Objective To study the expression of telomerase(TMA) in myocardial tissue of myocardial ischemia in rat. Methods Sixty Spraque-Dawley rats were randomly divided into A,B, C three groups,then each group was randomly divided into control group and testing group.Two myocardial ischemia groups were injected isoprenaline(8 mg/kg),and two control groups were injected distilled water(8 mg/kg) in abdominal cavity.For the last myocardial ischemia group the coronary artery was ligated.The histological and immunohistochemical method were used for observing cardiac structural changes,with HE staining,and changes of TMA in myocardial tissue after myocardial ischemia in rat.The TMA contents were measured by image analysis. Equisetin (EQST) belongs to polyketide (PKS)-nonribosomal peptide synthetase (NRPS) type compound with an inhibitory effect of 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1) enzyme activity. This study investigated anti-obesity effect and insulin resistance improvement effect of EQST on high-fat diet (HFD)-induced ob/ob mice model. EQST treatment effectively reduced the body weight gain, fat weight gain and blood lipid content of model mice. All animal experiments were approved by the Medical Ethics Committee of Capital Institute of Pediatrics. EQST alleviated adipose tissue expansion and hepatic ballooning degeneration of model mice, and also effectively controlled the blood glucose content after glucose load and insulin load, showed a significant improvement in obesity and insulin resistance. EQST inhibited adipogenic proteins fatty acid-binding protein 4 (FABP4) and peroxisome proliferators-activated receptor γ (PPARγ), and upregulated thermogenic protein (uncoupling protein 1, UCP1) through suppressing 11β-HSD1 protein expression. In addition, EQST widely upregulates mitochondrial respiratory metabolism related proteins in adipose tissue and may improve insulin resistance through phosphatidylinositol-3-kinase (PI3K) pathway. Therefore, EQST plays an anti-obesity role by promoting adipose tissue thermogenesis and improving insulin resistance, which may provide reliable clues for improving obesity and diabetes.

Objective:To investigate the effects of oxidation of titanium on bonding strength of titanium and porcelain. Methods: 24 specimens with a size of 28 mm ?3 mm?0.5 mm were cast from pure titanium and divided into 4 groups, 6 specimens each group. Group 2, 3 and 4 were degassed at 750, 800 and 900 ℃ respectively, group 1 not degassed as negative control, then a uniform thickness of 1mm of Noritake super porcelain Ti-22 was veneered on 8 mm?3 mm in the central portion of each specimen. The specimens were subjected to a three-point bending test on a load-test machine with a span of 20 mm, and the failure loads were recorded and analyzed statistically. The opposite side of specimens of each group were observed by thin-film X-ray diffraction. Results: The failure loads (N) of group 1, 2, 3 and 4 were 10.33, 9.98, 5.53, 3.37. The oxide of group 1 was Ti6O only, the oxide of the gropu 2, 3, and 4 were Ti6O and TiO2 and the amount of TiO2 increased significantly as the temperature iss increasing. Conclusion: The oxide adherence strength value for pure titanium decreased while the oxide temperature is increasing. The higher of the degassed temperature of the specimens, the more TiO2 can be found in the oxide film and the smaller the failure load is, which indicats that oxidation of titanium has a negative effect on the bonding strength of porcelain and titanium.

In order to improve the optimum temperature of lipases, The Penicillum expansum lipase (PEL) gene was mutated by site-directed mutagenesis using overlap extension PCR technique. The recombinant plasmid pPIC3.5K-lip-E83V containing matant gene was expressed in Pichia pastoris GS115. The comparison experiments of the mutant PEL-E83V-GS with the wild-type PEL-GS showed that: the optimum temperature of the mutant (45℃) was higher by 5℃ than that of the wild type. The thermostability of the mutant was similar to the wild type. The enzymatic activity of the mutant was 188 U/mL at 37 ℃, which was 80% that of the wild type at the same conditions. The hydrophobic interaction may be enhanced in the surface region by the hydrophilic amino acid, Glu substituted with the hydrophobic amino acid, Val, and be responsible for the improvement of optimum temperature.

Objective To discuss the MRI features of pyometra and its correlation with pathologic.Methods MRI and DWI data of 12 cases pyometra proved by surgery and pathology were analyzed retrospectively, and a comparision with pathologic findings was made.Mean ADC values were calculated.Results According to the homogeneity of MRI signal: Uniform signal type in 2 cases,T1WI showed slightly low signal,T2WI showed slightly high signal,DWI showed high signal,and the distribution of pus compositions is relatively uniform;Signal mixed type in 5 cases, T2 showed at the bottom of the pus or peripheral mixed slightly short signal, DWI showed mixed high signal, there was some sediment in the bottom of the pus;5 cases of liquid layer type, the upper displayed signal of water,and the lower signal was lower than the upper signal on T2WI, of which 3 cases showed uniform signal of the lower, DWI showed a uniform high signal, the distribution of pus compositions is uniform, the other 2 cases showed mixed signal of the lower, which was pyometra with bleeding,DWI showed high signal and low signal mixed together.The average ADC value of the 12 patients were 0.532×10-3 mm2/s.12 cases of uterine volume were increased, including 10 cases of uterine wall thinning, the other 2 cases with inflammatory invasion, the uterine wall thickening.8 cases with cervical cancer.5 cases with pelvic effusion.Conclusion MRI findings of pyometra are characteristic, its MRI manifestations and pathological components are highly correlated,and the ADC value is of great value in the diagnosis of pyometra.

The treatment for adult refractory /relapsed acute lymphoblastic leukemia is a major challenge in clinical practice. Therapeutic strategies including high-dose single agent,intensified induction,new drugs,targeted therapy,and immunotherapy etc. may be of benefit to some patients. The post-remission treatments remain to be further developed.

Acute promyelocytic leukemia (APL), a subtype of acute myeloid leukemia, is characterized by rapid progress, prone to developing DIC, and high mortality. Typical chromosome translocation with PMLRARα fusion gene occurs in more than 95% of APL cases. Since 1986, the outcome of APL has been significantly improved in our country by firstly using ATRA and ATO for treating APL, making APL of AML curable by chemotherapy only. Based on our limited experiences, we discussed the related problems in the treatment of APL.
