Objective To observe the effects of recombinant human erythropoietin(rh-EPO) on immune function of cyclophosphamide-treated mice.Methods Six-week-old mice were randomly divided into 4 groups.Cyclophosphamide plus normal saline group(CTX(+NS));cyclophosphamide plus lower and higher dosage rh-EPO group(CTX+LDrh-EPO and CTX+HD rh-EPO);normal saline control group(NS).The changes of Hb,WBC,red blood cell immune function((C_3b)-R%),T lymphocyte proliferative responsiveness optical density index A(A score),IL-2,TNF-? of 4 groups were observed.Results The CTX + NS group showed lower levels on Hb,WBC,C_(3b)-R%,A score,IL-2 and TNF-? compared with NS normal control group(P0.05).There were positive correlation between C_(3b)-R% and Ascore,C_(3b)-R% and IL-2.Conclusions 1.Cyclophosphamide treated mice have lower Hb,WBC,C_(3b)-R%,A,IL-2 and TNF-? secretion.2.rh-EPO administration have improvements with Hb,RBC immune function,T lymphocyte proliferative responsiveness and IL2 production.

As the fourth member of the globin family, cytoglobin ( CYGB) is a hypoxia-induced gene, and its expression is upregulated under hypoxia.CYGB is widely distributed in the living body and has a variety of physiological functions, such as anti-oxi-dative stress, anti-fibrosis, and neuroprotection.It also plays a protective role in cardiovascular disease, cancer, and other hypoxia-re-lated diseases.This article presents an overview on the structure and distribution of CYGB, its physiological functions of anti-fibrosis, anti-cancer and muscle regeneration, and the regulation and the mechanism of its expression under hypoxia.

5.Effect of Purpura Decoction and Its Disassembled Prescriptions on Reticulated Platelet and ;Thrombopoietin of Thrombocytopenic Purpura Model Mice

Objective To observe the effects of Purpura decoction and its disassembled prescriptions on peripheral blood reticulated platelets (RP) and serum thrombopoietin (TPO) of idiopathic thrombocytopenic purpura (ITP) model mice, to explore the action mechanism and core structure of the prescription. Methods Totally 84 BalB/c mice were randomly divided into normal group, model group, prednisone group, Purpura decoction group, Buyi Qixue group, Buqi Huoxue group and Buxue Huoxue group. ITP mouse model was made by injecting intraperitoneally guinea pig-antimouse platelet serum. From the 5th day of modeling, mice were intragastrically administered. The normal group and model group were fed with normal saline 0.2 mL per mouse. Purpura decoction group was fed with Purpura decoction 0.43 mL per mouse. Buyi Qixue group was fed with Buyi Qixue decoction 0.36 mL per mouse. Buqi Huoxue group was fed with Buqi Huoxue decoction 0.27 mL per mouse. Buxue Huoxue group was fed with Buxue Huoxue decoction 0.3 mL per mouse. Prednisone group was fed with prednisone acetate suspension 0.2 mL per mouse. Ten days later, took the blood from the eyeball and isolated serum, the mouse peripheral platelet, RP and TPO were observed. Results After treatment, the platelet count of treatment groups was improved. Compared with before treatment, Purpura decoction group,Buqi Huoxue group and prednisone group had significant difference in platelet count (P<0.05). The RP of model group was significantly higher than the normal group (P<0.05), but RP absolute value was lower than the normal group (P<0.05). Compared with the model group, RP of treatment groups reduced (P<0.05), and RP was negatively correlated with platelet count (P<0.05). TPO level of the model group was higher than that of the normal group (P<0.01). TPO of treatment groups declined significantly, and had significant differences with that of the model group (P<0.05). Conclusion Core structure of Purpura decoction is qi tonifying and blood activating drug. The mechanism of treating ITP is related with lowering RP and TPO.
6.Relationship between methylation status in promoter region of Runx3 gene and children malignant lymphoma

Objective To study methylation status of the Runx3 gene in occurrence and development of children malignant lymphoma.Methods The bone marrow specimens of 48 children diagnosed as malignant lymphoma were included into experimental group.20 children with non-malignancy were included into control group.Methylation-specific polymerase chain reaction (MS-PCR) was used to detect methylation status of Runx3 gene in bone marrow cells.The reverse transcription-polymerase chain reaction (RT-PCR) was used to detect mRNA expression of Runx3 gene.Results MS-PCR assay results showed that 31 cases expressed Runx3 gene methylation in experimental group,the positive rate was 64.6 % (31/48),but no one was detected in control group (20 cases),the difference between two groups was significant (x2 =15.7,P <0.05).Dynamic observation of 42 cases in experimental group showed the declining of Runx3 methylation positive rate.RT-PCR assay results showed that all 20 cases in the control group expressed Runx3 gene mRNA,all cases with methylation status of the Runx3 gene in the experimental group didn’ t expressed Runx3 gene mRNA.In experimental group,9 cases of clinical remission expressed Runx3 gene mRNA,and 1 case of relapsed didn’ t expressed Runx3 gene mRNA.Conclusion Runx3 gene shows a high methylation status in bone marrow cells of malignant lymphoma children,so that blocked the expression of Runx3 gene,which is closely related to the occurrence and development of children malignant lymphoma.
7.ZO-1 gene methylation status and its clinical significance in children with non-Hodgkin lymphoma.

