Objective To understand the expression of mRNA for two Toll-like receptors (TLRs) (TLR2 and TLR4) and two T helper (Th)1 and Th2 cytokines [ interleukin (IL)-4, interferon (IFN)-gamma] in splenic macrophages of ovalbumin-induced allergic asthma mice model. Methods The expression of mRNA for TLB2、TLR4 and the house-keeping gene HPRT in both splenic macrophages of BALB/c allergic asthma mice and the control mice were analysed by real-time polymerase chain reaction (PCR). We examined the threshold cycle times (Ct) of cDNA of TLR2、TLR4 and HPRT gene to get Ct2、Ct4 and CtH respectively. The value of relative quantity of mRNA level was calculated by the ratio of Ct2/ CtH、Ct4/CtH. After stimulating cells with PMA and Ionomycin with anti-CD3/CD8 in the presence of Brefeldin A, intracellular levels of Th1 cytokines (IFN-?) and Th2 cytokines (IL-4) were determined for CD3+ CD8- and CD3+ CD8+ T cells. Results We observed Ct2/CtH (1. 26?0. 04,1. 35?0. 11, P 0.05) in splenic macrophages between the control mice and allergic asthma mice; the expression of TLR2 mRNA of allergic asthma mice was significantly reduced. 2.Chemical composition and antibacterial activity of the essential oil from leaves of Chimonanthus grammatus.

The chemical composition of essential oil, which from the leaves of Chimonanthus grammatus obtained by hydrodistillation were analyzed by GC-MS, and their possible antibacterial properties were screened. According to the results from GC-MS analysis, fifty-three components comprising 99.99% of the essential oil were identified. The major components of essential oil were 3-(4, 8-dimethyl-3,7-nonadienyl)(E) -furan (13.1%), bornyl acetate (12.66%), and 6,6-dimethyl-3-methylene-bicyclo[3.1.1] heptane (7.06%), etc. Antibacterial activity of essential oil was employed by two complementary test systems of disc diffusion and MIC/ MBC tests, which showed obviously antibacterial activity against all of the tested bacteria.

3.Clinical effect of fluticasone propionate combined with noninvasive positive pressure ventilation in the emergency treatment of AECOPD

Objective To explore the clinical effect of fluticasone propionate combined with noninvasive positive pressure ventilation in the emergency treatment of AECOPD .Methods 88 AECOPD patients were selected, and they were divided into two groups by digital random method ,44 cases in each group.The control group received conventional treatment, the observation group received fluticasone propionate and noninvasive positive pressure ventilation treatment.The curative effect of the two groups was compared .Results The total effective rate of the observation group was 100.0%,which was higher than 70.5% of the control group (χ2 =10.827,P =0.000).After treatment,in the observation group,the FVC was (2.50 ±0.32)L,forced expiratory volume in one second (FEV1 ) was (1.36 ±0.20) L,the first second forced expiratory solvent percentage of predicted value ratio (FEV1%) was (51.23 ±4.32),the arterial oxygen pressure(PaO2 ) was (10.51 ±2.10) kPa,arterial partial pressure of carbon dioxide(PaCO2 ) was (5.15 ±1.19) kPa,and in the control group,the FVC was (2.00 ±0.30) L,FEV1 was (1.08 ±0.12)L,FEV1% was (40.6 ±4.03),PaO2 was (9.32 ±2.11) kPa,PaCO2 was (6.06 ±1.23) kPa,the differences between the two groups were statistically significant (t =7.940,9.192,8.102,8.920,9.920,P =0.023, 0.006,0.011,0.008,0.005).The readmission rate of the observation group was 4.5%,which was significantly lower than 13.6% of the control group (χ2 =9.298,P =0.000).Conclusion Fluticasone propionate combined with noninvasive positive pressure ventilation in the treatment of AECOPD patients can effectively improve lung function , reduce the relapse rate of the disease.
4.Clinical significance of monitoring drainage fluid parathyroid hormone after thyroid surgery

Objective To explore the clinical significance of monitoring drainage fluid parathyroid hormone (dPTH) for estimating the in situ reserves and function of the parathyroid by analyzing the change of serum calcium,serum parathyroid hormone(sPTH) and dPTH after thyroid surgery.Methods According to the operative method,the total of 144 patients with thyroid disease were divided into five groups:unilateral lobectomy,unilateral lobectomy plus isthmectomy with unilateral lymph node dissection,total thyroidectomy,total thyroidectomy with unilateral lymph node dissection,and total thyroidectomy with bilateral lymph node dissection group.The blood calcium,sPTH and dPTH level of patients were tested before operation and on the 1st,2nd,3rd and 4th day after operation.The depression of serum calcium,hypocalcemia and hypoparathyroidism were observed after operation.The serum calcium,serum PTH and dPTH level were summarized and analyzed statistically in order to evaluate the in situ reserves and postoperative function of the parathyroid.Results Among the 114 cases,the decline of serum calcium level mostly happened on the 2nd day after operation(70 cases,61.4%).There were 36 patients with hypocalcemia (31.58%) and 34 patients with hypoparathyroidism (29.82%).Serum calcium level increased gradually in all of the patients.Although sPTH level swung,it had a rising trend on the whole.The level of serum calcium and sPTH was positively correlated.The level of dPTH was discrete and decreased along with time.The decline level of dPTH among different groups had statistical difference.Conclusions It is a promising method to evaluate the in situ reserves and function of the parathyroid by monitoring the level and changes of dPTH after thyroid surgery,and it is of value for preventive calcium supplementation after thyroid surgery.
5.Genetic polymorphism of D16S539, D7S820 and D13S317 loci in the Kazak ethnic population.

OBJECTIVETo analyze the genetic polymorphism of D16S539, D7S820 and D13S317 in Chinese Kazak ethnic population from Xinjiang.
METHODSOne hundred and two unrelated individuals and a sample of families (n=42) were investigated by multiplex amplification, 6% denaturing PAGE and silver staining. And, the obtained allele frequencies were compared with those of other populations.
RESULTSEight, seven, eight alleles were observed at the 3 STR loci respectively and the genotypes distributions were in accordance with Hardy-Weinberg equilibrium. The expected heterozygosities for these loci were 0.9439, 0.9356 and 0.9304; the calculated polymorphism formation content (PIC) was 0.9905; the discrimination power (DP), 0.9998; the paternity exclusion (PE), 0.9572. In addition, significant difference was found in comparison with other populations, and in the sample of families (n=42) no new mutations could be found.
CONCLUSIONThe multiplex examination of 3 STR loci can be used in forensic identification and population genetics research.
