1.Associations between partner-seeking activities through internet and sexual risk behaviours related to HIV/AIDS among men who have sex with men in China

Objective To explore the associations between partner-seeking activities through internet and HIV/AIDS related risk behaviours among men who have sex with men(MSM).Methods Target sampling.anonymous questionnaires were adopted and analyzed with SPSS 11.0.Results Data from 45.6% of the 2178 valid questionnaires showed that most partner-seeking activities was through internet.73.1% of them landed on gay website.in which 83.0% looked for sexual partners and 91.7% of them had intercourse with other men met through internet when compared with MSM who did not seek sex activities through internet.The internet group obviously had fewer numbers of partners(5.73 vs.10.3),oral sex partners(5.13 vs.8.71),passive anus sex partners(4.67 vs.8.26)and active anus sex partners (3.51 vs.6.06)in the past 6 months(P<0.01 respectively)and obviously had fewer group sex(16.7% vs.20.5%)activities in the past one year but had more activities on oral sex(94.7% vs.89.6%),anus sex(95.1% vs.86.5%),regular sexual partner(52.6% vs.46.7%)in the past 6 months(83.4% vs.75.5%)(P<0.01 respectively)and more likely to have finger intercourse(52.2% vs.38.5%),hemorrhage sexual intercourse(36.4% vs.21.9%,OR=2.04),non-local sexual intercourse(28.1% vs.22.7%)(P<0.01 respectively).Conclusion Seeking sexual partners through internet had positive and double effects on MSM's risk behaviours.suggesting that it is of urgent need to take the advange of internet and gay website to disseminate HIV/AIDS prevention message.
2.The impact of childhood sexual abuse on the development of AIDS related high risk behaviors and psychological appearances among men who have sex with men

Objective To study the AIDS related high risk behaviors and psychological appearances among men who have sex with men (MSM) who ever experienced childhood sexual abuse (CSA).Methods Target sampling for a cross-sectional study was developed and valid anonymous questionnaires were adopted to compare the differences of high risk behaviors related to AIDS and psychological appearances betwen those with or without CSA experiences among 2147 MSM from nine cities.Results Compared to corresponding ones without CSA experience,CSA group had a significant larger numbers in the following events: total sexual partners,anal sex episodes with same sex,female sexual partners and anal sex in the previous six months,with the figures of median as 20.0,10.0,3.0,3.0 respectively.In the previous year,30.8% of them had ever participated in 'group sex',19.2% ever exchanged money for sex,36.7% bled while having sexual intercourse,37.3% had sex with male partners away from his own region.All the above said figures were higher than non-CSA group,with significant differences.It also appeared that CSA experience had an impact on significant lower rate of condom use (67.3%) in the last anal sex.Those with CSA experience had more psychological problems which appeared as: 75.6% considered they would suffer from serious discrimination if their sexual orientation ever disclosed,34.7% had a strong intention of suicide and 24.3% ever having had suicidal attempts.The differences of the two groups showed statistical significance.Condusion CSA experience not only increased the number of AIDS related high risk behaviors in adulthood,but also had negative impact on their psychological appearances.It is of urgent need to carry out psychological intervention approaches to target on MSM with CSA experiences while childhood sexual education and rights assurance towards juvenile population should also not be neglected.
3.Study on pan-resistant Klebsiella pneumoniae harboring blaKPC-2 type carbapenemase gene from a hospital outbreak in Huzhou, Zhejiang

Objective To investigate the status of genotype of the KPC(Klebsiella pneumoniae carbapenemase)-encoding genes in Pan-resistant K. Pneumoniae, isolated from the 98th Hospital of People' s Liberation Army, Huzhou district, Zhejiang province, China. Methods 19 strains of Pan-resistant K. Pneumoniae were isolated from the inpatients between November, 2008 and July,2009. Phenotypic confirmatory test for suspected carbapenemases production were carried out by Modified Hodge test. Carbapenemase gene of blaKPC was analyzed by PCR and verified by DNA sequencing. Results In 19 strains of K. Pneumoniae, the positive rates of Modified Hodge test and gene of blaKPC were both 100.0%. These genes all belonged to blaKPC-2 subtype confirmed by nucleotide sequence analysis. Among them, the blaKPC-2 gene sequence of the HZ001 strain (its original serial number was HZ9871 ) had been registered in GenBank (GenBank Accession Number: GU086225).Conclusion All of the Pan-resistant K. Pneumoniae isolated from the inpatients harbored blaKPC-2 type carbapenemases gene and causing an outbreak in a hospital. Carbapenemases that producing type KPC-2 might be the major reason which causing the resistance to Carbapenems antibiotics.
5.Expression of matrix metalloproteinase-2 and insulin-like growth factor-1 in gastric carcinoma and their clinicopathological significance.

