2.Effect of estrone on hippocampal neuronal apoptosis induced by NMDA in rat primary culture

Aim To observe the action of estrone on the hippocampal neuronal apoptosis induced by NMDA in vitro and to analyse the mechanism underlying the neuroprotection of estrone. Methods The methods of both morphological analysis and cell viabilities were used, and Western blot was also used to analyce the role of antitoxiciy of estrone. Results The cultures were conformed with fluorescent dye of Hoechst 33258 and NeuN. About 31.6% neurons were induced into apoptosis by NMDA(300 ?mol?L~ -1 +glycine 5 ?mol?L~ -1 ,compared with control group, P0.05). MTT test also showed that estrone can protect cultures from apoptosis induced by NMDA, it can raise the cell viabilities exposed to NMDA. Western blot showed that activities of caspase-3 can be inhibited by estrone, the segments of caspase-3 can diminished after application of estrone together with NMDA. Conclusion Estrone had neuronal protection, this role maybe relate to the inhibition of caspase-3, a common pathway in apoptosis. The results can provide some clues to the explovation of the clinical use of estrone and to the development of a neuronal protective drug.
3.Role of voltage-dependent chloride channels in hippocampal neuronalapoptosis induced by 3-morpholinosydnonimine in rats

OBJECTIVE To explore the effect of chloride channels on the neuronal injury following cerebral ischemia. METHODS Tweleve day in vitro (12dIV) neurons in rats were randomly divided into normal control, 3-morpholinosydnonimine (SIN-1, 1.0 mmol·L~(-1) for 18 h) group, SIN-1+4-acetamido-4'-isothiocyanatostilbene-2,2'-disulphonic acid(SITS, 0.5 mmol·L~(-1)) group and SIN-1+4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS, 0.1 mmol·L~(-1)) group. Drugs were added with SIN-1 simultaneously and coincubated for 18 h. The neuronal apoptosis and morphological changes were detected with Hoechst 33258. Chloride channels(ClC)-2/ClC-3 were analyzed with immunofluorescence, the chloride channel currents were recorded with whole cell patch-clamp technique. RESULTS Hoechst 33258 staining showed that the apoptotic percentages were (18.61±0.59) %, (50.43±0.56)%, (23.37±0.52)% and (23.37±0.84)% in normal control group, SIN-1 group, SIN-1+SITS or SIN-1+DIDS groups, respectively. ClC-2/ClC-3 were positively expressed in normal neurons. The currents in neurons exposed to SIN-1 were increased about 55%-56%, SITS and DIDS, two kinds of chloride channel blockers could inhibited the currents about 50%-60% and 30%-40%, respectively. CONCLUSION Voltage-dependent chloride channel maybe participate in the neuronal apoptosis induced by NO, and the activities of chloride channels are perhaps involved in the cerebral ischemic injury.
4.The protective effects of chloride channel blockers on the cultured hippocampal neuronal apoptosis induced by NO in rats

Aim To observe the protective effects of SITS and DIDS,two kinds of chloride channel blockers,on hippocampal neuronal damage induced by NO in culture.Methods The cultures were divided into three groups:control group,NO treatment group,NO treatment plus chloride channel blocker group. The cultures were detected with the methods of morphological stain (Hoechst 33258),and the apoptotic neurons and neuronal viabilities were observed through MTT quantitative analysis. The activated caspase-3 was analyzed with western blot.Results There were significant protective effects of SITS and DIDS on neuronal damage with dose-dependence.Conclusions Chloride channel blockers have some protective effects against neuronal injury induced by NO.
5.The early diagnostic value of corneal laser confocal microscopy for small neuropathy in elderly patients with type 2 diabetes mellitus

