1.Changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan

Aim To study the changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan.Methods Wistar rats were randomly divided into 5 groups as follows:1% chitosan(W/V)group,2% chitosan(W/V)group,4% chitosan(W/V)group,bFGF(basic fibroblast growth factor) group and the control group.Rats were made for Ⅲ skin burns.The wound healing time was recorded,and the wound healing rate was calculated.Then the cell cycle and apoptotic dermal fibroblasts were determined and the amount of Hydroxyproline(HOP) in the skin tissue was analyzed.Results The wound healing rate of 4% chitosan(W/V) group was higher and the wound healing time of 4% chitosan(W/V) group was shorter than that of the control group.On the 7th,14th day post-injury,the content of protein of 4% chitosan(W/V) group was higher than that of the control group.The content of HOP of 2% and 4% chitosan(W/V) group was highest on the 7th day post-injury. Compared with that in control group,the percentage of cells of S stage in 4% chitosan(W/V) group was aboundant,and was reduced in apoptotic dermal fibroblasts.Conclusion The changes of cellular biological behaviors might be one of the mechanisms of that Chitosan could promote the wound healing of Ⅲ skin burns in rats significantly.
2.Apoptosis of macrophage induced by E_2 at high concentration

AIM To study the effect of 17?-estradiol(E 2) on apoptosis of macrophages at certain concentration.METHODS Fluorescent microscopy and scanning eletron microscopy(ESEM) were used to detect apoptosis changes of macrophages induced by E 2 at relatively high concentration and intracellular Ca 2+ and matochontrial transmembrane potential were examined by laser scanning confocal microscopy(LSCM). RESULTS Apoptosis of macrophages was induced by high concentration of E 2 (≥1 ?mol?L -1). Intracellular Ca 2+ elevated and matochontrial transmembrane potential decreased after E 2 treatment. CONCLUSION Apoptosis of macrophages is induced by certain concentration of E 2 (≥1 ?mol?L -1) and this may be related to elevated intracellular Ca 2+ and decreased matochontrial transmembrane potential.
3.Effect of 17?-estradiol on macrophages

AIM:To analyze the effect 17?-estradiol on activated macrophages and possibility of being a target of screening drugs for endometriosis. METHODS: Using scanning electron microscope, the surface of macrophages was observed. Nitric oxide (NO) in supernatant was determined by Griess's reagent. TNF-? was measured by MTT via L929 cells. Using Fluo-3/AM, i was measured by laser scanning confocal microscopy (LSCM). RESULTS: (1) Increased size, extension of more microvilli, expression of retraction fibers and elaboration of membrane ruffles were detected in 17?-estradiol treated macrophages. (2) 17?-estradiol induced NO release from peritoneal macrophages. (3)The more TNF-? was made by macrophages under the induction of 17?-estradiol. (4)Intracellular Ca 2+ elevated 39.8% in peritoneal macrophages after 17?-estradiol (100 nmol/L) treatment. CONCLUSIONS: Macrophages were activated by 17?-estradiol at a certain concentration. Activated macrophages by 17?-estradiol may participate in endometriosis and may be a target of screening drugs for endometriosis.
6.Anti-inflammatory constituents from Inula japonica.

Chemical constituents of Inula japonica were isolated and purified by repeated column chromatographies, over silica gel, and Toyopearl HW-40, and preparative HPLC. On the basis of spectral data analysis, including NMR and MS data, the structures of the isolates were elucidated and their anti-inflammatory activities were assayed. Fifteen compounds were isolated from the ethyl acetate extract of I. japonica, and their structures were elucidated as dihydrosyringenin (1), (3S, 5R, 6S, 7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one (2), (6R, 7E) -9-hydroxy-4,7-megastigmadien-3-one (3), arnidiol (4), taraxasterol acetate (5), 8,9,10-trihydroxythymol (6), taxifolin (7), luteolin (8), napetin (9), eupatin (10), spinacetin (11), quercetin (12), p-hydroxycinnamic acid (13), caffeic acid (14), and caffeoyl acetate (15). Compounds 1, 2, 7, 13 and 15 were isolated from the genus Inula for the first time, and compounds 3, 4, 9-11 and 14 were isolated from this plant for the first time. The anti-inflammatory activity result showed that compounds 3, 6-12 and 14 exhibited inhibition effect against leukotriene C4 (LTC4) synthesis and degranulation definitely in c-Kit Ligand (KL) induced mast cells, and compound 8 and 12 also had the suppression effect against lipopolysacharide(LPS) induced nitric oxide (NO) activity in RAW264.7 macrophages. It is firstly reported that compounds 7 and 9-11 possessed potent inhibition activities against LTC4 generation and degranulation in mast cells.

