Object To speed up the artificial propagation of the costly medicinal plant Dendrobium nobile Lindl. Methods The tender stems with lateral buds were tried as the explants and cultured on various culture media with different portions of hormon. Results The best medium for bud induction was the MS basic medium with addition of 6 BA (0.5 mg/L), NAA (0.2 mg/L), while MS with the addition of 6 BA (3.0 mg/L) NAA (0.5 mg/L), was suitable for propagation, MS with addition of IBA (0.3 mg/L), NAA (0.1 mg/L) was better for root growing, the striking growth rate was up to 95%. The young sprout was cultured fine on the base material mixed with coco crumbs, volcanic rocks and brick fragments (1∶1∶ 1) and covered by dried pteridophyte. Conclusion To provide the effective ways to reserve the resources of D. nobile and last its utlization.
3.Improvement of the Synthesis Technology of Sorafenib Tosylate

OBJECTIVE:To improve the synthesis technology of sorafenib tosylate (Ⅰ). METHODS:The intermediate N-methyl-(4-chlorpyridin-2-yl)carboxamide(Ⅳ)was obtained by chlorination and amidation with 2-picolinic acid as the starting com-pound. Meanwhile,N-[4-chloro-3-(triflouromethyl)phenyl]-N′-(4-hydroxyphenyl)urea (Ⅶ) was prepared from aminophenol and 4-chloro-3-trifluoromethyl phenyl isocyanate,which was obtained from 4-chloro-3-(trifluoromethyl)aniline(Ⅴ)by reaction with initiator triethylamine. Sorafenib tosylate was synthesized from Ⅳ and Ⅶ with potassium tert-butoxide by condensation and salt for-mation. The target compound was characterized by 1H-NMR. RESULTS:The target compound was confirmed as Ⅰ,with the over-all yield of 74% [based on 4-chloro-3-(trifluoromethylanilin)]. The purification of chromatogram was 94%. Optimized technology improves yield and simplifies multi-step intermediate decompression,distillation and purification process;the application of initia-tor shortens the duration of reaction. CONCLUSIONS:Ⅰ is prepared successfully,and raw material can be obtained easily and is easy to operate with high yield.
4.Analysis of 3 527 Male Infertile Patients’Semen

Objective To study the influence factors of infertility by analysis of semen sample and reference for clinical treat-ment.Methods 3 527 cases of semen sample were collected from Jan 2012 to Jun 2014.All samples were analysed by SQA-V analyzer and compared with 80 cases of normal semen.Results There were 358 normal samples (10.2%)and 3 169 ab-normal samples (89.8%).Among the abnormal samples low sperm motility had the highest ratio (2.7%)while abnormal pH had the lowest ratio (61.5%).All the indexes had significance difference to normal sample expect pH value (t=0.065, P =0.969).Among them,the comparison of rate had statistical significance(χ2 =3.214~24.712,P <0.05).The compari-son of mean also had statistical significance(t=2.523~15.324,P <0.05).Conclusion Infertility male almost has abnormal index of semen volume,liquefaction time,sperm motility,sperm density,sperm morphology,sperm viability.Accurately sperm analysis can provide objective basis to clinical diagnosis and treatment.
5.Effects of metformin on inflammatory status and phosphorylated Stat3 expression in lesions of a psoriasis-like mouse model

