1.The effect of Jiangu tablet on articular chondrocyte apoptosis in T-2 toxin poisoning rats

Objective To study the effects of Jiangu tablet on articular chondrocyte apoptosis in T-2 toxin poisoning rats. Methods According to random number table, fifty weaning male SD rats were divided into two groups by body mass, i.e., 10 for normal control group and 40 for T-2 toxin group (intragastric administration of distilled water or T-2 toxin 200 ng·g-1·d-1) for 30 days. Then the T-2 toxin group was divided into T-2 toxin group, Jiangu tablet low-dose group, middle-dose group and high-dose group treating with 3 ml of T-2 toxin 200 ng or different concentration of Jiangu tablet (contain 1.562 5, 3.125 0 and 6.250 0 g Jiangu tablet active compound). Each group had 10 rats and was given T-2 toxin or different amount of Jiangu tablet for 30 days. Then the rats were killed. The articular cartilage was removed from rats and RNA was extracted from the articular cartilage by Trizol. The mRNA expressions of p53, Bax, Bcl-2 and cysteinyl aspartate specific proteinase(caspase)-3 were detected by real-time PCR. Results The mRNA expressions of p53, Bax, Bcl-2 and caspase-3 in normal control group, T-2 toxin group, Jiangu tablet low-dose group, middle-dose group and high-dose group were: 1.00 ± 0.98, 200.37 ± 30.39, 180.19 ± 28.14, 120.25 ± 15.35, 50.34 ± 10.12;1.00 ± 0.98, 185.37 ± 10.15, 152.59 ± 15.23, 108.46 ± 9.14, 57.18 ± 1.31; 1.00 ± 0.99,0.22 ± 0.03, 0.28 ± 0.06, 0.43 ± 0.08, 0.58 ± 0.04; 1.00 ± 0.97, 209.55 ± 25.64, 152.38 ± 15.46, 120.14 ± 11.52 and 49.24 ± 8.69, respectively. Compared with the normal control group, the mRNA expressions of p53, Bax and caspase-3 were up-regulated in T-2 toxin group, low-dose group, middle-dose group and high-dose group while the mRNA expression of Bc1-2 was down-regulated(all P < 0.05). The mRNA expressions of p53 and caspase-3 in Jiangu tablet high-dose group and middle-dose group were significantly decreased than those in T-2 toxin group (all P<0.05). The mRNA expressions of Bcl-2 in Jiangu tablet high-dose group and middle-dose group were significantly increased than that in T-2 toxin group(all P<0.05). The mRNA expression of Bax in Jiangu tablet high-dose group was significantly decreased than that in T-2 toxin group(P<0.05). Conclusion The Jiangu tablet can significantly inhibit the apoptosis of articular chondrocyte in T-2 toxin poisoning rats.
2.Proliferation and identification of dendritic cells from peripheral blood of patients with bladder cancer in vitro

Objective To investigate the proliferation and identification of dendritic cells(DC)de- rived from peripheral blood of patients with bladder cancer in vitro.Methods The mononuclear cells were prepared from peripheral blood of patients with bladder cancer by Ficoll-Hypaque centrifugation method,and were induced by the recombinant cytokines hGM-CSF(50 ng/ml),hlL-4(10 ng/ml)and hTNF-?(50 ng/ ml)for 2 weeks.The growth and morphology of DC were observed through the phase contrast or electron mi- croscope,and their pheuotypes were determined by flow cytometry.The capacity of DC to activate T cell-de- pendent anti-tumor immune responses was tested by MTT method.Results The DC cultured in vitro turned into suspensive growth from adhesive situation on the 6th day,then the number of DC increased con- tinuously and the cells showed the irregular morphologic appearance of DC with veiled edges on the 8th day. Flow cytometry showed that the mature DC expressed high levels of specific markers such as CD_(1a),CD_(83), CD_(86)and HLA-DR.T cells activated by DC showed strong cytotoxicity to bladder cancer cell line BIU87 with a killing rate of(48.8?3.7)%,while the killing rate of T cells which were not activated by DC was(25.7?1.5)%;the difference of the rate between them was significant(P<0.01). Conclusions The DC can be cultured from peripheral blood of patients with bladder cancer by induction of rhGM-CSF,rhIL-4 and hT- NF-?in vitro.This may lay an experimental foundation for further research on DC vaccine.
3.Pre-treatment with melatonin inhibits oleic acid-induced acute lung injury in rats

