3.Relationship between expression of bcl-2 gene and neuronal apoptosis in neonatal rats with hypoxic-ischemic brain damage

Objective To observe the expression of bcl-2 gene in cell apoptosis of neonatal rats followed by hypoxic-ischemic brain damage(HIBD) and investigate the mechanism of neuronal apoptosis after HIBD.Methods Fifty-four neonatal SD rats were used in 1 sham-operated group and 8 trial groups. The models of HIBD were established in neonatal rats by inhaling the mixed gases of 92 % N 2 and 8 % O 2, the animals were sacrificed by dislocation their heads at different time points(0.5,1,3,6,12,24,48,72 h), the hippocampus were dissected for morphological analysis. The neuronal apoptosis and the expression of bcl-2 gene in hippocampus were detected by the methods of immunohistochemistry. Results The apoptotic cells appeared at the time point of 3 h, and reached the peak at 48 h, then decreased. The positive cell of bcl-2 protein increased from the time point of 30 min and reached the peak at 6 h and then decreased gradually following HIBD. Conclusion The expression of bcl-2 gene plays a role in the process of neuronal apoptosis following HIBD.
4.Inflammatory reaction changes with aging in kidneys of human TIMP-1 transgenic mice

Objective To explore the role of tissue inhibitor of metalloproteinase-1(TIMP-1) during renal senescence by using human TIMP-1 transgenic mice.Methods Renal histological changes of wild type mice and transgenic mice at the age of 3,12,24 months were observed by periodic acid-schiff(PAS)staining of paraffin sections.The numbers of F4/80 positive cells were detected by immunofluoreseence.The protein expressions of TIMP-1,TIMP-2,matrix metalloproteinase(MMP)-9,MMP-2,intercellular adhesion molecule-1(ICAM-1),transforming growth factor?1(TGF-?1),collagenⅢand collagenⅣwere detected by Western blot.The activities of gelatinases and TIMP-1 were examined by gelatin zymography and reverse zymography respectively.Results Focal renal fibrosis was found in two genotypes with aging.At the age of 24 months,compared with wild type,in kidneys of transgenic type,the expressions and activities of gelatinases were dowregulated (MMP-2:2.08?0.20 vs.3.39?0.43;MMP-9:4.02?0.82 vs.6.72?1.40,all P<0.05);the expressions of collagenⅢ,collagenⅣ,ICAM-1,and TGF-?1 were upragulated(0.72+0.11 vs.0.57?0.09;0.84?0.13 vs.0.6?0.11,0.72?0.12 vs.0.53?0.07; 0.69?0.12 vs.0.45?0.09,all P<0.05),and the numbers of F4/80 positive cells were increased (18.8?4.4 vs.12.7?3.6,P<0.05)with the upregulated expression and activity of TIMP-1(1.10?0.18 vs.0.62?0.09;50.75?7.25 vs.20.64?3.50,P<0.05).Conclusions TIMP-1 could promote age-related renal fibrosis through enhancing inflammation reaction by ICAM-1 upregulation.
5.Anatomic analysis of Wenchuan earthquake injuries with otorhinolaryngology symptoms

Objective To analyze the injuries after the earthquake with otorhinolaryngology symptoms and the characteristics of with iniuries and to provide some information when dealing with similar disasters in the future. Methods The basic information of earthquake-related iniuries with otorhinolaryngology symptoms was collected in 11 large and medium-sized hospitals (including foreign medical teams) in Mianyang City and the surrounding four hard-hit counties.Their characteristics were analyzed together. Results There were 289 patients that have otorhinolaryngology symptoms in the 11 included hospitals, among them there were 217 cases have deftnite records that could be analyzed.The data showed that damage to ear and head and neck injuries as the main symptoms was large proportion of injuries suffer multiple fractures and soft tissue injuries such as skin abrasions.Nose injury and facial fractures were also common.Orbital fracture and eye damage could be combined occurred.Brain injury and skull fractures often occurred while trachea and throat injury was rare happened. Conclusions Different disaster cause different damage.After the establishment of disaster-related injuries in professional spectrum more active and effective could be behaved in life rescue and the arrangement of personnel and material when dealing with similar situations.
6.Method of detection of soluble HLA-I and soluble HLA-I level alteration in storage blood.

Aim of this study was to develop the detection method of soluble human leukocyte antigens I (sHLA-I) and to explore sHLA-I level alteration in storage blood and its significance. sHLA-I level in sera of 60 Guangdong normal individuals and sHLA-I concentration in blood components from 20 donors quantitatively were detected by sandwich ELISA. The results showed that sensitivity of this assay was 2.84 ng/ml. Coefficients of variation were 5.80% within assays and 9.00% between assays respectively. The recovery rate was >/= 98.57%. The sHLA-I level of normal individuals in Guangdong was (699.54 +/- 360.10) ng/ml. sHLA-I in red blood cells stored for 28 days and in random-donor platelets were significantly higher than that in other blood components and their amount was proportionate to the number of residual donor leukocytes and to the length of storage. In conclusion, sandwich ELISA assay for detection of sHLA-I is a sensitive, specific and stable technique. Blood components with different concentration of sHLA-I may be chosen for clinical transfusion.

