International Journal of Laboratory Medicine 2015;(22):3265-3267,3270
Objective To evaluate the necessity and feasibility of nucleic acid test for donors blood screening .Methods From July 1 ,2011 to December 31 ,2014 ,a total of 170 316 blood samples which were negative in enzyme-linked immunosorbent assay (ELISA)and qualified in aianine aminotransferase detection ,were selected in this study stochastically .All the samples were detected hepatitis B virus(HBV) ,hepatitis C virus(HCV) ,human immunodeficiency virus(HIV) by nucleic acid amplification technology (NAT) .NAT positive samples were reconfirmed in National Center for Clinical Laboratories(NCCL) .Results A total of 160 cases of nucleic acid reactive samples were found out ,the total response rate was 0 .09% ,The response rate of Roche nucleic acid detec-tion system was 0 .10% ,response rate of David nucleic acid detection system was 0 .08% ,there was no significant difference be-tween the two methods(P>0 .05) .In 27 cases of specimens ,14 cases were confirmed as HBV DNA positive ,no HCV RNA and HIV RNA were detected ,the confirmed positive rate was 51 .85% .There were 2 samples detected by chemiluminescence HBsAg reactivity .Conclusion ELISA screening of blood donors has missing phenomenon ,nucleic acid detection method could be used as an effective supplement of the ELISA ,could improve the safety of blood for clinical use ,detection sensitivity is better than ELISA .

Objective To investigate the therapeutic effect of NSR siRNA on nerve regeneration following spinal cord hemi-transsection injury in rats. Methods Rats with T8 spinal cord hemi-trans-section were didded into 3 groups, ie, siRNA group, NS group and control group. SiRNA or NS was in-jected into lateral cerebral ventricle just after spinal cord injury. The therapeutic effect of NgR siRNA was evaluated by using BBB locomotor rating scale, retrograde horseradish peroxidase(HRP)tracing and HE staining. Results BBB locomotor rating scale showed that the recovery of the locomotor function of siRNA group seemed to be better than that of the other two groups from the 4th week, but there was no statistical difference. Retrograde HRP tracing showed a large number of positive cells in the anterior horn of spinal cord, with statistical difference compared with NS group and control group(P<0. 05). Eight weeks after spinal injury, HE staining showed disorderly distribution of the fibres in NS group and control group but serial fibres in the injury region in siRNA group. Conclusion NSR siRNA may promote the nerve regeneration following spinal cord injury.

Objective To evaluatethe expression and significance of RUNX3 gene and protein in hepatocellular carcinoma.Method Reverse transcription-polymerase chain resction (RT-PCR) and immunoitistochemistry SP method were used to detect the expressions of RUNX3 gene and protein in 23 cases of hepatocellular carcinoma and their adjacent non-cancerous tissue. Results The expression of RUNX3 gene and protein were significantly reduced in hepatocellular carcinoma tissue (26.1% and 17.4% respectively),which were prominently lower than those in adjacent non-cancerous tissue:91.3% (21/23) and 87.0% (20/23),P<0.01.Conclusion The lower-expression of RUNX3 gene and protein may play an important role in hepatocellular carcinoma.

Objective To explore the clinical effect of percutaneous osteoplasty with SKy bone expander system in the treatment of calcaneal fracture. Methods 63 feet of 55 patients with calcaneal fractures were involved in this study.According to Sanders fracture classification including 38 feet of Sanders Ⅱ,18 feet of Sanders Ⅲ,7 feet of Sanders Ⅳ.There were 6 to 16 days interval between the injuries and the surgeries. The patients were treated by percutaneous osteoplasty with the SKy bone expander system. The standard of operation was the satisfaction of reduction and Bohler's and Gissane's angles under X-ray.Results All of 63 feet of 55 patients were followed up for average 22 months.According to the criterion of therapeutic effect, the results were as follows:excellent in 30 cases, good in 26 cases, fair in 7cases,and no poor case. The excellent and good rate was 88.9%.Conclusion Percutaneous osteoplasty with SKy bone expander system in the treatment of calcaneal fracture, especially in reduction and fixation of Sanders type Ⅱ and Sanders type Ⅲ,can recover Bohler's and Gissane's angles, significantly shorten the duration of illness, and has fast recovery and can possess satisfactory curative effect, and it is worth popularizing.

OBJECTIVE:
To investigate the effect of benzo(α)pyrene on the ATPase activity and content of Ca²⁺ in the hippocampus of neonatal SD rats.
METHODS:
Sixty male and 60 female 4-days-old neonatal SD rats were randomly divided into 5 groups (n=24): a blank control group, a vehicle control group (peanut oil), 3 benzo(α)pyrene groups (0.02, 0.2 and 2 mg/kg, respectively). SD rats were given benzo(α)pyrene (dissolved in peanut oil) by gavage daily from postnatal day 4 (PND4) to PND20. The nerve reflex, the condition of neuro-muscle development and motion function were examined in the period of treatment. The colorimetric technique was used to detect the activity of Ca²⁺-ATPase and Ca²⁺-Mg²⁺-ATPase in hippocampus after the treatment. The concentration of Ca²⁺ of synapse in the hippocampus of rats was detected by fluorescent labeling.
RESULTS:
The results from the behavior tests showed that duration of surface reflex latency in rats with medium dose of benzo(α)pyrene was longer compared with that in the control group in PND12. The duration of surface reflex latency in rats with high dose of benzo(α) pyrene is longer in PND 14 and PND 16 compared with that in the control group (P<0.05). Compared with the rats in the control group, the activities of Ca²⁺-Mg²⁺-ATPase and Ca²⁺-ATPase in hippocampus in rats with high dose benzo(α) pyrene were significantly decreased, and the degree in the decrease of Ca²⁺-ATPase activity was dose-dependent (P<0.05). The contents of Ca²⁺ in the hippocampus in rats with medium or high dose of benzo(α) pyrene were significantly increased compared with that in the control group (P<0.05), which showed a dose-dependent manner (P<0.05).
CONCLUSION
Benzo(α)pyrene exposure led to the decrease in ATPase activity as well as Ca²⁺ overload of the synapse in the hippocampal tissue, which in turn results in the nerve damage of newborn SD rats.
