1.Effect of cisplatin combined with 5-fluorouracil on endocrine Ishikawa cells in endometrial cancer and its possible mechanism

Objective To investigate the effect of cisplatin and 5-fluorouracil on endocrine Ishikawa cells in the treatment of endometrial cancer. Methods The human endometrial cancer cell line Ishikawa were selected, at the logarithmic growth period, the cells were divided into three groups of 5-fluorouracil (5 mg/L) group, cisplatin (5 mg/L) group and cisplatin (5 mg/L) combined with fluorouracil (5 mg/L) group ( combined group).After treated with corresponding drugs treatment, the cell viability was detected by MTT, the apoptosis was detected by flow cytometry, and the Bcl-2 and p65 expressions cells were detected by Western blot.Results The inhibitory rate of combined group, cisplatin group and 5-fluorouracil group were (41.45 ± 3.13)%, (25.20 ±3.09)% and (23.19 ±4.10)% respectively, and the inhibitory rate in the combined group was significantly higher than that of the other two groups (P<0.05).The apoptosis rate of the combined group, cisplatin group and 5-group were (29.44 ±4.35)%, (5.74 ±1.12)% and (5.82 ±1.78)% respectively, and the apoptosis rate in the combined group was significantly higher than that of the other two groups (P<0.05).The Bcl-2 relative expression in the combined group, cisplatin group and 5-fluorouracil group were (0.31 ±0.11), (1.23 ±0.49) and (1.28 ±0.59), the p65 expression were (0.67 ±0.23), (1.67 ±0.56) and (1.71 ±0.71), combined group of Bcl-2 and p65 relative expressions were obviously less than that of the other two groups ( P <0.05 ) .Conclusion Cisplatin combined with 5-fluorouracil on endocrine Ishikawa cells in the treatment of endometrial cancer could promote cell apoptosis, inhibit the cell proliferation, and its mechanism may be related to the inhibition of Bcl-2 and p65 protein expressions.
2.Effect of IL-2 Gene Therapy on Immune and Hematopoietic Reconstitution after Bone Marrow Transplantation

In the present study, the effect of fibroblast-mediated IL-2 gene therapy on immune and hematopoietic reconstitution was observed after syngeneic bone marrow transplantation (BMT) in the mice which had received high-dose chemotherapy. The NK activity , LAK activity and the proliferation of BMT were augmented significantly , while there were no effects on the formation of CFU-GM, CFU-MK and CFU-E of bone marrow after IL-2 gene therapy. The results suggested that fibroblast-mediated IL-2 gene therapy can accelerate and prompt the process of immune reconstitution, then enchance the antitumor effect and reduce the complication such as infection after BMT. The experiment provide basis for the application of IL-2 gene therapy in BMT in the future.
3.Analysis on the clinical characteristics of the schizophrenia to make troubles and accidents likely

Objective To explore the clinical characteristics of the patients with schizophrenia to make troubles and accidents likely ,to provide scientific evidence for preventing and controlling these patients to make troubles and accidents effectively .Methods 100 patients with schizophrenia to make troubles and accidents likely were randomly extracted and assigned to the study group from psychiatric department of this hospital .100 ordinary patients were divided into control group .Their data on illness were sur-veyed and analyzed retrospectively .The difference of the general condition and illness data along with mental symptoms between the both group were compared .Results The proportion of patients to be man peasant ,in rural area ,no a spouse and medical insurance , to have a lower degree of culture and to be 20 - 39 years old in the study group was significantly higher than those in the control group(P< 0 .05) .The social support and economic conditions of family in the study group were significantly poorer than those in the control group(P< 0 .05) .The proportion of patients to be paranoid schizophrenia ,to have a complications and the positive score of the positive and negative syndrome scale (PANSS) in the study group were significant higher than those in the control group but the negative score of PANSS and the course of disease were significantly lower (P< 0 .05) .The proportion of patients to have posi-tive symptoms such as illusion and delusion in the study group were significantly higher than that in the control group (P< 0 .05) . But the proportion of patients to have negative symptoms such as avoid social and emotional dull in the study group were signifi -cantly lower than that in the control group(P< 0 .05) .Conclusion The positive symptoms contribute to making troubles and acci-dents in SP patients .The patients to be paranoid schizophrenia ,a man and peasant ,in prime of life ,in rural area ,no a spouse and to have a lower degree of culture maybe make troubles and accidents more likely .Having a poor social support and economic conditions of family and no medical insurance cause the probability of their making troubles and accidents increase .
4.Bone cement kyphoplasty for repair of rheumatoid arthritis combined with Kümmell’s disease:a follow-up addressing vertebral height and spinal stabilization

