Objective To improve the recognization of hip joint lesion in ankylosing spondylitis and it's related factors.Methods Pelvis computed radiography(CR) and HLA-B27 were examined in 100 cases of ankylosing spondylitis proved by clinical.Results 58% cases appeared differed degree of narrowing of the interticular space,osteoporosis on head of femur and acctabulum,cystis under the surface of acctabulum and joint inosculated.This probability was higner at thoe patients that had diaease at youthful or HLA-B27(+).Conclusion Hip joint lesions of ankylosing spondylitis appear characteristic signs in CR.It relates with the age that the disease come on and HLA-B27.

3.Effects and Mechanism of Atherosclerosis Prevention by DMARDs in Patients with Rheumatoid Arthritis

Objective To investigate the effect of DMARDs on the prevention of early-onset atherosclerosis in rheumatoid arthritis ( RA) patients for exploring an appropriate schedule to reduce cardiovascular events. Methods Seventy-two patients with early RA were included in this study. Erythrocyte sedimentation rate ( ESR) and high sensitivity C reactive protein ( hs-CRP) were detected before and after treatment to evaluate inflammation. Carotid intima-media thickness ( cIMT) and endothelium-dependent flow-mediated vasodilatation (ED-FMD) were monitored. Disease Activity Score in 28 Joints (DAS28) and health assessment questionnaire-disability index ( HAQ-DI) was used to assess severity degree of RA and life quality of the patients, respectively. The patients were treated with methotrexate (MTX), hydroxychloroquine (HCQ) and sulfasalazine (SSZ) alone or combined based on severity of RA. Results After 1 year of treatment, ESR, hs-CRP, DAS28 and HAQ-DI were significantly improved (all P<0. 01). At the same time, the serum lipid levels also had obvious changes. However, there was no obvious difference in body mass index ( BMI) after 1 year. The cIMT ratio value was significantly decreased compared to baseline (0. 43±0. 08 mm vs. 0. 50±0. 16 mm, P=0. 002), and FMD% also significantly improved from basic value of 5. 26 to 7. 57 after treatment (P=0. 041). Conclusion Early intervention of RA by using DMARDs can effectively control the disease and slow the progression of atherosclerosis, but also reduce cardiovascular mortality.
4.Research on mental status and coping style in aged cardre ward patients with long- term hospitalization

Objective To explore the prevalence of anxiety, depression, loneliness in elderly patients with long- term hospitalization, its coping style and the correlation between them. Methods 48 hospitalized elderly patients with long- term hospitalization were investigated by Standard Scale and self- administrated questionnaire, the Pearson's correlation was used to find the correlation. Results It was concluded that anxiety, depression and other negative emotions were commonly seen in elderly patients with long term hospitalization, these patients usually adopt the avoidance and yield coping style as well,and there was a positive correlation between them, the r value was 0.438, 1.473 amd 0.501 respectively. Conclusions This research confirmed the possibility of negative emotion in elderly patients with large long- term hospitalization, but also closely related to the choice of coping styles. The nursing staff should closely observe and assess the psychological status of elderly patients, planned, targeted to help elderly patients use more positive coping style.

Objective To determine blood/gas and tissue/gas partition coefficients of sevoflurane, isoflurane and halothane and evaluate the effects of pregnancy on them. Methods Ten 18-22 day pregnant and 10 non-pregnant SD rats were killed under pentobarbital anesthesia. 6.Effects of ceftriaxone on glutamate and glutamate transporter subtype-1 in hip-pocampus following traumatic brain injury in rats

Objective:To sduty the effect of β-lactam antibiotics efiriaxone on the levels of glutamate and glutamate transporter subtype-1 in hippocampus following traumatic brain injury in rats.Methods: All rats were divided randomly into three groups:sham group;TBI group and CTX group.The rat model of TBI were made by modified Feeney method ,and treated with ceftriaxone immediately after injury ( 200 mg/kg ).Wet-dry weight method was used to evaluate brain edema.The content of glutamate in hippocampus was measured with the high performance liquid chromatography.The expression of GLT-1 in hippocampus areas was tested by immunohisto-chemical and Western blot methods.Results:Compared with TBI group ,TBI-induced cerebral edema was significantly attenuated ( P<0.05 ) , the content of glutamate in hippocampus was were significantly decreased ( P<0.05 ) , the level of GLT-1 were significantly increased in CTX group ( P<0.05 ).Conclusion: β-lactam antibiotics ceftriaxone can block the excitatory neurotoxicity , reduce the extent of brain edema.

