1.THE EFFECT OF HIGH-DOSE ESTRADIOL ON THE THYMUS OF THE RATS——A HISTOLOGICAL STUDY

Rapid involution of the thymus was induced in SD rats after administration of high-dose estradiol benzoate. The prominent histological changes observed were as follows: (1) the thymic cortex was become thinner; (2) the corticomedullary ratio was decreased; and (3) a substantial loss of cortical lymphocytes occurred. As a consequence, marked reduction in thymus weight was caused. Under electron microscope, the epithelial reticular cells (ERCs) were found, to be a heterogenous cell group. Their responses to high-dose estradiol were different from each other, and the distinctive changes observed were: (1) cell degeneration changes in some pale ERCs were observed; (2) the other pale ERCs were found to be filled with lower density vacuoles which were very similar to secretory vacuoles; and (3) the dark ERCs were inereased obviously, and many collagen fibril bundles were seen in the vicinity of their perikarya or wrapped by their cytoplasmic processes. There were no striking ultrastructural changes in thymocytes. However, the number of ANAE positive cells in the experimental group was lower than that of the controls evidently. In addition, a large number of plasma cells were observed in the cortex of the experimental group. Based on the above mentioned observations, the possible mechanism of the estradiol effect on the thymus involution was discussed.
2.Analysis of radiation dose to patients during coronary angiography and intracoronary stenting using radial and femoral artery access

Objective To evaluate the radiation dose to patients using radial and femoral artery access in coronary angiography (CAG) and intracoronary stenting (IS) ,provide basis for clinical intervention path .Methods The data of 190 samples (43 by femo-ral and 147 by radial) underwent CAG and 54 samples (17 by femoral and 37 by radial) underwent CAG+IS were analyzed retro-spectively .All samples were divided into two groups (radial group and femoral group) by different approach ,and radiation dose in different approach were analyzed .Results There was no significant difference of Dose Area Product (DAP) and Cumulative Dose (CD) using femoral and radial access in CAG (P>0 .05) .Separating two samples which CD were much higher than others ,the mean DAP was 23 .93 Gy · cm2 and the mean CD was 358 .85 mGy using radial vs .27 .06 Gy · cm2 and 369 .57 mGy using femoral , not distinctive either(P=0 .734 ,P=0 .834) .In CAG+IS ,the mean DAP was 82 .64 Gy · cm2 using radial and it was 78 .11 Gy · cm2 using femoral ,and the mean CD was 1 286 .41 mGy using radial and it was 1 267 .76 mGy using femoral .There were no signifi-cant difference in both DAP and CD (P=0 .705 ,P= 0 .919) .Conclusion The radiation dose of DAP and CD were not different when using radial access and using femoral access in CAG and CAG +IS .
3.Purification Comparison and Influence of Ethanol Precipitation and Chitosan Flocculation Methods on Paeoniflorin Content in Shuyu Compound Granules

Objective To compare the refining effects between ethanol precipitation and chitosan flocculation methods on extracting solutions of Shuyu compound granules.Methods Orthogonal experiment was utilized to examine the influence of liquid concentration, alcohol concentration and standing time on refining of Shuyu compound granules extracting solution by ethanol precipitation method, and compare the impact of ethanol precipitation and chitosan flocculation methods on the contents of solids and paeoniflorin.Results The optimized condition of ethanol precipitation method was 0.8 g/mL solution concentration (crude drug) with 20% ethanol standing for 6 h. Both ethanol precipitation method and chitosan flocculation method reduced the extraction ratio of Shuyu compound granules decoction, but chitosan flocculation method caused a great loss of paeoniflorin.Conclusion Low concentration ethanol precipitation method can be used for the purification of Shuyu compound granules. Whether chitosan flocculation method is suitable requires further study.
4.Effect of Acupuncture plus Chinese Herbal Medication on ET-1 and CGRP in Child Patients with Mesenteric Lymphadenitis

Objective To investigate the effect of acupuncture plus Chinese herbal medication on plasma endothelin (ET-1) and calcitonin gene-related peptide (CGRP) in child patients with mesenteric lymphadenitis.Methods One hundred and eighty child patients with mesenteric lymphadenitis were randomly allocated to groups A, B and C, 60 cases each. Group A received acupuncture at Zusanli and pricking Sifeng points plus oral administration of Wudang Babao Zijinding; group B, oral administration of amoxicillin and clavulanate potassium granules; group C, oral administration of Wudang Babao Zijinding alone. ET-1 and CGRP contents were measured in the three groups before and after treatment.Results There were statistically significant pre-/post-treatment differences in ET-1 and CGRP contents in group A (P<0.01). There were statistically significant post-treatment differences in ET-1 and CGRP contents between group A and group B or C (P<0.01).Conclusions Acupuncture plus Chinese herbal medication is an effective way to treat mesenteric lymphadenitis in children. It can regulate ET-1 and CGRP in the patients.
5.Effectiveness of iterative metal artifact reduction for reduction of metal artifact in chest CT scanning

