1.Culture of rat hepatocytes in collagen sandwich system and its cytochrome P450 activity measurement

AIMTo establish a collagen sandwich system (CSS) wit h self-prepared collagen for rat hepatocytes culture[FQ(14*2。46,X-WZ]in vitro, and study the effect of CYP3A substrates on cyt o chrome P450 activities of hepatocyte cultured by CSS. METHODSS terilized type Ⅰ collagen was prepared from rat tail tendons. The morphology of hepatocyte cultured by collagen coated (CC) and CSS method was investigated and compared. Activities of ALT, AST, and LDH in aliquots of extracellular medium w ere measured by biochemical assay. CYP1A, CYP2E1 and CYP3A activities of hepatoc ytes were also determined. RESULTSBy CSS method, the shape of hepatocytes was maintained, and the structure of cells was integrated after cult ivated for 6 d. However, in CC method after 3 d, hepatocytes dropped from the co llagen matrix, and the activities of ALT, AST and LDH in aliquots were increased . As compared with control group, in CSS cultivated hepatocytes, the activities of CYP3A increased 33%, and CYP2E1 decreased 45% by nimodipine, and CYP3A was in duced 1 94-fold (P

Object To investigate the effects of ethanol sediments obtained from the tuber of Angelica sinensis (Oliv). Diels (ESA) and its litmusless component (ESA 1) on the secretion of TNF ? and IL 1 by mice peritoneal macrophages in vitro. Methods L929 cell line cytotoxicity was used for the assay of TNF ?. The proliferation of L929 cell line was used for the assay of IL 1. Results The secretion of TNF ? and IL 1 by mice peritoneal macrophages which were co cultured with ESA or ESA 1 in vitro can be significantly promoted. At the concentrations in range of 5~20 ?g/mL, there is a dose dependence in the action of ESA, while there is not the similar effect of ESA 1, even though it showed the marked effect. Conclusion ESA and ESA 1 can enhance the secreting TNF ? and IL 1 of mice peritoneal macrophages in vitro.
3.Culturing rat Kupffer cell with double collagen culture

AIM To explore a long-term Kupffer cell culture method, which can best maintain the function and activity of cell and decrease the cellular damage. METHODS Lactic dehydrogenase (LDH) in the supernatants were measured to observe the severity of cellular damage ,the function of Kupffer cell was reflected with the nitrogen monoxide(NO) content and the intracellular acid phosphatases(ACP) activity. In the conditions of common culture(CC),single collagen culture(SCC) and double collagen culture(DCC), with or without serum, study the maintenance of cellular function and the severity of cellular damage. This study introduced the development, qualification requirements, training contents and methods, practice authority and field, practice management and safety of specialist nurses' multi-sitepractice, elaborated the development status of multi-site practice of nurses in foreign countries, summarized international experience, combined with the national conditions, explored the development mode of multi-point practice of specialist nurses in China, with a view to providing domestic specialized nurses with multi-site practice. It also provides suggestions for managers to formulate relevant policies and develop localized multi-site practice.
6.Investigation and analysis of prevalence rate of inpatients′ nosocomial infections and risk factors and its prevention strategy research

Objective:To investigate the characteristics of nosocomial infection and relative risk factors, and provide the basis for the management and control of nosocomial infection.Methods:A cross-sectional survey was conducted on all inpatients with nosocomial infection in the First Affiliated Hospital of Shandong First Medical University from 0:00 to 24:00 on September 18, 2018.Results:There were 2 766 inpatients in the survey day, and the detection rate was 100%. The number of nosocomial infections was 57, with a current incidence rate of 2.06% (57/2 766), and the number of infections was 60, with a secondary incidence rate of 2.17% (60/2 766). The departments with the highest infection rates were neurosurgery department and intensive care units (ICU), which were 9.92% (13/131) and 9.38% (6/64) respectively. The lower respiratory tract at the infected site was the first, accounting for 50.00% (30/60). The main pathogens of nosocomial infection were gram-negative bacteria. The incidence of nosocomial infection was higher in patients with age ≥60 years ( χ2 value was 18.22 , P<0.001), ventilator( χ2 value was 37.84 , P<0.001), urinary tract intubation ( χ2 value was 180.57 , P<0.001), diabetic nephropathy ( χ2 value was 34.71 , P<0.001), and hemodialysis ( χ2 value was 368.60 , P<0.001) with statistically significant differences (all P<0.05). Conclusions:The focus should be on the prevention and treatment of lower respiratory tract infection inpatients in the intensive care unit of neurosurgery, and medic should develop preventive measures to avoid the occurrence of nosocomial infection according to the risk factors of patients with age ≥60 years, ventilator, urinary tract intubation, diabetic nephropathy, and hemodialysis to shorten hospital stay and costs.

