1.Influence of heating of laparoscopic pneumoperitoneum on recovery of patients after gastrointestinal operation

Objective To study the influence of carbon dioxide (CO2)gas temperature on recovery of patients after laparoscopic gastrointestinal operation.Methods 100 cases with gastrointestinal operation patients were selected and randomly divided into the experimental group and the control group with 50 patients in each.In the experimental group,pneumoperitoneum was established with CO2 gas heated to 37 ℃,and the control group used routine method to establish pneumoperitoneum.The items such as postoperative tracheal extubation time,anal exhaust time and extubation time were observed in two groups of patients.Results The endotracheal tube extubation time,anal exhaust time and extubation time in the experimental group were earlier than those in the control group.Conclusions Using CO2 gas heated to 37 ℃ to establish pneumoperitoneum during laparoscopic gastrointestinal operation can effectively promote the rehabilitation of patients after operation.
2.Determination of Related Substances in Donepezil Hydrochloride Tablets by HPLC

OBJECTIVE:To establish an HPLC method for the determination of related substances in Donepezil hydrochloride tablets.METHODS:The separation was performed on AlltimaTM C18 column with mobile phase consisted of 0.01 mol?L-1 dipotassium hydrogen phosphate(3% triethylamine,pH=4.3)-methanol(60 :40) and flow rate of 1.0 mL?min-1.The detection wavelength was set at 271 nm and the column temperature was maintained at 35 ℃.The content of related substance was calculated by self-control with main component.RESULTS:Related substances were completely separated from the main constituent and the impurities were separated well from each other.The determination limit was 0.05 ng and the contents of related substances were 0.42%.CONCLUSION:The method is specific,sensitive and reproducible for the determination of related substances in Donepezil hydrochloride tablets.
3.HCV NS5A and NS5B Enhance Expression of Human Ceramide Glucosyltransferase Gene

Host genes involved in lipid metabolism are differentially affected during the early stages of hepatitis C virus (HCV) infection.Here we demonstrate that artificial up-regulation of fatty acid biosynthesis has a positive effect on the replication of the HCV full-length replicon when cells were treated with nystatin.Conversely,the HCV RNA replication was decreased when fatty acid biosynthesis was inhibited with 25-hydroxycholesterol and PDMP(D-threo-1-phenyl-2-decanoylamino-3- morpholino-1-propanol).In agreement with these results,the expression level of GlcT-1(ceramide glucosyltransferase),a host glucosyltransferase in the first step of GSL (glycosphingolipid) biosynthesis,was found to be closely associated with the expression and replication of HCV RNA.On the other hand,the viral RNA can also activate GlcT-1 in the early stage of viral RNA transfection in vitro.To identify viral factors that are responsible for GlcT-1 activation,we constructed ten stable Vero cell lines that express individual HCV proteins.Based on the analyses of these cell lines and transient transfection assay of the GlcT-1 promoter regions,we conclude that HCV proteins,especially NS5A and NS5B,have positive effects on the expression of GlcT-1.It is possible that NS5A and NS5B stimulate transcription factor(s) to activate the expression of GlcT-1 by increasing its transcription level.

5.Comparative study on family cohesion and family education style of adolescents with internet addiction disorder

Objective To compare internet addiction disorders' family cohesion and their family educa-tion style, and to help prevent internet addiction disorder and interference treatment in families with internet addic-tion disorder. Methods Ninety adolescents with internet addiction disorder were investigated as the experimental group and one hundred normal adolescents as the control group. Questionnaire and in-depth interview was used to collect data,and the data was analyzed with SPSS 10.0. Results Average scores of control group were significant higher than those of experimental group in family cohesion ((51.75±8.86) vs (38.89±12.17), P<0.01). Av-erage scores of control group were significant higher than these of experimental group in family education style ((50.65±7.66) vs (41.44±10.84), P < 0.01). Average scores of experimental group were significant higher than those of control group in dissatisfaction of family cohesion and family education style ((20.87±4.66) vs (12.33±7.88); (23.30±11.06) vs (14.28±7.29), P < 0.01). Conclusion The adolescents with internet addiction disorder have low family cohesion and high dissatisfaction of family education style,wherefore during in-ternet addiction family intervention and prevention, it should pay particular attention to cultivating close and harmo-nious relations among families members. In aspect of family education, parents should pay special attention to chil-dren's healthy personality development, and guide their children to actively take part in social practice and train-ing.
6.Effects of herbal medicine extraction on bone destruction and pain behaviors in rats of bone cancer pain

Objective To evaluate therapeutic effects of Bushen-Zhuanggu medicine extraction on bone destruction in rats bone cancer pain.Methods Rat models of cancer-induced bone pain were established by inoculating MRMT-1 cells into tibia of rats.Behavioral signs indicative of pain including 50%paw withdrawal threshold(von Frey tactile sensitivity test)and thermal withdrawal latency were observed.Pathomorphological changes of tibia were monitored with HE staining.Results In the behavioral tests,herbal medicine treatment attenuated mechanical allodynia and thermal hyperalgesia.Histological examination showed that this treatment inhibited tumor proliferation and preserved the cortical and trabccular bone structure.Conclusion Bushen-Zhuanggu medicine extraction is an anti-nociceptive and bone-preserving agent in rats of bone cancer pain.
7.Expression of interferon-stimulated gene ISG20 and investigation of its antiviral effect on HCV replicon