Objective To assess the effectiveness of iterative metal artifact reduction (IMAR) on metal artifacts reduction in thorax scan.Methods Thoracic phantom with two pedicle screws implanted in both sides of the T5 vertebrae was used,with the scan parameters of 130 kV and CARE Dose 4D,the phantom was scanned with and without the screws respectively.Images without screws were reconstructed with FBP.Images with screws were reconstructed with FBP and IMAR respectively.Three ROIs were selected on tissues including aorta,pulmonary and paravertebral soft tissue on image slice adjacent to the screws.The CT value and standard deviations (noise) of ROIs were measured,and the deviation of CT value (△HU) was calculated as the difference between CT values in images with and without screws.Twenty-six cases who received chest CT examination and with pedicle screw implant in scanning range were collected.The scanning parameters and image reconstruction methods were the same as phantom scan.The CT value (HU) of metal artifacts adjacent to vertebrae and dorsal soft tissue was measured,and the image quality of reconstructed image by two skilled radiologists independently was evaluated.Results In the phantom after implanted screws,the noise were significantly reduced by IMAR compared to FBP in all the three ROIs of aorta,pulmonary and paravertebral soft tissue (P<0.05),and the △HU was significantly smaller in IMAR compared to that with FBP (P<0.01).In 26 patients,there were significant differences in CT value of vertebral bone tissue and dorsal soft tissue between FBP and IMAR (P<0.05),and the subjective evaluation scores of the two image reconstruction methods showed a statistically significant difference (P<0.05).Conclusion IMAR can significantly reduce streak artifacts of metal implant and adjuste the CT values of artifact affected tissues to make it more close to the true value without metal implant.
6.Effect of Tiam 1 on cell proliferation and migration in head and neck squamous cell carcinoma cells.

OBJECTIVE:
To investigate the effect of T lymphoma invasion and metastasis 1 (Tiam 1) overexpression in head and neck squamous cell carcinoma (HNSCC) cells.
METHOD:
Endogenous expression of Tiam 1 in 8 head and neck squamous cell carcinoma cell (HNSCC) lines was investigated by real-time RT-PCR. A lentivirus vector containing Tiaml was transfected into UM-SCC-47 cells, a head and neck squamous cell carcinoma cell line with little endogenous Tiaml expression. Stable clone, obtained by G418 screening, were assayed by RT-PCR and Western blot to validate the gene expression efficiency. The biological behaviors of the transduced cells were determined by cell counting, MTT and in-vitro migration assay.
RESULT:
Tiam 1 gene was highly expressed in M2 cell line and it's low level expression was found in UM-SCC-47. Cell counting and MTT assay showed that over-expression of Tiaml significantly promoted cell proliferation (P < 0.05). The cell monolayers overexpressed Tiaml that resulted in a significant increasment of cell migration in infected head and neck squamous cell carcinoma cell lines (P < 0.05).
CONCLUSION
Tiam 1 gene plays an important role in the growth and migration in head and neck squamous cell carcinoma cell lines. It may be a useful marker for metastasis of head and neck squamous cell carcinoma.

7.Prevalence and Relative Factors of Cognitive Disfunction in Old Adults in Conmmunity of Luzhou, Sichuan, China

Objective To investigate the prevalence and relative factors of cognitive disfunction among the community- dwelling old adults. Methods 766 participants over 60 years old dwelling in Luzhou, Sichuan, China were enrolled with Multi-stage stratified sampling. Information about sociodemographic characteristics, lifestyle, health condition, nutrition were interviewed, and their cognitive function was evaluated with Mini-Mental State Examination (MMSE). Results The prevalence of cognitive disfunction was 19.7% among them. Gender (male vs. female, OR=0.577), age (over 90 years vs. 60- 64 years, OR=29.857), administrator (administrator vs. not administrator, OR= 0.360), farmer (farmer vs. not farmer, OR=2.121), nutrition (malnutrition vs. eutrophia, OR=2.460), body weight (underweight vs. normal weight, OR=3.222), living status (UN-Habitat vs. living alone, OR=0.658), marital status (widowed vs. not widowed, OR=2.208), education attainment (junior college vs. illiteracy, OR=0.328), income (over 1000 yuan vs. no income, OR=0.596) significantly related with cognitive disfunction (P<0.05). Multiple Logistic regression analysis showed that old age, occupation (farmer) and malnutrition were independent risk factors for cognitive disfunction. Conclusion The cognitive disfunction related with multi-factors, and need more effective and available intervention in the old adults.
8.Disease burden of leukemia in Jiangsu Province from 1990 to 2019

