1.Effects of Buyang Huanwu Decoction on Contents of PCⅢ and Ⅳ-C in Blood Serum of Pulmonary Fibrosis Rats

Objective To discuss intervention effects of Buyang Huanwu decoction on pulmonary fibrosis (PF) rats. Methods Totally 144 healthy male SD rats were randomly divided into six groups:sham-operation group, model group, prednisone group, Buyang Huanwu decoction high-, medium-, low-dose groups, 24 rats in each group. Except for sham-operation group, the rest groups were given injection of BLM via tracheal intubation for the establishment of PF model, while sham-operation group was instilled the same amount of NS. From the second day, all the rats received a gavage by related medicine. Eight rats in each group were killed respectively on day 7, 14 and 28 after intratracheal instillation. The contents of PCⅢ and Ⅳ-C in blood serum of experimental rats were tested by radioimmanoassay method. Results Compared with the sham-operation group, the contents of PCⅢ and Ⅳ-C in blood serum of other groups increased gradually, and that of the model group was the most obvious (P<0.01). Compared with the model group, each medicine group had different decreases in the contents of PCⅢ and Ⅳ-C in blood serum (P<0.01). Compared with the prednisone group, the contents of PCⅢ and Ⅳ-C in blood serum of Buyang Huanwu decoction medium-dose group obviously decreased on the 14th and 28th day (P<0.05), as well as that of Buyang Huanwu decoction low-dose group on the 28th day (P<0.05). Conclusion Buyang Huanwu decoction could effectively reduce proliferation of collagen fibers and excess deposition of ECM in lung tissue of PF, and played an anti-fibrosis effect on lungs.

3.Detection of CK-19 mRNA in peripheral blood from breast cancer patients by reverse transcription polymerase chain reaction

Purpose: Detect cytokeratin 19 mRNA(CK-19 mRNA) in nucleated cells in peripheral blood from breast cancer. To establish a diagnostic method for breast cancer metastasis in peripheral blood. Methods: Peripheral blood samples in breast cancer patients (test group, n = 66) and benign tumour in breast patients ( control group, n = 37) were taken. Then, the nucleated cells were separated and total RNA extracted, and CK-19 mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) technique. Results: Samples were diagnosed CK-19 mRNA positive when 460 bp band appeared in RT-PCR end-product. The positive rate of CK-19 mRNA is 36. 36 % (24/66) in test group . 4.Early clinical analysis of angina after acute myocardial infarction

Objective To investigate the related factors of early angina after acute myocardial infarction,and to provide basis for the disease prevention and control.Methods 1 32 cases with acute myocardial infarction were selected.The clinical data were collected.The occurrence rate of early angina pectoris after acute myocardial infarc-tion,and clinical characteristics were analyzed.The related factors of acute myocardial infarction angina were explored.Results The incidence rate of early angina pectoris after acute myocardial infarction was 29.55%.Within 7 days after acute myocardial infarction occurred angina,the highest rate for 64.1 0%,followed by 7 -1 4 days in the occurrence of angina pectoris,28.21 %.34 cases were the original location of myocardial infarction ischemia and 5 cases were the far part of ischemia,21 cases showed ST segment elevation,1 8 cases showed ST lack blood group downward.Anterior wall,inferior wall infarction composite,successful thrombolytic therapy recanalization,myocardial infarction before episodes of angina pectoris,history of hypertension,heart function classification more than or equal to grade III in patients with acute myocardial infarction occurred after the proportion of early angina pectoris were signifi-cantly higher (all P <0.05).Conclusion Anterior wall,inferior wall infarction composite,successful thrombolytic therapy recanalization,myocardial infarction before episodes of angina pectoris,history of hypertension and cardiac functional grading more than or equal to grade III are related to early angina pectoris after acute myocardial infarction. We should actively take measures aimed at early prevention and treatment of early angina pectoris after acute myocar-dial infarction to avoid illness aggravating,improve the patients'prognosis.
5.Effects of pioglitazone on the expressions of HIF-1α and VEGF in renal tissues of diabetic rats

