1.Effects of cleavage-stage biopsy on in vitro development of human embryos

Objective To evaluate the effects of biopsy methods, biopsy timing and the number of cell removed on in vitro development of embryos Methods One hundred and fifty four embryos of good morphology from in vitro fertilization patients were studied Sixty six embryos were allocated to the following three groups: chemical drilling biopsy group (26), mechanical drilling biopsy group (26) and control group (20) One cell was removed from the embryos of the two biopsy groups The remaining 88 embryos were allocated to two groups: biopsy group (44) and control group (44) Two cells were removed from biopsy group by chemical drilling method The stage of the embryo before biopsy, biopsy time, lysed blastomere, growth potential and hatching capacity of the biopsied embryos, total cell number at the blastocyst stage were recorded and evaluated Results The mean time of biopsy in the chemical drilling group (231?20) seconds was significantly shorter than that in the mechanical drilling group (262?23) seconds ( P 0 05) However, the proportion developing to the blastocyst stage was reduced after the removal of two cells from the 6 cell stage in comparison to the control (1/8 versus 5/8, P
2.Observation on the Therapeutic Effect of Vertigo Treated by Pinching the Spine

Objective: To observe the therapeutic effect of vertigo treated by pinching the spine. Methods: In the treatment group, 80 cases of vertigo were treated by pinching the spine and An-pressing and Rou-stationary circular pressing. In the control group, 66 cases were treated by An-pressing and Rou-stationary circular pressing. Results: The total effective rate was 95.0% in the treatment group, and 66.7% in control group. The therapeutic effect in treatment group was better than that in control group (P<0.01). Conclusions: Pinching the spine to treat the vertigo could get rapid and good therapeutic effect, and may avoid the toxic and side effect of drug treatment.
4.Construction of recombinant modified vaccinia virus Ankara with Ag85A and ESAT-6 gene and examination of their immunogenicity in mice

Objective To construct five types of recombinant modified vaccinia virus Ankara (MVA) carrying genes encoding antigen 85A (Ag85A), early secretory antigenic target (ESAT-6) or IL-2 and to investigate the immunogenicity of these recombinant MVA in mice. Methods The genes encoding Ag85A and ESAT-6 were amplified by PCR from Mycobacterium tuberculosis H37Rv genomic DNA. The am-plified DNA fragments were sub-cloned into vector p18. The recombinant plasmids were introduced respec-tively into MVA by homologous recombination. The five types of recombinant MVA clone were selected in the MPA selection medium and their expressions of TB antigens were confirmed by Western blot. The recombi-nant MVA were used to immunize BALB/c mice. Ag85A-ESAT-6 fusion protein induced proliferation of spleno-lymphecytes from immunized mice was detected by MTT test. The concentration of IFN-γ in superna-tant of spleno-lymphocytes cultures was also analyzed by ELISA. The delayed type hypersensitivity (DTH) response to tuberculosis antigens was measured after injection of tuberculin purified protein derivative (PPD) in hind footpads of immunized mice. Results The five types of recombinant MVA were successfully con-structed by confirming their expression of TB antigen with Western blot. After co-cultured with the fusion protein Ag85A-ESAT-6 in vitro, the spleno-lymphocytes from recombinant MVA immunized mice showed sig-nificant proliferation activity (P<0.01) and increased IFN-γ secretion (P<0.05) while the same cells showed no response when co-cultured with saline. The spleno-lymphocytes from recombinant MVA immu-nized mice also demenstrated significant proliferation activity (P<0.01) and increased IFN-γ secretion (P<0.01) under Ag85A-ESAT-6 inducement, compared with the spleno-lymphocytes from mice immunized with wild type MVA or saline. The significant DTH response to PPD was detected in recombinant MVA-immunized mice (P<0.05). Conclusion The five types of recombinant MVA carrying genes of TB anti-gens were constructed. The constructed recombinant MVA could induce specific cellular immunity against tu-berculosis antigen in mice.
5.Nursing care for patients with malignant obstructive jaundice receiving biliary stenting combined with linear 125I seeds strand implantation

