1.Antigenic Localization of Specific Allergen in the Body of Dermatophagoides pteronyssinus by Immunohistochemistry

Objective To study the localization of specific allergen of Dermatophagoides pteronyssinus. \ Methods\ Through optical microscope,the specific allergens of D.pteronyssinus were observed in paraffin sections using D.pteronyssinus\|specific IgE antibodies from the patient sera. \ Results and Conclusion \ The digestive system was found occupying large parts of body cavity of D.pteronyssinus by HE staining, while the specific allergens of D.pteronyssinus were mostly occurred in the midgut tissue, gut contents, cuticle and reproductive system in the immunostained sections. The results also showed that many parts of D. pteronyssinus were recognized by the specific IgE antibodies obtained from allergic individuals to D.pteronyssinus, which provided a theoretic base for further study of isolation and purification of the specific allergen.
2.Effects of Sp1 on IP3R1 protein expression in human mesangial cells

Objective To investigate the effects of specific protein 1 ( Sp1 ) on the TNF-αin-duced expression of inositol 1, 4, 5 trisphosphate receptor type 1 ( IP3R1 ) in human mesangial cells ( HMCs) and to further elucidate the molecular mechanism regarding the decreased glomerular filtration rate ( GFR ) during hepatorenal syndrome .Methods Quantitative real-time polymerase chain reaction and Western blot assay were used to analyze the effects of TNF-αon the expression of IP3R1 at mRNA level and the expression of IP3R1 and Sp1 at protein level in HMCs , respectively.HMCs were transfected with a re-combinant plasmid PGL3-IP3R1 promoter to determine the effects of TNF-αon the activity of IP3R1 promot-er.HMCs were treated with Mithramycin A , an inhibitor of Sp1 binding, and transfected with Sp1-siRNA plasmid respectively to evaluate the expression of IP 3R1 regulated by TNF-α.The role of TNFR1 and TNFR2 in the TNF-αinduced expression of Sp 1 and IP3R1 proteins were detected by Western blot .Results TNF-αincreased the expression of IP3R1 at mRNA level and the expression of IP3R1 and Sp1 at protein lev-el in HMCs.Moreover, the activity of IP3R1 promoter in HMCs was remarkably increased by TNF-αas well.TNF-αinduced expression of IP3R1 was inhibited by Mithramycin A in a concentration dependent manner.HMCs transfected with Sp1-siRNA plasmid showed a significantly decreased expression of IP 3R1 protein.Both anti-TNFR1 and anti-TNFR2 antibodies blocked the TNF-αinduced IP3R1 expression, while only anti-TNFR1 antibodies inhibited the TNF-αinduced Sp1 expression.Conclusion TNF-αmight in-crease the expression of IP3R1 through the TNFR1/Sp1 signaling pathways in HMCs .
3.Auxiliary fixation of proximal humerus fractures with medial support

There is a considerable increase in incidence of proximal humerus fractures.Locking plates are widely applied in the treatment of proximal humerus fractures due to their superior advantages.However,they may still lead to a high incidence of complications,especially in complex proximal humerus fractures.Reconstruction of medial support for a proximal humerus fracture can reduce complications effectively.At present,there is no consensus on the optimal method for medial support.The methods commonly used for medial support as an auxiliary fixation of proximal humerus fracture include screw support,bone transplantation,bone substitute and dual plates,all of which are discussed in this review.
4.Analysis of urinary iodine monitoring results of school children aged from 8 to 10 in Nanchang City, Jiangxi Province

Objective To survey the urinary iodine (UI) status of school children aged from 8 to 10 in Nanchang City, and to provide a scientific basis for preventing and controlling the iodine deficiency disorders (IDD). Methods From 2009 to 2012, Donghu, Xihu, Qingshanhu, Qingyunpu, Wanli, Nanchang, Xinjian, Jinxian and Anyi 9 counties (areas) were chosen in Nanchang City as monitoring areas, and five townships were selected according to the five directions as east, west, south, north and centre in each county (area), one school was selected in each township, 20 school children aged from 8 to 10 (10 males and 10 females) were chosen as respondents. Ammonium persulfate digestion-arsenic cerium catalytic spectrophotometry (WS/T 107-2006) was used to detect UI. The monitoring data on UI of 8 to 10 years old school-age children were collected and analyzed. Results From 2009 to 2012, a total of 3 600 urine samples were collected, the median of urinary iodine (MUI) was 257.35 μg/L. In the 4 years, the MUI of school children aged from 8 to 10 was 315.30, 314.80, 262.92 and 112.73 μg/L, respectively, the MUI decreased year by year, the difference was statistically significant (χ2=631.129, P<0.05). Compared with 2009 and 2010, the proportion [15.22% (137/900), 14.67% (132/900), 25.11% (226/900), 30.22%(272/900)] of MUI of 100 -199μg/L (moderate intake) in 2011 and 2012 increased year by year; the difference was statistically significant (χ2=93.977, P< 0.05). The MUIs between different counties (areas) were statistically significant (χ2=36.520, P<0.05). The MUI of children aged 8 (280.10 μg/L), 9 (255.11 μg/L) and 10 (249.20 μg/L) decreased with increasingage (χ2 = 7.813, P < 0.05). The MUI of male students (269.70 μg/L) was higher than that of female students (247.60μg/L), the difference was statistically significant (Z = - 3.704, P < 0.01). Conclusions Iodine nutrition status of 8 to 10 years old school-age children in Nanchang City is good, iodine intake meets the body's need. It is suggested that the monitoring for UI of school-age children should be strengthened in order to prevent the potential harmful effects of inappropriate iodine intake.
5.Exploratory study on the role of tumor microenvironment-related gene CCR7 in Cuproptosis and its potential as a prognostic marker for liver cancer

