4.APPLICATION OF DYNA-LOK SYSTEM IN THERAPY OF LUMBAR SPONDYLOLISTHESIS

From July 1998 to May 2000, 31 cases of lumbar spondylolisthesis accompanied with lumbar stenosis were treated with spinal cord decompression, autograft interbody fusion and fixed by Dyna lok system. All the patients were followed up for 6 to 28 months, averaged 12 7 months. 29 of the 31 patients were recovered completely. 2 cases of cauda equina were recovered within 2 months. Lowback pain remained in 1 case of multi segements spondylolisthesis which had been fixed for 3 vertebrae. 1 patient accompanied with cervical spinal stenosis had some residual claudication. In conclusion, spondylolisthesis fixed with the Dyna lok system can achieve a satisfactory result.
6.Gene Coloning,Expression and Enzymatic Assay of Human sPLA2-IIA

Objective: To clone the cDNA of human sPLA2-IIA,construct the engineered Escherischia coli expressing human sPLA2-IIA and identify the expressed human sPLA2-IIA. Methods: Total RNAs were purified from human fetal spleen. The cDNA of human sPLA2-IIA was cloned by RT-PCR and inserted into plasmid pET32a(+) between NcoI and EcoRI sites for expressing the recombinant human sPLA2-IIA in Escherischia coli BL21(DE3). The recombinants were screened by SDS-PAGE. The engineered Escherischia coli expressing trxA-human sPLA2-IIA fusion protein was established. The expressed human sPLA2-IIA exists in the form of inclusion body and accounts for about 25% of the total proteins of Escherischia coli BL21(DE3). Conclusion: the engineered E. coli methods are suitable for preparing plenty of human sPLA2-IIA which has laid base for the large-scale expression,purification and basic studies of human sPLA2-IIA.
7.The effect of non-linear blending function for dual-energy CT on CT image of pulmonary angiography

Objective To evaluate the effect of non-linear blending function for dual-energy CT on image of pulmonary angiogra-phy.Methods 27 patients underwent dual energy CT pulmonary angiography(CTPA).Data obtained with 100 kVp,140kVp,and non-linear blending were divided into group A,B,and C respectively.CT value of emboli were measured.Corresponding signal to noise ratio(SNR )and contrast to noise ratio(CNR)were calculated.One-way Anova analysis and Friedman test were used to ana-lyze statistical significance among these values.Results On quantitative analysis of 27 patients,there was no statistical difference for CT value of CTPA among the three groups(P >0.05).For the noise,CNR and SNR,there were statistically significant among the three groups(P <0.05).Conclusion The non-linear blending function has certain advantages in improved image signal-to-noise ratio and it can be used in CT pulmonary angiography examination for patients suspected pulmonary embolism clinically .

Traditional treatments for malignant tumor are far from meeting the clinical demands.Recently,research on anti-tumor targeted drugs has made a significant breakthrough,which brings new hope for the treatment of malignant tumor.Anti-tumor targe-ted drugs can specifically target malignant tumor and directly in-hibit the growth of tumor cells,showing no toxicity to the normal tissues and organs.Herein we reviewed the research progress of small molecular targeted drugs and antibody targeting new drugs.
9.THE HYPOGLYCEMIC EFFECTS OF SOYBEAN HYPOCOTYL EXTRACT IN DIABETIC RATS AND THEIR MECHANISM

Objective: To study the hypoglycemic effects of soybean isoflavones (SI) and soyasaponins (SS) in diabetes, and their inhibitory activities on alpha-glucosidase (EC 3.2.1.20) and alpha-amylase (EC 3.2.1.1). Methods: GK/Jcl type 2 diabetic rats were given diet containing 20 g/kg of soybean hypocotyl extracts (SHE) for 20 weeks, then the blood glucose and oral glucose tolerance test were observed. SI and SS were isolated by ODS column chromatography and high-performance liquid chromatography (HPLC) from SHE. The inhibitory activities of each component of SI and SS against alpha-glucosidase and alpha-amylase were tested by colorimetric method. Results: SHE decreased blood glucose significantly in type 2 diabetic rats and improved glucose tolerance in both normal and diabetic rats. In alpha-glucosidase inhibitory assay, Group B, Group E and DDMP (2,3-dihydro-2,5-dihydroxy-6-methyl-4H-pyran-4-one) saponins showed potent inhibitory activities with IC50 values of 10-40 靘ol/L. Isoflavone aglycones also showed potent inhibitory activities against alpha-glucosidase with IC50 values of 20-150 mol/L, while isoflavone glycosides showed a little lower potencies. Inhibition of SI and SS on alpha-amylase was low, and their inhibitory activities were about 10%~20% at the concentration of 1g/L. Conclusion: SI and SS may decrease blood glucose and improve oral glucose tolerance in diabetic rats, probably via the inhibitory effects on alpha-glucosidase and alpha-amylase.