The effects of pioglitazone on the expressions of hypoxia-inducible factor-1 α (HIF-1 α) and vascular endothelial growth factor (VEGF) in renal tissues of diabetic rats were observed. Diabetic rat model was established by feeding high-carbonhydrate-fat diet and injecting streptozotocin. After the treatment with pioglitazone, the kidney index, 24 h urinary albumin, blood urea nitrogen, serum creatinine, fasting blood glucose, fasting insulin, homeostasis model assessment for insulin resistance (HOMA-IR), total cholesterol,triglycerides, and low-density lipoprotein-cholesterol of diabetic rats were significantly lower than those of untreated ones, high-density lipoprotein-cholesterol was increased, the expressions of HIF-1α and VEGF in renal tissue were decreased ( all P<0. 01 ). It suggested that pioglitazone may improve renal function and the balance of glucose-lipid metabolism in diabetic rats via down-regulating HIF-1/VEGF pathway.
6.Aspirin for the prevention of colorectal adenomas: a systematic review

Objective To assess the effect of aspirin for the chemoprevention of colorectal adenomas by meta analysis of the published literature.Methods Cochrane strategy in combination with manual search was used to identify previously published randomized controlled trials by searching PubMed,EMBase,Cochrane Library,China Journal Full-text Database(CNKI),Chinese Scientific Journal Full-text Database (CSJD) and Chinese Biomedical Literature Database (CBM).Results Six randomized controlled trials involving a total of 2 858 patients were studied.Of the six trials,two trials were performed in China,four trials were in the Europe and the United States.Some sufficient evidence were found to support that aspirin could prevent of colorectal adenomas compared with placebo group ( P =0.003,RR =0.66,95% CI:0.50-0.86).No adaquate evidence supported the role of aspirin in the prevention of development of colorectal cancer ( P =0.29,RR =0.65,95% CI:0.30-1.44).High-dose aspirin ( P =0.10,RR =0.85,95% CI:0.71-1.30 ) and low-dose aspirin could prevent colorectal adenomas compared with placebo group( P =0.02,RR =0.57,95% CI:0.36-0.90),and a dose-dependent associtation was found.The risk of stroke was higher in any dose of aspirin compared with placebo group ( P =0.04),and the risks of adverse events had no significant differences in all groups.Conclusion Aspirin might prevent the development of colorectal adenomas in individuals,but could not prevent the colorectal cancer.
7.Cancer stem cells and their isolation and identification

Cancer stem cells are a subclass of stem-cell-like tumor cells.They have an apparently higher tumorigenicity than other cell populations within the same tumor.They play a Very important role in genesis,development and maintenance of cancer.The data from research in leukemia and solid timors suggest that in each cancer tissue,only a small fraction of the cells have the ability to initiate tumor,which are called cancer stem cells.In recent years,with in-depth research on stem cells and cancer stem cells,accumulating evidence suggests that cancer stem cells are the cause of malignant tumor metastasis and recurrence and are resistant current callcer drugs.Drugs targeting cancer stem cells are urgently needed. Flow cytometry and magnetic cell sorting using the identified tumor stem cell surface markers are mainly used to separate cancer stem cells or stem cell-like cells. In this article,the concept of cancer stem cells and the commonly used approaches to isolate and identify cancer stem cells,especially for gastric cancer stem cells will be reviewed.
8.The detection of γδT lymphocytes of peripheral blood in patients with lung cancer

Objective To study the function of γδT lymphocytes in patients with lung cancer.Methods γδT lymphocytes in peripheral blood of the 124 preoperative patients with lung cancer(experiment group) and 56 healthy donors(control group) were detected by Flow Cytometry. Results The percentage of γδT cells(CD+3 γδTCR+) was (6.54±2.90) % in control group and (3.76±2.81) % in experiment group (t =3.568,P <0.001). The γδT cells in peripheral blood of patients with lung cancer was less than that of healthy donors (t =3.568, P <0.001). There was significant difference in expression of γδT cells in the peripheral blood between the all test groups and healthy control group (P =0.000). Compared with control group, γδT cells of the peripheral blood in experiment group were correlated with TNM stage, general types, pathological changes position and histology grades (P <0.05). The expression of γδT cells in the peripheral blood were not associated with age, gender (P >0.05). Conclusion The results suggested that the dysfunction of γδT had important function on the development of lung cancer.