Objective:To explore the interplay between tumor microenvironment (TME)-associated genes, cuproptosis, and the prognosis of liver cancer through transcriptome sequencing and functional genomics analysis.Methods:Employing a hybrid approach that integrates bioinformatics with fundamental experimental research, we utilized the TCGA database to acquireexpression profiles and clinical-pathological information from 424 liver hepatocellular carcinoma patients. We evaluated ImmuneScore and StromalScore to categorize patients into high and low groups, subsequently identifying differentially expressed genes (DEG) at the intersection of these groups. Core DEG were identified through univariate Cox regression analysis and protein-protein interaction (PPI) network analysis. The association between the expression levels of core genes and the survival time of liver cancer patients was analyzed using the R language and Kaplan-Meier analysis, and verified using the Kaplan-Meier Plotter online database. We established a cuproptosis cell model and performed RNA-seq to examine gene expression alterations during copper-induced cell death, followed by in vitro cell experiments for verification.Results:A total of 1 701 and 2 041 DEG were llinked t ImmuneScore and StromalScore, respectively, encompassing 1 134 commonly upregulated genes and 60 commonly downregulated genes. The top 30 core genes from the PPI network's dominant nodes were cross-referenced with univariate Cox regression results, leading to the identification of the pivotal immune gene CCR7. CCR7 mRNA expression levels were higher in hepatocellular carcinoma tissues than in normal tissues ( P<0.05). Patients with high expression of CCR7 in liver cancer had a longer overall survival compared to those with low expression ( P=0.003). Treatment with elesclomol-CuCl2significantly curtailed the survival of hepatocellular carcinoma cel ( P<0.001). RNA-seq data from the cuproptosis model indicated a downregulation of CCR7 expression during the onset of cuproptosis [|log 2(FC)|=2.27, P<0.001], and downregulation of CCR7 expression enhanced the sensitivity of hepatocellular carcinoma cells to cuproptosis inducers. Conclusion:The TME-related gene CCR7 is implicated in cuproptosis, and its downregulation might facilitate the process in liver cancer.CCR7 holds potential as a biomarker for liver cancer prognosis.
6.Prediction of postoperative visual acuity in cataract patients with idiopathic macular epiretinal membrane by hand-held retinal optometer and optical coherence tomography.

OBJECTIVE:
To investigate the value of hand-held retinal optometer and optical coherence tomography (OCT) in predicting postoperative visual acuity in patients with age-related cataract and idiopathic macular epiretinal membrane.
METHODS:
We retrospectively analyzed the data of patients undergoing phacoemulsification combined with intraocular lens implantation for age-related cataract in our hospital from January, 2019 to April, 2020.Preoperative examination detected idiopathic macular epiretinal membrane in 45 of the patients (52 eyes) with lens opacity grade C2N2P1 according to LOCSⅡ lens opacity classification criteria.Based on the thickness of the macular fovea, the eyes were divided into group A (9 eyes) with macular thickness < 300 μm by OCT examination, group B (25 eyes) with macular thickness of 300 to 400 μm, and group C (18 eyes) with macular thickness >400 μm.The best corrected visual acuity and retinal visual acuity before operation and the best corrected visual acuity on the first day and at 3 months after the surgery were compared among the 3 groups.The consistency between the preoperative retinal vision and the best corrected vision at 3 months after the surgery was analyzed.
RESULTS:
The best corrected visual acuity at one day and 3 months after the surgery differed significantly from that before the surgery in all the 3 groups (
CONCLUSIONS
For patients with cataract and idiopathic macular epiretinal membrane, phacoemulsification combined with intraocular lens implantation can improve postoperative vision.Hand-held retinal optometer can accurately assess postoperative vision in patients with stage C2N2P1 cataract.Patients with a macular thickness >400 μm caused by idiopathic macular epiretinal membrane are likely to have poor postoperative visual outcomes.
