Objective To investigate the relationship of fasting serum FFA composition with central obesity, hypertension, insulin resistance, levels of serum zinc and age, gender-related differences. Methods Serum FFA were determined by high performance liquid chromatogram in 232 commuity|based subjects aged between 35 and 55 years: 109 hypertensives, 123 normotensives. 2.Isolation of Strain LW-3 Capable of Degrading Chlorimuron-ethyl and Its Biological Characteristics

A bacterium(designated as LW-3), capable of degrading Chlorimuron-ethyl, was isolated from the long-term contaminated soil by Chlorimuron-ethyl. Based on physiological and biochemical analyses and sequences comparison of 16S rDNA, strain LW-3 was identified as Pseudomonas sp. LW-3 could use Chlorimuron-ethyl as sole nitrogen source for growth. The optimum pH and temperature for degradation of Chlorimuron-ethyl were pH 6.5~7.2 and 30?C~35?C, at same temperature the pH change to the Chlorimuron -ethyl degrading influence is large. When the pH and temperature were pH 6.5 and 32?C, 50 mg/L Chlori- muron-ethyl could be degraded to 70%~80% level within 7 days.
3.Investigation on economic status of psychiatric patients and their families in Shaoxing area in Zhejiang Province

ObjectiveTo investigate the economic status of psychiatric patients and their families in Shaoxing area in Zhejiang Province.MethodsA self-edited questionnaire was used to survey the annual income and expenses for medical treatment among psychiatric patients and their families.ResultsThe average annual income of per person in psychiatric family was (3823.07±3354.99) Yuan while average annual income of per psychiatric patient was (1918.20±5188.42) Yuan, which were only 62.3% and 31.2% of the rural controllable annual income, 20.9% and 14.2% of urban controllable annual income in Shaoxing area. In the meantime, 30.43% psychiatric patients did not have any incomes and 47.43% were below 500 Yuan per year. Engel coefficient was (59.65±24.36). The average expense for medical treatment was 1052.54 Yuan per year, which was 54.9% of annual income of the psychiatric patients. In some illness, this rate was as high as 90% or above.ConclusionThe incomes of psychiatric family members and psychiatric patients themselves are much lower than the local average level, while they would be spent more than a half of the income on their medical treatment. Psychiatric patients as well as their family members are the extremely low income and weak social class and need help by the government and society.
5.Analysis of pulsed-field gel electrophoresis molecular subtyping of Shigella strains in Shenzhen.

OBJECTIVETo analyze the genetic relations of Shigella isolated from Shenzhen in 2001-2006 and develop primary molecular subtyping surveillance network of Shigella.
METHODSChromosomal DNAs from 55 isolated in agarose were digested with the restriction enzyme Xba I, and then were analyzed by pulsed-field gel electrophoresis. Pulsed-field gel electrophoresis (PFGE) patterns were clustered using BioNumerics software.
RESULTSAll 41 distinctive PFGE patterns were identified among 55 strains. 32 strains belonged to one cluster. Differences were observed in other strains.
CONCLUSIONBoth genetic-related clones and non-related clones of Shigella existed in Shenzhen. The development of PFGE molecular subtyping surveillance network would contribute to the active surveillance, outbreak investigation and source tracking for Shigellosis.

6.Effects of FLAG protocol in treatment of the first time induced non-remission acute myeloid leukemia.

In order to evaluate the efficacy of FLAG protocol (fludarabine, cytosine arabinoside and granulocyte colony-stimulating factor) in treatment of the first time induced non-remission acute myeloid leukemia (AML), 19 patients with first time induced non-remission acute myeloid leukemia were treated with FLAG protocol. The results showed that out of the 19 patients 13 patients obtained complete remission (CR) and the CR rate was 68.4%, 2 patients obtained partial remission (PR) and the PR rate was 10.5%, the overall remission rate was 78.9%. Among the patients in CR 5 patients had been received allogeneic stem cell transplantation, 3 patients from them survived without disease, including 1 patient has survived 26 months and still remains in CR. Main toxicities of this protocol were gastrointestinal side effects, myelosuppression and neutropenia, slight abnormality of liver function and so on. It is concluded that the FLAG protocol should be employed for the the first time induced non-remission patients as early as possible, and provides conditions for the hematopoietic stem cell transplantation.

7.Clinical pathological features of the 8p11 myeloproliferative syndrome.