Objective To evaluate the effect of metformin on an imiquimod-induced psoriasis-like mouse model,and to explore its related mechanism.Methods A total of 50 male Balb/c mice were randomly divided into several groups:control group,imiquimod group (IM group) and imiquimod combined with metformin group (IM-Met group).The IM-Met group were classified into 3 subgroups (100,150 and 200 g/L IM-Met groups) according to the concentration of metformin.Mice in the control group were treated with topical vaseline cream on the back and intraperitoneal injection of 0.9% sodium chloride physiological solution of 0.3 millilitres every day.Mice in the IM group were treated with topical imiquimod cream on the back and intraperitoneal injection of 0.9% sodium chloride physiological solution of 0.3 millilitres every day.Mice in the 100,150 or 200 g/L IM-Met groups were treated with topical imiquimod cream on the back and intraperitoneal injection of 100,150 or 200 g/L metformin of 0.3 millilitres respectively every day.The changes of the erythema area,scales and psoriasis area and severity index (PASI) over time in each group were observed by naked eyes.These mice were sacrificed after 7 days of treatment.Western blot analysis was performed to determine the protein expression of interleukin (IL)-17 and phosphorylated signal transducer and activator of transcription 3 (p-Stat3) in the dorsal skin tissues of these mice,and enzymelinked immunosorbent assay (ELISA) to detect serum levels of IL-17,IL-6 and tumor necrosis factor-α (TNF-α) in these mice.Comparison among these groups was carried out by using one-way analysis of variance,and means of two groups were compared by using least significant difference (LSD)-t test.Results Compared with the IM group,the 100,150 and 200 g/L IM-Met groups showed alleviated erythema and scaling severity,and significantly decreased PASI score,protein expression of IL-17 and pStat3,and serum levels of inflammation factors such as IL-17,IL-6 and TNF-α (all P < 0.05).Seven days later,the PASI score and protein expression of IL-17 and p-Stat3 were all significantly lower in the 150 g/L IM-Met group than in the 100 g/L IM-Met group (t =4.18,-5.95,-2.80 respectively,P < 0.05,≤ 0.01,≤ 0.01 respectively),but no significant differences were observed between the 150 g/L IM-Met group and 200 g/L IM-Met group (t =0.29,-0.42,-0.32 respectively,P > 0.05,=0.69,=0.76 respectively).The serum levels of IL-17,IL-6 and TNF-α in the 150 g/L IM-Met group were 33.388 ± 1.556 ng/L,210.741 ±4.440 ng/L and 249.434 ± 8.594 ng/L respectively,which were significantly higher than those in the 200 g/L IM-Met group (26.720 ± 2.156 ng/L,174.997 ± 9.501 ng/L,193.034 ± 6.661 ng/L respectively;t =7.93,10.79 and 16.403 respectively,all P < 0.05),but significantly lower than those in the 100 g/L IM-Met group (44.008 ± 1.722 ng/L,260.926 ± 7.724 ng/L,271.409 ± 3.037 ng/L respectively;t =-14.47,-17.81 and -7.62 respectively,all P < 0.05).Conclusion Metformin can alleviate psoriasis-like skin lesions and inflammation status in the imiquimod-induced psoriasis-like mouse model to a certain extent,which may be related to the decrease of p-Stat3 protein expression.
6.Disposition and tissue distribution of ML12 in rats.

To investigate the disposition and tissue distribution of ML12 after intravenous (iv) administration in rats, the compound in plasma or in tissue was extracted into ethyl acetate under basic condition and was determined by HPLC after extracted by dilute sulfuric acid. Excitation wavelength and emission wavelength of fluorescence detection were 278 nm and 307 nm, respectively. The data were processed with the software 3P97 to calculate the main pharmaceutical parameters of ML12. At dose of 5 and 10 mg/kg, the elimination of the drug from plasma was found to be kinetically linear, but when the dosage was 20 mg/kg, a non-linear feature was observed. The highest level of ML12 was found in the kidney. Distribution of ML12 after iv administration was extensive and the concentration-time profile was found to be fitted to an open two-compartment model.

7.Prevention of postoperative cerebrospinal fluid leakage with absorbable hemostatic fluid gelatin.

OBJECTIVETo explore the effectiveness of absorbable hemostatic fluid gelatin in preventing postoperative cerebrospinal fluid leakage.
METHODSThe clinical data of 17 patients with dura mater tear were retrospectively analyzed from March to September in 2003. There were 16 males and 1 female, aged from 16 to 67 years old with an average of (39.6 ± 15.4) years. The injury site was at cervical vertebrae in 1 case, thoracic vertebrae in 9 cases, thoracolumbar junction in 4 cases, lumbar vertebrae in 3 cases. There were burst fracture in 4 cases and fracture-dislocation in 13 cases. According to ASIA grade, 12 cases were grade A, 2 cases were grade B, 2 cases were grade D, 1 case were grade E. Two cases caused by traffic accident, 10 by high falling, 4 by heavy parts crash, 1 by stairs fell during the earthquake. Absorbable hemostatic fluid gelatins were used to plug the dura mater tear,in order to prevent postoperative cerebrospinal fluid leakage. Postoperative drainage were recorded every day.
RESULTSOf 17 patients, 15 cases did not develop with cerebrospinal fluid leakage. Two cases develop with cerebrospinal fluid leakage after operation and their drainage were removed at 6 to 7 days after operation. In all cases, no complications related with cerebrospinal fluid leakage occurred, such as headache, dizzy, fever,neck resistance, rash, incision disunion, incision infection, hematoma, neurologic symptoms aggravation. No abnormal phenomena was found on incision surrounding at follow-up of 9 months.
CONCLUSIONUsing absorbable hemostatic fluid gelatin to plug the dura mater tear during operation is an effective method in preventing postoperative cerebrospinal fluid leakage.