AIM:To assess the protective role of melatonin(MEL)in a rat model of oleic-induced acute lung injury.METHODS:Twenty-four rats were randomly allocated to three groups as follows:saline(NS)injection group,oleic acid(OA)injection group and MEL plus OA injection group,the lavage protein,lung wet-to-dry weight ratio,malondialdehyde(MDA)content,superoxide dismutase(SOD)activity and lung histopathology were examined.RESULTS:(1)Injection 0.15 mL/kg of OA led to a severe acute lung injury(ALI),characterized by significantly increasing in lavage protein,lung coefficient(P<0.01),and by histopathological alterations which presented hemorrhage,edema.thickened alveolar septum and the existence of inflammatory cells in alveolar spaces;(2)Infusion of MEL(20 mg/kg,intraperitoneally for 60 min before the oleic acid)markedly alleviated above-mentioned symptom induced by OA,consistent with decrease of MDA level(P<0.01) and the increase of SOD activty(P<0.01).CONCLUSION:Pre-treatment with MEL can attenuate the OA -induced ALI in rats via cleaning and preventing the formation of free radicals and further lessening the increase of alveolocapillary membrane permeability,these data suggest that MEL may be effective in the prevention of ALI.
4.Blood vessels and nerves surrounding the seminal vesicles: A clinical anatomic study.

OBJECTIVETo investigate the precise locations of the blood vessels and nerves surrounding the seminal vesicles (SV) in men and provide some anatomical evidence for SV-related minimally invasive surgery.
METHODSWe observed the courses and distribution of the blood vessels and nerves surrounding SVs and obtained the data for positioning the SV neuroplexes in 20 male pelvises.
RESULTSOne branch of the neuroplexes was distributed to the SVs bilaterally with the neurovascular bundles, (2.85 ± 0.18) cm from the median sulcus of the prostate (MSP), while another branch ran through the Denonvillier fascia behind the SV, (0.81 ± 0.06) cm from the MSP. The arterial SVs (ASV) originated from the inferior vesical artery and fell into 4 types, 55% going directly to the SVs as one branch, 15% running between the SV and the ampulla of the deferent duct as another branch, 25% downward as 2 branches to the SV and between the SV and the ampulla of the deferent duct respectively, and 5% as the other ASVs. The shortest distance from the ASV through the prostatic neuroplexus to the posterior SV was (1.08 ± 0.09) cm.
CONCLUSIONIn SV resection, neuroplexus injury can be reduced with a bilateral distance of < 2.85 cm and a posterior distance of < 0.81 cm from the MSP, and so can bleeding by vascular ligation between the SV and the ampulla of the deferent duct.

5.Treatment of non-infective nonunion with locking plates following surgery for long bone fractures

Objective To investigate the cause of fracture nonunion and discuss the experience of locking plates fixation combined with autoallergic cancellous bone in the management of non-infective long bone nonunion.Methods A retrospective analysis was made in 38 patients(29 males and 9 females)with fracture nonunion treated with locking plates fixation combined with autoallergic cancellous bone from August 2006 to August 2010.Of all,22 patients were treated with plates and 16 with interlocking intramedullary nails.Results All the patients were followed up for a mean of 12 months(range,6-24 months).The bone union time for all the fractures averaged 5.3 months(range,3-7 months),with no complications like implant loosening or fracture.Conclusions Iatrogenic factor is the main cause for fracture nonunion after open reduction and internal fixation of long bone fractures and the locking plates combined with autoallergic cancellous bone are a safe and effective treatment procedure.
6.Effect of osthol on memory impairment of mice in AlCl3-induced acute senile model.

AIMTo study the protective effect and mechanism of osthol on learning and memory impairment of mice with acute senile model induced by AlCl3.
METHODSAfter s.c. AlCl3 60 mg.kg-1 for 7 d and i.p. osthol 15 and 7.5 mg.kg-1 for 12 d, using step-through test and step-down test, the effect of osthol on learning and memory was observed and the glutathione peroxidase (GSH-PX) activities in blood and superoxide dismutase (SOD) activities in plasma and cerebrum were measured.
RESULTSOsthol 15 and 7.5 mg.kg-1 significantly improved the capability of memory and enhanced the activities of GSH-PX and SOD in AlCl3 treated mice.
CONCLUSIONOsthol shows protective effect on brain memory impairment of mice in acute senile model induced by AlCl3. Perhaps the mechanism is involved in enhancing the activities of GSH-PX and SOD, clearing away the free radical, protecting the brain neuron from the harm of lipoperoxide.

