OBJECTIVETo determine the relationship between viral burden in urine and hearing loss in neonates with cytomegalovirus (CMV) infection.
METHODSTwenty-two neonates with CMV infection between April 2006 and January 2010 were enrolled. Their viral burden in urine and hearing loss information were studied. The receiver operating characteristic curve (ROC) was constructed and the cutoff was determined based on their medical information. The hearing levels were evaluated by brain stem auditory evoked potential (BAEP) during the age of 3 to 6 months in 20 patients.
RESULTSThe viral burden in urine in neonates with abnormal BAEP was higher than that in neonates with normal BAEP (5.06 ± 1.50 vs 3.73 ± 0.86, P<0.05). Hearing loss was predicted with a sensitivity of 0.545 and a specificity of 1.0 by using ROC at the cutoff point of 5.1 which were defined after logarithmic conversion at 1.27×10(5) copies/mL of CMV burden in urine. The incidence of hearing loss during the age of 3 to 6 months was strikingly higher in high viral burden group than that in low viral load group (P<0.05).
CONCLUSIONSThe viral burden in urine can predict the possibility of hearing loss in neonates with CMV infection. Hearing loss is likely to be developed when viral burden in urine ≥1.27×10(5) copies/mL in neonates with CMV infection.

4.Analysis of pulsed-field gel electrophoresis molecular subtyping of Shigella strains in Shenzhen.

OBJECTIVETo analyze the genetic relations of Shigella isolated from Shenzhen in 2001-2006 and develop primary molecular subtyping surveillance network of Shigella.
METHODSChromosomal DNAs from 55 isolated in agarose were digested with the restriction enzyme Xba I, and then were analyzed by pulsed-field gel electrophoresis. Pulsed-field gel electrophoresis (PFGE) patterns were clustered using BioNumerics software.
RESULTSAll 41 distinctive PFGE patterns were identified among 55 strains. 32 strains belonged to one cluster. Differences were observed in other strains.
CONCLUSIONBoth genetic-related clones and non-related clones of Shigella existed in Shenzhen. The development of PFGE molecular subtyping surveillance network would contribute to the active surveillance, outbreak investigation and source tracking for Shigellosis.

6.Effects of storage time on quality of Desmodium styracifolium seeds.

The dynamic changes of germination percentage, germination potential, thousand-seed weight, antioxidase activity in Desmodium styracifolium seeds with different storage time were tested, and electrical conductivity, contents of soluble sugar, soluble protein, starch in seed leach liquor were also determined in order to reveal the mechanism of seed deterioration. The results as the following. (1) The germination percentage, germination potential and thousand-seed weight of D. styracifolium seeds declined, while the seed coat color darkened with the extension of storage time. (2) The activities of superoxide dismutase (SOD) and peroxidase (POD) decreased with the prolongation of storage period. The SOD activity declined fastest in 1,095-1,185 d of storage, while the POD activity declined significantly in 365-395 d of storage. (3) The electrical conductivity and the contents of soluble sugar, starch in seed leach liquor increased, while the content of soluble protein declined with the extension of storage time. (4) Correlation analysis indicated that the germination percentage, germination potential and thousand-seed weight of D. styracifolium seeds have a significantly positive correlation with SOD and POD activity, while have a significantly negative correlation with the electrical conductivity, contents of soluble sugar and starch. It can be concluded that during the storage of D. styracifolium seeds, physiological and biochemical changes including decrease in antioxidase activity, rise in electrical conductivity, degradation effluent of soluble sugar and starch, degradation of soluble protein were the main factors leading to the seed deterioration.

7.Effect of ischemic postconditioning on the expression of myocardium matrix metalloproteinase-2 induced by ischemia/reperfusion in rats.