The constituents in 95% ethanol extract of the root of Rosa cymosa Tratt were purified by column chromatography techniques, leading to isolation of eleven triterpenes. Their structures were elucidated by spectroscopic data as pomolic acid (1), fupenzic acid (2), ursolic acid (3), euscaphic acid (4), arjunic acid (5), tomentic acid (6), 3β-E-feruloyl corosolic acid (7), 1β-hydroxyeuscaphic acid (8), myrianthic acid (9), cecropiacic acid (10), and ilexoside B (11). Among them, compounds 3, 6-8, 10 and 11 were obtained from this plant for the first time, and compounds 7 and 10 were obtained from this genus for the first time.

8.Effect of oxidative stress in development of acute high altitude response during the process of strong physical work at high altitude.

OBJECTIVETo assess the effect of oxidative stress in development of acute high altitude response (AHAR) during the process of strong physical work at high altitude and its change after return to lower altitude.
METHODSNinety-six officers and soldiers of rapid entering into high altitude (3 700 m) with strong physical work were analyzed, all subjects were male, aged 18-35 years. According to the symptomatic scores of AHAR were divided into 3 groups: severe AHAR (group A, n = 24), mild AHAR (group B, n = 47) and without AHAR (group C, n = 25). Levels in serum 8-iso prostaglandinF2alpha(8-iso-PGF2alpha), superoxide dismutase (SOD) and malonaldehyde (MDA) were measured at higher altitude stayed 50 d and after return to lower altitude (1 500 m) 12 h and 15 d, and 50 healthy volunteers (group D) at 1 500 m altitude served as controll.
RESULTSLevels of serum 8-iso-PGF2alpha and MDA [(9.53 +/- 0.47) microg/L, (8.91 +/- 0.39) micromol/L] were significantly higher in group A than those in group B [(8.34 +/- 0.42) microg/L, (7.31 +/- 0.32) micromol/L] , group C [(7.02 +/- 0.48) microg/L, (6.41 +/- 0.23) micromol/L] and group D [(5.13 +/- 0.56) microg/L, (5.48 +/- 0.33) micromol/L], (all P < 0.01), and serum SOD [(52.08 +/- 3.44) micro/ml] was significantly lower in group A than that in group B [62.27 +/- 2.54) micro/ml], group C [(71.99 +/- 3.35) micro/ml] and group D [(80.78 +/- 3.44) micro/ ml] (all P < 0.01), there were significant differences between group B and C, C and D (all P < 0.01). At altitude 3 700 m 50 d, AHAR scores was positively correlated with serum 8-iso-PGF2alpha and MDA (all P < 0.01), negatively correlated with SOD (P < 0.01). Serum 8-iso-PGF2alpha and MDA were negatively correlated with SOD (all P < 0.01). Levels of serum 8-iso-PGF2alpha and MDA were significantly higher at altitude of 3 700 m 50 d than those at altitude of 1 500 m 12 h,15 d in group D (all P < 0.01), and serum SOD was significantly lower than that at 1 500 m 12 h,15 d in group D (all P < 0.01), there were significantly difference between at 1 500 m 12 h and 15 d (all P < 0.01), there were no difference between at 15 d in group D (all P > 0.05).
CONCLUSIONThe more serious of oxidative stress and oxidative/antioxidative imbalance, the more serious of AHAR, oxidative stress and oxidative/antioxidative imbalance may be involved in the development of AHAR. The changes were obviously improved after return to lower altitude 12 h, and recovered to normal after 15 d.

9.Prevalence and clinical characteristics of orthostatic hypotension in the elderly and retired population

objective To determine the prevalence and clinical characteristics of orthostatic hypotension(OH)in the elderly and retired population.Methods A total of 1174 elderly and retired people underwent health screening physical examination in Guangzhou military region were included.The orthostatie blood pressure and heart rate were measured in supine position after resting for more than 5 minutes and at 0 and 2 rain after standing.Subjects were divided into OH positive group and OH negative group.Orthostatic hypotension was defined as 20 mm Hg(1 mm Hg=0.133 kPa)or greater decrease in SBP and/or 10 mm Hg or greater decrease in DBP after standing.Results The prevalence of OH in this cohort was 25.6%at either 0 or 2 min after standing(21.6%or 20.7%respectively).Incidence of hypertension,myocardial infarction(MI),heart failure(HF),ischemic stroke and diabetes was significantly higher in OH positive group than in OH negative group(all P<0.05),however,antihypertensive medication was similar between the two groups.Conclusions Orthostatic hypotension is comnlon in the elderly and retired population and is associated with increased risk of hypertension,diabetes mellitus and cardiovascular disease.
10.Application and nursing of right ventricular outflow tract high septum pacing

Objective To explore the feasibility and nursing strategy of active-fixation lead in patients with right ventricular outflow tract high septum ( RVOTHS)pacing. Methods Forty patients were divided into two groups randomly and use VVI pacing mode. One group underwent the right ventricular outflow tract high septum(RVOTHS) with the active fixation lead. The other group underwent the right ventricular apex (RVA)pacing with the passive-fixation lead. The parameters and complication after operation of two groups were recorded and compared accordingly. Results Operations went smoothly with few complications in two groups.The active-fixation lead group had one wire dislocation. while the passive-fixation lead group also had one. There was no serious complication during all operations. The exposure time during operation for RVOTHS prolonged obviously (t=4.036,P<0.01). The comparative difference of ventricular spacing threshold , feeling threshold and electrode impedance between the two groups during operation had no statistic meaning (P>0.05), the pacing electrocardiogram QRS wave width of RVOTHS team was more narrow than that of RVA team, but the difference has no statistical meaning (t = 1. 613, P > 0. 05 ). Conclusions It is safe and feasible to pace in RVOTHS with active electrode. The complication after operation can be well decreased by nursing care and individual nursing.
