1.Influence of different disinfection solution on infection and pain of puncture hole in patients with open fracture of both tibia and fibula treated with external fixation

Objective To discuss influence of different disinfection solution on infection and pain of puncture hole in patients with open fracture of both tibia and fibula treated with external fixation.Methods 41 patients were divided into group A(21 cases)and group B(20 cases).Group A adopted 75% ethanol to sterilize the puncture holes.While the group B used 5%iodide instead.The incidence of infection and pain of puncture holes was observed separately at the 1st week,6th week after operation and removing of fixation pin.Results The incidence of infection of puncture holes was not apparently different from each other(P>0.05)at any other time points.The incidence of paint in group B was lower than group A at the 1st week and 6th week after operation(P<0.05).The incidence of pain was higher in patients with puncture hole infection than those without puncture hole infection(P<0.01).Conclusion No difference was seen in puncture hole infection with either 75% ethanol or 5% iodide.Infection of puncture hole was correlated with pain only at the 1st week after operation.Pain could be alleviated with application of 5% iodide.

Sodium Ferulate(SF)is sodium salt of ferulic acid.Experimental studies showed that SF can reduce ischemical reperfusion injury of brain,heart,and liver.Clinically,SF was widely used in cardio-cerebrovascular diseases and immune system disorder.In the article,the experimental studies on SF preventing ischemical reperfusion injury of tissues were reviewed.
3.Application of nursing process in the management of mother and child care among floating population

Objective To explore the management mode for mother and child care service among floating population. Methods Nursing process was applied for assessment, diagnosis, plan, implementation and evaluation of management mode for mother and child care service among floating population in Shiqiao street, Xiacheng district, Hangzhou city. Results Compared with those three years before implementing the nursing process, themanagement of mother and child care was enhanced, for example, the record rate of pregnant woman increased by 208.82% and the systematic management rate increased by 32.70%, the record rate of child care increased by 931.73%, the systematic management rate of child care increased by 31.85%. Conclusions If used properly, nursing process can help to promote the management of mother and child care among floating population greatly.

6.The Relationship of Polymorphism of TNF-? and Type 2 Diabetes Mellitus in Elderly Patients

Objective To study the relationship of TNF-? G308A and G238A polymorphism with type 2 diabetes mellitus in elderly patients.Methods Blood specimens were collected from 60 elderly patients with type 2 diabetes mellitus and 30 normal elderly volunteers.Genomic DNA was extracted.The primers of PCR for the SNPs of G308A and G238A were optimized and the polymorphism of amplicons was analyzed by DNA sequencing.Results There was a significant difference of TNF-?-308 gene in DM patients compared with control group,while no difference of TNF-?-238.Conclusions The 308 G-A polymorphism of TNF-? promoter gene may be associated with type 2 diabetes mellitus in elderly patients.
7.LOCALIZATION OF NMDA RECEPTORS IN THE VENTRAL HORN =OF SPINAL CORD OF ADULT RAT AND DURING POSTNATAL DEVELOPMENT

Objective To investigate the cellular localization and developmental patterns of NMDA receptor subunits(NMDAR1 and NMDAR2A/B) in the ventral horn of rat spinal cord during postnatal stages. Method Immunohistochemical staining method was used in the present study. Results NMDAR1-or NMDAR2A/B-immunorectivity is abundantly observed in the lamina Ⅷ and Ⅸ of ventral horn.The immunopositive product is localized mainly in the neuronal somata,dendrites and axon terminals.Dynamic changes of NMDA receptor expression occur in early postnatal stages.NMDA receptor subunits express weakly at postnatal 7 days (P7),goes up at P14,reaches maximum level at P21,then remains this level until adult. Conclusion Neuronal functional NMDA receptors are composed of NMDAR1 and NMDAR2A in heteromeric configuration in the ventral horn of spinal cord;NMDA receptors may be involved in motor neuron maturation and plasticity during postnatal development.;
8.CONSTRUCTION OF THE MUTANTS OF HUMAN PEPTIDE ANTIBIOTICS hPAB-? AND ITS RECOMBINANT BACULOVIRUS

The aim was to determine the antimicrobial activities of peptide antibiotics hPAB ? and construct the recombinant baculovirus of its mutants. Four mutants of hPAB ? were designed based on molecular autosyndetic modeling and its genes were cloned by PCR. The transfer plasmid and recombinant baculovirus were constructed. The results showed that the hPAB ? cloned previously, is a good peptide antibiotics. Four mutant genes of hPAB ? were inserted into pFAST HTa plasmid and the recombinant pFAST hPAB ? were screened by restriction enzymes analysis and DNA sequencing. The recombinant baculovirus was obtained after transforming pFAST hPAB ? into Escherichia coli DH10Bac. Our work lays a good foundation for further research.
9.Expression, purification and bioactivity of a recombinant fusion protein consisting of anti-HBs single chain Fv and interleukin-2

Objective To study the expression, purification and bioactivity of a recombinant fusion protein consisting of anti-HBs single chain Fv and interleukin-2. Methods The engineering bacterium M15[pQE-ScFv-IL-2] which can express the fusion protein consisting of anti-HBsAg single chain Fv and interleukin-2, was induced by IPTG, then a 43kD recombinant protein was identified by SDS-PAGE and Western-blot analysis. Results The ratio of target protein to total protein of host reached 18%. Further analysis confirmed that the recombinant protein formed inclusion body in the cytoplasm of bacteria. 95% purity could be achieved after two-step purification of ScFv-IL-2, including Ni metal chelating chromatography (the first) and ion-exchange chromatogram (the second). The bioactivity assay of the purified product showed that the antibody-cytokine fusion protein could bind to HBsAg specifically and stimulate the proliferation of CTLL-2. Conclusion These results suggest that the fusion protein retains the bioactivity of its parental molecules, and may be a potential gene-engineering targeting drug for the treatment of chronic hepatitis B and other relevant diseases.
10.Expression of recombinant humanized anti-HBsAg Fab fragment in genetically engineered Pichia pastoris in high-density fermentation

Objective To study the expression in high density fermentation of anti HBsAg Fab fragment in Pichia pastoris , and the purification and activity detection of expressed target protein. Method The high density fermentation of genetically engineered Pichia pastoris was proceeded in a 5L bioreactor using fed batch fermentation. The fermentation temperature was set at 28-30℃,the pH was 5 0-5 5, and the DO was kept over 20%. When the absorbance (OD 600 ) of the broth reached 400-450 (the first time of fermentation), 200-250 (the second time), and 300-350 (the third time), the induced phase was initiated, and the methanol concentration was 0 5%-1%. The fermentation ended after 96h's induction, the target protein was purified by affinity chromatography, and its activity was assessed by ELISA. Results It showed that the optimum initial cell density during methanol induced phase should be 300-350, which was good for control of the fermentation process and the expression of recombinant Fab. At the end of the fed batch phase, a yield of about 245mg/l of Fab was reached, and 98% purity could be reached as demonstrated by affinity chromatography. The results of ELISA showed that the supernatant of fermentation and the purified recombinant Fab could bind to HBsAg specifically. Conclusion The success of high density fermentation lays a sound foundation of mass production and clinical applications of recombinant humanized anti HBsAg Fab fragment
