1.A novel HLA-derived peptide induces the immune tolerance of rats renal allografts

Objective To observe the immunosuppression function of a novel HLA-derived peptide, RDP1258, after rat renal transplantation in vivo.Methods Peptides were synthesized by Fmoc chemistry method. The rat renal heterotopic transplantation model (n=32) was established. The allograft rats were divided into 4 group: group A (n=8) receiving subtherapeutic CsA+RDP1258 peptide; group B (n=8) receiving RDP1258 alone; group C (n=8) receiving subtherapeutic CsA alone; and group D (n=8) receiving no immunosuppression. The renal allograft survival days of receptor rats were observed. Serum creatinine was measured and color Doppler flow imaging was checked. Renal tissues were harvested for light microscopic examination. The immunologic tolerance status of rat renal allografts was detected by MLR of 60th day receptor rat spleen cells cultured with the donor spleen cells and the third party.Results The allograft survival time in groups A, B, C and D were ( 63.4?30.6)days, (18.3?7.3)days, (16.9?6.4)days and (9.4?2.6)days, with the difference being significant between group A and other three groups. The results of MLR revealed that a hyporesponsiveness to donor occurred while the normal reaction to the third parity was retained.Conclusion A novel HLA derived peptide therapy combined with a sub-therapeutic dose of CsA significantly prolonged allograft survival days, and the mechanism might be that RDP1258 induced a donor-specific immunologic tolerance status in the recipients.
2.Color Doppler ultrasonography of tranient synovitis of the hip in children

Objective To investigate the value of color Doppler ultrasonography in early diagnosis and subsequent follow-up of the transient synovitis of the hip in children. Methods Color Doppler ultrasonography was performed on 80 children with transient synovitis of the single hip, and the characteristics of the blood signal changes in the synovial membrane of the hip joint were observed before treatment, 4 days and 8 days after treatment, while the normal hip joints were examined as the controls. Results Abnormalities in the injured side of the hip joints were displayed in all 80 children. Obviously increased blood signal of the injured side was detected with injured before treatment, but gradually decreased 4 days after treatment, and became similar to the healthy side after 8 days. Conclusion CDFI has an important role in monitoring changes of blood supply, and affords clinical application value in the therapeutic effect observation in transient synovitis of the hip in children.
3.Effects of neonatal repeated intermittent inhalation of sevoflurane on learning-memory and Tau protein, p-Tau protein in juvenile and adult rats

Objective Learning and memory function is the form of brain higher nervous activity .Hippocampus is the main parts responsible for learning and memory function .Once damaged , it will seriously affect the quality of life in patients .The purpose of this paper was to observe the effects of neonatal repeated intermittent sevoflurane inhalation on learning-memory function and Tau protein , p-Tau protein in brain hippocampus in juvenile and adult rats . Methods Twenty-four healthy SD rats ( n=24 ) were randomly di-vided into juvenile sevoflurane inhalation group ( n=6) , juvenile con-trol group ( n=6);adult sevoflurane inhalation group ( n=6) , and adult control group ( n=6) .Rats in juvenile sevoflurane inhalation group and adult sevoflurane inhalation group inhaled 2.6%sevoflurane at the postnatal 7th day, 14th day, 21th day ( P7,P14,P21) for 2 hours.Rats in juvenile control group and adult control group inhaled the carrier gas (1L/min Air+1L/min O2) at the same time for 2 hours.During P31~37, Morris water maze test was conducted in juvenile sevoflurane inhalation group and juvenile control group to detect the behavior.During P91~97, Morris water maze test was conducted in adult sevoflurane inhalation group and adult control group to detect the behavior .Then hippocampi were taken out to detect the expression levels of Tau protein and p -Tau protein. Results ( 1) Comparison of escape latency at the same time ① Juvenile period: no statistical difference between sevoflurane inhalation group (52.04±41.90,29.77±14.23, 19.87±5.71,22.74±13.73,21.91±9.07) and control group (47.82±8.06,25.26±12.53,23.79±9.49, 20.00±10.10, 14.03±7.55) had (P>0.05).② Adult period: no statistical difference between sevoflurane inhalation group (42.00± 14.12, 26.87±16.93, 19.80±13.76, 15.06±8.45, 8.66±4.82) and control group (41.97±25.66,22.88±10.04,15.88±5.20,9.26± 3.98,11.33±6.05 (P>0.05).(2) Comparison of spatial probe test results:no statistical difference in the swimming times from original area,swimmingresidencetime,swimmingdistanceandspeedbetweengroups(P>0.05).(3)Tauproteinexpressiondetection ①Juvenile period:In the hippocampal CA1 region, CA3 region, DG region, the expression level of Tau in the sevoflurane inhalation group (0.237±0.015, 0.324±0.024,0.226±0.019) was higher than the control group (0.185±0.024,0.232±0.040, 0.184±0.018) (P>0.01).②Adult peroid:no statistically significant difference between sevoflurane inhalation group and control group (P>0.05). (4)p-Tau(Ser396)proteinexpressiondetection ①Juvenileperiod:nostatisticallysignificantdifferencebetweensevofluraneinhala-tion group and control group ( P>0.05) .②Adult period:The expression level in the hippocampal CA3 region of sevoflurane inhalation group (0.170±0.005) was higher than control group (0.158±0.011) (P<0.05), but in the CA1 and DG regions there was no statisti-cally significant difference (P>0.05). Conclusion Neonatal repeated intermittent sevoflurane inhalation has done no harm to learn-ing and memory function of juvenile and adult rats , however , it can result in the significant increase of hippocampal Tau protein expres-sion level in juvenile rats and the increase of hippocampal p-Tau protein expression level in adult rats .

