Professor Nie Li-fang achieved good clinical efficacy in treating chronic kidney disease by regulating the spleen and stomach. This paper introduces Professor Nie’s specific therapies to adjust the spleen and the stomach, such as Invigorating the spleen and benefiting qi, Tongfu Jiangzhuo, Regulating activities of qi.
2.Evaluation of the efficacy of botulinum toxin type A in the treatment of trigeminal neuralgia

Objective:To review the clinical efficacy of botulinum toxin type A(BTX-A)in the treatment of trigeminal neuralgia (TN).Methods:The main bibliographic databases were searched from English and Chinese literatures on the topic and the refer-ences of the identified articles were also searched for additional studies.2 reviewers assessed the quality of the included studies and extracted data duplicate.Results:3 Chinese and 1 English reports with 203 participants were included.However,substantial hetero-geneity(I2 =77%)precluded meta-analysis and the results were individually described.3 trials reported that BTX-A was more effec-tive than the control and 1 showed no difference between groups.Adverse effects of BTX-A were reported in 4 reports and were mild to moderate.Conclusion:BTX-A is effective in treating TN with few adverse effects.However,the evidence is weak because of a few studies included and most of them with low methodological quality.Further studies with high quality are needed to testify the evidence.
3.Constructing autologous composite skin around the expander for the repair of skin defects

BACKGROUND:The construction of tissue engineered skin needs a long time and high price, and the repair effects are poor. Moreover, the problems such as antigen elimination and disease propagation are not thoroughly solved. How to solve the tough problem of wound surface repair in patients lacking of autologous skin using current mature technology before the occurrence of ideal tissue engineered skin.
OBJECTIVE:To investigate the effects of autologous composite skin constructed around expanders on the repair of the wound surface.
METHODS:A total of 10 rabbits were selected. Two globular silica gel expanders were embedded subcutaneously in the symmetrical sites on the back of rabbits. After the expanders were covered by fiber kystis, cellsuspension of primary cultured skin epithelial cells (experimental group) or physiological saline (control group) were infused into lacuna between the expander. Four weeks later, the expanders were obtained. Experimental group presented epithelization, i.e., autologous composite skin. The skin and some fiber members on the top of the expanders were resected around the encystations. The fiber kystis on the bottom and surrounding the expanders was left to form the wound surface covered by autologous composite skin. However, the wound surface was covered by non-epithelization fiber kystis in the control group. The healing of wound surface was observed until recovered.
RESULTS AND CONCLUSION:In the experimental group, the wound surface was ruddy and pure, with less secretion;the average healing time was (14.0±0.4) days;the microscopic appearance indicated that epithelial layer was thick and regular. In the control group, there was more secretion on wound surface;the average healing time was (27.0±0.7) days;the microscopic appearance indicated that epithelial layer was thin and irregular. These results suggested that the construction of autologous composite skin around the expanders could noticeably promote the healing of wound surface.
4.INFLUENCE OF TRACE ELEMENTS As, Se, Te, Re, Ir ON CELL PROLIFERATION OF TETRAHYMENA PYRIFORMIS

As an excellent unicellular biological model, Tetrahymena pyriformis S1 has been employed in studying the biological effects of the chemical elements. The nutritious and/or toxic effects on T. pyriformis S1 induced by As, Se, Te, Re, Ir have been observed. Trace amounts of As, Se, Re and Ir added in the peptone-glucose culture medium could stimulate the cell proliferation of T. pyriformis in the following range of concentration respectively: As 0.03-0.05 ppm, Se 0.01-1.0 ppm, Re 0.02-0.08 ppm, Ir 1.0-3.0 ppm, whereas Te only showed its inhibitory effect on the growth of cells.
5.Activity identification of anti-caspase-3 mRNA hammerhead ribozyme in both cell-free condition and BRL-3A cells