OBJECTIVETo investigate the methylation status of zonula occludens-1 (ZO-1) gene promoter and its clinical significance in children with stage IV non-Hodgkin lymphoma (NHL) and to provide a basis for further etiological study and early diagnosis of this disease.
METHODSFifty-five children with a confirmed diagnosis of stage IV NHL (40 cases of T-NHL and 15 cases of B-NHL) were selected as the case group, and 20 children with diseases other than hematologic malignancies were selected as the control group. Bone marrow samples were collected from these subjects. Methylation-specific PCR (MS-PCR) was applied to evaluate the methylation status of ZO-1 gene promoter, and the integrated optical density (IOD) was determined. RT-PCR was used to measure the mRNA expression of ZO-1.
RESULTSMS-PCR showed that the methylated bands of ZO-1 gene promoter were found in 39 (70.9%) of 55 patients in the case group before treatment, while no ZO-1 gene promoter methylation was detected in the control group. With close tracking of 47 cases in the study group, consisting of 32 cases of T-NHL and 15 cases of B-NHL, the rates of ZO-1 gene promoter methylation prior to treatment were 72% and 67%, respectively, (P>0.572). The cases of T-NHL and B-NHL showed no significant changes in methylation rate in the early and middle phases of chemotherapy (P>0.05), but they showed significant changes in methylation rate in the late phase of chemotherapy (P<0.05). RT-PCR showed that the NHL cases carrying methylated ZO-1 gene had no mRNA expression of ZO-1, while all children in the control group had mRNA expression of ZO-1. There was no linear relationship between the total number of peripheral blood leukocytes and ZO-1 gene IOD (r=0.093, P=0.575); a positive correlation was found between the number of malignant cells in bone marrow and ZO-1 gene IOD (r=0.669, P<0.001).
CONCLUSIONSZO-1 gene shows a hypermethylation status in children with NHL, and the methylation level is positively correlated with the number of malignant cells in bone marrow. ZO-1 may be used as a novel molecular marker in early diagnosis, outcome assessment, prognostic evaluation, and detection of minimal residual disease.

8.Rapid quantification of total nitrogen and end-point determination of hide melting in manufacturing of donkey-hide gelatin.

Hide melting presents itself as one of the most critical processes in the production of donkey-hide gelatin. Here a NIR-based method was established for the rapid analysis of in-process hide melting solutions as well as for end-point determination of this process. Near infrared (NIR) spectra of hide melting solutions were collected in transflective mode. With the contents of total nitrogen determined by the Kjeldahl method as reference values, partial least squares regression (PLSR) was employed to build calibration models between NIR spectra and total nitrogen. Model parameters including wavelength range and PLS factors were optimized to achieve best model performance. Based on the contents of total nitrogen predicted by calibration model, end point of hide melting was determined. The constructed PLS model gave a high correlation coefficient (R2) of 0.991 3 and a root mean square error of prediction (RMSEP) of 0.807 g x L(-1). With the predicted total nitrogen and predefined limit, decisions concerning the proper times of melting were made. This research demonstrated that NIR transflectance spectroscopy could be used to expeditiously determine the contents of total nitrogen which was subsequently chosen as the indictor for determining the end-point of hide melting. The proposed procedure may help avoid unnecessary raw material or energy consumption.

9.The development of a polarized vernier optometer for tonic accommodation measurement.

A vernier optometer, based on the Scheiner principle, that makes use of polarized light, is designed and developed. It is specialized in measurement of tonic accommodation, accommodation and accommodation response. The vernier optometer has been proved by chinical testing results, to be a reliable and effective instrument with fine stabilily, good repeatability and sensitivity for short sight study relating to oculogyral parameters.

10.The long-term excessive alcohol consumption on sperm quality and the impact of germ cell apoptosis

Objective To investigate the long-term excessive drinking semen nitric oxide (NO) content in their sperm quality and spermatogenic cell apoptosis and infertility. Methods Nitrate reductase method was used to specific reduce the NO metabolites nitrate (NO3-) to nitrite (NO2-), which was used on behalf of the total NO level. Terminal deoxynucleotidyl transferase (TdT) and mediated nick end labeling (TUNEL) method and binocular optical microscope were used to detect and observe the rate of apoptosis of spermatogenic cells and the morphological structure. The SQA-V sperm quality automatic analyzer was used to measure sperm quality. Results In not drinking semen fertility group, NO content was (54.81±11.45)μmol / L, the rate of spermatogenic cell apoptosis was (4.52±1.23)%, sperm motility was (80.24±0.17)%, energy a+b (78.32±0.12)%, deformity rate (5.30±0.13)%, and long-term excessive alcohol infertility C group was [(128.83±22.73)μmol/L,(17.34±2.53)%,(51.18±0.58)%,(21.45±0.26)%,(21.12±3.24)%] respectively. Compared to a very significant difference (t=10.04,17.38,6.69,15.59,17.02,P<0.01) . In long-term excessive drinking group, the levels of NO and spermatogenesis cell apoptosis rate was significantly positive correlated (r=0.93,P<0.01).Apoptosis of spermatogenic cell nucleus chromatin was condensed in the formation of crescent-shaped perinuclear, nuclear was cleavaged to form apoptotic bodies. Conclusions In the long-term excessive drinking semen, NO content and spermatogenic cell apoptosis rate was increased, the sperm had poor quality. The results showed that the long-term excessive drinking can cause germ cell apoptosis in male infertility and promote the body to overproduce NO, whichmay be one of the reasons for male infertility.