5.Comparison of costs of combined intravenous and inhalation general anesthesia and total intravenous anesthesia

ObjectiveTo compare costs of combined intravenous and inhalation anesthesia (CIIA) and total intravenous anesthesia (TIA) when lower limbs orthopedics (LLO) performed on cerebral palsy children (CPC).MethodsTo analyze data of 49 CPC who received LLO under CIIA and TIA,and compare costs of two anesthetic styles.ResultsTIA had a significantly lower cost than CIIA (P<0.001),and showed a lower incidence of post-operation nausea and vomiting.ConclusionTIA can decrease cost of anesthesia and post-operation nursing compared with CIIA.
6.Improvement of the Synthesis Technology of Sorafenib Tosylate

OBJECTIVE:To improve the synthesis technology of sorafenib tosylate (Ⅰ). METHODS:The intermediate N-methyl-(4-chlorpyridin-2-yl)carboxamide(Ⅳ)was obtained by chlorination and amidation with 2-picolinic acid as the starting com-pound. Meanwhile,N-[4-chloro-3-(triflouromethyl)phenyl]-N′-(4-hydroxyphenyl)urea (Ⅶ) was prepared from aminophenol and 4-chloro-3-trifluoromethyl phenyl isocyanate,which was obtained from 4-chloro-3-(trifluoromethyl)aniline(Ⅴ)by reaction with initiator triethylamine. Sorafenib tosylate was synthesized from Ⅳ and Ⅶ with potassium tert-butoxide by condensation and salt for-mation. The target compound was characterized by 1H-NMR. RESULTS:The target compound was confirmed as Ⅰ,with the over-all yield of 74% [based on 4-chloro-3-(trifluoromethylanilin)]. The purification of chromatogram was 94%. Optimized technology improves yield and simplifies multi-step intermediate decompression,distillation and purification process;the application of initia-tor shortens the duration of reaction. CONCLUSIONS:Ⅰ is prepared successfully,and raw material can be obtained easily and is easy to operate with high yield.
7.Effects of chloride channel blockers on the contractile and relaxative responses in rat aorta rings

