1.Imprinting Status of H19 Gene in Normal Villi Tissue During First Trimester:A Preliminary Study

OBJECTIVE To study the imprinting status of H19 gene in normal villi tissue during the first trimester,and its relation to the invasion of trophoblast. METHODS Using PCR-RFLP methods to examine the imprinting status of H19 gene in 93 cases of normal villi tissue during the first trimester. RESULTS Among 93 cases, heterozygous genotypes were found in 42 cases. And 11 cases of biallelic expression were found among these 42 cases of heterozygous genotypes from 5 to 9 weeks, however no biallelic expression existed from 10 to 12 weeks. CONCLUSIONS During the first trimester, H19 is expressed biallelically at the first 10 weeks. The H19 gene may dynamicly change in the trophoblast, and the dynamic change may have close relationship with the invasion of the trophoblast.
3.Protecting mechanism of heat stress in treatment of acute lung injury

Objective To investigate the protecting mechanism of heat stress pretreatment on acute lung injury(ALI). Methods The oleic acid ALI mouse model was built to dynamically observe the binding capacity and the binding affinity of glucocorticoid receptor(GR),the levels of GR,heat shock protein 90(Hsp90)and Hsp70 before and after hyperthermic stress pretreatment. Results Heat stress pretreatment had significant protective effect on ALI.Western blotting showed insignificant changes of GR levels but progressive increase of level of Hsp70 and Hsp90.Heat stress pretreatment exerted insignificant effect on Bmax and Kd of GR,shown by radio ligand binding assay after ALI. Conclusion The protective effects of heat stress pretreatment on ALI of mouse may relate to its ability of keeping stable GR level and increasing levels of Hsp70 and Hsp90.
4.Mechanism of dexamethasone inhibiting U937 cell adhesion and phagocytose function

ObjectiveTo investigate the mechanism of dexamethasone (Dex) in inhibiting monocyte adhesion and phagocytose function.Methods Under the stimulation of phorbo1-12-myristate-13-acetate (PMA),U937 monocytes cultured in vitro were treated with Dex and Fasudil respectively.The adhesion rate of U937 monocles to human umbilical vein endothelial cells (HUVECs) and their phagocytic ability of India ink were studied.The protein content and activity of rho-associated coiled-coil protein kinase 1 ( ROCK1 ) as well as the effects of mifepristone and cycloheximide on Dex were determined.ResultsBoth DEX and Fasudil could significantly inhibit the adhesion tate and phagocytosis of U937 cells stimulated by PMA and suppressed the activity of ROCK1.While mifepristone and cycloheximide could not alter these effects of DEX.ConclusionDEX interferes with the adhesion and phagocytosis function of U937 cells by inhibiting ROCKI activity.
5.Effects of traumatic brain injury on learning and memory ability and hippocampal CREB protein in rat

Objective To observe the change ofbrain water content,levels of cAMP response element binding protein(CREB)and phosphorylated CREB(p-CREB) in the early stage of traumatic brain injury(TBI) and to investigate the effect of TBI and p-CREB on learning and memory.Methods Fifty-four male adult Wistar rats were randomized into the normal group(18 cases),control group(18 cases) and TBI group(18) according to the random number table method.The TBI model was built according to the modified Feeney method and previous experimental parameters.At 12 h after TBI,Western blot analysis were performed to measure the expressions of hippocampal tissue CREB and p-CREB,the Morris water maze test was used to detect the behavior of rats in each group and the wet-dry method was applied to test brain water content.Results The brain water content at 12 h after TBI in the TBI group was remarkably risen compared with the normal group and control group;the expression levels of hippocampal CREB and p-CREB at 12 h after TBI in the TBI group were significantly decreased compared with the normal group and control group,the latent stage was increased and the frequency searching the accuracy within 2 min was decreased.Conclusion Brain edema is obvious after TBI and the levels of CREB and p-CREB are decreased,which maybe one of the reasons for the impairment of learning and memory function after TBI.
6.The expression of aquaporin-4 in the ischemic penumbra tissues after acute cerebral ischemia in rats

