1.Improvement of the Optimum Temperature of Penicillium expansum Lipase by Site-directed Mutagenesis

In order to improve the optimum temperature of lipases, The Penicillum expansum lipase (PEL) gene was mutated by site-directed mutagenesis using overlap extension PCR technique. The recombinant plasmid pPIC3.5K-lip-E83V containing matant gene was expressed in Pichia pastoris GS115. The comparison experiments of the mutant PEL-E83V-GS with the wild-type PEL-GS showed that: the optimum temperature of the mutant (45℃) was higher by 5℃ than that of the wild type. The thermostability of the mutant was similar to the wild type. The enzymatic activity of the mutant was 188 U/mL at 37 ℃, which was 80% that of the wild type at the same conditions. The hydrophobic interaction may be enhanced in the surface region by the hydrophilic amino acid, Glu substituted with the hydrophobic amino acid, Val, and be responsible for the improvement of optimum temperature.

Amblyopia is a developmental abnormality that results from physiological alterations in the visual cortex and impairs form vision. Based on multimedia, visual perceptual learning was widely applied in amblyopia treatment. Perceptual learning provided an impor-tant new method for treating amblyopia both in children and adults, and it reflects alterations of neural responses in the visual pathway. The therapeutic efficacy of perceptual learning was related to age, amblyopic types and degrees, training time and programs closely.
3.Neuroprotection of estrogen against injury induced by ?-amyloid protein (25-35) in rat cortical neurons

AIM: To study the effect of estrogen on injury induced by ?-amyloid protein (A?) in primary cultures of rat cortical neurons. METHODS: The effect of 17?-E_2 on A?(25-35)-induced cell death in primary rat cortical neurons was observed by phase contrast light microscopy, Giemsa staining and determination of lactate dehydrohenase (LDH) release rate. RESULTS: A?(25-35) induced cell death in rat primary cortical neurons. Forty eight hours pretreatment with 17?-E_2 protected rat primary cortical neurons from A?(25-35)-induced injury. CONCLUSION: A? evokes toxicity in rat primary cortical neurons. Estrogen can protect the rat primary cortical neurons against injury induced by A? (25-35).

5.Research advances in herpesviruses glycoprotein N gene and its encoded protein.

Glycoprotein N is encoded by glycoprotein N (gN) gene of herpesviruses. The amino acid composition and expression level of this protein vary among difference species of herpesviruses. According to present studies, gN protein is expressed in cytoplasm of host cells, mainly in endoplasmic reticulum. The gN forms a complex with glycoprotein M in host cells. The complex is involved in the processes of viral replication and inter-cellular infection. Moreover, this protein plays a role in immune evasion from host immune system. The study will provide a theoretical basis for further study of herpesvirus gN gene and its encoded protein.

6.Forecast of the indicators on maternal and child health of China in 2020 using auto-regressive model

Objective:To forecast the indicators on maternal and child health of China in 2020.Methods: Based on Surveillance data of the indicators on the maternal and child health in China since the 1990s,forecasting models were found using auto-regressive method,and the indicators on maternal and child health in China in 2020 were forecasted using the models after they had been tested and valued.Results: Auto-regressive models on infant mortality rate(IMR),under-5 mortality rate(U5MR) and maternal mortality(MMR) were found.The models and their parameters passed statistical tests,and their mean absolute error was 5% or so and determination coefficients were all more than 90%.Conclusion: The IMR of China in 2020 was forecasted to be 6.35‰,the U5MR 7.37‰ and the MMR 22.21/100 000.
8.Comparison analysis of cellular morphology and immune phenotypic in Uygur and Han patients with myelodysplastic syndrome

Objective To investigate the cell morphology and immune phenotypic characteristics in Uygur and Han patients with myelodysplastic syndrome (MDS) in Xinjiang Uygur Autonomous Region.Methods Bone marrow smears of 67 cases diagnosed as MDS were systematicly observed and recorded,flow cytometry (FCM) was used to test the immunophenotyping.Results There were abnormal hematopoiesis in different extent in granulocyte series,erythron series and megakaryocytic series of all patients.Occurence of pathological hematopoietic performance in Uygur patients with MDS was similar to that in Han,granulocyte series [52 cases (77.6 %)] >megakaryocytic series [44 cases (65.7 %)] > erythrocyte series [36 cases (53.7 %)],the differences in two groups were not significant (x2 values 1.02,0.30,0.02,respectively,all P > 0.05).The changes type of pathological hematopoietic performance in two groups were similar,single garden nuclear megakaryocyte [36 cases (53.7 %)],false Pelger nuclear anomaly granulocyte [36 cases (53.7 %)],erythroid gigantic young cell change [33 cases (49.3 %)],granulocyte cell particles reduced or absent [27 cases (40.3 %)] etc.The result of FCM showed that along with the progressive change of RA/RAS to RAEB/RAEB-t,the expression ratios of mature CD15 were decreased,while the expression ratios of early stage CD117 and CD34 were raised gradually (x2 values 6.23,12.06,8.95,7.37,8.95,8.08,respectively,all P < 0.05),the differences in two groups were not significant (x2 values 0.715,0.024,0.146,respectively,all P > 0.05).Meanwhile,Uygur patients with MDS has great expression of CD56 and Han had HLA-DR,there were significantly differences in two groups (x2 values 3.91 and 3.90,respectively,both P < 0.05).Conclusion Occurence of pathological hematopoietic performance in Uygur patients with MDS are similar to that in Han.Most MDSs has two or more levels of pathological hematopoietic performance.The differences of immunophenotype characteristics in two groups are partially significant and test of immunophenotype are helpful in the diagnosis,classification,prognosis of MDS.

The lengths and depths of occlusal rest base slope were calcukted accurately when the inclinations of occlusal rest base were 10?,15?and 20?at 1/2, 1/3 and 1/4 crown width respectively. The data will be usefull in the process of removable denture design with computer and provide a suitable way for the preparation of tilt abutment.