7.Investigation of knowledge, attitude and practice of Hepatitis B vaccine among general physical examination people in Changchun

Objective To investigate knowledge,attitude and practice of Hepatitis B vaccine among general physical examination people in Changchun and improve the rate of hepatitis B vaccine.Methods 732 cases of general medical groups were investigated with the KAP of Hepatitis B vaccine questionnaire.Results 69% of the general medical groups had very little knowledge about hepatitis B vaccine,including an annual minimum antibody level of awareness (44.7% ).Male was different form female about the knowledge (79% vs 85.1%,and x2 =4.648,P<0.05).88.1% people want learn the relative knowledge about the vaccine,and 75.1% people have get vaccinated Hepatitis B.Conclusions The hepatitis B vaccine knowledge,attitudes and behavior of physical examination population in Changchun,in general,are satisfactory.Education is the main influential factors.Therefore,it is the focal point for community nurses to carry out targeted health education in the future to improve hepatitis B vaccination rate.
8.Experience and coping styles of pulmonary tuberculosis college students

Objective To understand the experience and coping styles of college students with pulmonary tuberculosis.Methods By purposive sampling method,15 tuberculosis college students were selected to do an indepth interview in Xi' an city.The data were analyzed by the thematic framework.Results The experience of students with pulmonary tuberculosis included self-stigma,other students keep them at a distance,being shunned by neighbours and being treated differently by some policise.And the coping styles of them included self-isolation,concealing the disease,not satisfied with the policy and rationalization.Conclusions Health education about pulmonary tuberculosis should be strengthen,and more care should be paid on the pulmonary tuberculosis college students.We should try to make a good learning,living and treatment environment for them so as to promote them to cope tuberculosis more scientifically.
9.Methylation of FHIT gene promoter region in DNA from plasma of patients with myelodysplastic syndromes and demethylating effect of decitabine.

This study was aimed to detect the methylation status of FHIT gene promoter region in the DNA from plasma of patients with myelodysplastic syndrome (MDS), and to investigate the demethylating effect of decitabine. Methylation-specific PCR method was used to detect the methylation status of FHIT gene promoter region in the DNA from plasma of 4 patients with MDS before and after treatment with decitabine plus semis CAG therapy (among them, 1 case of newly diagnosed MDS, 3 cases progressed into acute leukemia). The results indicated that 3 cases were found to have an increased methylation in the promoter region. After treatment with decitabine plus semis CAG, increased methylation was reversed in 2 cases. In 4 cases, 2 cases displayed clinical response. It is concluded that FHIT gene hypermethylation is associated with MDS pathogenesis. Decitabine has demethylating effect on the FHIT gene hypermethylation of plasma from MDS patients. Detecting the methylation status of FHIT gene in DNA from plasma may play a role in MDS auxiliary diagnosis or prognosis.

10.Role of miRNA-126/miRNA-126* in the fetal lung development of rats.

OBJECTIVETo investigate the expression and role of miRNA-126/miRNA-126(*) in the fetal lung development of rats.
METHODSTwelve pregnant Sprague-Dawley rats were randomly divided into 3 groups and the fetal rats were removed at 16, 19 and 21 days of gestation respectively. Hematoxylin and eosin staining was performed to observe lung morphology of fetal rats. Then microRNA (miRNA) microarray was used to study the expression patterns of miRNA-126/miRNA-126(*) in fetal lungs at the three time points. And miRNA-126(*) was selected for further study by real-time PCR.
RESULTSThere was no evident difference in the expression of miRNA-126 among the three groups, however the expression level of miRNA-126(*) increased gradually as the fetal lung developed. The real-time PCR result further showed that expression of miRNA-126(*) increased gradually with lung development, displaying significant differences among the three groups (P<0.05).
CONCLUSIONSmiRNA-126(*) may play an important role in development of the fetal lung in rats.