AIM To study the effects of DIDS and furosemide on the contraction induced by phenylephrine and the endothelium-dependent relexation induced by ATP. METHODS Measurement of isometric force of rat thoracic aortae rings with and without endothelium. RESULTS DIDS(1~300 ?mol?L -1 ) and furosemide(10~320 ?mol?L -1 ) inhibited the contractile response induced by phenylephrine in a concentration-dependent manner. The inhibitory rate was different between aortic ring endothelium-intact and that endothelium-denuded. The values of IC_ 50 for DIDS were (12.0?8.0) ?mol?L -1 and (28.3?7.3) ?mol?L -1 respectively,while the values of IC_ 50 for furosemide were (17.9?6.6) ?mol?L -1 and (41.0?15.6) ?mol?L -1 respectively. DIDS(10 ?mol?L -1 ) did not chang ATP-induced vasodilative effect at the concentrations of 10 ?mol?L -1 and 100 ?mol?L -1 , but enhanced the relaxation induced by 1 mmol?L -1 ATP. Aim To study the changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan.Methods Wistar rats were randomly divided into 5 groups as follows:1% chitosan(W/V)group,2% chitosan(W/V)group,4% chitosan(W/V)group,bFGF(basic fibroblast growth factor) group and the control group.Rats were made for Ⅲ skin burns.The wound healing time was recorded,and the wound healing rate was calculated.Then the cell cycle and apoptotic dermal fibroblasts were determined and the amount of Hydroxyproline(HOP) in the skin tissue was analyzed.Results The wound healing rate of 4% chitosan(W/V) group was higher and the wound healing time of 4% chitosan(W/V) group was shorter than that of the control group.On the 7th,14th day post-injury,the content of protein of 4% chitosan(W/V) group was higher than that of the control group.The content of HOP of 2% and 4% chitosan(W/V) group was highest on the 7th day post-injury. Compared with that in control group,the percentage of cells of S stage in 4% chitosan(W/V) group was aboundant,and was reduced in apoptotic dermal fibroblasts.Conclusion The changes of cellular biological behaviors might be one of the mechanisms of that Chitosan could promote the wound healing of Ⅲ skin burns in rats significantly.
9.Simultaneous Determination of Two Components in Furazolidone, Indometacin and Cuscohygrinolis α-Acetylbenzoacetate Suppositories by HPLC with Dual Wavelength Detection

Objective:To establish an analytical method for the determination of furazolidone and indometacin in furazolidone, in-dometacin and cuscohygrinolis α-acetylbenzoacetate suppositories by HPLC. Methods: The separation was performed on a ZORBAX Extend-C18 (250 mm × 4. 6 mm,5μm) column. The mobile phase was acetonitrile and 0. 035 mol·L-1 potassium phosphate monobas-ic aqueous solution (adjusting pH to 3. 0 with acetic acid) with gradient elution. The flow rate was 1. 0 ml·min-1, and the detection wavelengths were set at 364 nm and 318 nm. The column temperature was 30℃ and the injection volume was 20 μl. Results:Under the selected chromatographic conditions, the two components showed good linearity within the range of 0.005-0.05 mg·ml-1(r=0. 9999). The limit of detection was 20 ng·ml-1 and 26 ng·ml-1, respectively. The limit of quantitation was 70 ng·ml-1 and 90 ng·ml-1, respectively. The average recovery was 99. 4%(RSD=0. 6%, n=9)and 99. 4%(RSD=0. 3%,n=9),respectively. Conclusion:The method is simple, rapid and specific, and the results are accurate and reliable. The method can be used for the de-termination of the two components in furazolidone, indometacin and cuscohygrinolis α-acetylbenzoacetate suppositories.
10.A survival analysis of primary duodenal carcinoma after radical resection

Objective To investigate survival for primary duodenal carcinoma patients after radical resection and identify risk factors associated with overall survival.Methods Data of 51 patients with primary duodenal carcinomas who underwent radical resection between December 2003 and December 2012 at Beijing Cancer Hospital were included.Survival analysis was drawn by Kaplan-Meier method,univariate and multivariate analyses were performed to identify variables associated with survival after resection by COX regression model.Results The median overall survival time was 67 months,and the median disease-free time was 40 months,1-,2-and 3-year overall survival rates were 88%,84% and 67%,respectively.Multivariate analysis revealed that regional lymph-node positive (P =0.032) and CA199 > 37 U/ml (P =0.037) were independent risk factors of patients' overall survival.Conclusions Radical resection improves survival for primary duodenal carcinoma patients.Regional lymph-node positive and CA199 >37 U/ml were the most important risk factors of patients'overall survival.
