1.Inoculation of murine bone marrow mesenchymal stem cells induces tumor necrosis in mouse with orthotopic hepatocellular carcinoma

Objective:To observe whether murine bone marrow mesenchymal stem cells(MSCs)implantation improves the survival of hepatocellular carcinoma(HCC)-bearing mice,and to investigate whether MSCs can differentiate to hepatocytes in HCC microenvironment in a mouse model of orthotopic HCC and its effects on tumor cells.Methods:Murine bone morrow MSCs were obtained through adherent culture method combined with magnetic cell sorting CD45-,CD11b-cells,labeled with CFSE in vitro.Phenotypes of MSCs were analyzed by flow cytometry.A murine model of orthotopic HCC was induced by intrahepatic injection of 5?105 murine H22 hepatoma cells in a BALB/c mouse.In this experiment,twelve BALB/c mice were randomly divided into MSCs implantation and saline administration groups on the 7th day after establishment of orthotropic HCC.CFSE labeled MSCs were injected into tumor and/or normal liver tissue in MSCs implantation group.The life span of HCC-bearing mice was observed.After three weeks,albumin was determined by immunohistochemistry.The livers of HCC-bearing mice were observed by light microscopy.Results:In MSCs group,the mean survival time was 25 days(95%Confidence interval:22-28 d),while,in the control group,mean survival time was 21 days(95%CI:20-23 d).However,the statistical difference was not obvious between the two groups(P=0.0713).It showed that the CFSE labeled cells mainly localized in the border of tumor,also a few in the tumor bed,and expressed albumin.Interestingly,there was a larger area of necrosis in the tumor bed,compared with that in the controls.Conclusion:It can be concluded that MSCs not only could engraft in livers of carcinoma-bearing BALB/c mice,but also could differentiate to hepatocyte-like cell.At the same time,MSCs might induce tumor cells necrosis.Further mechanism need to be studied.
2.Study in effect of exercise intervention on the renal transplant recipients during convalescence stage

Objective To explore the effects of exercise nursing intervention on renal transplant recipients.Methods A convenient sample of 64 renal transplant recipients during rehabilitation period were divided into the experimental group and the control group with 32 cases in each group.The experimental group received routine care and rehabilitation exercise,while the control group just received routine care.The body mass index(BMI),blood pressure and heart rate were measured in the two groups.Results Before intervention the differences of BMI,heart rate,blood pressure between the two groups were not statistically significant.However,after intervention,the BMI and blood pressure of the experimental group were lower than those of the control group,but there was no significant differences in heart rate.Conclusions Rehabilitation exercise nursing intervention contributes to raise recipients' quality of life and promote whole rehabilitation.
3.Radiation therapy for pancreatic carcinoma

Surgery is the treatment of choice for pancreatic carcinoma.However,only 20% of patients are eligible for surgery,and local control of disease and survival are not good.Although non-surgical treatments result in poor outcome,chemotherapy has been shown to be an effective treatment.The role of irradiation has not been well-established.Modern irradiation technology has shown promising results but there is still lack of strong prospective research evidence to support its widespread use.Irradiation brings along problems which need to be resolved,including target motion,definition of irradiation target,optimal fractionation,total treatment time and the treatment sequence of surgery,chemotherapy and radiation therapy.

Objective To explore whether the dopamine D1 and D2 receptors were involved in pathogenic mechanism of Parkinsonian mice induced by paraquat. Methods The models of Parkinson's mice were induced by oral paraquat. The levels of dopamine D1 and D2 receptor proteins and the expression of receptor mRNAs in striatum were examined by immunohistochemistry and in situ hybridization, respectively. Aim The pharmacokinetic parameters and relative bioavailability of capsule A(Maiwei Pharmaceutical Co Ltd, Beijing, China) and capsule B (Xianjing Pharmaceu-tical, Hunan, China) were studied. Methods A single oral dose of 600 mg gemfibrozilof these two kinds of capsules was given to 12 chinese healthy male volunteers in anopen, randomized crossover study. Plasma levels were determined with HPLC-UVmethod. Results The plasma concentration-time curve was fitted to 1-compartmentopen model with a first order and lag time absorption and the major pharmacokineticparameters of capsules A and B were shown respectively as following: C max(32. 69?5. 67 )and (29. 41?2. 60) mg?L-1; Tmax (1. 01?0. 14) and (1. 13?0. 37) h, t1/2ka(0. 46 ? 0. 18) and (0. 62 ? 0. 20) h; C max (1. 11 ? 0. 32) and (1. 32 ? 0. 26) h; MRT(2. 14 ? 0.27) and (2. 37 ? 0.26) h; AUC (91.7 ? 13.2) and (82.2 ? 7. 38) mg?h? L-1. There were no significantly differences between the pharmacokinetic parame-ters of capsule A and B. The relative bioavailability of the capsule A was (110 ? 9) % ascompared to the capsule B. Conclusion The two kinds of capsules have the equivalentbiological effects.
7.Influences of Astragalus and Hurido effective components on apoptosis of rat glomerular mesangial cells

AIM To investigate the influences of Astragalus and Hurido effective components on apoptosis of rat glomerular mesangial cells.METHODS The proliferation of rat mesangial cells was induced by LPS and then intervened with Astragalus (astragalus polysaccharide,astragaloside Ⅳ,calycosin) and Hurido (hirudin).The cells were collected after 24 h.4',6-Diamidino-2-phenylindole (DAPI) staining was used to observe cell nuclear changes.Quantitative and qualitative expressions of Bcl-2 were detected by streptavidin-biotin complex (SABC).RESULTS Astragalus and Hurido effective components could induce the apoptosis of rat glomerular mesangial cells,and decrease the expression of Bcl-2.CONCLUSION Astragalus and Hurido effective components can induce the mesangial cell apoptosis by down-regulating Bcl-2 expression,which can be used for the treatment of mesangial proliferative glomerulonephritis.
8.Study on oriented differentiation of bone marrow mesenchymal stem cells by fibroblast in rat uterine ligament with mechanical stretch

Objective To investigate the effect on the differentiation of bone marrow mesenchymal stem cells (BMSC) with non-contact co-culture with mechanical stimulated ligament fibroblasts. Methods A cyclic 10% uniaxia strain at 1 Hz was applied on rat pelvic ligament fibroblasts, then were co-cultured with BMSC for 3, 6 and 12 days in non-contact condition. The protein expression of collagen Ⅰ ,Ⅲ in BMSC were detected by SP method and revealed by the mean gray value. The mRNA expressions of collagens type Ⅰ and type Ⅲ in the BMSCs were measured with real-time (RT)-PCR ,and the results were indicated by the ratio between the mRNA and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) . Results (1) Protein expression; after 3 days co-culture with pelvic ligament fibroblasts, expression of collagen Ⅰ and Ⅲ in BMSC are 82. 4 ± 3. 4 and 76. 8 ± 2. 5. When compared with 80. 2 ± 2. 6 and 74. 6 ± 1. 1 in BMSC without co-culture, there was no significant difference (P > 0. 05) . Conclusion Non-contact co-culture with mechanical stretch stimulated ligament fibroblasts, it might promote synthesis of types Ⅰ and Ⅲ collagen in rat BMSCs and induced BMSC differentiated into pelvic ligament fibroblasts.