Objective To investigate the early diagnostic value of cornel confocal microscopy for the screening of small neuropathy in elderly patients with type 2 diabetic mellitus.Methods In the prospective study,96 elderly patients with diabetes as study group and 46 patients with non-diabetes as the control group were continuously collected from our hospital endocrinology and ophthalmology out patients during May 2014 to February 2016.The 96 cases of type 2 diabetes were subdivided into 47 patients with diabetic peripheral neuropathy (DPN)and 47 patients with nowdiabetic peripheral neuropathy(non DPN).Results The diabetes course was shorter in non-DPN group than in DPN group(P=0.000).The levels of glycosylated hemoglobin and urine albumin were lower in the nonDPN than in the DPN(P =0.072,0.007,respectively).The corneal nerve fiber density was lower in the DPN group than in NDPN group (P =0.000).Corneal nerve fiber density was higher in control group than in DPN and NDPN group.The differences in number of corneal nerve fibers showed no statistical significance between DPN and NDPN group (x2 =2.391,P =0.314).But the number of corneal nerve fibers was significant less in DPN and NDPN group than in control group(x2 =16.014,P =0.000).The negative correlation was found between the course of disease and corneal fibrous density by using single factor linear regression analysis.The number of corneal nerve fibers was lower in smoking group than in non-smoking group(P=0.003).The multiple linear regression analysis showed that duration of diabetes was a risk factor for diabetic neuropathy.Conclusions In some elderly diabetic patients with non-neuropathy,corneal nerve fiber density and number have been significantly decreased before nerve conductive velocity is reduced.Therefore,corneal confocal microscopy can be used to detect and diagnose small diabetic neuropathy in elderly patients with diabetes mellitus.
6.The diagnostic value of ProGRP for small cell lung cancer

Objective To explore the clinical significance of serum ProGRP in diagnosis of SCLC.Methods Serum ProGRP were detected in twenty-three SCLC patients,forty NSCLC patients and forty-three patients with benign pulmonary disease whose were definite diagnosis collected from April 15,2016 to July 19,2016,and a contemporary cohort of 40 healthy controls were while recruited.Results The levels of ProGRP in SCLC group were significantly higher than those in NSCLC group,benign pulmonary disease group and healthy control group(P <0.05).The levels of ProGRP in extensive disease group were significantly higher than those in 1imited disease group(P <0.05).There was no significant difference in the positive rates of ProGRP between the sex group and age group (P > 0.05 ).The boundary value of diagnosis of SCLC through ProGRP identified through ROC curve was 65.66 ng/L.The sensitivity and specificity of the ProGRP for the diagnosis of SCLC were respectively 90.9% and 89.9%.Conclu-sion Serum ProGRP can be use as a sensitive and specific index for diagnosis of SCLC,and the level of ProGRP can also be used for the clinical staging of SCLC.
7.Clinical efficacy of benadryl cream and metronidazle cream in treatment of topical corticosteroid dependent dermatitis

Objective To assess the clinical efficacy and safety of 1% benadryl and 3 % metronidazle cream in the treatment of topical corticosteroid dependent dermatitis. Methods The study was conducted in a single-blind way. The 86 patients were divided into 3 groups: 1% benadryl and 3 %metronidazle cream (group A), loratadine (group B) and gluco-corticoids (group C). All preparations were applied twice daily for 8 weeks. Study visits took place at baseline and weeks 2, 4 and 8. Results The clinical response rate in the groups A, B and C was 86.20%, 73.33%, and 66.67%, respectively; the clinical response rate in group A was significantly higher than that in groups B and C (P<0.05). Conclusions 1% benadryl and 3 % metronidazle cream are effective and safe in the treatment of topical torticosteroid dependent dermatitis.
8.Expression of GABA and glutamate vesicular transporter in depressed mice