10.Effects of quercetin on nuclear factor-κB p65 expression in renal ubiquitin-proteasome system of diabetic rats

Objective To investigate the protective effect of quercetin on diabetic nephropathy and to explore its possible mechanism.Methods Type 2 diabetes mellitus rat model was established by feeding high-carbonhydrate-fat diet and injecting with streptozotocin.At 72 hour after injection,blood samples were collected from the tail veins of all rats.Those rats with blood glucose level ≥ 16.7 mmol/L were considered as the diabetes model been successfully established.The model rats were randomly divided into type 2 diabetic group ( group DM,n =9 ) and quercetin group ( group QUE,n =9 ).Other rats were used as normal controls (group NC,n =8).All rats were performed by intragastric administration for 8 weeks.At the end of experiment,the rats were sacrificed and fasting plasma glucose( FPG),fasting insulin( Flns),serum creatinine (SCr),blood urea nitrogen(BUN),TG,TC,LDL-C,24 h urine protein (24 h UP),and kidney index ( KI ) were evaluated.Pathological changes of kidney were observed by periodic acid-silver metheramine( PASM ).The expressions of ubiquitin and NF-κB p65 on glomeruli w ere examined by immunohistochemical method,and its association with the incidence of proteinuria was analyzed.Results In groups DM and QUE,the level of FPG [ ( 25.45 ± 1.23 ) mmol/L and ( 19.99 ± 1.20 ) mmoL/L],FIns [ ( 25.67 ± 2.58 ) mU/L and ( 19.29 ± 1.80 ) mU/L ],SCr[ ( 44.00 ± 2.53 ) μmol/L and ( 34.43 ± 2.23 )μmol/L],BUN[ ( 11.60 ± 0.39 )mmol/L and (8.20 ± 0.37) mmoL/L],TG [ (3.32 ± 0.22 ) mmol/L and (2.43±0.25)mmol/L],TC[(2.95 ±0.21) mmol/L and (2.24 ±0.17)mmol/L],LDL-C[(2.03 ±0.22 ) mmol/L and ( 1.49 ± 0.13 ) mmol/L ],24 h UP [ ( 46.67 ± 2.50 ) mg/24 h and ( 25.57 ± 2.82 )mg/24 h]and KI[ (9.76 ±0.30) × 103 and (8.44 ±0.26) × 103 ] were significantly increased than the indexes of group NC [ (6.56 ± 0.41 ) mmol/L,( 12.63 ± 1.41 ) mU/L,( 22.88 ± 2.36 ) μmol/L,( 5.45 ±0.51 ) mmoL/L,( 1.64 ± 0.1 1 ) mmol/L,( 1.33 ± 0.17 ) mmol/L,(0.46 ± 0.05 ) mmol/L,( 12.38 ±1.19)/24 h and (6.78 ±0.12) × 103].Moreover,the above indexes in group QUE were obviously lower than group DM.There was evidence of pathological changes associated with diabetes,such as focal and segmental sclerosis and thickened basement and mesangial expansion.The expressions of ubiquitin and NF-κB p65 in renal tissues of group DM increased significantly ( P < 0.01 ).The expression of ubiquitin and NF-κB p65 were positively related with the level of 24 h UP ( r =0.893,0.879,P < 0.01 ).Compared with group DM,all above indexes in group QUE were markedly alleviated ( P < 0.01 ).The expression of ubiquitin and NF-κB p65 was reduced but didn't reach level in group NC ( P < 0.01 ).Conclusion The increased expression of NF-κB induced by ubiquitin-proteasome system may participate in the pathogenesis of proteinuria in diabetic nephropathy.Quercetin has renal protective effects partly through reducing NF-κB p65 expression.