Objective:To study the antiviral effect of ISG20 on HCV replicon.Methods:Wild type ISG20/mutated ISG20 cDNAs were obtained by RT-PCR/two step-PCR directed mutagenesis, and wild type ISG20 and dominant negative mutated ISG20 mammal expression vectors were consuructed. The constructed pISG20wt and pISG20m expressing vectors were transfected into Huh7 cells or Huh7 cells containing HCV replicon to investigate its effects on HCV replicon replication.Results:The ISG20wt/ISG20m expression vectors were constructed and the expressions of these two vectors were confirmed at both mRNA and protein levels. The effects of ISG20wt on HCV replicon replication were evaluated by Northern blot and Western blot. The results showed that expression of ISG20wt had significant inhibitory effect on HCV RNA replication.Conclusion:ISG20 participates in the anti-HCV action of IFN-? on HCV replicon system.
8.Application of early warning model based on decision tree for medical equipment quantity and quality

Objective To propose a decision tree-based early warning model so as to predict the medical equipment quantity and quality under special conditions to quantize maintenance and detection staffs allocation,components supply,emergency planning and etc.Methods A data set was established based on the verification report,data on performance parameters detection and daily management log,and then underwent pretreatment.A decision tree came into being with the algorithms of ID3,CHAID and etc.The data rule corresponding to the decision tree was transformed into a series of early warning information.The model was verified by applying it to medical equipment usage management.Results The decision tree developed was applied to analyzing a vehicle-mounted water purifying device maintenance and usage records in some logistics support vehicle,and it's found the main factors contributing to the failures included migration distance along non-paved road,ambient temperature and service time.Conclusion The decision tree-based early warning model for medical equipment quantity and quality gains high feasibility and practical values.
9.The changes of the ratio of T helper 17 and regulatory T cells in the peripheral blood of patients with anti-tuberculosis drug-induced liver injury and its significance

Objective To observe the changes of the peripheral blood T helper (Th)17 cells/regulatory T cells (Treg) ratio in patients with anti‐tuberculosis drug‐induced liver injury ,and to explore the relationship between Th17/Treg ratio and liver function . Methods Ninety inpatients with the diagnosis of secondary pulmonary tuberculosis (TB) from Affiliated Hospital of Zunyi Medical College from Feb .2013 to Mar .2015 were included in this study .All these patients had normal base line liver function .After 2 weeks of anti‐TB chemotherapy ,21 patients with alanine transaminase (ALT) and/or aspartate aminotransferase (AST) ≥2 × upper limit of normal (ULN) were defined as TB with liver injure group ,the other 69 patients with ALT and/or AST<2 × ULN were defined as TB with non‐liver injured group .Before treatment and two weeks after anti‐TB therapy ,the peripheral blood Th17/Treg ratios and liver functions of all patients in both groups were detected in order to compare the changes of Th17/Treg ratios pre and post anti‐TB chemotherapy and to explore the relationship between Th17/Treg ratio and ALT/AST levels .Student′s t test was used for statistical analysis ,and Pearson correlation analysis was used for correlation test .Results Before treatment ,there was no statistical significance of Th17 ,Treg and Th17/Treg ratio between groups (P> 0 .05) .The Th17 levels in TB with liver injured group ([2 .522 ± 0 .388]/mL vs [2 .075 ± 0 .369]/mL ,t=3 .633 ,P<0 .01) and TB with non‐liver injured group ([2 .326 ± 0 .348]/mL vs [1 .929 ± 0 .402]/mL ,t= 6 .468 , P< 0 .01) ,and Th17/Treg ratios in both groups(0.618±0.104vs0.489±0.107,t=3.553,P<0.01 ;0.554±0.108vs0.450±0.098,t=6 .353 ,P<0 .01) were all increased after anti‐TB chemotherapy ,with statistically significant differences . However ,there was no statistical significance of Treg levels pre and post anti‐treatment in both groups (both P>0 .05) .The Th17 level and Th17/Treg ratio in TB with liver injured group were significantly higher than those in TB with non‐liver injured group (t= 2 .203 , P< 0 .05 and t= 2 .345 , P< 0 .05 , respectively) .In liver injured group ,the Th17/Treg ratio was positively correlated with ALT level (r=0 .849 ,P=0 .044) ,but not with AST level (P>0 .05) .Conclusion The changes of Th17/Treg ratio are related with anti‐TB drug‐induced liver injury in patients with secondary pulmonary tuberculosis .
10.Study on fluorescent quantum dot immunofiltration assay for quantitative detection of C-reactive protein

Objective To study the feasibility of using fluorescent immunofiltration test based on quantum dots (QDs)for rapid and quantitative detection of C-reactive protein.Methods Based on homemade QDs and QDs-antibody bioconjugates,an immune de-tection method was established via the double antibodies sandwich technique on the immunochromatography card.The test results could be read under the irradiation of UV light,and quantitative results could be measured through the combination of a laser and fluorescent spectroscopy.Results Under UV light irradiation,the minimum detection concentration of CRP was 0.156 mg/L.U-sing the quantitative detection method,the fluorescent intensities on the cards could be established a linear relationship with the con-centration of CRP,and the linear equation was that log(Y )=0.563 log(X)+4.570,r2 =0.958.Conclusion The fluorescent quan-tum dot immunofiltration assay can be used for quantitative detection of CRP;The quantum dots immuno-labels have the potential to develop new type of immune-diagnostic reagents.
