serum and urine TGF?1 le vel in bladder cancer patients with ELISA technique. Results:The serum TGF?1 level(72.64?14.31 ng/ml) in bladder c ancer patients was significantly higher than that in normal controls(62.07?11.3 6 ng/ml). With the progression in pathological stage, grade and tumor advancemen t, the serum TGF?1 level in patients changed in a decreasing trend. The urine T GF?1 level(803?552 pg/ml) in patients was significantly lower than that in nor mal controls(2995?1891 pg/ml). The differences of urine TGF?1 level in patient s with different pathological stages and grades were not significant. Conclusions:The elevated serum TGF?1 level may be the cause of damaged cellular immune function in bladder cancer patients. The reduction of u rine TGF?1 level in patients could lead to the acceleration of tumor cell proli feration and may be the risk factor for tumor recurrence.
2.Clinical manifestation of Churg-Strauss syndrome

Objective Churg-Strauss syndrome (CSS) is a rare multisystem vasculitis of unknown aetiology. To describe the clinical features of the disease, the treatment and long-term follow-up. Methods Eight CSS patients were selected from rheumatology department of guangdong province peoples hospital between 2000-2005. Data were obtained retrospectively. The patients′ clinical manifestation and laboratory result were studied. Results All patients had asthma and hypereosinophilia. The lung,skin and peripheral nervous system were the most commonly involved organs. The majority of patient received intravenous injection pulses of methylprednisolone 0.5 g/d?3 d, followed by per oral prednison 1 mg?kg-1?d-1 and intravenous injection pulses of cyclophosphamide 750 mg/m2. The outcome and long-term follow-up were good. Conclusion Churg-Strauss syndrome is a systemic vasculitis occurring in patients with a history of asthma, corticosteroids and immunosuppressive treatment may received good outcome.

Objective To determine the clinical efficac y of nuclear matrix protein 22(NMP22) as a novel urine marker for bladder cance r. Methods Urine NMP22 values were determined for 18 bla dder cancer patients and 20 urological benign disease patients by an enzyme-lin ked immunosorbent assay (ELISA). Objective To investigate the clinical features of genitourinary invasion by malignant lymphoma.Methods Three cases of genitourinary invasion by malignant lymphoma were reported.Case 1,a 68-year-old man,complained of right flank malaise for 1 month.CT showed a soft tissue shadow of 4.0 cm?5.5 cm with homogeneous density at right renal hilus,and diffusely thickened right greater psoas muscle at the level from lower pole of the kidney to internal iliac muscle with low-density soft tissue shadow inside.Case 2,a 72-year-old man,presented with distended left leg for 2 weeks.CT indicated left seminal vesicle mass with soft tissue density of 2.5 cm in diameter,and a soft tissue mass of 6.5 cm?4.5 cm at the left iliac artery furcation.Case 3,a 48-year-old woman,complained of fever for 1 month.CT showed well-defined,solid masses of 8.0 cm?6.0 cm and 7.0 cm?6.5 cm,respectively,in diameter with inhomogeneous density at bilateral adrenal gland areas.The incidence,clinical features,prognosis and surgical treatment of this disease were summarized in combination with review of the literature.Results All 3 cases underwent operations.The primary foci originated from pelvic or retroperitoneal lymph nodes.The kidney,seminal vesicle and adrenal gland were involved in each of the 3 cases.The pathological types were all non-Hodgkin diffusive big B cell lymphoma.One case died 2 months after operation.The other 2 received CHOP chemotherapy after operation,and were followed for 4 months and 2 years,respectively.They were both alive with no recurrence at last time of the follow-up.Conclusions Genitourinary invasion by malignant lymphoma is an uncommon disease with atypical clinical presentations and poor prognosis.Surgical treatment has little effect,while radiotherapy and chemotherapy should be the first choice.Explorative surgery has positive significance for definite diagnosis.
5.Screening of proteins interacting with RSK4 in breast cancer by affinity purification and liquid chromatography/mass spectrometry