Objective:
To investigate the trends in the disease burden of leukemia in Jiangsu Province from 1990 to 2019, so as to provide insights into leukemia control in Jiangsu Province.
Methods:
The prevalence, incidence, death and disability adjusted life years (DALYs) of leukemia in Jiangsu Province from 1990 to 2019 were retrieved from the 2019 Global Burden of Disease Study (GBD 2019) database, and standardized by the age structure of Chinese populations in 2000. The trends in the disease burden of leukemia in Jiangsu Province from 1990 to 2019 were analyzed using average annual percent change (AAPC).
Results :
The average annual standardized prevalence, incidence, mortality and DALY rate of leukemia were 63.63/105, 9.76/105, 4.10/105 and 194.83/105 in Jiangsu Province from 1990 to 2019, respectively. The average annual standardized prevalence (AAPC=1.420%, t=5.644, P<0.001) and incidence (AAPC=0.806%, t=3.505, P<0.001) of leukemia appeared a tendency towards a rise, while the average annual standardized mortality (AAPC=-1.589%, t=-14.714, P<0.001) and DALY rate (AAPC=-1.849%, t=-9.046, P<0.001) of leukemia appeared a tendency towards a decline in Jiangsu Province from 1990 to 2019. Higher average annual standardized prevalence (65.27/105 vs. 62.38/105, P<0.001), incidence (10.32/105 vs. 9.29/105, P<0.001), mortality (4.69/105 vs. 3.57/105, P<0.001) and DALY rate of leukemia (216.94/105 vs. 172.80/105, P<0.001) were estimated among men than among women. The crude prevalence of leukemia peaked among patients at ages of 0 to 14 years and 60 to 74 years, and the crude incidence, DALY rate and mortality of leukemia peaked at ages of 0 to 14 years and 75 years and older.
Conclusions
The mortality and DALY of leukemia appeared a tendency towards a decline and the prevalence and incidence appeared a tendency towards a rise in Jiangsu Province from 1990 to 2019; however, there were high disease burdens of leukemia among men, children and the elderly.
9.Cloning the differentially expressed genes in the retina of rds mouse during the development of retinitis pigmentosa

Objective To clone the differentially expressed genes in the retina of rds mouse (the animal model of congenital retinitis pigmentosa) during the disease development. Methods The retinal mRNA of rds mouse during the development of retinitis pigmentosa was analyzed by the mRNA differential display. The differentilly expressed mRNA fragments were cloned and sequenced. Results There was obvious difference of gene expression between rds mouse and the control during the development of retinitis pigmentosa. Five differentially expressed bands were cloned and sequenced. One of those had 86% identity (132/154) with the sequence of the human cDNA DKFZp434D1227 from adult testis in GenBank, which was submitted lately (15-Oct-1999) and without much information. The other had lower identity with the sequeces in GenBank. A highly expressed clone in the rds mouse on postnatal day 25 had the same length as another clone in the normal on postnatal day 37, which was not expressed in the rds mouse on day 37. The sequences of the two clones were identical in all but two base pairs. Conclusion These results indicate that there are a lot of novel differentially expressed genes in the chronic processing diseases, such as retinitis pigmentosa.
10.Losartan regulates oxidative stress via caveolin-1 and NOX4 in mice with ventilator- induced lung injury.

OBJECTIVETo investigate the effect of losartan in regulating oxidative stress and the underlying mechanism in mice with ventilator-induced lung injury.
METHODSThirty-six male C57 mice were randomly divided into control group, losartan treatment group, mechanical ventilation model group, and ventilation plus losartan treatment group. After the corresponding treatments, the lung injuries in each group were examined and the expressions of caveolin-1 and NOX4 in the lung tissues were detected.
RESULTSThe mean Smith score of lung injury was significantly higher in mechanical ventilation model group (3.3) than in the control group (0.4), and losartan treatment group (0.3); the mean score was significantly lowered in ventilation plus losartan treatment group (2.3) compared with that in the model group (P<0.05). The expressions of caveolin-1 and NOX4 were significantly higher in the model group than in the control and losartan treatment groups (P<0.05) but was obviously lowered after losartan treatment (P<0.05). Co-expression of caveolin-1 and NOX4 in the lungs was observed in the model group, and was significantly decreased after losartan treatment.
CONCLUSIONLosartan can alleviate ventilator-induced lung injury in mice and inhibit the expression of caveolin-1 and NOX4 and their interaction in the lungs.
