1.Effect of propofol on hepatic cell apoptosis induced by ischemia-reperfusion in rats

Objective To investigate the sinusoidal endothelia) cell (SEC) apoptosis induced by hepatic ischemia-reperfusion (I/R) and the effect of propofol on the hepatic cell apoptosis in vivo. Methods For hepatic I/R study we utilized a rat model of 70% liver ischemia according to Kohli, et al . Twenty-four male SD rats weighing 250-350 g were randomly assigned to 3 equal groups of eight animals : group A was subjected to 30 min of liver ischemia followed by 6 h of reperfusion and normal saline was infused iv at the onset of reperfusion, at a rate 10 ml ? kg-1 ? h -1 for 60 min; group B was subjected to the same J/ R as in group A but instead of NS propofol 20 mg " kg was given iv followed by continuous infusion of 0.5% propofol at 50 mg? kg-1 ? h-1 for 60 min; group C underwent sham operation followed by intravenous NS infusion at 10 ml kg h for 60 min. Blood samples and liver biopsies were obtained at the end of 6h of reperfusion, for determination of plasma alanine aminotransferase (ALT) activity and hepatic malondialdehyde (MDA) content, apoptosis and microscopic examination (light and electron microscopy) . Apoptosis was determined both qualitatively and quantitatively by DNA laddering and TUNEL methods. 2.Study on Function of JAK2/STAT3 Pathway in Promoting Angiogenesis with Angelica Microcapsules for Replenishing Blood

OBJECTIVE To investigate the function of JAK2/STAT3 pathway in promoting angiogenesis with Angelica mi-crocapsules for replenishing blood (Ang-Mc).METHODS Establish the human umbilical vein endothelial cell (HUVEC) model,and then cell counting kit(CCK-8) and flow cytometry were used to test cell viability and cell apoptosis.Western blot was used to detect expression of p-STAT3 and VEGF.RT-PCR was used to detect expression of vascular endothelial growth factor(VEGF) and miRNA-21.RESULTS When Ang-Mc with a concentration of 20 μg/mL acted on HUVEC for 24 h,it could effectively promote the HUVEC proliferation by up-regulating the expression of p-STAT3,VEGF and miRNA-21. AG490 can effectively inhibit Ang-Mc promoting HUVEC proliferation effect and reduce the expression of p-STAT3,VEGF and miRNA-21.CONCLUSION The effect of Ang-Mc in promoting angiogenesis is related to JAK2/STAT3 signaling path-ways.
3.Rapidly Determination of Major and Minor Elements in Different Processed Products of Pyrite by X-Ray Fluorescence Spectrometry

OBJECTIVE A comprehensive understanding of the major and minor elements in different processed products of Pyrite,providing scientific basis for the clinical rational use of Pyrite.METHODS The determination method of 10 compo-nents including Fe,S,Si,Al and other major and minor components in different processed products of Pyrite by X-ray fluores-cence spectrometry was established.RESULTS The detection limit of Fe,S,Mn,Si,Al,Ti,Ca,P,Zn and Pb was 13.37, 36.92,7.56,68,29,77.47,12.46,22.54,21.64,4.55,3.24 mg/kg.Determination of standard material composition of GBW(E)070088 (iron ore)and GBW 07267 (pyrite)for 5 repetitions.The accuracy were 94%~107% and the RSD were 1. 0%~10.1%.CONCLUSION The method can be used to determine the major and minor elements in different processed prod-ucts of Pyrite,and it is accurate and convenient.
