1.Neutrophil extracellular traps induce production of MPO-ANCA in chronic bronchitis rats

Objective:To investigate the mechanism of chronic bronchitis with recurrent long duration infections in the production of MPO anti-neutrophil cytoplasmic antibodies(ANCA). Methods:Rats were gave intratracheal injection of LPS and smoked to established chronic bronchitis ( CB ) model. Rat neutrophils were incubated with phorbol-12-myristate-13-acetate ( PMA ) to induce Neutrophil extracellular traps(NETs). NETs were then used to immune chronic bronchitis rats. Serum concentrations of myeloperoxidase anti-neutrophil cytoplasmic antibodies(MPO-ANCA),Citrullinated Histone H3(CitH3),Interferon-α(IFN-α)were evaluated. Results:(1)The serum levels of MPO-ANCA in chronic bronchitis rats immuned with NETs(CB/NETs)were significantly higher than those in chronic bronchitis rats(CB)and healthy control rats,(62. 4975±19. 63764)ng/ml vs (17. 3806±4. 88649)ng/ml,(20. 8884± 4. 39607)ng/ml(both P<0. 01). (2)The serum levels of CitH3 in CB/NETs rat were significantly higher than those in CB rat and healthy control rats,(32. 5974±20. 2915)ng/ml vs (8. 8568±2. 36692)ng/ml,(5. 6225±0. 83738)ng/ml(P<0. 05,P<0. 01). (3)The serum levels of IFN-α were found increased sharply in two CB/NETs rats(3735.7,3428.4 pg/ml). (4)Renal function among three groups showed no significant difference yet. ( 5 ) Serum CitH3 level showed a positive correlation with serum MPO-ANCA level(r=0. 61,P<0. 05). Conclusion:Chronic bronchitis with recurrent infections could induce the production of ANCA probably by the stimulation of NETs,but whether it might develop to AAV remains need a duration.

Objective To explore the effect of Melatonin(MT) on CD4+CD25+ regulatory T cell (CD4+CD25+Tr)and airway inflammation in asthmatic rat.Methods Thirty-two SD rats were randomly divided into 4 groups,8 rats in each group.Asthmatic group:rats were immunized on day 1 and 7 by intraperitoneal inject of mixture of ovalbumin(OVA) and aluminumhydroxide.From day 14,the animals were allenged with aerosolized OVA for 20 min per day for 7 consecutive days.MT group:OVA-sensitized rats were injected intraperitoneally with 0.1 mg/kg MT 30 min before each OVA challenge.Dexamethasone group:OVA-sensitized rats were injected intraperitoneally with 0.5 mg/kg Dexamethasone 30 min before each OVA challenge.Control group:OVA for inhalation and MT for intraperitoneal injection was replaced with saline.After the last challenge,peripheral blood was stained to count the percentage of eosinophil(EOS).Then the rats were lavaged and total leukocytes counts in bronchoalveolar lavage fluid(BALF) were performed after staining with Wright-Giemsa staining.The EOS counts around the airway was counted after the histological section of lung staining with hematoxylin and eosin staining.The serum level of immunoglobulin E(IgE) was detected by immunoenhancement.The change of CD4+CD25+Tr was assessed with flow cytometry.SPSS 10.0 software was applied to analyze data. OBJECTIVETo evaluate the effect of a novel blockade technique for gastric cancer on blood-borne metastasis of gastric cancer cells to portal vein.
METHODSTwenty-three cases of gastric cancer were divided into routine operation group (8 cases intraoperatively without blockade technique) and blockade group (15 cases with blockade technique). Blood samples from portal vein pre- and intraoperatively, as well as gastroepiploic vein limited within the blockade area were obtained to detect CK19 mRNA expression by using RT-PCR technique.
RESULTSBefore the dissection of gastric lesion, the overall positive rate of CK19 mRNA expression in portal vein blood is 34.7% (9/23), including 37.5% (3/8) in routine operation group and 33.3% (5/15) in blockade group. While the course of tumor resection, those positive rates were 87.5% (7/8) in routine operation group and 6.7% (1/15) in blockade group respectively (P < 0.05). CK19 mRNA expression in the right gastroepiploic venous blood limited within the blocking area was all positive in 15 cases of blockade group.
CONCLUSIONThis blockade technique can be used effectively to block the intraoperative spread of gastric cancer cells, thus prevent blood-borne metastasis due to operative manipulation.

