Objective To study the effect of shenfu injection on the neuron apoptosis in hippocampal CA1 region of newborn with hypoxic-ischemic brain damage(HIBD).Methods The experiment included 2 parts.One was to measure the apoptosis rate by the flow cytometry,and the other was to investigate the expression of Bcl-2 and Bax of neurons in left hippocampal CA1 region.Models of postnatal 7-day Sprague-Dawley(SD)rats with HIBD were established,and were equally divided into 4 groups:sham operation(group S),control group(group C),shenfu injection pretreatment(group P),and shenfu injection treatment(group SF).The neuron apoptosis rate in hippocampal CA1 region in every group was measured at 2 h before and 2,12,24 h,3,7,14 and 28 d after hypoxic ischemic(HI) insult.The expressions of Bcl-2 and Bax were performed by immunohisochemistry.Results The apoptosis of neuron in hippocampal CA1 region in group P and group SF after HI insult was significantly less than that of group C(P
2.Simulation and experimental study of cone artifact in spiral CT.

OBJECTIVETo study regular patterns of cone artifact resulted from interpolation algorithm of spiral CT.
METHODSBased on the principle of interpolation algorithm and back-projection reconstruction, a mathematical model of the reconstructed image was established to clarify the relation of the scanning parameters and the characteristics of the scanned object with the cone artifact. Experiments were carried out by a set of acrylic phantoms on siemens plus 4 CT scanners.
RESULTSThe artifact in the image was directly proportional to the table increment per gantry rotation of the scanner, and was positively correlated to the tangent of half cone-angle and inversely to the radii in the reconstruction plane of the phantom. The theoretical analysis was validated by experimental results.
CONCLUSIONThe cone artifact is related to the scanning parameters and the characteristics of the scanned object.

3.Association between hypertriglyceridemic waist?to?height ratio phenotype and chronic kidney disease in a community population in South China: a cross-sectional study.

OBJECTIVETo investigate the relationship between hypertriglyceridemic waist to height ratio phenotype (HWHtR) and chronic kidney disease (CKD) in a community population in South China.
METHODSA cross sectional study was conducted among 2142 residents in Zhuhai (Guangdong Province, China) from June to October of 2012. The HWHtR phenotype was defined as a waist to height ratio(WHtR) ≥0.55 and triglyceride level ≥2.0 mmol/L, based on which the participants were divided into HWHtR group and nonHWHtR group. CKD was defined as an eGFR<60 mL/min per 1.73 m2 or an ACR ≥30 mg/g. A logistic regression model was established to investigate the relationship between chronic kidney disease and HWHtR phenotype.
RESULTSCompared with the nonHWHtR group, the HWHtR group had a higher prevalence of chronic kidney disease (11.1% vs 33%, P<0.001). Analysis using the logistic regression model showed that HWHtR was significantly associated with CKD in the unadjusted analyses (OR=3.23, 95% CI: 2.32-4.48, P<0.001). After adjustment for age, sex, history of hypertension, history of diabetes, systolic blood pressure, diastolic blood pressure, drinking, physical exercise, education and current smoking, HWHtR was significantly associated with CKD (OR=2.36, 95% CI: 1.52-3.67, P<0.001); the association of HWHtR and CKD was still significant after further adjustment for BMI (OR=2.12, 95%CI: 1.34-3.35, P<0.001).
CONCLUSIONOur finding suggests that HWHtR is associated with CKD in this community population.

6.Effect of MiR-200b on retinal endothelial cell function in high-glucose condition and the mechanism.

OBJECTIVETo investigate the effect of MiR-200b on human retinal endothelial cells (hRECs) cultured in high glucose and explore the mechanism.
METHODShRECs cultured in high glucose or in normal media were examined for MiR-200b mRNA expression using real-time PCR. The effect of MiR-200b transfection on hREC proliferation in high-glucose culture was evaluated with MTT assay, and real-time PCR and Western blotting were performed to determine vascular endothelial growth factor (VEGF) and transforming growth factor β1 (TGFβ1) expression in the transfected cells.
RESULTSThe cells in high-glucose culture showed significantly decreased MiR-200b expression and active proliferation. Compared with those in normal control cells, VEGF and TGFβ1 mRNA and protein expressions increased markedly in cells cultured in high glucose (P<0.05). MiR-200b transfection of the cells caused significantly increased cellular expression of MiR-200b but decreased expression levels of VEGF and TGFβ1 mRNA and protein, and suppressed hREC proliferation in high glucose culture (P<0.05).
CONCLUSIONMiR-200b can regulate REC growth and proliferation by changing VEGF and TGFβ1 expressions and thus play a role in the pathogenesis and progression of diabetic retinopathy.

7.Antiproliferative effects of LY294002 on MCL Jeko-1 cell line and its mechanism.

