2.Expression and distribution of tight junction proteins in colorectal cancer

Objective To investigate the expression and distribution of tight junction proteins in colorectal carcinoma.Methods Real time reverse transcription PCR, Western blot and immunohistochemistry were used to check the expression of claudins(CL-1 to-4),occludin(OC),zona occludens proteins(ZO-1 and ZO-2)in 35 colorectal carcinomas and matched adjacent normal tissues.Results The expression of claudin-1 and claudin-2 in cancer tissues was 0.97 and 0.34 respectively.Either of them was higher than that in normal tissues.Compared with adjacent normal tissues,it increased by 26.3-and 30.4-fold respectively at the mRNA level.Immunohistochemistry analysis showed different distribution patterns for occludin,ZO2,claudin 1,claudin 2 and claudin 4 between the cancer and normal tissues.Normal colonic epithelium exhibited n cytoplasmic staining for claudin-1,whereas the strong plasma membrane staining was presented in carcinoma cells. The claudin-2 proteins were detected mainly under the epithehal cells and presented spot granular staining in normal epithelium,whereas much more spot granular staining existed in the whole epithelial cells of carcinomas.Conclusions There was close association between abnormal expression and distilbution of tight junction proteins and colorectal cancer.And these proteins may be good candidates for the diagnoses and therapy for colorectal cancer.
3.The ocular pharmacokinetics of a new antibody (MIL60) against vascular endothelial growth factor ( VEGF) following subconjunctival injection in rats

Objective To evaluate the ocular pharmacokinetics of a novel antibody, MIL60, tar-geting vascular endothelial growth factor ( VEGF) after subconjunctival injection. Methods After subcon-junctival injection of MIL60 in rabbits, aqueous humour, vitreous from both eyes and peripheral blood were collected of each rabbit to analyze the concentration of MIL60 with ELISA. Results After subconjunctival administration, MIL60 could penetrate into the injected eye′s anterior chamber and vitreous, and maintained at an effective concentration in the aqueous or vitreous for about 10. 7 or 6. 8 days. Moreover, MIL60 could be found in the uninjected eye. Conclusion After subconjunctival injection, MIL60 could maintain at a therapeutic concentration in injected eyes. The pharmacokinetics analysis of MIL60 might provide some basis and guidance for its application in humans in the near future.
4.Treatment of residual neuralgia of herpes zoster by ear point taping and pressing therapy combined with acupoint-injection.

OBJECTIVETo Assess therapeutic effect of ear point taping and pressing therapy combined with acupoint-injection on residual neuralgia of herpes zoster.
METHODSOne hundred and sixteen cases were randomly divided into a comprehensive group (n = 60) and a medication group (n = 56). The medication group were treated with routine western medicine, and the comprehensive group with ear point taping and pressing therapy combined with acupoint-injection besides the routine western medicine. Auricular points selected for ear point taping and pressing were Shemen, Neifenmi (endorine), Pizhixia (subcortex), Gan (liver), Dan (gallbladder), Fei (lung) and corresponding auricular points to the lesion parts, with the two ears alternatively used, pressing each day; points selected for point-injection of VitB12 were Zusanli (ST 36), Neiguan (PC 6), Quchi (LI 11), Taichong (LR 3). The pain degrees, the time of pain alleviation and pain ceasing of the patient were regularly recorded.
RESULTSThe average time of pain alleviation and pain ceasing of the patient in the comprehensive group were significantly shorter than those in the medication group (P < 0.01). The cured rate and the cured and markedly effective rate were 60.0% and 83.3% in the comprehensive group, and 28.6% and 50.0% in the medication group, with significant difference between the two groups (P < 0.05).
CONCLUSIONEar point taping and pressing therapy combined with acupoint-injection is effective and safe for treatment of residual neuralgia of herpes zoster.

5.Microvascular study of the transmidline scapular flap vascularized by the contralateral circumflex scapular artery.