7.Kinetics of serum albumin in patients with severe sepsis

Objective Albumin is well known to decrease in response to sepsis, However, the.degradation and distribution in patients with severe sepsis to explore the mechanism of hypoalbuminemia in sepsis. Methods 10 volunteers and 10 patients with severe sepsis. 125I labeled albumin was administered intravenously to 10 healthy volunteers and 10 patients with severe sepsis. Each subject had frequent blood samples taken at 0,1,2,4,8,12,24 hours and on day 2, 3, 4, 5, 6, 7, 9, 11, 13, 15, 18, 22, 25 to measure 125I concentration and draw the curve of concentration over time. Plasma was regarded as the central pool and body fluid as side pool, The curve of albumin concentration vs time was expected to follow two compartment model. Results Radioactivity of blood samples was counted and the results were graphically expressed. The half-life time(t1/2), apparent volume of distribution(Vd) and transportation rate(K12) of albumin from the central pool to the side pool were calculated. The half-life time in sepsis was obviously shorter than that in control group (8.2 1.4 vs 12.5 1.7days, P<0.001). The transportation rate in sepsis group was quicklier than that in control group [(4.4±1.9)× 10-2/h vs (2.4±0.6)×10-2/h, P<0.005]. There was no significant difference in apparent volume of distribution between two groups. Conclusions In patients with severe sepsis, the distribution rate of albumin from plasma to body fluid was obviously elevated and the decomposition rate of albumin was markedly increased.
8.Comparative analysis of results about four calculating methods used to determine the obesity in 2825 adults

Objective To explore the difference of four calculating methods used to determine the obesity. Methods Two thousand four hundred and forty six (2446) men and three hundred and seventy nine (379) women were measured height and body mass, Standard body mass, BMI, body fat ratio and obesity index(OI) were calculated with formula. According to the BMI≥ 25 kg/m2 , BMI ≥ 27 kg/m2 and BMI≥28 kg/m2, the obese adults were 1419,680 and 435 respectively;there were 649 adults that their body mass was over 20%standard body mass; there were 639 adults that their body fat ratio was over 25%(male)and 33%(female). Results For obesity determination, the adults who were 20%overweight and over standard body fat ratio were significantly different from those whose BMI were over 25 kg/m2 ( P<0. 05 ) ;Those who were 20%overweight and over standard body fat ratio were not significantly different from those whose BMI were over 27 kg/m2 ( P>0.05 ) ; Those who were 20%overweight and over standard body fat ratio were significantly different from those whose BMI were over 28 kg/m2 ( P<0. 05 ). Conclusion Determining obesity with BMI≥27 kg/m2 is feasible and rational.
9.Clinical observations on patients with surgical treatment after heart valve prosthesis implantation

Objective To evaluate the safety of surgical procedures for patients after heart valve prosthesis implantation.Methods Clinical data of 12 cases with heart valve prosthesis implantation undergone other surgical treatment from November 1996 to December 2005 were retrospectively analyzed.All the cases had routine oral warfarin with prothrombin time (PT) of 20.0—28.3 s averaged 23.5 s, international normalized ratio (INR) for prothrombin of 1.79—2.23 averaged 1.95 and heart functional class Ⅰ—Ⅲ.Among them,appendectomy was performed in three cases with acute appendicitis,reposition and repair in one with inguinal hernia,radical gastrectomy in two with gastric carcinoma,left hemicolectomy in one,cholecystectomy in three,left femoral head replacement in one,and bilateral high ligation and ablation of great saphenous vein in one.Elective surgical operation was performed in seven cases,and emergency operation in five.In those with elective surgery,warfarin was stopped 2—3 days before operation,while 5—10 mg vitamin K_1 was injected intramuscularly 6—8 hours before emergency surgery with preoperative median PT of 15.1 and 15.3 s and median INR of 1.24 and 1.30,respectively.In operation,5—10 mg vitamin K_1 were injected intravenously into the patients by drip depending on their bleeding on the surface of wound.ECG,blood pressure,hemoglobin and oxygen saturation were routinely monitored for all the cases intraoperatively and postoperatively.For the cases with heart function above class Ⅱ,fluid infusion was adjusted based on intubated central venous pressure,and for those with general anesthesia,analyses of blood gases and electrolyte were monitored routinely in operation.Results OPeration time averaged 20—160 rain in all the 12 patients,with blood loss 5—280 ml in average and without complications of massive hemorrhage,thrombosis and heart failure.Conclusions Surgical operation was safe for patients with heart valve prosthesis implantation,if preoperative PT and INR were adjusted to about 15 s and 1.30,respectively by cessation of warfarin or application of vitamin K_1,combined with careful manipulation and strengthened perioperative management.
10.ISOLATION AND IDENTIFICATION OF XJ STRAIN OF CANINE DISTEMPER VIRUS

A canine distemper virus strain was isolated from the lung of dog coming from Aksu in Xing Jiang using lung primary M cell during the CDV molecular epidemiological study. It was demonstrated to be a virulent strain of CDV by a series of systematic identification such as morphology , serology neutralization test, canine infection test, and molecular virology test.
