1.Study on extraction process of total flavones of Pollen Typhae

Objective: To study the extraction process of the total flavones of Pollen Typhae ( Typha angustifolia L.). Methods: The optimum extraction process was selected with the orthogonal design. The content of total flavones was determined by UV spectrophotometry. Results: The significant effects of alcohol concentration, extraction time and extraction times on the extraction of total flavones were discovered. Conclusion: The optimum extraction process was as follows: Pollen Typhae was extracted with 70% alcohol for 3 times, with the solvent volume 8, 6, 6 times amount and extraction time 60, 60, 30 minutes in turn.
2.Study on adscription of plasma effective constituents of rat after administrated with alkaloid fraction of Nelumbo nucifera

Objective: To study the adscription of plasma effective constituents of rat after oral administration of alkaloid fraction of Nelumbo nucifera.Methods: Based on the established HPLC analytical method of plasma effective constituents,analysis and comparison were carried out among HPLC profi les of plasma samples obtained after oral administration of alkaloid fraction of Nelumbo nucifera,blank plasma,alkaloid fraction of Nelumbo nucifera.Results: Sixteen compounds were detected under the method,three of them were architypes of compound contained in alkaloid fraction of Nelumbo nucifera,and the others were metabolites.Through the comparison of the UV spectra,seven metabolites remain essential structure because the max wavelength was same to the architypes’,the other six metabolites’ structure changed greatly.Conclusion: Through the study of the adscription of plasma effective constituents of rat after oral administration of alkaloid fraction of Nelumbo nucifera,the structure of metabolites can be conjectured,which may be helpful to study the ADME and mechanism of action.
3.Research on quality control method of stilbene fraction of Polygonum multiflorum Thunb

Objective:To establish a method for controlling the quality of the stilbene fraction of Polygonum multiflorum T hunb. Methods: The contents of total stilbene and 2,3,5,4,-tetrahydroxy-stilbene-2-O-?-D-glucoside were determined by UV and HPLC respectively. Results: As a reference, 2,3,5,4,-tetrahydroxy-stilbene-2-O-?-D-glucoside had a good linear relationship in the range of 3.04-15.20?g/ml and 0.1032-0.6192?g, the linear regression equation was Y=0.0588X+0.0026(r=0.9998) and Y=3614089.84X+14475.21(r=0.9997), the average recovery was 99.95% with RSD=2.11%(n=6)and 100.08% with RSD=2.83%(n=6)for the determination of total stilbene and 2,3,5,4,-tetrahydroxy-stilbene-2-O-?-D-glucoside respectively. Conclusion: The determination results of 3 batches of sample showed that the method was simple, convenient and accurate, and could be used for the quality control of the stilbene fraction of Polygonum multiflorum Thunb.
4.A new aporphine alkaloid in leaves of Nelumbo nucifera

Objective To study the chemical constituents in the leaves of Nelumbo nucifera. MethodsCompounds were repeatedly purified by chromatography and structures were elucidated by physicochemical properties and spectroscopic analyses. Results Nine alkaloids were isolated and identified as 2-hydroxy-1-methoxy-6-methyl-6a,7-dehydroaporphine (1),armepavine (2),dehydroroemerine (3),dehydronuciferine (4),2-hydroxy-1-methoxyaporphine (5),liriodenine (6),pronuciferine (7),roemerine (8),and nuciferine (9). Conclusion compound 1 is a new aporphine alkaloid named nelumnucine.
5.The prevalence of human bocavirus among children with acute respiratory tract infection in Guangdong

Objective To investigate the prevalence of human bocavirus(HBoV)among children with acute respiratory tract infection(ARTI)in Guangdong Province.Methods Four hundred and forty-seven nasopharyngeal aspirates or swabs samples from children with ARTI in Guangdong Province were collected from June 2007 to May 2008.HBoV capsid protein VP gene fragments were detected using polymerase chain reaction(PCR).Positive PCR products were sequenced.The DNA and translated amino acid sequences were aligned with known HBoV sequences in GenBank and phylogenetic analysis was also done.Results The positive rate of HBOV was 5.1%of samples from 447 ARTI cases.Ten samples were positive for both HBoV and other respiratory virus,which was 43.5%of positive samples.The main diagnosis for HBoV positive children included wheezing pneumonia,bronchiolitis and bronchial pneumonia.HBoV positive children ranged from 42 days to 6 years old,and most of them were younger than one year.HBOV infection was more common during summer,early autumn and late spring.Through sequence alignment and phylogenetie analysis,the DNA sequences and amino acid sequences of VP gene fragments of isolated HBoV strains showed 97.8%-98.8%and 99.3%-100.0%identity with ST1,respectively.Conclusions HBoV is one of the important pathogens of lower respiratory tract infection in children in Guangdong Province,which is more prevalent in infants younger than one year.Although VP gene fragment of HBoV is conservative in general,there are still some missense mutations.
6.Application feasibility analysis of modified sentinel lymph node biopsy for acral malignant melanoma

