Purpose]Research on otolaryngology disease properties and the classical prescription method, explore the mechanism and the application of the treatment of otolaryngology disease. [Methods]Selection of swel ing ear, ear vertigo, slowly closing pus ent common diseases such as ear, combined with the Treatise on Febrile Disease and Synopsis provisions, analysis of the disease by syndrome differentiation thinking, expound the classical prescription principles and the range for the treatment of the disease. [Results]Treating most otolaryngology disease can use the classical prescription, the curative effect of treatment of otolaryngology disease has obvious advantages. [Conclusion]It has been verified by classical prescription and clinical use, efficacy, and the system of subtlety, ingredients to streamline, familiar with the classical prescription treatment of otolaryngology disease, wil improve the level of physician treatment based on syndrome differentiation and patients to reduce the economic burden, has broad prospects.
3.Mechanisms of rosuvastatin in treatment of elderly patients with coronary heart disease complicating hyperlipidemia

Objective To observe the changes of serum lipids ,hs‐CRP ,MFG‐E8 and Klotho gene levels after rosuvatatin treatment in elderly patients with coronary heart disease complicating hyperlipidemia for investigating the action mechanisms of ro‐suvatatin and its application value .Methods Totally 129 elderly patients with coronary artery disease complicating hyperlipidemia in our hospital were randomly divided into two groups .The control group received only conventional treatment ,while on this basis the rosuvatatin group was given rosuvastatin 10 mg everyday ,with 2 months as a treatment cycle .Blood lipids ,hs‐CRP ,MFG‐E8 and Klotho gene levels before and after treatment were compared between the two groups .The regulation effect of rosuvatatin was investigated Results The blood lipod ,hs‐CRP ,MFG‐E8 and Klotho before treatment had no obvious difference between the two groups .The levels of TC ,LDL‐C and TG after treatment in the rosuvatatin group were significantly decreased compared with the control group ,while the HDL‐C level was significantly increased ,the differences were statistically significant (P<0 .05) .In addi‐tion ,the hs‐CRP level after treatment in the rosuvatatin group was significantly lower than that in the control group (P<0 .05) . Compared with the control group ,the levels of MFG‐E8 and Klotho after treatment in the rosuvatatin group were increased ,the difference was statistically significant (P<0 .05) .Conclusion Rosuvastatin could decrease the blood lipid and hs‐CRP levels ,up‐regulates the MFG‐E8 and Klotho levels ,alleviates the inflammatory reaction and has the anti‐vascular aging effect ,thus effectively treats the patients with coronary heart disease complicating hyperlipidemia .
4.Construction of recombinant eukaryotic expression vector pEGFPCI-HIF-1α and its expression in eukaryotic cells PC12

Objective To construct the eukaryotic expression vector with the mutant of hypoxia inducible factor 1α (HIF-1α) that could over express nonhypoxic dependent HIF-1α and investigate its expression in eukaryotic cells PC12. Methods The coding sequence of HIF-1α was amplified from total RNA from the injured spinal cord of rats by means of RT-PCR. Then, overlapping extension PCR was employed to obtain one complete sequence encoding HIF-1α mutant without oxygen-sensitive degradation domain (ODD). By using gene recombination technique, the gene HIF-1α△ODD was inserted into eukaryotic expression vector _pEGFPC1 that contained a reporter gene of enhanced green fluorescent protein. The recombinant plasmid was successfully constructed and transfected into the neural cell line PC12. The expression of HIF-1α△ODD was analyzed by fluorescent observation and Western blot. Results A 2kb gene fragment encoding H1F-1α△ODD was successfully obtained by overlapping extension PCR. On this basis, the recombinant plasmid _pEGFPC1-HIF-1 α△ODD was constructed and testified with restriction enzyme digestion and DNA sequencing. After this recombinant plasmid was transfected into the neural cell line PCI2, both fluorescent observation and Western blot demonstrated that HIF-1α△ODD was over expressed under nonhypoxia conditions. Conclusion The eukaryotic expression vector _pEGFPC1-HIF-1α△ODD is successfully constructed and HIF-1α△ODD over expresses in transfected PCI2 cell lines.
5.Effect of RNA silencing of transforming growth factor ?1 on the growth of colon cancer cells by RNA interference

Objectives To observe the effect of inhibition of transforming growth factor (TGF)?1 on the growth and aggression of colon cancer cells by RNA interference. Methods HCT116 colon cancer cells were transfected with small interfering dsRNA (siRNA), the effect of the inhibition of TGF?1 was tested by semi-quantitative RT-PCR, and changes in cell cycle distribution were analyzed by flow cylometry, the ability of aggression was detected by soft agar colony assay. Results The inhibition of the expression of TGF?1 was significantly detected after the transfection of siRNA against TGF?1, which induced the increase of S phase by 59.0%-82. 5% and decrease in G2 phase, colony assay further demonstrated that siRNA against TGF?1 led to a significant reduction in colony formation as compared with the control group. Conclusions The present data suggested that the expression of TGF?1 was conveniently and rapidly blocked by siRNA transfection, and this may lead to the cell cycle arrest and inhibition of the growth and tumorigenesis of colon cells.
6.Prevention and treatment of rectal complications after ~(125)I brachytherapy for prostate cancer