BACKGROUND:For patients with rheumatoid arthritis and Kümmel ’s disease, how to effectively control back pain, to recover patient’s locomotor activity and to avoid a vicious cycle of disuse osteoporosis is a key therapeutic target. Kyphoplasty is a recently developed new technology of minimal y invasive spine surgery. Few reports concerned the kyphoplasty for rheumatoid arthritis and Kümmel ’s disease. OBJECTIVE:To assess the clinical outcome of bone cement kyphoplasty for the treatment of Kümmel ’s disease combined with rheumatoid arthritis. METHODS:From June 2012 to July 2013, 11 female patients at the age of 65.4±5.1 years with Kümmel ’s disease combined with rheumatoid arthritis, who suffered from severe back pain, were treated with bone cement vertebroplasty. Back pain and imaging indexes were compared and observed before surgery and during fol ow-up. Imaging indexes contained preoperative and postoperative anterior height of vertebral body after fractures, the ratio of anterior height to posterior height of the vertebral body, and local kyphosis angle (Cobb method). RESULTS AND CONCLUSION:No patients were lost to fol ow up. 11 patients were fol owed up for 6 to 12 months. Significant differences in fol ow-up and preoperative Visual Analogue Scale scores, anterior height of vertebral body after fractures, the ratio of anterior height to posterior height of the vertebral body, and local kyphosis angle were detected (P<0.05). Two patients experienced bone cement leakage. No severe complications appeared such as pulmonary embolism or neurological dysfunction. These data confirmed that bone cement vertebroplasty for rheumatoid arthritis combined with Kümmel ’s disease can effectively lessen back pain, partial y restore the height of vertebral body after fracture, rebuild spinal stabilization, reduce local kyphosis, and is a safe effective repair method.
5.Establishment of a zebrafish model of thrombosis and the intervention effect of Guanxinning tablet

Objective To establish a zebrafish model of thrombosis induced by three kinds of inducers and observe the anti?thrombotic effect of a Chinese traditional medicine, Guanxinning tablet ( GXN) . Methods The zebrafish models of thrombosis was induced by using 1?5μmol/L phenyl hydrazine, 80μmol/L arachidonic acid and 5 mg/L ponatinib, re?spectively, and were treated with various concentration of GXN, clopidogrel or asprin. The thrombus in the tail vein was observed under microscope, Erythrocytes in the zebrafish heart were stained with o?dianisidine and the erythrocyte staining intensity was assessed with a NIS?Elements DTM image analyzer, and the anti?thrombolic effect of GXN was calculated. Results Venous thrombus was significantly increased and the staining intensities of erythrocytes in the heart were signifi?cantly decreased after induction by phenyl hydrazine, arachidonic acid or Ponatinib ( P <0?001 ) , respectively. At the same time, GXN showed an incresing anti?thrombolic effect in the zebrafish models (P<0?001) in a dose?effect manner, with a IC50 of GXN of 44?32 mg/L,138?5 mg/L and 459?5 mg/L, respectively. Conclusions The zebrafish models of thrombosis are successfully established by phenyl hydrazine, arachidonic acid or Ponatinib, respectively, by different for?mation mechanisms. GXN has been shown to have an anti?thrombosis effect, probably, by multiple target effects.
6.FINE DISTRIBUTION OF THE LYMPH VESSELS IN THE STOMACH AS STUDIED BY ENZYME-HISTOCHEMICAL METHOD

By using 5'-nucleotidase-alkaline phosphatase (5'-Nase-AlPase) double staining method, the fine distribution of the lymph vessels in the stomach wall of the rabbit, guinea pig and rat has been observed. Under light microscope lymph capillaries showed strongly 5'-Nase positive reaction with brown or dark brown staining. However blood capillaries revealed significantly A1Pase activity with blue staining. The lymph capillaries were seen in all layers of the stomach wall in three species, but the lymph vessels were only shown in the submucosa, muscularis and serosa. In the mucosa lymph capillaries could be found in the deep layer of the lamina propria. In the submucosa there were lymph vessels and capillaries. In the muscularis lymph capillaries and vessels not only could be found between oblique and circular muscle or between circular and longitudinal muscle, but also among muscle fiber bundles of each layer. In the serosa there were larger lymph vessels situated near the muscularis. The observation on semithin and ultrathin sections also shown the similarity in the lymph vessels distribution. There was no difference in the distribution of lymph vessels in the stomach wall between rabbit, guinea pig and rat.
7.Effects of Atomic Oxygen Radical Anion on Escherichia coli Cells

The effects of atomic oxygen radical anion (O-) on the inactivation and morphological changes ofEscherichia coli (E. coli)on the surface of bio-indicator carrier were investigated. The O- flux was generated from a novel developed O- generator where the Oinactivation ofE. coli was sensitive to the O- intensity and the cell mortality was enhanced to more than 3-logarithm reduction with the exposure to 1.5 mA/cm2 O- flux for 120 min. Field emission scanning electron microscopy (FESEM) observations showed that O- flux destroyed cellular structures. Lipid peroxidation reaction induced by atomic oxygen radical anion for E. coli cells was also observed using product of malondialdehyde (MDA) as an index. The concentration of MDA increased to 1.2 μmol/g(dry weight) of cells when E. coli suspension (5.6×107 cfu/ml) was treated by the O- flux (1.5 μA/cm2) for 15 min. The findings revealed that the atomic oxygen radical anions, with strong oxidation power, was effective in inactivating E. coli and caused lipid peroxidation reaction at the first time,which would be potential useful to develop a novel approach for the microbial decontamination and for the study on the interactions between microorganisms and O-.
8.Construction of MuSK-mCherry fusion fluorescent protein and used for detection of MuSK antibodies in myasthenia gravis

