1.Effect of rhodiola on lipidperoxides and ultrastructure of mitochondria in the brain of senile mice

BACKGROUND: Senility is the degenerative change of every tissue and organ in biological body,which is the result due to comprehensive actions of various pathological and physiological processes. Rhodiola is a kind of natural medical plant,acting on delaying organic senility,preventing and treating senile disorders,etc. But the definite mechanisms on those have been unknown yet.OBJECTIVE: To probe into the mechanism of rhodiola extract( nuodikang capsule) on anti-senility.DESIGN: Randomized,controlled experimental research based on the experimental animals.SETTING: Neurological internal department in a military hospital affiliated to a university of Chinese PLA.MATERIALS: The experiment was performed in Pathology Experiment Room of Fourth Military Medical University of Chinese PLA from March to August 2002. Sixty Kunming mice were randomized into normal control group,model group,3 dosage groups of 1,2,4 g/kg of nuodikang successively and Vitamin E(Vit. E) positive control group 0. 1 mg/kg,10 mice in each group.INTERVENTIONS: In the model,nuodikang groups and Vit. E positive control,physiological saline solution of D-galactose 30 g/Lwas applied in subcutaneous at the back of neck,150 mg/kg per day,continuously for 8weeks. Simultaneously,in nuodikang group and Vit. E positive control,the medicine was applied by gastric perfusion,once daily,continuously for 8weeks.MAIN OUTCOME MEASURES: Effects of nuodikang capsule on activity of lactate dehydrogenase(LDH),the content of lipidperoxides(LPO),the activity of superoxide dismutase(SOD) in the brain,the content of malondialdehyde (MDA) and ultrastructure of mitochondrion in hippocampal neurons.RESULTS: In nuodikang capsule groups of 1,2,4 g/kg,the significant differences(P < 0.05) presented in LPO content in brain tissue[(30. 1±2.9),(27.8±3.1),(26.9±1.9) nmol/g] and MDA content [(110.2±13.5),(95.4±20.1),(90.2±16.5) nmol/g],1 compared with the model group[ (33.4 ± 2.2),(126.5 ± 17.2) nmol/g],indicating significant differences(P < 0.05),and LDH activity[(1.74±0. 14),(1.92 ±0. 23),(2.04 ±0.21) NU/g] and SOD activity[ (119. 1 ±2.2),(121.3 ± 0.9),(126.4 ± 2.0) NU/g] were remarkably increased,compared with the model group[ (1.68 ±0. 19),(115.9 ±2. 1) NU/g],indicating significant differences( P < 0.05) . It was indicated by electro-microscope that nuodikang capsule groups of 2,4 g/kg acted on obvious protection on degenerative change of mitochondrion in hippocampal neurons caused by galactose.CONCLUSION: Nuodikang capsule acts significantly on anti-senility,which is related to its anti-oxidative injury and protection on neurons.
4.Diagnostic value of proton magnetic resonance spectroscopy in multiple sclerosis

Objective To evaluate the diagnostic value of proton magnetic resonance spectroscopy (1H MRS) in multiple sclerosis (MS).Methods MRI and 1H MRS were carried out in 29 patients with MS (MS group) and 26 matched normal control subjects(NC group). By calculating the area below peak, the N-acetylasparatate(NAA), creatine (Cr), choline (Cho) in brain were quantified. The NAA/Cr and Cho/Cr ratios were compared between the two groups. Every patient was measured by EDSS. The correlation analysis was done between NAA/Cr, Cho/Cr ratios and EDSS scores.Results The ratios of NAA/Cr, Cho/Cr were 1.38?0.43, 1.99?0.84 in MS group, 1.89?0.49, 1.48?0.36 in NC group. NAA/Cr decreased in MS group. The difference between two groups was statistically significant ( P0.05). Conclusions The metabolic ratios measured by 1H MRS in MS patients are changed significantly. The ratio of NAA/Cr can reflect the severity of the clinic neurological disability in MS patients.
5.Construction of the eukaryotic expression plasmids of Mycobacterium tuberculosis ESAT-6 antigen and its immunogenicity