The sacrum is a common site of stress injury. The occurrence of pressure ulcers not only leads to the aggravation of the patient′s condition, the prolongation of hospital stay, the increase of medical costs, the decline of life quality, but also aggravates the burden of society, family and medical staff. Early reasonable and effective application of sacral dressing can prevent the occurrence of pressure injury. In this study, the characteristics and effectiveness of different types of sacral dressings were reviewed by reviewing relevant literatures at home and abroad and summarizing the existing relevant studies, so as to provide guidance for clinical application.
8.The perception and feeling of nursing staff in neonatal palliative care: a Meta-synthesis of qualitative research

Objective:To systematically evaluate qualitative studies on the perceptions and feelings of nursing staffs implementing neonatal palliative care, aiming to provide insights for advancing clinical practice in China.Methods:The databases including China National Knowledge Infrastructure, Wanfang, VIP, Chinese Biomedical Literature Database, PubMed, CINAHL, Embase, Cochrane Library, Web of Science, and PsycINFO were included to retrieve the literature on the perceptions and feelings of nursing staffs from inception until March 28, 2024. The literature quality was assessed utilizing the Joanna Briggs Institute Australian Centre for Evidence-Based Health Care Quality Assessment Criteria for Qualitative Research (2016), and the findings were synthesized through Meta-integration techniques.Results:A total of 12 studies were included, yielding 46 themes were extracted; 8 categories were summarized and 3 synthesis results were obtained: nursing staffs experience both negative and positive aspects, the real-life challenges faced by nursing staffs in implementing neonatal palliative care, and the practical experiences and needs of nursing staffs regarding neonatal palliative care.Conclusions:It is crucial to continually address nursing staffs′ negative emotions and tackle challenges related to staffing, training, communication, and ethical dilemmas to ensure appropriate end-of-life symptom management in neonatal palliative care.
9.Investigation on good death status of terminally ill cancer patients and the influence factor

Objective To explore the good death status of terminally ill cancer patients from the nurses' perspective and identify associated factors. Methods A cross-sectional survey was conducted among the nurses in charge of the patients within 3 months after their death to evaluate the status quo of patients with end-stage cancer from the perspective of nurses, using anonymous questionnaire. Using convenient sampling method, 101 competent nurses from 209 patients with end-stage tumors who died in a hospital from October 2017 to January 2018 were selected. The nurses were investigated with nurses' general information questionnaire, patient information questionnaire and Good Death Inventory (GDI). Single factor analysis, Pearson correlation and multivariate linear regression analysis were used to analyze the influencing factors of good death in patients with end-stage cancer. Results The total score of GDI was (243.00±39.21). Among GDI, the three lowest scores were independence (7.43±4.25), physical and psychological comfort (8.17±4.82), and religious and spiritual comfort (8.53±4.72);while the three highest scores were being respected as an individual (18.32±2.76), good relationship with medical staff (18.23±2.59), and natural death (17.97±3.42). There were significant differences in patients' death status among different departments, medical payment methods, professional training experience of competent nurses and treatment methods within 3 months before death (t=17.351, 2.158, -2.679, -2.993;P<0.05). Pearson correlation analysis showed that nurse working life was positively correlated with the total score of GDI (r=0.953, P<0.01). Regression analysis showed that the factors influencing the good death of patients with end-stage cancer were the department of oncology medicine, the way of treatment and the professional training experience of nurses (P<0.05). Conclusions The overall status of good death of terminally ill cancer patients is poor. To strengthen the palliative care training about the knowledge and skills helps to improve the nursing quality of end-of-life care and achieve the goal of good death.
10.Construction of core theoretical curriculum of hospice care for professional nursing postgraduates

Objective:To construct the core theoretical curriculum of hospice care for professional nursing postgraduates.Methods:From May to October 2019, totally 24 experts in clinical care, academic associations, clinical medicine and nursing education from 15 affiliations in 7 provinces and cities were selected for correspondence consultation by convenient sampling. After literature review, the consultation questionnaire was designed by according to the opinions of the expert coordination group. Two rounds of Delphi expert consultations with were conducted.Results:The established core theoretical curriculum of hospice care for professional nursing postgraduates included basic courses of nursing, compulsory courses of hospice care and elective courses of hospice care, with a total of 24 courses in 3 modules.Conclusions:The curriculum framework of hospice care for professional nursing postgraduatesprovides a basis for the future development of advanced nursing education for senior nursing care providers.