Background and purpose:As a tumor suppressor gene, ribosomal S6 kinase 4 (RSK4) plays important roles in inhibiting cell proliferation, migration and inducing cell apoptosis. However, the proteins interacting with RSK4 are still unknown. This study aimed to screen proteins interacting with RSK4 in breast cancer cell line MDA-MB-231 by lfag-tag affnity puriifcation and LC-MS/MS (liquid chromatography/mass spectrometry).Methods:The pcDNA3.1/EGFP-RSK4-Flag eukaryotic expression vector was constructed by inserting full lengthRSK4 gene into vector pcDNA3.1/EGFP-Flag. And then the recombinant plasmids were transferred into MDA-MB-231 cells. Real-timelfuorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot were used to detect the expression of RSK4 in MDA-MB-231 cells. Affnity puriifcation and LC-MS/MS were applied to screen proteins interacting with RSK4, and the related action mechanism of RSK4 with its interacted proteins was detected based on bioinformatics gene ontology (GO) and ingenuity pathway analysis (IPA).Results:Twenty-four proteins, such as serine/threonine-pro-tein kinase 38 (STK38)/serine/threonine-protein kinase 38-like (STK38L), MOB kinase activator 2 (MOB2) and protein arginineN-methyltransferase 5 (PRMT5), were successfully identiifed by Flag-tag affnity puriifcation followed by LC-MS/MS analysis, which probably interacted with RSK4. Bioinformatics analysis of the identiifed proteins suggested the proteins interacting with RSK4 were involved in diverse biological pathways, such as apoptosis and cell migration. Conclusion:According to bioinformatics results of proteins interacting with RSK4 identiifed by affnity puriifcation and LC-MS/MS, biological networks of RSK4 are involved in apoptosis and migration in breast cancer cells.
6.Combined mesenchymal stem cells and allogenic bone marrow transplantation in treatment of MRL/Ipr mice

In order to study the role of the bone marrow-derived mesenchymal stem cells(MSCs)transplanted with or without bone marrow(BM)in the treatment of lupus mice and the effect of MSCs in the onset of systemic lupus erythematosus(SLE).Method Twenty 12-week-old female MRL/lpr mice were randomly divided into four groups,including simple bone marrow transplantation group(SG,BM 1×107),united group-1(UG1,BM 1×107+MSCs 1×104),united group-2(UG2,BM 1×107+MSCs 1×106),the positive control group(PG,no transplantation).BALB/c mice were used as the negative control group(NG,no transplantation).MSCs which were amplified from the bone marrow of male BALB/c mice in vitro were transplanted into the female MRL/lpr mice with or without BM.One month later Y chromosome was detected to confirm if the transplantation was successful or not.The change of weight, white blood cells,urine protein,anti-dsDNA antibody,the pathology and immunofluorescence of renal were observed to evaluate the therapeutic efficacy.Results Y chromosome was detected in all transplanted female mice.Compared with PG,urine protein concentration in SG,UG1 and UG2 significantly decreased 30 days after transplantation(P<0.05).40 days after transplantation,the rite of anti-dsDNA antibodies in sG(0.91±0.27)was still higher than NG,which OD value wag 0.47 s0.10(P<0.05),but there was no statistical difference among UG1(0.76±0.28),uG2(0.73±0.10)and NG(P>0.05).However,50 days after transplantation,there was no marked difference of the tite of anti-dsDNA antibodies in SG(0.55±0.15),UG1(0.57±0.14)and UC2(0.58±0.05)compared with NG(P>0.05).After transplantation there was no vasculitis.no inflammatory cell infiltration in matrix and no obvious intercapillary cells proliferation in the kidney.The immunofluoroscence became negative or weakly positive.Conclusion MSCs transplantation with or without BM Can both improve the pathogenetic condition of MRL/lpr mice.MSCs can accelerate the clearance of anti-dsDNA antibody and promote the restoration of injured organs.We presume that MSCs are important immunological regulation cells in SLE.