This study was aimed to investigate the effects of LY294002, a specific inhibitor of phosphatidylinositol 3-kinase, on growth and apoptosis of MCL Jeko-1 cell line and its mechanism. The proliferation inhibitory rate of Jeko-1 cells treated by different doses of LY294002 was assayed by MTT method; the level of apoptosis of Jeko-1 cells was detected by flow cytometry; the expression level of apoptosis-related protein Cyclin D1, Bcl-2, procaspase-3 and PI3K/Akt signaling pathway protein phosphorylated-Akt (p-Akt), phosphorylated-TOR (p-mTOR), phosphorylated-P70S6K (p-P70S6K) phosphorylated-Akt (p-Akt) in Jeko-1 cells were determined by Western blot. The results showed that the growth of Jeko-1 cell line was inhibited by LY294002. The apoptosis rates of Jeko-1 cells treated with 0, 5, 10 and 20 µmol/L of LY294002 for 24 hours were (3.25 ± 1.27)%, (11.34 ± 2.35)%, (22.81 ± 2.74)%, (43.61 ± 3.48)% respectively, the difference between them was statistically significant (P < 0.01). Phosphorylation levels of PI3K/Akt signaling pathway protein p-Akt, p-mTOR, p-P70S6K decreased, the expression of apoptosis-related protein cyclin D1, Bcl-2, procaspase-3 was down-regulated.It is concluded that the LY294002 can inhibit Jeko-1 cell proliferation, which may be realized through down-regulating the phosphorylation level of p-Akt, p-mTOR, p-P70S6K, inhibiting the P13k/Akt signaling pathway, and promoting the cell apoptosis.

8.Expressions of DAPK3 and c-Myc in endometrial cancer and their relationship with the patients' prognosis.

OBJECTIVETo investigate the expressions of DAPK3 and c-Myc and their prognostic value in endometrial carcinoma (EC).
METHODSThe expressions of DAPK3 and c-Myc were detected immunohistochemically in 132 surgical specimens. The relationship between DAPK3 and c-Myc protein expressions and the clinicopathological features of the patients was evaluated.
RESULTSImmunohistochemical analysis revealed low DAPK3 expression in 60.61% (80/132) and high c-Myc expression in 53.79% (71/132) of the specimens of EC. Both DAPK3 expression and c-Myc expression were significantly correlated with FIGO stage (P=0.034 and 0.015, respectively) and lymph node metastasis (P=0.022 and 0.031, respectively). DAPK3 expression was closely correlated with the histological grade (P=0.027) and depth of myometrial invasion (P=0.011). Kaplan-Meier survival analysis indicated that patients with low DAPK3 expressions had a shorter overall survival rate than those with high DAPK3 expressions (P=0.023), while patients with high c-Myc expressions had poorer prognoses than those with low c-Myc expressions (P=0.002). Spearman correlation analysis showed that DAPK3 and c-Myc expressions were negatively correlated (P<0.001, r=?0.310). Multivariate analysis identified a high c-Myc expression as the independent predictor of the prognosis of EC (P=0.007).
CONCLUSIONSA low expression of DAPK3 and a high expression of c-Myc are associated with aggressive and metastatic behaviors of EC, and their detection may help to predict the prognosis of the EC patients.

9.Expression of nuclear export factor CRM1 and p27 in glioma.

OBJECTIVETo investigate the expression of nuclear export factor CRM1, Ser10-phosphorylated p27 and p27 in human gliomas.
METHODSThe expression of CRM1, Ser10-phosphorylated p27 and p27 were investigated in 70 cases of human gliomas and 10 specimens of the normal brain tissue by immunohistochemical technique and Western blot.
RESULTSThere were significant differences on the expression levels of CRM1, Ser10-phosphorylated p27 and p27 among normal brain tissue, gliomas of grades II and gliomas of grades III plus IV (P < 0.01). The expression of CRM1 in gliomas was inversely correlated with the expression of p27 (r(s) = -0.727, P < 0.01) and positively correlated with the expression of Ser10-phosphorylated p27 (r(s) = 0.954, P < 0.01) and Ki-67 (r(s) = 0.799, P < 0.01). Moreover, the expression of Ser10-phosphorylated p27 was inversely correlated with p27 (r(s) = -0.744, P < 0.01) and positively correlated with Ki-67 (r(s) = 0.785, P < 0.01).
CONCLUSIONSCRM1, through recognizing and binding with Ser10-phosphorylated p27, may promote moving of p27CRM1 from its original locating sites; act as a critical signaling component in the proliferative process of glioma cells and then, plays an important role in the development of gliomas.

10.Methylglyoxal inhibits human umbilical vein cell migration in vitro by down-regulating integrinβ3.

OBJECTIVETo explore the effects of methylglyoxal on endothelia cell migration.
METHODSHuman umbilical vein endothelial cells (HUVECs) were stimulated by serial concentrations of methylglyoxal (MGO, 0, 25, 50, 100 and 200 µmol/L) for 24 h, and the cell migration was assessed by scratch wound and Transwell assay. The expression of integrin β3 in the treated cells was examined by immunoblotting, and the effect of an anti-β3 antibody, LM609, on cell migration was investigated.
RESULTSMethylglyoxal significantly inhibited HUVEC migration in a concentration-dependent manner (P<0.05). Methylglyoxal decreased the expression of integrin β3 in a time- and concentration-dependent manner (P<0.05). LM609 also significantly inhibited HUVEC migration (P<0.05).
CONCLUSIONMethylglyoxal inhibits HUVEC migration in vitro by down-regulating integrin β3 expression.