Objective To study the transcription effects and cleavage activities of rat caspase-3 specific hammerhead ribozyme (Rz107) in both cell-free conditions and BRL-3A cells. Methods Rat caspase-3 gene fragment was cloned into the pGEM-T EASY vector under the T7 promoter control. The 32 P-labeled caspase-3 transcript was the target-RNA. Rz107 genes designed against caspase-3 mRNA were cloned into vector p1.5 between 5'-cis-Rz and 3'-cis-Rz. 32 P-labeled ribozyme transcripts were incubated with target-RNAs at different conditions and autoradiographed after denaturing gel-electrophoresis. Rz107 was electroporated into BRL-3A cells and the Rz107 expression was analyzed by RT-PCR. Results In cell-free conditions, Rz107 was active at 37℃. The optimal temperature was 50℃. The Km and kcat were 14.13?nmol/L and 2.31*min-1 respectively. Intracellular cleavage efficiency of Rz107 was 37%, as analyzed by RT-PCR. This indicated that the design of Rz107 was correct, and Rz107 had the activity of common enzymes. Conclusions Rz107 in cell-free conditions possessed perfect specific catalytic cleavage activity, and it can also cleave the target RNA successfully in cells. The results illustrate the feasibility of ribozyme therapy as a potential alternative approach for treating liver disease caused by apoptosis.
6.Relationship between dental calcification stages of the third molar and ages among teenagers in Chengdu.

OBJECTIVETo investigate the relationship between the stages of calcification of the third molar and age among children in Chengdu.
METHODSThe study subjects consisted of children between 2009 and 2011 at the Department of Orthodontics, West China Hospital of Stomatology, Sichuan University. Panoramic radiographs were obtained. The stages of calcification of the third molar were analyzed by Demirjian's method. The development of the third molar between the male and female was evaluated by independent samples t-test analysis. The Spearman rank correlation was applied to analyze the stages of calcification of the third molar and chronological age.
RESULTSThe development of the third molar of the male was different from that of the female at stage A of calcification of the third molar(P=0.026). The Spearman rank correlation coefficient revealed highly significant relationships between the stages of calcification of the third molar and chronological age of the three groups (male, r=0.808, P=0.000; female, r= 0.729, P=0.000; all, r=0.760, P=0.000). Furthermore, the linear regression equations were established as follows: Yall= 4.898+2.036X: Ymale=4.987+2.046X; Yfemale=4.819+2.036X.
CONCLUSIONThe stages of calcification of the third molar may be an available indicator in judging chronological age of teenager.

7.Diagnosis and treatment of posterolateral elbow dislocation in children

Objective:To investigate the diagnosis and treatment of posterolateral elbow dislocation in children.Methods:A total of 19 children with posterolateral elbow dislocation were diagnosed and treated at Department of Children's Orthopedics, Sichuan Orthopedic Hospital from April 2018 to April 2021. They were 12 boys and 7 girls, aged from 7 to 14 years (average, 10.9 years). Internal epicondyle avulsion fracture of the humerus was complicated in 11 cases, external epicondyle avulsion fracture of the humerus in 5 cases, capitulum avulsion fracture of the humerus in 3 cases, and fracture of the ulna coronoid process in 4 cases. After evaluation of the elbow stability, stable elbows were treated nonsurgically while the unstable ones complicated with fracture were treated with open reduction and internal fixation. After treatment, the injured limbs were fixated with elbow flexion 90° in forearm pronation position for 4 to 6 weeks.Results:In this group, 4 children received nonsurgical treatment and 15 ones were treated surgically. Internal epicondyle fractures of the humerus were treated by open reduction and internal fixation with Kirschner wire and hollow screws, and external epicondyle avulsion fractures or capitulum avulsion fractures of the humerus by suture fixation with Kirschner wire tension band or absorbable anchors. Three coronal process fractures were treated with plate internal fixation but one coronal process fracture was not treated with internal fixation. All the children were followed up for 6 to 30 months (average, 13.3 months). The last follow-up showed that fractures got united in all children, with no ectopic ossification, fine anatomical relationship of the elbow, and no recurrence of elbow dislocation or instability. The last follow-up showed that the Mayo elbow performance scores (MEPS) ranged from 75 to 100 points, averaging 93.2 points.Conclusions:In the treatment of posterolateral elbow dislocation in children, the key point is to rebuild the stability of the elbow. Non-surgical treatment is recommended for cases of a stable elbow with posterolateral rotation and dislocation while surgical treatment for cases of an unstable elbow because surgery leads to fine functional recovery and efficacy in children.
8.Exploration on the medication law of TCM compound patents and new prescriptions for diabetes kidney disease in recent 20 years