Objective:To construct a fusion protein of extracellular domain peptide fragment of muscle specific kinase ( MuSK) and fluorescent protein mCherry ,and used as antigen in the detection of antibodies against MuSK ( MuSKAb ) in the sera of patients with myasthenia gravis ( MG).Methods:The mCherry gene was amplified by PCR from vector pRSET-B and cloned into pGEM-T Easy Vector,and furthermore, cloned into Eukaryotic expression vector pMT /BiP/V5-His ( MuSK), which contains MuSK extracellular domain 22-452 amino acid peptide fragment gene to construct the fluorescent fusion protein gene MuSK -mCherry.The recombinant vector was subsequently transfected into drosophila S 2 cells for expression.The expressed fusion proteins were verified in confocal mi-croscope ,and used as antigen in the detection of MuSKAb in sera of MG patents in fluorescence immunoprecipitation test .Results:The fluorescent fusion protein MuSK-mCherry was successfully constructed and expressed.The MuSKAb in sera of patents with MG could be detected in fluorescence immunoprecipitation test using the constructed MuSK-mCherry fusion protein as antigen.Conclusion: It is available to use the constructed fluorescent fusion protein MuSK-mCherry as antigen in fluorescence immunoprecipitation test for the detection of MuSKAb in sera of patents with MG.
9.Association between SLC10A1 genetic variations and suspectbility to hepatitis B virus infection by mother-to-child transmission

Objective To investigate the association between SLC10A1 gene mutations in c.800G>A mutation and c.356 +1098C >T mutation, and the susceptibility to HBV infection by mother-to-child transmission ( MTCT) .Methods Totally 306 individuals born to HBeAg-positive mothers with high load HBV and without receiving nucleotide analogues treatment, including 247 HBV-infected cases and 59 non-HBV-infected ones were enrolled from Southwest Hospital during May 2011 and July 2015.Blood samples were collected from all the subjects, then genomic DNA was extracted and c.800G>A mutation and c.356+1098C>T mutation of SLC10A1 were genotyped .Chi-square test (Pearsonχ2or continuity correctionχ2) was performed to identify the difference in genotypes between two groups.Results Among vaccinated individuals (55 HBV infected and 56 not infected), the frequency of genotype GA of c.800G>A mutation in non-infected ones was 14.3%(8/56), there was a tendency of increasing compared with HBV infected ones (5.5%, 3/55), but the difference was not statistically significant (χ2 =2.424, P =0.119). Similarly, the frequencies of genotypes CC, CT and TT of the c.356+1098C>T mutation in HBV infected ones were 20.0%(11/55), 47.3%(26/55) and 32.7%(18/55), while those in non-infected ones were 12.5% (7/56), 69.6% (39/56) and 17.9% (10/56), and the difference was not of statistical significance (χ2 =5.766, P=0.056).In all subjects (vaccinated and non-vaccinated), the frequency of genotype GA of c.800G>A mutation in non-HBV infected group had an increasing tendency compared with HBV-infected offspring (13.6% vs.6.9%), but the difference was not statistically significant (χ2 =2.010, P=0.156);the frequencies of genotype CC, CT and TT of c.356+1098C>T mutation in HBV infected patients were 20.2%(50/247), 49.8%(123/247) and 30.0%(74/247), while those in non-HBV-infected group were 11.9%(7/59), 69.5%(41/59) and 18.6%(11/59), and the difference was statistically significant (χ2 =7.436, P =0.024 ) .Within the HBV infected group, the frequencies of genotype GA of c.800G>A mutation were 5.5%(3/55) in vaccinated individuals and 7.3%(14/192) in non-vaccinated individuals, and the difference was not of statistical significance (χ2 =0.030, P=0.863);Similarly, the frequencies of genotype CC, CT and TT of c.356 +1098C >T mutation in vaccinated individuals were 20.0%(11/55), 47.3%(26/55) and 32.7%(18/55), while those in non-vaccinated individuals were 20.3%(39/192), 50.5%(97/192) and 29.2%(56/192), and the difference was not of statistical significance (χ2 =0.274, P=0.872).Conclusion c.356+1098C>T mutation in SLC10A1 may be associated with susceptibility to HBV infection of child born in HBeAg positive pregnant women infected with high load HBV.
10.Overview of genetic and biochemical loci in Mesocricetus auratus and its albino mutant

Objective To establish the genetic and biochemical loci in Mesocricetus auratus and its albino mua-tant.Methods Protein isozyme cellulose acetate electrophoresis was used to determine the genetic and biochemical loci in Mesocricetus auratus and its albino mutant, using the genetic and biochemical loci of mice and rats.Results 25 biochemi-cal markers of Mesocricetus auratus and albino mutant were established, and polymorphism of their genetic biochemical loci was analyzed.Conclusions Polymorphism of biochemical loci is present in Mesocricetus auratus.Some differences exist between the genetic biochemical loci of Mesocricetus auratus and their albino mutant.These results laid the foundation for further study on genetic mechanism of albino mutation in Mesocricetus auratus.