OBJECTIVETo investigate the expression of matrix metalloproteinase-2 (MMP-2) and insulin-like growth factor-1 (IGF-1) in gastric carcinoma and their clinicopathological significance.
METHODSExpressions of MMP-2 and IGF-1 were examined by using immunohistochemical SP staining and cross-compared with clinicopathological features of gastric carcinoma.
RESULTSHigh expression of MMP-2 and IGF-1 were observed in 70.4% (307/436) and 49.5% (216/436) of gastric carcinoma tissues respectively, significantly higher than those in non-tumor gastric mucosa (3.3% and 5.4%, respectively; all P < 0.05). The high expression rate of MMP-2 and IGF-1 were significantly associated with the patient age, tumor size, tumor location, Lauren classification, TNM staging, depth of tumor infiltration, presence of vessel invasion, lymph node and distant metastasis (all P < 0.05). In addition, the expression of MMP-2 was positively linked with the expression level of IGF-1 (P < 0.05). Univariate analysis showed that high expression of MMP-2, was significantly associated with poor prognosis of tumor of TNM stage I and II (all P < 0.05), high expression of IGF-1 was significantly correlated with poor prognosis of patients with TNM stage I, II and III tumor (all P < 0.05). Cox multivariate analysis indicated that the high expressions of MMP-2 and IGF-1 could be independent prognostic indices for gastric carcinoma.
CONCLUSIONSHigh expression of MMP-2 and IGF-1 proteins are significantly correlated with the invasion and metastasis of gastric carcinoma, it is helpful to simultaneously detect the expressions of MMP-2 and IGF-1 proteins in predicting prognosis of patients with gastric carcinoma.

6.Study on the association of partner-seeking and high risk behaviors related to AIDS among men who have sex with men in the different sites

Objective To explore the situation of AIDS-related high risk behaviors among men who have sex with men (MSM) ever experienced partner-seeking in different sites. Methods Target sampling for cross-sectional study ( "snowball" ) and valid anonymous questionnaires were adopted to compare the differences of high risk behaviors related to AIDS. 1834 MSM were studied in nine cities who sought sex partners through internet, bars or bath ponds. Results Compared to the other corresponding groups, those MSM from the bath pond group had a significant larger number of total sexual partners, chances of having anal and oral sex with same sex, and ones of anal sex in the previous six months with the median number as 40, 16, 20 and 4 respectively. Interact group had a higher rate of bleeding (36.4%) when having sexual intercourse in the previous year and a lower proportion of 8.7% exchanged sex for money, and 14.9% and 13.3% in both bar and bath pond groups respectively. Bar group had significant lower rates of condom use with 79.6% while having sex with men and 45.3% with women, but higher rates(28.9% ) ever participated in group sex in the last year and 4.2% with H1V infection. Conclusion MSM had high risk behaviors related to AIDS appeared in different sites when seeking partners. Intervention strategy should be taken, according to the characteristics of different subpopulations, especially on the bath pond group.
7.Differential susceptibility of naïve versus cloned CD4+ T cells to antigen-specific and MHC-restricted anergy induction.