Immunoelectron microscopic technique was used in the present study to observe the serotonin (5-HT)-, substance P (SP)-, and leucine-enkephalin (L-Enk)-like immunoreactive ultrastructures in the nucleus raphe magnus (NRM) of the rat. 5HT-like immunoreactive (5-HT-LI) axonal terminals were found to form axosomatic, axo-dendritic, and axo-axonic synapses with non-5-HT-LI neuronal cell bodies, 5-HT-LI and non-5-HT-LI dendrites, and non-5-HT-LI axonal terminals respectively. Non-5-HT-LI axonal terminals formed axo-somatic and axo-dendritic synapses with 5-HT-LI neuronal cell bodies and dendrites. SP-LI (or L-Enk-LI) axonal terminals formed axo-somatic, axo-dendritic synapses with SP-LI (or L-Enk-LI) and non-SP-LI (or non-L-Enk-LI) neuronal cell bodies and dendrites, respectively. L-Enk-LI axonal terminals constituting axo-axonic synapses with L-Enk-LI axonal boutons were observed less frequently. The most common synaptic type made by 3 kinds immunoreactive profiles mentioned above was axo-dendritic synapses made by non-immunoreactive axonal terminals with immunoreactive dendrites. The majority of the immunoreactive axonal boutons were mainly filled by clear spherical vesicles, but sometimes were mixed with small number of flat and granular vesicles. The immunoreactive products were irregular electron-dense substances, and were located on both inner and outer surfaces of the vesicles, or on the surface of the membranous cell organelles in the cytoplasm, etc.

Objective To investigate the responses of neurons and astrocytes within rat′s spinal trigeminal nucleus(Sp5C) following nocuous pain stimulation induced by subcutaneous formalin injection into the unilateral upper lip. Methods By means of anti\|phospholipase(PLC),anti\|Fos protein and anti\|glial fibrillary acidic protein(GFAP) immunohistochemical method,the distribution of reactive neurons and astrocytes in the Sp5C was observed at different time after formalin injection. Results No immunohistochemical staining was found in normal rat Sp5C.After formalin injection,PLC like immunoreaction(\|LI),Fos\|LI and GFAP\|LI were found in astrocytes while PLC\|LI and Fos\|LI in neurons.The labeled astrocytes and neurons showed similar distribution and close relationship.PLC\|LI and Fos\|LI were found earlier than that in neurons.Conclusion\ The results suggested that the astrocytes in the Sp5C might participate in pain modulation in CNS and actively regulate neuron′s function.

Objective To evaluate the inhibitory effect of cationic Liposomes complexed to plasmids encoding endostatin and/or angiostatin on the growth and metastasis of Lewis lung cancer in mice model. Methods C57BL/6j mice were established as mice model. Cationic liposome complexed plasmids encoding angiostatin and endostatin were administrated intratumorally to inhibit the growth and metastasis of the implanted tumor. The size change of the tumor; metastasis in lung, the activity, nourishment, survival period of the mice were observed to evaluate the function of cationic liposome complexed plasmids. Results The treatment group could inhibit the growth and metastasis of the implanted tumor. 10.Construction of recombinant plasmid harboring human peptide antibiotic gene hPAB-? and its expression in E. coli

Objective To construct the recombinant plasmid with a human peptide antibiotic hPAB-? gene and to make it expressed in E. coli. Methods To replace the CNBr cleavage site in plasmid pFAST-hPAB-? (CNBr), a pair of primers containing the hydroxylamine cleavage site were designed, and the amplified PCR fragments were cloned into pFAST-HTa plasmid to produce pFAST-hPAB-? (HA), which was then transformed into E. coli DH10B. The constructed plasmid was identified by Ehe Ⅰ/Hind Ⅲ digestion and direct DNA sequencing. An Ehe Ⅰ/Hind Ⅲ digested fragment from pFAST-hPAB-? (HA) was subcloned into pQE32-CP to construct pQE32-CP-hPAB-?, which was transformed into E. coli JM109. The bacteria containing the expression plasmid were induced to express the fusion protein by IPTG. SDS-PAGE was carried out to analyze the molecular weight, expression quantity and expression form of the target fusion protein. After captured by Ni-NTA affinity column, the fusion protein was subjected to hydroxylamine cleavage analysis. Results An expected 230bp fragment was obtained by digesting pFAST-hPAB-? with Ehe Ⅰ/Hind Ⅲ. After this fragment was cloned into pQE32-CP, the recombinant plasmid was confirmed to contain the correct target sequence by DNA sequencing. The recombinant plasmid pQE32-CP-hPAB-? could express a desired protein with a relative molecular weight about 27kD, and its expression level reached 43 percent of the total bacterial proteins. The inclusion bodies were lysed by 8mol/L urea, and the fusion protein could then be captured by Ni-NTA column and cleaved by 2mol/L hydroxylamine at pH9.0. Conclusion The recombinant plasmid pQE32-CP-hPAB-? has been successfully constructed, and it can express the desired hPAB-? fusion protein in E. coli JM109 at high level. These results provide the foundation for future research.