OBJECTIVETo observe the microvascular distribution of the transmidline scapular flap supplied by the contralateral circumflex scapular artery.
METHODSThe integument and deep tissues of 6 fresh cadavers were dissected and radiographed after vermilion mixture was injected to the unilateral circumflex scapular artery.
RESULTSThe vascular tree passed the midline and reached to the contralateral acromion. The vessel density was the highest in the irrigating side of the back, which was higher in the middle area. In the contralateral side,the high vessel density concentrated in the upper part of the back.
CONCLUSIONSThe result revealed the direct evidence for the clinical application of the transmidline scapular flap. In design and elevating of the transmidline scapular flap, it should be slanting to the upper part of the contralateral back.

6.Hemodynamic study of an over-midline axial flap.

OBJECTIVETo investigate the hemodynamic changes of an over-midline axial flap.
METHODSAn over-midline axial flap, based on the iliac artery and veins, was raised on the back of each selected Wistar rat (n = 30), vascularied by contralateral. The flap size is 7 cm x 3 cm. The over-midline part is 3 cm x 3 cm. A laser doppler flowmetry (LDF) was used to evaluate the hemodynamic changes preoperatively, and 6 hours, 24 hours, 48 hours, 72 hours, 7 days, and 14 days after the operation respectively.
RESULTSAll of the flaps survived well. But, in the first three days after the operation, the blood perfusion of the flap in the areas of the midline and the distal part was significantly lower than the preoperation's level. Thereafter, the values of the blood perfusion started to increase till to the maximum 7 days after the operation. On the 14 days after the operation, the vascular structures was axially matched to the base of the flap. In the distal part of the flap, the choke vessels were gradually opened and resulted in the blood perfusion to the normal level.
CONCLUSIONThe hemodynamic changes of the over-midline axial flap transmidline flap could occur to match the need of the base of the flap.

7.Expansion of CD4~+ CD25~+ FoxP3~+ regulatory T cell pool in patients with active tuberculosis

Objective To investigate the frequency of CD4~+CD25~+FoxP3~+regulatory T cells (Treg)and the expression of the functional protein,FoxP3,in patients with active tuberculosis and the relationship between Treg and the pathogenesis of tuberculosis.Methods Forty-five patients with active tuberculosis(including 25 cases of pulmonary tuberculosis and 20 tuberculous lymphadenitis), 20 healthy controls,20 recovered tuberculosis patients and 6 patients with reactive hyperplasia in cer- vical lymph node were enrolled.The frequency of CD4~+ CD25~+ FoxP3~+ Treg in the peripheral blood was measured by flow cytometry.FoxP3 mRNA expression was determined by real-time reverse transcriptase-polymerase chain reaction(RT-PCR)and the expression of FoxP3 protein in lymphoid tissues was measured by immunohistochemistry.Results The frequency of natural Treg in the peripheral blood from the patients with active tuberculosis was 2.91%?0.23%,which was signifi- cantly higher than that of healthy control group(1.22%?0.18%)and recovered tuberculosis patients(1.50%?0.17%,P
8.Biosynthesis of a new psoralidin glucoside by enzymatic glycosylation.

OBJECTIVETo modify the structure of psoralidin using in vitro enzymatic glycosylation to improve its water solubility and stability.
METHODSA new psoralidin glucoside (1) was obtained by enzymatic glycosylation using a UDP- glycosyltransferase. The chemical structure of compound 1 was elucidated by HR-ESI-MS and nuclear magnetic resonance (NMR) analysis. The high-performance liquid chromatography (HPLC) peaks were integrated and sample solution concentrations were calculated. MTT assay was used to detect the cytotoxicity of the compounds against 3 cancer cell lines in vitro. Results Based on the spectroscopic data, the new psoralidin glucoside was identified as psoralidin-6',7-di-O-β-D- glucopyranoside (1), whose water solubility was 32.6-fold higher than that of the substrate. Analyses of pH and temperature stability demonstrated that compound 1 was more stable than psoralidin at pH 8.8 and at high temperatures. Only psoralidin exhibited a moderate cytotoxicity against 3 human cancer cell lines. Conclusion In vitro enzymatic glycosylation is a powerful approach for structural modification and improving water solubility and stability of compounds.

