2.Effects of sex, day-night rhythm on the concentration of the growth hormones in normal people

Growth hormone (GH) concentration in the blood of healthy adult was measured by RIA (radioimmunoassay) method. The results are as follow: Concentration of GH in the blood of male is lower than in female(p<0.05). There is no significant difference in concentrations of GH at 8-10 AM and 6-9 PM.

It is evident that an elevation of airway albumin excreation rate without clinical proteinuria strongly predicts a later progression on diabetic renal disease. So we studied the correlation between Microalbumin checkly RIA & Micral-Test®. We collected urine between 08:00 h and 08:00 h next day and then checked microalbuminuria by radioimmunoassay method and Micral-Test® The results are as follows: 1. There was significant correlation between microalbuminuria checked by RIA & Micral-Test® 2. There was poor correlation between diabetes duration or HV-A1c and maximal change in albumin excreation rate. 3. So we concluded that Micral-Test® can be used in laboratory instead of RIA.

4.The relation of the thyroid cancer with the tumor marker quantified by the radioimmuno assay.

Using the radioimmuno assay to quantify the serum concentrations thyroglobulin and calcitonin in the normal people and patients with the thyroid cancer to evaluate the relation between them and apply this technique for diagnosing the cancer. The comparison found that the serum calcitonin in the patients with the thyroid cancer was lower than this in normal people during the severe or treated phase of the disease. The serum calcitonin concentration was only applied in the phase that the thyroid cells has not been destroyed, phase of relapse or remote metastasis.

5.Evaluation of ES-300 for the Detection of Anti-HCV Antibody.

BACKGROUND: A fully automated enzyme-immunoassay (EIA) analyzer, Enzymun System, ES-300 (Boehringer Mannheim, Germany) uses streptavidin technology and performs single test or panels of up to 12 tests per run. We evaluated the results of ES-300 for anti-HCV by comparing the results with microplate-EIA, radioimmunoassay (RIA), and confirmatory test. METHODS: Total 79 sera (51 positive, 24 negative, 4 indeterminate results confirmed by Lucky HCD Confirm) were analysed. ES-300 with Enzymun-Test(R) Anti-HCV (Boehringer Mannheim, Germany) and microplate-EIA (Green Cross Center Innotest HCV 3.0(R)) were used. Fifty one sera were examined additionally by 2nd-generation RIA method, NANBDINE 125C(General Biologicals Corp., R.O.C.). And all results were compared to the results of Lucky HCD Confirm. RESULTS: The overall concordance rate of ES-300 and Innotest(R) was 72/79 (91.1%). The results of Lucky HCD Confirm on seven discrepant samples were five negative and two indeterminate. The results of ES-300 and NANBDINE 125C showed concordance rate of 90.2%. The sensitivity and specificity of ES-300 with regard to Lucky HCD Confirm were 94.5%, and 87.5%, respectively, and that of Innotest(R) were 98.2% and 66.7%, respectively. Clear distinction of positive and negative results by signal/cut off ratio was available in both EIAs. The positive predictive values of ES-300 and Innotest(R) were 94.5%, and 87.1%, respectively. CONCLUSIONS: ES-300 showed relatively good results in sensitivity and positive predictive value with regard to confirmatory test. In EIA-positive persons, however, follow-up study would be necessary for reliable evaluation of HCV infection.

7.Postictal Serum Prolactin Values and Its Significance in Convulsive Disorder.

The authors analysed 196 cases of convulsive disorder in children who were admitted to the pediatric department of Kyunghee Hospital from June 1993. Blood samples were drawn immediately following seizures on arrival. Serum prolactin values were measured by radioimmunoassay. The results obtained were as follows: 1) Mean serum prolactin value was 55.4+/-20.8ng/ml in generalized seizures within 1 hr following seizures, 22.5+/-3.9ng/ml in partial seizures. There was a significant difference between generalized scizures and partial scizures (p<0.05). 2) Mean serum prolactin value was 23.5+/-9.7ng/ml in generalized seizures within 1~3 hrs following seizures, 10.4+/-4.9ng/ml in partial seizures. There was a significant difference between generalized seizures and partial seizures (p<0.05). 3) Mean serum prolactin value was 9.5+/-4.4ng/ml in generalized seizures 3 hrs following seizures, 9.8+/-4.8ng/ml in partial seizures. There was no significant difference between generalized seizures and partial seizures (p>0.05). We found transient hyperprolactinemia following generalized seizures but a little change following gartial seizures. Postictal elevation of serum prolactin may represent a biochemical marker of generalized and partial seizures.