4.Influence of the number of removed axillary lymph nodes on the prognosis of node-negative primary breast cancer.

OBJECTIVETo analyze the relationship between the number of removed axillary lymph nodes and prognosis of axillary node-negative breast cancer.
METHODSThe clinicopathological data of 655 patients with breast cancer were analyzed retrospectively. The disease-free survival curves were generated according to the number of removed axillary lymph nodes using Kaplan-Meier plots. The correlation between the co-variables and rate of breast cancer-related events was analyzed using Cox model.
RESULTSThe overall five year-disease free survival rate of the 655 cases was 94.4%. The rate of patients with lymph node number ≤ 12 was 90.3%, and that of lymph node number > 12 was 96.5%, with a statistically significant difference (P = 0.009). Significantly less breast cancer-related events were observed in patients with lymph node number > 12 (15/426, 3.5%) than that in patients with lymph node number ≤ 12 (22/229, 9.6%) (P = 0.009).
CONCLUSIONSWhen axillary node dissection is indicated, dissection of lymph nodes >12 leads to much less breast cancer-related events than that in patients with dissected lymph node ≤ 12. The more lymph nodes are dissected, the more accurate prognosis can be estimated.

5.Correlation between elevated plus and zero mazes as two animal models of state anxiety in Kunming mice

Aim To explore the correlation between elevated plus maze (EPM) and elevated zero maze (EZM),as two type animal models of state anxiety in Kunming mice. Methods The behavior of adult,♂/♀,Kunming mice in EPM and EZM was recorded by sequence,for 5 min,with a one-week inter-trial interval. The following parameters were evaluated:percentage of time in open arms (Otime%) and total entries into open and closed arms(Entries). Subsequently,descriptive analysis,fac-tor analysis,cluster analysis,correlation analysis and consisten-cy test were calculated for these parameters. Results T test showed that, compared with EPM, Otime% of EZM (male/fe-male/both sex) decreased, while Entries (male/both sex) in-creased significantly;Fiedman test showed that Otime% (male/female/both sex) and Entries(female/both sex) of both EPM and EZM had great difference between the 5 repeated fragments;Wilcoxon test showed that, compared with EPM, Otime% of EZM in 1st min (male/female/both sex), 2nd min (male/fe-male/both sex),3rd min (female/both sex) decreased, while the Entries in 1st min (male/both sex), 4th min (male/fe-male/both sex),5th min(both sex) increased markedly. Clus-ter analysis showed that EPM and EZM parameters could be clustered into two groups:EPM group and EZM group(male/fe-male/both sex). Factor analysis indicated that the EPM and EZM parameters could be extracted as two factors: EPM factor and EZM factor (male/female/both sex). Correlation analysis displayed either general(in Otime%) or poor(in Entries) cor-relation(male/female/both sex) between EPM and EZM param-eters. Consistency test displayed general consistency (male/fe-male/both sex) in Otime% between EPM and EZM. Conclu-sions Despite of the similar internal principles to induce state anxiety,the different external environment(structure) results in different behavioral patterns,different structure dimensions,fair correlation and concordance between EPM and EZM, and the stable index of EPM and EZM might be preferred to Otime%.
7.Relationship Between Body Fat Indices and Hypertension Level by Quantitative CT: A Multicenter Study in Chinese Population

Objectives: To explore the relationship between body fat indexes and hypertension levels by quantitative CT (QCT) study. Methods: A total of 1488 participants from "prospective urban-rural epidemiology (PURE) study" were enrolled. The participants' age were from 42 to 82 years; based on blood pressure (BP) they were divided into 2 groups: Non-hypertension group, n=783 and Hypertension group, n=705. QCT fat measurement software was used to examine L2 level abdominal visceral adipose tissue (VAT) and subcutaneous adipose tissue (SAT); body mass index (BMI), waist circumferences (WC), hip circumferences (HC) and waist-to-height ratio (WHtR) were measured and calculated; liner Logistic regression analysis and 2-classified Logistic analysis were conducted to study the relationships between hypertension and anthropometric indices, abdominal VAT, SAT respectively. Results: Compared with Non-hypertension group, Hypertension group had increased BMI, WC, HC, WHtR, abdominal VAT and SAT, P<0.05. BMI, WC, HC, WHtR, abdominal VAT and SAT were related to both systolic BP (SBP) and diastolic BP (DBP), P<0.05. Multivariable Logistic regression analysis indicated that WHtR, abdominal VAT, SAT were the independent impact factors of SBP and WC, abdominal VAT were the independent impact factors of DBP; further investigation showed that abdominal VAT and WC were the independent impact factors of hypertension. Conclusions: Abdominal VAT was robustly related to hypertension in Chinese population, it might be helpful for risk stratification in hypertension patients.
8.Application value of CT examination of lymph node short diameter in evaluating cardia-left gastric lymph node metastasis in thoracic esophageal squamous cell carcinoma.