Objective:To analyze the medication and compatibility law of TCM compound patents in the treatment of diabetic nephropathy (DN) based on data mining method; To provide basis for research and development of new drug in clinic.Methods:TCM compound patents for DN treatment were retrieved from national patent platform. Excel 2019 was used to conduct statistical analysis on drug frequency, property and taste and meridian. SPSS Modeler 18.0 and SPSS Statistic 26.0 were used for drug association rules and clustering analysis. The complex network of co-occurrence of core drugs was constructed with Cytoscape 3.9.0, and the potential of the correlation between new prescriptions and drugs was demonstrated.Results:A total of 261 TCM compound patents were included, including 438 kinds of Chinese materia medica. High-frequency drugs included Astragali Radix, Rehmanniae Radix, Salviae Miltiorrhizae Radix et Rhizoma, Lycii Fructus, etc. Drug categories were mainly deficiency tonic drugs. The properties and tastes were mainly cold and sweet, and the meridians were mainly liver and kidney meridians. The commonly used medicinal pair was Ganoderma-Rehmannine Radix. The commonly used triple medicinal combination was Notoginseng Radix et Rhizoma-Angelicae Sinensis Radix-Ganoderma. There were 7 groups of clustering medicines, including Notoginseng Radix et Rhizoma, Ganoderma, Angelicae Sinensis Radix, Euryales Semen, Rehmanniae Radix and Lycii Fructus. There were 5 groups of potential medicines, including Campsis Flos-Caulis Tinosporae Sinensis-Kalopanacis Radix-Fimbristylis Rigiduta Nees-Padicularisdis Dissectae Radix -Korshinsk Peashrub-Alismatis Fructus-Cynanchi Wallichii Radix. The core new prescriptions for treating DN were obtained through topological attribute analysis and screening.Conclusions:The national TCM compounds patents treatment for DN is based on the pathogenesis of this disease, which is characterized by deficiency in nature and excess in superficiality. It often uses methods such as tonifying qi and spleen, nourishing yin and tonifying kidney, promoting blood circulation and resolving blood stasis to improve clinical efficacy, providing ideas for the development of new drugs.
9.Correlation studies in goats about the membrane thickness and pressure of the maxillary and frontal sinuses

Objective This study aimed to explore the differences of membrane thickness and pressure on the paranasal sinus membrane in goats and analyze their causes. The results can provide theoretical basis and guidance for the issues of the maxillary sinus floor augmentation related to the membrane. Methods The membrane was cut into two sizes from every sinus membrane. The membrane was fixed in formalin to obtain tissue specimens for the membrane thickness study and pressure study. The correlation between the two parameters was then analyzed, and appropriate statistical methods and software were selected. Results The top of maxillary sinus, the bottom of maxillary sinus and the frontal sinus membrane thickness were (410.03±65.97), (461.33±91.37), (216.90±46.47) μm. The pressure were (260.08±80.12), (306.90±94.37),(121.72±31.72) kPa. The mean differences of the membrane thickness between the top of the maxillary sinus and the frontal sinus, bottom and frontal, and top and bottom were statistically significant (P<0.05). The mean differences in membrane pressure were also statistically significant (P<0.05). Conclusion The membrane thickness and pressure of the top and bottom of the maxillary sinus are higher than those of the frontal sinus membrane. However, the thickness and pressure of the bottom membrane are slightly higher than those of the top membrane. Pressure and membrane thickness are positively correlated in the sinus membrane.
10.A comparative study of enzymatic digestion methods for the preparation of single-cell suspensions from the lamina propria of the mouse intestine

Objective To investigate the optimal digestion method for the preparation of single-cell suspensions from mouse small intestinal lamina propria.Methods Ten mouse small intestines of uniform length were collected and randomly divided into two groups.Each group was used to prepare lamina propria single-cell suspensions by enzymatic digestion with collagenase A or collagenase Ⅷ.We compared the effects of these two enzymatic digestion method on the cell yield,cell activity,and cell surface antigens of the single-cell suspensions.Single-cell suspensions prepared by the superior enzymatic digestion method were then subjected to flow cytometry assay.Results Compared to collagenase-Ⅷ-based enzymatic digestion,collagenase-A-based digestion result ed in a higher cell yield(9.48±1.10)× 109vs(4.18±1.02)×109and higher proportions of live cells(86.36±3.32)%vs(61.62±10.93)%,active CD45+cells(57.19±5.11)%vs(26.01±11.44)%,active CD3+cells(8.73±2.89)%vs(4.52±2.49)%,active CD4+cells(6.19±2.09)%vs(3.22±1.91)%,and active B220+cells(15.06±4.27)%vs(5.07±2.20)%,providing high-quality cells for subsequent flow assays.Conclusions The collagenase A-based enzymatic digestion method is more suitable for the preparation of ingle-cell suspensions from the lamina propria of ouse small intestines.