8.Relationships among Ambulatory Plasma Renin Activity, Blood Pressure and Urinary Microalbumin Excretion Rate in Essential Hypertension.

OBJECTIVE AND METHODS: To determine correlations among ambulatory renin activity, ambulatory blood pressure and microalbumin excretion rate, 66 Korean essential hypertensives were studied after 4 week wash-out period. The ambulatory blood pressure was monitored every 30 minutes and mean BP were calculated automatically. Urinary microalbumin excretion rate(UAER) and ambulatory plasma renin activity(aPRA) collected at mid-day were measured by radioimmunoassays. Subjectives were divided into 2 groups by aPRA value(2ng/ml/hr). RESULT: 14 cases were high renin group and 52 cases low renin group. The mean BP were 148.83/94.69mmHg in low renin group, and 146.57/98.07mmHg in high-renin group without difference. UAER were not different also between both groups. 23.07%(4/14) of non-dippers were included in high renin group and 25.58%(12/52) in low renin group without statistical difference. The aPRA was significantly related to UAER and systolic and diastolic mean blood pressure. Also UAER was related significantly to day mean blood pressures. CONCLUSION: Thus aPRA is thought to be a meaningful indicator to predict hypertensive renal target organ damage as well as blood pressure measured with 24-hr ABPM.

9.Influence of the iodized salt program in community on the determination of thyroid gland function by using radioimmunoassay and immunometric assay

The study was conducted on 82 simple goiter patients and 91 Basedow patients after at least one year using iodized salt daily. The results showed that the uptake of radioactive iodine (RAIU) by the thyroid gland of simple goiter and Basedow patients decreased after 2 and 24 hours taking an oral dose of radioactive iodine. Especially, the decrease was 40-50% in simple goiter patients. The concentration of T3, T4, TSH in the sera was as following: in the simple goiter patients, the concentrations of T3 and TSH were slightly decreased, and the T4 serum concentration was increased. However, all the values were still within the normal range of euthyroidism. In Basedow patients, the concentrations of T3, T4, and TSH have not changed.

10.Dehydroepiandrosterone sulphate (DHEAS) levels in premenopausal women with systemic lupus erythematosus.

OBJECTIVE: Dehydroepiandrosterone sulphate (DHEAS), the major steroidal product of the human adrenal, is abnormally low in patients with SLE. Moreover, a recent study confirms a positive effect of the precursor DHEA on the disease course in SLE, which supports an etiologically important role of the hormone on SLE. The aim of this study is to search for an interrelation between clinical manifestations, laboratory findings, and disease activities and DHEAS in patients with SLE. METHOD: DHEAS were measured by radioimmunoassay kit using 125I-labeled DHEA-SO4 antibody-coated tube in the serum of 48 patients with SLE and in 46 control subjects. Laboratory findings, clinical symptoms, signs and SLE disease activity index (SLEDAI) in SLE patients were evaluated at blood sampling time. RESULTS: DHEAS was lower in patients with SLE compared to controls (45.60+/-42.62 mug/dL vs 101.55+/-56.54 mug/dL, p<0.005). The serum DHEAS levels were significantly negative correlation with SLEDAI (r=-0.333, p<0.05). There were significantly negative correlations between daily steroid dose and DHEAS (r=-0.384, p=0.012), and the amount of steroid used during previous 2 months and DHEAS (r=-0.011, p=0.011). CONCLUSIONS: Patients with SLE have low levels of DHEAS and significant negative correlation between SLEDAI, steroid dose and DHEAS.