Objective To evaluate the prevention and treatment of rectal complications following 125I brachytherapy for prostate cancer. Methods A total of 90 patients with locally advanced prostate cancer was treated by transrectal ultrasound guided prostate 125I brachytherapy combined with maximal androgen blockage from November 2001 to June 2006. The patients were followed every 1~3 months postoperatively, and rectal complications was analyzed and treated. Results Eighty-nine patients were followed for 1~55 months (mean, 22 months). The incidences of rectal complications at grade Ⅰ, Ⅱ, Ⅲ, and Ⅳ were 11.2% (10/89), 6.7% (6/89), 0%, and 2.2% (2/89), respectively. Symptomatic therapy was conducted in 16 patients with grade Ⅰ or Ⅱ complications, which were symptomatically relieved 6 months later. The grade Ⅳ complications occurred in 2 patients, one of whom underwent repeated examinations and treatment for proctitis, and the other of whom was complicated with severe diabetes and received a laparocolostomy with cystostomy. Conclusions The optimal medical treatment for radiation proctitis is follow-up checkups and observations. To reduce the incidence of urethrorectal fistula, excessive examinations and treatment should be avoided. Accompanying diabetes should be treated promptly and actively.
7.A proposed criterion for clinical diagnosis of internet addiction

Objective To establish a clinical diagnostic criterion(referred as criterion hereinafter) for patients with social dysfunction and abnormal mental behaviors caused by internet addiction,so as to provide guidance in future mechanism study,treatment and prevention for such disorder.Methods A rough draft of symptom parameters was used to evaluate the symptoms among the patients with a chief complaint of wallowing in internet which caused damned influence on study,work and interpersonal communication.The 1st draft of the criteria was drawn up according to the incidence and correlation of the symptoms.408 patients with internet addiction(IA) diagnosed according to the 1st draft were followed up for 12 months after being discharged from hospital,and the diagnostic coincidence and recurrence rate were calculated and analyzed,based on which the 1st draft was then revised and the 2nd draft was established.Clinical tests were performed on 150 patients with the same chief complaints by psychiatrists from many mental hospitals in China.Descriptions in the 2nd draft were not modified based on the test findings and the final draft was formulated.Results The proposed criterion for IA symptom included the following 7 items: ① craving for using internet;② withdrawal reactions after internet use reduced or stopped;③ tolerance;④ unable to control the intention of using internet;⑤ regardless of hazardous consequences;⑥ pay no attention to other activities;and ⑦ avoidance or alleviation of unhealthy emotions.Items ① and ② combined with any one of the remains made the final diagnosis of IA.The criterion for illness duration was suggested as greater than or equal to 6 hours of continuous internet use one day in average and 3 months at least after meeting the criteria for IA symptom.It was found by following-up that one year recurrence rate among the discharged patients from hospital was 70.6%,and the diagnostic coincidence was 96.1%.The diagnostic consistency on symptom evaluation between two psychiatric raters was 89.3%-98.0%(Kappa 0.73-0.87,P

AIM: To study the effects of Shenmai injection on cardiomyocytes apoptosis after acute anoxia-reoxygenation (A/R) and the possible mechanism. METHODS: In this experiment, cultured cardiomyocytes isolated from neonatal rat were used. Model of myocardial anoxia-reoxygenation injury was produced by depriving oxygen for 5 min and then restoring oxygen for 15 min. The apoptotic cells was detected by flow cytometry to detect labbled Annexin V-FITC/PI. The intracellular calcium level was observed by laser scanning confocal microscopy markered Fluo-3/AM. AIM: To study the protective effects of Shenmai injection, a Chinese medicine, on angiotensin Ⅱ (AngⅡ)-induced rat cardiomyocyte apoptosis in vitro and the probable mechanism. METHODS: Cultured cardiomyocytes from neonatal rats were stimulated with AngⅡ. 10.Research progress of patient-derived xenografts in precision cancer medicine

Cancer is a group of heterogeneous disease caused by diverse genomic alterations in oncogenes and tumor suppressor genes .Despite recent advances in high-throughput sequencing technologies and development of targeted therapies, novel cancer drug development is limited due to the high attrition rate from clinical studies .Patient-derived xenografts ( PDX) models are generated by implanting sectioned patient tumor fragments into immunodeficient mice .PDX models retain many of the key characteristics of patients ' tumors including histology , genomic signature , cellular heterogeneity , and drug responsiveness .These models cannot only serve as a platform for co-clinical trials by enabling the integration of clinical data , genomic profiles , and drug responsiveness data to determine precisely targeted therapies , but also be applied to the development of biomarkers for drug responsiveness and personalized drug selection .This review summarizes our current knowledge of this field , including methodologic aspects , applications in drug development , challenges and limitations , and utilization for precision cancer medicine .