8.Protective effect of asiatic acid from Potentilla chinensis on alcohol hepatic injury in rats.

To study the protective effect and the mechanism of asiatic acid (AA) from Potentilla chinensis on alcohol hepatic injury in rats. Male Wistar rats were randomly divided into six groups: the normal control group, the AA control group (8 mg · kg(-1) AA), the model group (5.0-9.0 g · kg(-1) alcohol) and high, medium and low-dose AA-treated groups (alcohol + 8, 4, 2 mg · kg(-1) AA). Each group was orally administered with the corresponding drugs once a day for 24 weeks. Approximately 1. 5 hours after the final administration, all rats were killed, and their blood samples and hepatic tissues were collected. The AST and ALT in rat serum and the contents of MPO, TNF-α, IL-1β, SOD, GSH-Px, GSH-Rd and MDA in hepatic tissues were detected. The expressions of NF-κB, TLR4, CD14, MyD88, TRIF and protein expression in hepatic tissues were measured by western blot. The pathological changes in liver tissues were observed by histological examination. The results showed that compared with the model group, the AA-treated groups showed significant decreases in serum ALT, AST and MDA and increases in the activities of SOD, GSH-Px, GSH-Rd and MPO. Moreover, AA markedly inhibited the expressions of TNF-α, IL-1β, TLR4, CD14, MyD88 and NF-κB. The histological examination showed alleviated hepatic issue ijury to varying degrees. In short, asiatic acid (AA) from P. chinensis could protect alcohol-induced hepatic injury in rats. Its mechanism may be related to the inhibition of NF-κB inactivation and the reduction of inflammatory response.

9.Study on antibiotic resistance of Neisseria gonorrhoeae in Nanning for 5 years

Objective To study the antibiotic resistance of Neisseria gonorrhoeae and the prevalence of penicillinase-producing Neisseria gonorrhoeae (PPNG) and high-level tetracycline resistant Neisseria gonorrhoeae (TRNG) in Nanning. Methods Minimum inhibitory concentrations (MIC) were determined by agar dilution method and ?-lactamase production was comfirmed by acidometric method.Results Among 458 strains, 33 strains(7.20%) were found to be PPNG and 231 strains(50.44%) were TRNG. The resistance rate to ciprofloxacin, ceftriaxone and spectinomycin were 87.33%, 1.31% and 0.22% respectively. Conclusion The situation of resistance to ciprofloxacin was very serious, and it was also found that a few of gonococcal isolates were resistant to ceftriaxone and spectinomycin. The study showed that it is important to successively survey the antibiotic resistance of Neisseria gonorrhoeae.
10.Molecular epidemiology of urogenital Chlamydia trachomatis infection in patients attending sexually transmitted diseases clinics

Objective To determine the prevalence and genotype profiles of urogenital Chlamydia trachomatis and their related factors in patients attending sexually transmitted diseases (STD) clinics in Guangxi Zhuang Autonomous Region. Methods C. trachomatis was screened by a plasmid PCR in 598 patients attending STD clinics. Then, positive specimens underwent nested-PCR to amplify the major outer membrane protein 1 (ompl) gene. The amplicons of ompl gene were digested by restriction endonucleases Alu I plus Hinf I and Cfol . C. trachomatis was differentiated according to the restriction fragment length polymorphism (RFLP) patterns. Results Out of the 598 samples, 83 were positive for plasmid-PCR. The prevalence of chlamydial infection was 13.9% with no significant difference between male and female patients. Nested-PCR based RFLP analysis showed that genotype E amounted to 27.7% (23/83), F 20.5% (17/83), D 13.2%(11/83), G 12.0%(10/83), K 7.2%(6/83), H 4.8%(4/83), I 3.6% (3/83), J 3.6%(3/83)and uncertain types 7.2% (6/83). Visible symptoms were observed less frequently in infections with C. trachomatis genotypes E and F compared with the other genotypes, while low abdominal pain occurred in 80% of infections with type G. Conclusions A certain proportion of out-patients attending STD clinic are infected with various types of C. trachomatis in Guangxi Zhuang Autonomous Region. The polymorphism of ompl gene may serve as a useful tool in molecular epidemiological studies of C. trachomatis.
