Malachite green (MG) is a dye which has been widely used as a topical fungicide and antiseptic in aquaculture throughout the world. Now,MG has become a highly controversial compound due to its potential highly residual and toxic characteristics in recent years. In the paper, the advances in toxicity, toxicological mechanism, detection methods, metabolism and removal of malachite green and its metabolite were reviewed, and the problems in aquaculture and the safety of aquaculture technology were discussed,in order to make us be aware of potential hazard of malachite green and provide a dependable evidences of safely breeding in aquatic products.

Video-assisted thoracic surgery (VATS) is high-skilled operation, which is tacit,scene-related,hard to transfer and individualized. The action formation of VATS is implemented by action orientation, imitation, integration, training and automation. The skill acquisition of VATS involves four overlapping steps:studying examples to do analogy, developing abstract rules, slowly moving to the use of production rules and retrieving specific examples.Research on teaching methods of VATS in the view of tacit knowledge helps to learn this skill better and more quickly.

Objective To construct five types of recombinant modified vaccinia virus Ankara (MVA) carrying genes encoding antigen 85A (Ag85A), early secretory antigenic target (ESAT-6) or IL-2 and to investigate the immunogenicity of these recombinant MVA in mice. Methods The genes encoding Ag85A and ESAT-6 were amplified by PCR from Mycobacterium tuberculosis H37Rv genomic DNA. The am-plified DNA fragments were sub-cloned into vector p18. The recombinant plasmids were introduced respec-tively into MVA by homologous recombination. The five types of recombinant MVA clone were selected in the MPA selection medium and their expressions of TB antigens were confirmed by Western blot. The recombi-nant MVA were used to immunize BALB/c mice. Ag85A-ESAT-6 fusion protein induced proliferation of spleno-lymphecytes from immunized mice was detected by MTT test. The concentration of IFN-γ in superna-tant of spleno-lymphocytes cultures was also analyzed by ELISA. The delayed type hypersensitivity (DTH) response to tuberculosis antigens was measured after injection of tuberculin purified protein derivative (PPD) in hind footpads of immunized mice. Results The five types of recombinant MVA were successfully con-structed by confirming their expression of TB antigen with Western blot. After co-cultured with the fusion protein Ag85A-ESAT-6 in vitro, the spleno-lymphocytes from recombinant MVA immunized mice showed sig-nificant proliferation activity (P<0.01) and increased IFN-γ secretion (P<0.05) while the same cells showed no response when co-cultured with saline. The spleno-lymphocytes from recombinant MVA immu-nized mice also demenstrated significant proliferation activity (P<0.01) and increased IFN-γ secretion (P<0.01) under Ag85A-ESAT-6 inducement, compared with the spleno-lymphocytes from mice immunized with wild type MVA or saline. The significant DTH response to PPD was detected in recombinant MVA-immunized mice (P<0.05). Conclusion The five types of recombinant MVA carrying genes of TB anti-gens were constructed. The constructed recombinant MVA could induce specific cellular immunity against tu-berculosis antigen in mice.

0.05).Conclusion The hypoxic exposure can accelerate the production of erythrocyte, and treat the exercise-induced anemia effectively. Improvement of some haematopoietic factors and enhancement of hematopoiesis in marrow is thought to be the possible mechanism.

Objective To determine the feasibility of achieving local transgenic expression in the rat lung using bone marrow mesenchymal stem cells ( MSCs) transfected with Lac-Z gene. Methods Primary cultures of bone marrow MSCs from Lewis rats were transfected with the pSV-?-galactosidase control vector and labelled with a fluorescent, membrane impermeable dye DAPI. The transfected and labelled MSCs (5?105 cells/animal) were injected into the jugular vein of syngenetic recipient rats. The animals were killed at 48 h and 8 wk after injection respectively. The lungs, spleens, livers, kidneys and skeletal muscle were then excised and examined under fluorescene microscope. The transgenic expression of Lac-Z gene was detected by incubating with the X-gal chromogen. Results Only a few DAPI labelled MSCs could be identified in the spleen, liver, kidney and skeletal muscle, whereas a large amount of DAPI labelled MSCs could be identified in the lung and most of them lodged in the lung parenchyma and air sac at 48h and 8wk after intravenous injection of transfected MSCs. After incubation with the X-gal chromogen, microscopic examination showed that a large number of MSCs with multiple intense blue staining were scattered throughout the lung. On the contrary only a few cells with blue staining could be identified in the spleen and kidney and no MSCs with blue staining could be seen in the liver pancreas and skeletal muscle. Conclusion Genetically modified MSCs injected into the jugular vein can target the lung effectively and achieve local transgenic expression for a long time.