T cell anergy has been successfully induced under different conditions in cloned CD4(+) T cells, but induction of T cell anergy in vivo has been difficult and controversial. Due to the low frequency of naturally occurring T cell population with specificity to a defined antigen, it is very difficult to study anergy of naïve T cells without prior in vivo priming which complicates the interpretation of experimental data. To solve this problem, we adopted the HNT-TCR transgenic mice which have homogeneous antigen specific CD4(+) T cell population. In this study, we generated an influenza virus hemagglutinin (HA) peptide-specific CD4(+) T cell clone from the HNT-TCR transgenic mice and induced anergy using APCs which were treated with the crosslinker, ECDI (1-ethyl-3-3(3-dimethylaminopropyl) carbodiimide). The proliferative response of the cloned or freshly purified naïve CD4(+) transgenic T cells after treatment with ECDI-treated APCs and the HA peptide antigen was monitored as the index of anergy induction. The results showed that anergy was successfully induced in the cloned HNT-TCR transgenic CD4(+) T cells. It was determined that the induced anergy was antigen- and MHC-specific. By contrast, anergy was not observed in freshly purified naïve CD4(+) transgenic T cells under the same conditions. The results suggest that naïve CD4(+) T cells may have different anergy inducing requirements, or that cloned CD4(+) T cells may have certain priming or in vitro cloning artifact which makes them more susceptible to anergy induction. We propose that induction of T cell anergy may depend on the T cell growth, activation and differentiation state or cloning conditions. The results from the present study may have important implications for the study of the mechanism(s) underlying T cell anergy induction in vivo and for applications of immune tolerance based therapy.

8.Different antihypertensive drugs preventing early hematoma expansion in patients with hypertensive cerebral hemorrhage

Objective To determine whether different antihypertensive drugs may have different effects on the hematoma and prognosis of intracerebral hemorrhage (ICH) patients to provide reference for drug choices in the clinical blood pressure control of patients with ICH.Methods Ninety three ICH patients,admitted to our hospital from January 2010 to January 2012,were chosen in our study and divided into nimodipine treatment group,urapidil treatment group and control group.Hematoma volumes of the patients were recorded at admission,24 and 72 hours after admission,respectively.The neurological disability grades of the patients were also recorded at discharge.Results The hematoma expanded in all of the three groups at 24 and 72 hours of admission,but not enjoying difference as compared with that before admission (P>0.05); the hematoma expansion in the nimodipine group was obviously restricted as compared with that in the urapidil group and control group at 24 and 72 hours of admission (P=0.026,P=0.019; P=0.016,P=0.022).Significant differences on the prognosis in the three groups were noted (P<0.05); the function recovery in the nimodipine group was significantly better as compared with that in the urapidil group and control group (P<0.05).Conclusion Nimodipine have better effect on the early hematoma expansion and function recovery of ICH patients as compared with urapidil; nimodipine should be first chosen in clinic.
9.Downregulation of Notch-regulated Ankyrin Repeat Protein Exerts Antitumor Activities against Growth of Thyroid Cancer.

BACKGROUNDThe Notch-regulated ankyrin repeat protein (NRARP) is recently found to promote proliferation of breast cancer cells. The role of NRARP in carcinogenesis deserves extensive investigations. This study attempted to investigate the expression of NRARP in thyroid cancer tissues and assess the influence of NRARP on cell proliferation, apoptosis, cell cycle, and invasion in thyroid cancer.
METHODSThirty-four cases with thyroid cancer were collected from the Department of General Surgery, Xinhua Hospital, Shanghai Jiao Tong University School of Medicine between 2011 and 2012. Immunohistochemistry was used to detect the level of NRARP in cancer tissues. Lentivirus carrying NRARP-shRNA (Lenti-NRARP-shRNA) was applied to down-regulate NRARP expression. Cell viability was tested after treatment with Lenti-NRARP-shRNA using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Apoptosis and cell cycle distribution were determined by flow cytometry. Cell invasion was tested using Transwell invasion assay. In addition, expressions of several cell cycle-associated and apoptosis-associated proteins were examined using Western blotting after transfection. Student's t-test, one-way analysis of variance (ANOVA), or Kaplan-Meier were used to analyze the differences between two group or three groups.
RESULTSNRARP was highly expressed in thyroid cancer tissues. Lenti-NRARP-shRNA showed significantly inhibitory activities against cell growth at a multiplicity of infection of 10 or higher (P < 0.05). Lenti-NRARP-shRNA-induced G1 arrest (BHT101: 72.57% ± 5.32%; 8305C: 75.45% ± 5.26%) by promoting p21 expression, induced apoptosis by promoting bax expression and suppressing bcl-2 expression, and inhibited cell invasion by suppressing matrix metalloproteinase-9 expression.
CONCLUSIONDownregulation of NRARP expression exerts significant antitumor activities against cell growth and invasion of thyroid cancer, that suggests a potential role of NRARP in thyroid cancer targeted therapy.