Objective:To explore the application feasibility of modified sentinel lymph node biopsy (SLNB) for acral malignant melanoma.Methods:The data of 60 patients with acral malignant melanoma in the Affiliated Tumor Hospital of Xinjiang Medical University from January 2017 to January 2020 were retrospectively analyzed. According to the sentinel lymph node (SLN) detection method, they were divided into observation group (30 cases) and control group (30 cases). The observation group used contrast-enhanced ultrasound combined with subcutaneous injection of methylene blue around the wrist or ankle joint to detect SLN; the control group used peritumoral injection of methylene blue to detect SLN. The patients were regularly followed up to evaluate the postoperative effect. The detection number, detection rate, sensitivity, false negative rate and the size of SLN were compared between the two groups.Results:In the observation group, the detection rate of SLN was 100.0% (30/30), the sensitivity was 87.5% (7/8), and the false negative rate was 3.3% (1/30); in the control group, the detection rate of SLN was 83.3% (25/30), the sensitivity was 62.5% (5/8), and the false negative rate was 12.0% (3/25); the differences were statistically significant (all P < 0.05). The number of SLN detected in the observation group (3.5±1.2) was significantly more than that in the control group (2.0±1.1), and the difference was statistically significant ( t = 7.121, P < 0.05). The minimum long-axis diameter of SLN detected in the observation group was (5.4±2.2) mm (range, 1.5-12.3 mm), and that in the control group was (11.8±5.4) mm (range, 10.0-16.8 mm), the difference between the two groups was statistically significant ( t = 6.353, P < 0.05). Conclusion:The modified SLNB for acral malignant melanoma has a higher application value in the detection of acral SLN than the peritumoral injection method, and a higher accuracy rate can be obtained.
7.Drug Quality Characterization Consideration of Preparations of Oil Tea Camellia Defatted Cake based on Antithrombotic Effect

This study was aimed to select the best preparation form of medicinal in camellia cake,based on the optimal antithrombotic agents in oil tea camellia defatted cake to establish the foundation of research.The content of effective index component of the oil tea camellia defatted cake preparation was detected by HPLC-PDA.The quality of medicinal preparation from oil tea camellia defatted cake was characterized based on the content,relative quantity,relative ratio of content,yield rate and antithrombotic effect.The results showed that the effect of antithrombotic of three medicinal preparations from oil tea camellia defatted cake was listed in the following descending order of enrichment,ethanol extract,and water extract.The enrichment was the best preparation form of medicinal in oil tea camellia defatted cake preparation with the highest content of effective index components,the lowest extract rate and the lowest dosage.But it had the best effect of antithrombotic.It was concluded that the significant improvement of efficacy was due to the increase of the content of the components in the preparation and the rationality of the proportion.At the same time,analysis of component quality based on basic composition of drug system through the consideration of effective components of type and quantity,composition and other quality factors can further guide the preparation of drugs.
8.Studies on metabolism of Chinese materia medica of radix from Paeonia lactiflora Pall

ObjectiveTo research the metabolism of components in the Radix of Paeonia lactiflora Pall. Methods(①) we established the HPLC fingerprint of water extract of Paeonia lactiflora Pall. and real-time monitored the chemical composition. (②) We established the HPLC fingerprint of rats' serum samples from hepatic portal vein, serum samples from aorta abdominalis and samples of intestinal absorption of Paeonia lactiflora Pall. (③) On this basis, using the established methods, I-IPLC fingerprint spectrum of serum samples,the sample of herb, the sample after intestinal metabolism, rats' serum samples from hepatic portal vein and rats'serum samples from aorta abdominalis were analyzed and compared in order to infer the metabolism of components in the Radix of Paeonia lactiflora Pall. Results24 compositions were detected, seven of which were metabolized by intestinal flora and could not be absorbed into blood; six of them could not be absorbed directedly into intestinal; eight new compounds were absorbed into blood after bowel metabolism while they were not detected in water extract in Paeonia lactiflora Pall. ConclusionWe could infer the metabolic processes of chemicals of Paeonia lactiflora Pall. in oral administration with this method.
9.Studies on chemical components of Lobelia chinensis.

OBJECTIVETo study on the chemical constituents of Lobelia chinensis.
METHODThe coloumn chromatographic techniques were applied to isolate constituents, and their structures were elucidated by means of spectral data analysis.
RESULTSixteen compounds were isolated and identified as daucosterol (1), diosmetin (2), apigenin (3), chrysoeriol (4), loteolin (5), hesperidin (6), loteolin-7-O-beta-D-glucoside (7), apigenin-7-O-beta-D-glucoside (8), linarin (9), diosmin(10), 5,7-dimethoxy-8- hydroxycoumarin (11), palmitinic acid (12), lacceroic acid (13), stearic acid (14), beta-sitosterol (15), daucosterol (16).
CONCLUSIONAll of these compouds were obtained from L. chinensis for the first time.

10.Quantitative determination of contents of three components in Brucea javanica by HPLC.

OBJECTIVETo develop an HPLC method for quantitative determination of three quassinoids in Brucea javanic.
METHODThe determination was carried out on a phenomenex C18 column (4.6 mm x 250 mm, 5 microm) with gradient elution program of methol-water at a flow rate of 1.0 mL x min(-1), and the detection wavelength was 270 nm.
RESULTLinearites of bruceine D, brusatol and bruceine H were good (r = 0.9996, 0.9996, and 0.9998) in ranges of 2.52-12.60, 2.19-10.95, and 2.91-14.55 microg, respectively. The average recoveries of bruceine D, brusatol and bruceine H were 100.01%, 100.95% and 100.43% respectively, and RSD of the above three compounds were 0.31% (n = 6), 1.7% ( n = 6) and 1.7% (n = 6), respectively.
CONCLUSIONThe determination results of three batch samples showed that the method was simple, accurate and could be used in the quantitative determination of three components in the B. javanica.