This study was aimed to investigate the clinico-pathological features, diagnosis and treatment of the 8p11 (eight p11) myeloproliferative syndrome (EMS). Morphological changes of cells were evaluated by bone marrow smear and biopsy. The cell immunophenotypes were analysed by flow cytometry. Karyotypes were determined by conventional cytogenetic method, and bcr/abl fusion gene was detected by reverse transcription-polymerase chain reaction (RT-PCR). The results indicated that EMS was a relatively rare disease characterized by the occurrence of a bcr/abl-negative myeloproliferative disorder and a T-cell lymphoblastic lymphoma (T-LBL). Bone marrow examination showed myeloid hyperplasia or myeloproliferative neoplasm, often accompanied by eosinophilia. Flow cytometric immunophenotyping showed increased myelomonoblasts; cytogenetic analysis showed a translocation at the 8p11 locus; RT-PCR demonstrated non bcr/abl fusion gene. At the molecular level, all cases carried a chromosomal abnormality involving the fibroblast growth factor receptor 1 (FGFR1) at chromosome 8p11. Up to now, 11 partner genes have been identified and associated with FGFR1 rearrangements. The most common partner is ZNF198 on chromosome 13q11-12. Majority of patients terminate in acute myeloid leukemia which is resistant to conventional chemotherapy. Currently, the only curative option appears to be allogeneic hematopoietic stem cell transplantation. In conclusion, EMS is myeloid and lymphoid neoplasm, associates with FGFR1 rearrangements. It is usually misdiagnosed as T-LBL, atypical chronic myeloid leukemia (aCML) or chronic myelogenous-monocytic leukemia (CMML). Timely cytogenetic and molecular biological examination is vital in order to avoid misdiagnosis and mistreatment.

8.Preliminary study on difference of Id4 gene methylation in various types of myelodysplastic syndromes.

The aim of this study was to investigate the difference of Id4 gene promoter methylation in patients with different subtypes of myelodysplastic syndromes (MDS). By using MS-PCR method, the methylation status of Id4 gene was detected in 50 patients with different subtypes of MDS. Id4 methylation was also detected in bone marrow samples from patients with iron deficiency anemia which served as control. The results showed that Id4 gene was unmethylated in all of bone marrow samples from controls. In various subtypes of MDS patients, the rate of Id4 gene methylation was different. No Id4 methylation was found in 6 cases of RA, 2 cases of RARS and 4 cases of MDS-U. Id4 methylations was found in 2 out of 18 patients with RCMD. Id4 methylation in 3 out of 12 patients with RAEBI and other 3 out 8 patients with RAEBII were found. In groups with blast ratio lower or higher than 5%, the incidence of Id4 gene methylation were 6.7% and 30% respectively, so that there was significant difference. In tentative conclusion, Id4 gene methylation possibly is found in MDS patients with higher ratio of blast cells.

9.Significance of id4 promoter methylation in monitoring AML patients with completely remission.

The study was purposed to investigate the significance of id4 gene promoter methylation in monitoring AML patients with complete remission (CR). Methylation specific-PCR (MS-PCR) were used to detect the status of promoter methylation of id4 gene in bone marrow samples from AML patients with CR who had accepted induction with DA or IA and 4 to 5 consolidation chemotherapy with Ara-C. The results showed that in the all 32 patients, 15 were found to show id4 promoter methylation and 7 out of the 15 were found relapsed or tendency to relapse in the following-up period. While all the 17 patients with id4 unmethylation were still in their CR status in the same period. In conclusion, id4 promoter methylation might be a predictor for relapse of AML patients with CR in certain degree.

10.Significance of id4 gene promoter methylation detection in lymphoma.

The aim of this study was to evaluate the significance of id4 gene promotor methylation detection in NHL patients. MS-PCR method was used to detect the status of id4 gene methylation in health donors and newly diagnosed NHL patients. The results indicated that the id4 gene was unmethylated in bone marrow samples from health donors. Among 18 newly diagnosed NHL patients, including one NHL patient with bone marrow cells involved, 4 patients were found in id4 gene methylation by MS-PCR. The 14 patients with id4 gene unmethylation were in their stable status and no bone marrow involvement were found by bone marrow biopsy during the 8-month follow-up. During the follow-up, the patient with both bone marrow involvement and id4 gene methylation turned to leukemia, in 2 out of the 3 patients with id4 gene methylation but without bone marrow involvement at diagnosis, the bone marrow involvement was found at last. It is concluded that the id4 gene methylation may be an indicator for MRD in NHL patients without bone marrow involvement.