Objective To discuss the nursing points for patients with malignant obstructive jaundice who are receiving biliary stenting combined with linear 125I seeds strand implantation. Methods A total of 41 patients with malignant obstructive jaundice were enrolled in this study. Biliary stenting combined with linear 125I seeds strand implantation was performed in all the 41 patients. Sufficient preoperative preparation and psychological intervention were carefully implemented. During the operation, active cooperation with the surgical procedures and close observation of patient’s vital signs as well as adverse reactions such as nausea and vomiting were strictly carried out. After the treatment the management of PTCD drainage tube, the protection of radiation and the early detection of complications were further emphasized. Results The technical success rate of biliary stenting together with linear 125I seeds strand implantation was 100%. The serum total bilirubin levels, determined at one week, one and 3 months after the treatment, were significantly decreased when compared with the preoperative data. Clinically, the jaundice basically subsided. Bile-cardiac reflex occurred in one case and displacement of PTCD drainage tube in 2 cases. Twelve patients developed chills and fever after operation. Acute renal insufficiency occurred in one patient, which was relieved after effective management. The median survival time was 10.9 months. Conclusion Careful and comprehensive preoperative preparation can ensure a successful operation. Intraoperative close observation of patient’s condition and skilled cooperation with the operator, and high quality postoperative nursing measures and close observation for early signs of complications are essential nursing points to ensure that the patients can safely get over the rehabilitation period.
6.BIOAVAILABILITY OF Se IN Se ENRICHED MUSHROOM IN RATS

Bioavailability of selenium in Se enriched mushroom, Pleurstus ostreatus, was assayed with Se depleted rats. The selenium status in rats including blood, liver Se levels and erythrocyte glutathione peroxidase (GSH-Px) activity was evaluated. Young male SD rats were first fed with basal Se deficient diet for 4 weeks, followed by either continued depletion or repletion for another 3 weeks with 0.51, 0.75, 1.91, 3.18?g Se as sodium selenite, and 0.45, 0.72, 1.59, 3.41?g Se as Se enriched mushrooms per gram of diet for each group of rats respectively. Both Se levels in blood and in liver and erythrocyte GSH-Px activity were enhanced significantly with the increasing dietary Se contents. The relative bioavailability of selenium (taken selenite as 100%) in the Se enriched mushroom (P.ostreatus) was 66.66% based on the criteria of blood Se level, or 125.0% of liver Se level, or 123.42% of erythrocyte GSH-Px activity respectively. These results demonstrated that Se enriched mushroom (P.ostreatus) was an excellent dietary Se source.

To propose the hypothesis of the requirement for optimum ratio of various active cons tituents of Chinese materia medica with desired effects, which suggests that both single drug and compound drug prescriptions have their own target effects corresponding to their main indications and their active constituents must be in optimum ratio This hypothesis was initially tested in an ischemic animal model The neuroprotection of Yushen Oral Liquor (YSOL, an oral liquor made of Chinese materia medica) on neuronal injury in CA1 region of rat hippocampus following transient forebrain ischemia was used as an index to evaluate the drug's effect By combination of HPLC and pharmacological method, the relationship of HPLC and pharmacodynamical results of each sample made of YSOL was studied to observe the chromatogram peaks representing the drug's neuro protective effects There are three peaks named as R1, R2, and R3 for YSOL, which closely correlate to the drug's neuronal protective effects and the relative weight but not the absolute quantity of those peaks plays a critical role in producing neuronal protective effects of YSOL on ischemia induced neuronal damage The results provide experimental data supporting our proposed hypothesis

10.Crystallography of ATP hydrolysis mechanism in rat brain kinesin.

Rat brain kinesin is a conventional kinesin that uses the energy from ATP hydrolysis to walk along the microtubule progressively. Studying how the chemical energy in ATP is utilized for mechanical movement is important to understand this moving function. The monomeric motor domain, rK354, was crystallized. An ATP analog, AMPPNP, was soaked in the active site. Comparing the complex structure of rK354 x AMPPNP and that of rK354ADP, a hypothesis is proposed that Glu237 in the Switch II region sensors the presence of gamma-phosphate and transfers the signal to the microtubule binding region.