9.Clinical application of alpha1 adrenoceptor antagonist Naftopidil to the treatment of chronic non-bacterial prostatitis.

OBJECTIVETo study the efficacy and safety of alpha1 adrenoceptor antagonist Naftopidil in the treatment of chronic non-bacterial prostatitis.
METHODSAn opened, self-controlled, multicentral clinical trial was conducted. One hundred and six cases of patients who had been diagnosed as chronic non-bacterial prostatitis (NBP) were treated with Naftopidil (25 mg once a day) for 4 weeks. The efficacy was evaluated by the NIH Chronic Prostatitis Symptom Index (NIH-CPSI) and the WBC in the examination of prostatic secretion (EPS) after the treatment.
RESULTSAfter 4 weeks therapy, 105 cases were evaluable. After treatment, NIH-CPSI total score were averagely decreased 12.0 points (P <0.001), symptom score 7.9 points (P <0.001) and QOL score 4.1 points (P <0.001). There was a statistically significant difference in WBC count ([(15.2 +/- 15.1)/HP vs (9.5 +/- 12.0)/HP, P < 0.01] and max flow rate(MFR) [(19.2 +/- 4.8) ml/s vs (22.7 +/- 4.9) ml/s, P < 0.01]. The total effective rate were 84.8% in the whole group. The clinical adverse rate was 3.81%, including 3 cases of mild dizziness and 1 case of mild inappetence.
CONCLUSIONalpha1 adrenoceptor antagonist Naftopidil is effective and safe for the treatment of chronic non-bacterial prostatitis.

10.Polymorphism K469E of intercellular adhesion molecule-1 gene and restenosis after coronary stenting in Chinese patients.

BACKGROUNDInflammation is a major cause of restenosis after coronary stenting. Intercellular adhesion molecule-1 (ICAM-1) is an important adhesion molecule that plays a key role in the tight adhesion between leukocytes and vascular endothelium. The object of this study was to investigate the association between the K469E polymorphism of the ICAM-1 gene and restenosis after coronary stenting in North Chinese population.
METHODSThe ICAM-1 K469E polymorphism was genotyped using polymerase chain reaction-restriction fragment length polymorphism method in 124 patients who had undergone coronary stenting and coronary angiography at least 3 months earlier. Information on clinical risk factors and procedure-related data were also collected.
RESULTSOf 124 enrolled patients in total, there were 72 cases of in-stent restenosis. The restenosis rate in this population was 58.1%. The frequencies of the three possible genotypes of the ICAM-1 K469E polymorphism were: KK genotype 50.8%, EE genotype 41.9%, and EK genotype 41.9%. Among restenosis patients, the frequency of the KK genotype was 58.3% and the frequency of E allele carriers was 41.7%. Among non-restenosis patients, the frequency of the KK genotype was 40.4%, and the frequency of E allele carriers was 59.6%. The distribution of these two genotype groups between restenosis and non-restenosis patients was significantly different (P = 0.049). Using multivariate logistic regression, the difference between the two groups was more apparent. The odds ratio of KK homozygotes vs E allele carriers was 2.6, with 95% confidence interval 1.2 - 5.8 (P = 0.018). After grading of risk factors, we found that the KK genotype was a stronger predictor of in-stent restenosis in obesity or hyperlipemia patients, with an odds ratio of 9.3 and 3.7, respectively (P < 0.05).
CONCLUSIONIn our study population, KK homozygotes of the ICAM-1 codon 469 mutation had a higher risk of restenosis after coronary stenting, especially in the case of obese or hyperlipemia patients.