Objective The aim of this study was to investigate the aquaporin-4(AQP4) expression in the ischemic penumbra tissues.Methods Thirty-six Wistar rats were divided into 7 groups randomly, including control group(n=6) and occluded groups(n=30). The occluded groups were studied after the right middle cerebral artery of the rats unilaterally occluded(MCAO) at an interval of 15 min, 30 min, 1 h, 3 h, 6 h and 24 h, respectively(n=5 for each group). The operation process of the control group was the same as the occluded group except occluded MCAO. Then all rats were imaged with T_1WI, T_2WI and diffusion weighted-imaging(DWI). The brain tissue, according to the method by LIU Meili reported, was regarded as the area of the graphic penumbra. The relative apparent diffusion coefficient of the graphic-penumbra (rADC_1) and the center infarction(rADC_2)(ratios between the values of the occluded side and the opposite side) were calculated. The animals were sacrificed and perfused with the mixture solution consisting of TTC at different time intervals. The graphic-penumbra of the biggest layer of the ischemic cerebral tissue which corresponded to the DWI was examined with immunohistochemistry and RT-PCR. Meanwhile, histologic examination was performed at same site of the lesion. Results There were no significant changes on MRI, the relative apparent diffusion coefficient and the expression of the AQP4. The abnormal high intensity was found on DWI at 15 min after MCAO. T_2WI detected the lesion at 1 h after MCAO. The value of the rADC_1 decreased within 24 h after MCAO in ischemic penumbra, especially, it descended quickly within 1 h after MCAO, from(70.4?6.9)% at 15 min to(53.5?10.9)% at 1 h. Whereas, in the infarct tissue, the changes of the rADC_2 had a rule of decrease from(71.5?6.6)% at 15 min to(45.7?10.5)% at 3 h at first time, and then follow an increasing up to(78.7?11.5)% at 24 h after MCAO. The expression of AQP4 increased gradually within 24 h after MCAO, from 0.42?0.05 at 15 min to 1.18?0.12 at 24 h, it showed negative relationship with the rADC_1 in the ischemic penumbra (r= -0.966,P
7.Survey on child health service capacity of community health institutions in Shenzhen city

Objective:To survey the child health service capacity of community health service institutions in Shenzhen city.Methods:This was a cross-sectional study. An online survey was conducted among 559 community health service centers, stations and clinics in Shenzhen from January 23, 2024 to February 3, 2024. The questionnaire contents included the development of child health management, vaccination, diagnosis and treatment of common diseases in children, management of children′s chronic diseases, appropriate technology, availability of children′s special drug dosage forms, provision of special Chinese patent medicine and appropriate technology of traditional Chinese medicine for children, referral and remote consultation in 2019 and 2023. The reasons of unavailable service items were asked in the questionnaire and the questionnaire also contained an open-ended question about the suggestions for improving the capacity of child health services.Results:A total of 559 valid questionnaires were collected, accounting for 64.48% (559/867) of the community health service centers, stations and health clinics in Shenzhen in 2023. Compared to 2019, there was a significant increase in the rate of pediatric health services provided in 2023, including the diagnosis and treatment of common diseases in children under 6 years (96.06%, 537 institutions), nebulized inhalation therapy (96.60%, 540 institutions), influenza and other respiratory pathogen detection (90.70%, 507 institutions), rotavirus and other intestinal pathogen detection (34.53%, 193 institutions), allergen detection (81.75%, 457 institutions), blood oxygen saturation monitoring (84.44%, 472 institutions), pediatric-specific formulations of Western medicine (90.52%, 506 institutions), pediatric-specific formulations of traditional Chinese medicine (89.27%, 499 institutions), appropriate Chinese medicine techniques (88.19%, 493 institutions), relatively fixed referral hospitals (95.17%, 532 institutions), and remote consultation service (19.14%, 107 institutions); and the differences were statistically significant ( P<0.01). The reasons for unavailable service items were lack of space and personnel for the basic public health services, lack of space, personnel and a deficiency in knowledge and technical capabilities for diagnostic tests, and lack of equipment and personnel for remote consultation service. Suggestions made by 394 respondent institutions (70.48%) for further improvement included: conducting and strengthening various training (44.42%, 175/394), consultation and teaching by expert visiting (18.53%, 73/394), increasing the space and equipment (9.39%, 37/394), and further study at higher-level units (98.88%, 35/394). Conclusions:The child health services have been greatly improved in community health service institutions in Shenzhen, but there are still rooms for further improvement such as the ability of child health management, vaccination, chronic disease managements and provision of remote consultation.