Objective To study the expression of vesicular GABA transporter and vesicular glutamate transporter 1 in de‐pression .Methods Mice was divided into control group and defeat group stochastically .By social defeat model and social avoidance , the defeat group was divided into two groups:susceptible group and unsusceptible group .Synaptic proteins were extracted respec‐tively from the 3 groups .We detected the expression abundance of VGAT and VGLUT1 by Western blot .Results Compared with the control group ,in susceptible group ,the residence time in the contact area was significantly reduced ,and the residence time in the corner area was significantly increased ,with statistical difference(P<0 .05) .In the prefrontal cortex and hippocampus ,Com‐pared with the control group and the unsusceptible group ,the expression levels of VGLUT1 and VGAT were increased in the sus‐ceptible group(P<0 .05) .there were no statistically significant in VGLUT1 and VGAT leveles between control group and the un‐susceptible group(P>0 .05) .In the striatum ,although the expression levels of VGAT and VGLUT1 were increased in susceptible group ,but in unsusceptible group ,the expression of these proteins also increased significantly .Conclusion The prefrontal cortex and hippo‐campus excitability and inhibitory vesicle transport were changed in depression ,which may relate to the transcription disorder .
9.Study of multiple valve replacement in patients with giant left ventricle

Objective: To summarize the experience of valve replacement for multiple valve insufficiency in patients with giant left ventricle and the operative indication. Methods: Multiple valvular operations were performed in 62 patients with giant left ventricle between 1991 and 2002. Combined mitral and aortic valve replacement was performed in 56, tricuspid valve annuloplasty in 43, left atrium placation surgery in 12, and mitral valve replacement in 5. Results: The early postoperative complication and mortality rate were 45.2% and 17.7%, respectively .The late mortality rate was 6.5%. The main factors influencing the early surgical results were preoperative severe left ventricular enlargement (ESD ≥6.0cmand EDD ≥8.0cm) and systolic dysfunction (EF ≤0 40 and FS ≤0 25), perioperative ventricular fibrillation, postoperative low cardiac output and multiple organal failure. The main factors affecting long term survival were postoperative severe ventricular arrhythmia and left ventricular enlargement with depressed systolic performance. Conclusion: The keys to improve the early and late results of multiple valve replacement in these patients with giant left ventricle are the choice of optimum surgical timing, the proper management of the high risk factors mentioned above during perioperative and follow up periods.
10.Comparison of normal standard value of glomerular filtration rate in different types of SPECT

Objective To evaluate and compare the normal value of glomerular filtration rate (GFR) in different modalities of single photon emission computed tomography (SPECT). Methods Fifty normal volunteers were divided into 4 groups (aged 20-29, n=14;30-39, n=10;40-49, n=14;50-, n=12). All of the volunteers underwent renal dynamic imaging with MPR SPECT, and single renal GFR value as well as total GFR value was calculated, respectively. The data were compared with GFR values obtained from Starcom 400AC/T SPECT (aged 20-29, n=6;30-39, n=11;40-49, n=7;50-, n=9). Single t test was used to testify the difference of the mean value between the two modalities of SPECT. Results The mean value of single renal GFR in each group of MPR SPECT was (51.03±11.12) ml, (48.44±11.13) ml, (47.79±7.71) ml and (46.54±12.28) ml,while of Starcom 400AC/T SPECT was (57.92±9.03) ml, (57.34±10.33) ml, (55.30±8.51) ml and (44.12±7.94) ml, respectively, and the total renal GFR in each group of MPR SPECT was (102.13±18.33) ml, (96.62±19.90) ml, (95.54±13.04) ml and (92.85±22.83) ml, of Starcom 400AC/T SPECT (115.86±16.51) ml, (113.14±17.71) ml, (110.54±11.13) ml and (88.12±14.41) ml, respectively (all P<0.05). The mean value of single renal GFR was (48.51±10.50) ml and (52.92±10.61) ml, respectively, while the mean value of total renal GFR was (97.04±18.16) ml and (105.63±18.72) ml, respectively (all P<0.05). Conclusion GFR standard value obtained from different SPECT renal dynamic imaging is varied. It is necessory to establish GFR standard value for each type of SPECT.