Objective: To investigate the application value of computed tomography (CT) examination of lymph node short diameter in evaluating cardia-left gastric lymph node metastasis in thoracic esophageal squamous cell carcinoma (ESCC). Methods: A total of 477 patients with primary thoracic ESCC who underwent surgical treatment in the Affiliated Cancer Hospital of Zhengzhou University from January 2013 to December 2017 were collected. All of them underwent McKeown esophagectomy plus complete two-field or three-field lymph node dissection. Picture archiving and communication system were used to measure the largest cardia-left gastric lymph node short diameter in preoperative CT images. The postoperative pathological diagnosis results of cardia-left gastric lymph node were used as the gold standard. Receiver operating characteristic (ROC) curve was used to evaluate the efficacy of CT lymph node short diameter in detecting the metastasis of cardia-left gastric lymph node in thoracic ESCC, and determine the optimal cut-off value. Results: The median short diameter of the largest cardia-left gastric lymph node was 4.1 mm in 477 patients, and the largest cardia-left gastric lymph node short diameter was less than 3 mm in 155 cases (32.5%). Sixty-eight patients had cardia-left gastric lymph node metastases, of which 38 had paracardial node metastases and 41 had left gastric node metastases. The lymph node ratios of paracardial node and left gastric node were 4.0% (60/1 511) and 3.3% (62/1 887), respectively. ROC curve analysis showed that the area under the curve of CT lymph node short diameter for evaluating cardia-left gastric lymph node metastasis was 0.941 (95% CI: 0.904-0.977; P<0.05). The optimal cut-off value of CT examination of the cardia-left gastric lymph node short diameter was 6 mm, and the corresponding sensitivity, specificity and accuracy were 85.3%, 91.7%, and 90.8%, respectively. Conclusion: CT examination of lymph node short diameter can be a good evaluation of cardia-left gastric lymph node metastasis in thoracic ESCC, and the optimal cut-off value is 6 mm.

10.The Clinical Features of Metabolic Syndrome in Receptors of Hematopoietic Stem Cells.

OBJECTIVE:
To evaluate the incidence and clinical characteristics of metabolic syndrome (MS) within one year after hematopoietic stem cell transplantation (HSCT) in order to screen the risk factors for HSCT-MS, provide early intervention and improve the long-term quality of survival of patients.
METHODS:
The clinical follow-up data of 64 HSCT patients (survival time > 1 year) who received HSCT in our center from January 2007 to August 2018 were collected. Among them, 50 cases were allogeneic hematopoietic stem cell transplantation (allo-HSCT) and 14 cases were autologous hematopoietic stem cell transplantation (auto-HSCT). The changes of MS-related indexes and clinical characteristics before and 1, 3, 6 and 12 months after HSCT were analyzed retrospectively.
RESULTS:
In allo-HSCT group, 14 cases were diagnosed as MS before operation, including high-density lipoprotein cholesterol (hypo-HDL-C)> hyper triglycerides(hyper-TG)> hyper fasting glucose(hyper-FBG)> abdominal obesity (AO) > hypertension. The preoperative diagnosis of MS in the auto-HSCT group was 5 cases, in the order of hyper-FBG> hyper-TG> AO> hypo-HDL-C> hypertension. Incidence of MS at 1, 3, 6 and 12 months after transplantation: 19, 26, 24 and 20 cases in the allo-HSCT group, respectively; auto-HSCT group were 7, 7, 6 and 6 cases, respectively. Hyper-TG and hypo-HDL-C were prominent in both groups.
CONCLUSION
The incidence of HSCT-MS is significantly higher within 1 year after HSCT. Regardless of allo-HSCT and auto-HSCT, the prevention and control of HSCT-MS is emphasized as an important guarantee to improve the long-term survival quality of HSCT patients.