OBJECTIVETo investigate the effect of ischemic postconditioning on the expression of rat myocardium matris metalloproteinase-2 (MMP-2) induced by ischemia/reperfusion (I/R) and relationship between its expression and interstitium and the effect on left ventricular function.
METHODSTwenty-four rats were randomly divided into 3 groups (n = 8): sham control (SC) group, ischemic/reperfusion (I/R) group and ischemic postconditioning (IPTC) group. The left ventricular peak systolic pressure and its derivate (+/- dp/dt) were calculated; The amount of myocardium collagenous were determined; The vitality of superoxide dismutase (SOD) and content of malondialdehyde (MDA) of plasma were detected; The activity of myocardium MMP-2 was measured by Western blot and RT-PCR.
RESULTSAs compared with I/R group, IPTC could lower the expression of MMP-2, ameliorate left ventricular function and increase the content of myocardium collagenous. In the meantime, the vitality of superoxide dismutase (SOD) of plasma were greatly enhanced and the content of malondialdehyde (MDA) of plasma were reduced in IFC group.
CONCLUSIONProtective effect of IPIC on myocardium may be due to reduce free radical, lower expression of MMP-2 and protect myocardial interstitium. MMPs plays an important role in the myocardial protection provided by IPTC.

8.Statistical analysis of clinicopathologic characteristics of 1075 cases with colonic cancer.

OBJECTIVETo investigate the clinicopathologic characteristics of colonic cancer.
METHODSAll clinical data of surgical cases from hospital registry from 1982 to 2002 were analyzed using SPSS 11.0 software.
RESULTSThere were 1075 patients with colonic cancer including 573 males and 502 females. The median age of the patients was 58 years. 90.8% of the patients aged over 40 years . The proportion of right colonic cancer was 64.7%. Well-differentiated and moderately differentiated adenocarcinoma accounted for 69.8%, and mucous carcinoma 13.6%. The proportions of early stage I, advanced stage II, III and IV cancer were 8.1%, 45.0%, 26.5% and 20.3% respectively. The 5-, 10-year survival rates were 61.9% and 53.0% respectively.
CONCLUSIONSThe risk for colonic cancer is significantly increased in patients over 40 years. The frequency of right colonic cancer is higher than that of left colonic cancer. The overall survival rate is high.

9.Autologous hematopoietic stem cell transplantation for patients with diffuse large B-cell lymphoma: a retrospective study.

The purpose of this study was to investigate the efficacy of autologous hematopoietic stem cell transplantation (auto-HSCT) for patients with diffuse large B-cell lymphoma (DLBCL), and analyse the factors influencing prognosis. The clinical data of 67 patients with DLBCL received auto-HSCT from 1996 to 2011 and cumulative overall survival (OS), progression-free survival (PFS), transplant-related mortality (TRM), and relapse rate were retrospectively analyzed. The results showed that the median follow-up time was 40 months after transplantation. Three-year cumulative OS and PFS were 70.6% and 66.4% respectively, 5-year cumulative OS and PFS were 70.6% and 63.8% respectively, and TRM was 7.2%. One-year and three-year cumulative relapse rate were 16.5% and 23.7% respectively. Univariate analysis revealed that the age and pre-transplant disease status were significantly associated with poor prognosis (P < 0.05). It is concluded that auto-HSCT is a safe and effective therapeutic option for patients with DLBCL, especially for the young patients or patients with better remission.

10.Construction of a retroviral vector for RNA interference targeting human telomerase reverse transcriptase.

OBJECTIVETo construct a recombinant retroviral vector for RNA interference targeting human telomerase reverse transcriptase (hTERT).
METHODSThe sequences coding for enhanced fluorescence protein (EGFP), U6 promoter and a small interfering RNA (siRNA) targeting hTERT were amplified by PCR, respectively, and sub-cloned sequentially into the retroviral shuttle plasmid pLXSN to construct the plasmid pLXSN-EGFP-U6-siTERT. The recombinant expression plasmid was identified by restriction enzyme digestion and sequencing. Fluorescence microscopy and flow cytometry were employed to analyze EGFP expression in NIH3T3 transfected with the recombinant plasmid, and MMT assay was performed to evaluate the growth inhibition of Hela cells resulting from RNA interference mediated by the plasmid.
RESULTSSequence analysis and restriction enzyme digestion showed that the recombinant expression plasmid pLXSN-EGFP-U6-siTERT was constructed successfully. Twenty-four hours after transfection of NIH3T3 cells with the recombinant plasmid, the expression rate of EGFP reached 24.1% as shown by flow cytometry. MTT assay demonstrated a cell death rate of 53.2% 72 h after transfection of Hela cells with the plasmid.
CONCLUSIONThe successful construction of the recombinant retroviral plasmid mediating potent cell growth inhibition suggests the great potential of RNA interference technique in suppressing hTERT expression in mammalian tumor cells.