Objective To construct the eukaryotic expression plasmids of Mycobacterium tuberculosis ESAT-6 antigen and to investigate its immunogenicity in mice. Methods The sequence encoding ESAT-6 was amplified by PCR from Mycobacterium tuberculosis H37Rv genomic DNA. Then, the amplified fragment was sub-cloned into pVAC expression vector. The eukaryotic expression plasmid pVAC-ESAT-6 was transfected into Vero E6 cells with liposome. The mRNA expression of pVAC-esat-6 in Vero E6 cells was detected by RT-PCR and its expression product was analyzed by Western-blot. The plasmid pVAC-esat-6 was introduced into the mice by gene gun injection. The specific antibody to ESAT-6 and the level of IFN-? in supernatant of spleno-lymphocyte cultures were detected by ELISA methods. Results The recombinant plasmid pVAC-esat-6 was successfully constructed, and the expression of ESAT-6 was also detected in vitro. 6.The Value of Vertical to Clivus CT Scanning in Diagnosing Tentorial Hernia in the Patient with Brain Trauma

Objective To explore the value of vertical to clivus CT scanning in diagnosing tentorial hernia and to analyze the CT findings of tentorial hernia in the patient with brain trauma. Methods Vertical to clivus CT scaning were performed in 156 cases with brain trauma.Results Vertical to clivus scanning method directly demonstrated the temporal transtentorial hernia in 72 of 156 patients with unilateral high intracranial pressure, but the orbitomeatal line scanning method was not able to show this direct finding. The CT findings of hernia were the tissue of temporal lobe appeared in the cisterna ambiens inside the tentorium accompanied by enlarged cistern on the involved side. No change of cistern were seen in 84 cases.Conclusion The scanning method with vertical to the clivus is effective in detecting tentorial hernia in the patient with cerebral trauma, it should be performed in the patient with cerebral trauma and increased intracranial pressure.

Rheumatoid arthritis (RA) is a kind of chronic, progressive, multiple, invasive autoimmune disease with two chief cclinical manifestations arthrosynovitis and ex-arthrosis, easy to occur in middle-aged women, also occur in children and the elderly, is characterized by progressive and break out repeatedly. RA pathogenesis is complex, there is no special treatment, used in treatment of R drug varied, new drugs and new therapies also emerge in endlessly, main including non-steroidal anti-inflammatory drugs (NSAIDs), slow action anti-rheumatism medicine (SAARDs), glucocorticoids (GCs), biological agent, traditional Chinese medicine and traditional Chinese medicine preparations, domestic market for rheumatoid main drug treatment are NSAIDs, SAARDs, GCs, traditional Chinese medicine and traditional Chinese medicine preparations. Traditional Chinese medicine and traditional Chinese medi- cine preparations for the treatment of RA have its unique advantages, show the characteristics of overall adjustment, multi-level and multiple targets, and also can alleviate and against side effects of western medicine. 10.Study of Experimental Parkinsonian Animal by Using High Performance Liquid Chromatography with Polymer-Overoxidized Polypyrrole Film Chemically Modified Electrode

A novel polymer- overoxidized polypyrrole film chemically modified electrode (CME) as the liquid chromatography-electrochemical detector (LC-ECD) was fabricated and applicated, which could be used to determine the monoamine neurotransmitters. The experimental model of parkinsonian animal was established by using medicine. Microdialysis sampling coupled with LC-ECD was used to monitor the monoamine neurotransmitters and their metabolites in experimental animal brain in vivo under different situations. The mechanism of experimental animal's Parkinsonian disease was studied elementarily. An accurate and reliable analysis method was supplied for filting the new and the more effective medicine to cure parkinsonian disease.