Objective To observe the effect of Glycyrrhizic acid phospholipid complex self emulsifying preparation against carbon tetrachloride induced acute and chronic liver injury in rats and C57 BL/6J mouse.Method A cute hepatic injury rat model and a chronic hepatic injury were established by intraperitoneal (ip) injection with carbon tetrachloride (CCl4),the level of ALT and AST in serum were determined.The pathologic changes of hepatic tissue were observed with light microscope.Results The liver function results showed that Glycyrrhizic acid phospholipid complex self emulsifying preparation can significantly lower ALT and AST after acute and chronic liver injury (P < 0.01).Hepatic pathology study showed Glycyrrhizic acid phospholipid complex self emulsifying preparation could alleviate hepatic inflammation and effectively protect liver cells.Conclusion It was revealed that Glycyrrhizic acid phospholipid complex self emulsifying preparation could prevent carbon tetrachloride from inducing liver injury.
9.Features of intestinal flora in patients with nonalcoholic fatty liver disease and Helicobacter pylori infection

ObjectiveTo investigate the features and mechanism of action of intestinal flora in patients with nonalcoholic fatty liver disease (NAFLD) and Helicobacter pylori (HP) infection by comparing the changes in intestinal flora between the healthy population, the patients with HP infection, the patients with NAFLD, and the patients with NAFLD and HP infection. MethodsThis study was conducted among the 19 patients with NAFLD (NAFLD group), 19 patients with HP infection (HP group), and 19 patients with NAFLD and HP infection (NAFLD+HP group) who were admitted to The Second Affiliated Hospital of Henan University of Science and Technology from March 1, 2023 to April 30, 2024, and 20 individuals undergoing physical examination were enrolled as control group. Fecal samples were collected, total DNA was extracted for PCR amplification, and 16S rDNA sequencing was performed to compare the features of intestinal flora between the four groups. An analysis of variance was used for comparison of continuous data between multiple groups, and the chi-square test was used for comparison of categorical data between multiple groups. The Mann-Whitney U test or the Kruskal-Wallis H test was used for comparison of the species in intestinal flora. ResultsThe NAFLD+HP group showed a tendency of reduction in flora abundance compared with the other three groups. There was a significant difference in flora distribution between the NAFLD+HP group and the NAFLD group and between the NAFLD group and the control group (P<0.05). At the phylum level, the top three species in the NAFLD+HP group were Firmicutes (59.94%), Proteobacteria (17.00%), and Actinobacteria (14.75%), with an increase in the proportion of Proteobacteria and a reduction in the proportion of Actinobacteria compared with the other three groups. At the genus level, the top five dominant bacteria in the NAFLD+HP group were Bifidobacterium, Streptococcus, Escherichia-Shigella, Agathobacter, and Ruminococcus gnavus_group. Compared with the NAFLD group, the NAFLD+HP group had increases in the abundance of Streptococcus, Veillonella, and Rothia and reductions in the abundance of Dialister and Ruminococcus toraues_group. Compared with the HP group, the NAFLD+HP group had reductions in the abundance of Collinsella, Subdoligranulum, Catenibacterium, and Porphyromonas and increases in the abundance of Citrobacter and Olsenella (all P<0.05). ConclusionPatients with NAFLD and HP infection have changed in intestinal flora. These flora may be the intestinal microecological factors for HP infection in promoting the development and progression of NAFLD.
10.Alterations of β-γ coupling of scalp electroencephalography during epilepsy.

Uncovering the alterations of neural interactions within the brain during epilepsy is important for the clinical diagnosis and treatment. Previous studies have shown that the phase-amplitude coupling (PAC) can be used as a potential biomarker for locating epileptic zones and characterizing the transition of epileptic phases. However, in contrast to the θ-γ coupling widely investigated in epilepsy, few studies have paid attention to the β-γ coupling, as well as its potential applications. In the current study, we use the modulation index (MI) to calculate the scalp electroencephalography (EEG)-based β-γ coupling and investigate the corresponding changes during different epileptic phases. The results show that the β-γ coupling of each brain region changes with the evolution of epilepsy, and in several brain regions, the β-γ coupling decreases during the ictal period but increases in the post-ictal period, where the differences are statistically significant. Moreover, the alterations of β-γ coupling between different brain regions can also be observed, and the strength of β-γ coupling increases in the post-ictal period, where the differences are also significant. Taken together, these findings not only contribute to understanding neural interactions within the brain during the evolution of epilepsy, but also provide a new insight into the clinical treatment.
