1.Analysis of Shuanghuanglian Injection Powder by HPLC-ESI-mass/mass spectrometry

AIM: To analyse the main chemical constituents of Shuanghuanglian Injection Power(Flos lonicerae japanicae,Radix scutellariae and Fructus forsythiae) by high performance liquid chromatography-electrospray ionization-mass/mass spectrometry. METHODS: The sample solution was analysed by a Zorbax C_18 column with a gradient mobile phase comprised of acetonitrile and water.Electrospray ionization mass spectrometry detector was used,full-scan detection mode was evaluated for the identification of the liquid chromatography peaks. RESULTS: To analyse the mass spectrum of every liquid chromatography peaks,we identified 18 molecular mass from the ion chromatogram of Shuanghuanglian Injection Powder,9 compounds were identified,including quinic acid,caffeic acid,three monocaffeoylquinic acids,dicaffeoylquinic acid,forsythin,forsythoside and baicalin. CONCLUSION: Quinic acid is identified from Shuanghuanglian Injection Power for the first time.High performance liquid chromatography-mass/mass spectrometry is emerging as an important tool in traditional Chinese medicine.

Objective To investigate effects of the complex therapy on severe inhalation injury. Methods The cases with inhalation injury and acute respiratory failure (ARF) of 4172 consecutive burn patients from Jan. 2000 to Feb. 2003 in our institute were studied retrospectively. Objective To study the effect of acute serious smoke inhalation injury to unilateral lung on contralateral lung and the therapeutic efficacy of massive bronchoscopic lavage with saline to the primary injured bronchoalveolus at early stage after smoke inhalation on mitigating the secondary damage of the other lung and so as to confirm the therapeutic validity of lung lavage to smoke inhalation injury. Methods Fifteen mongrel dogs were insufflated with sawdust smoke into left lung and then randomized into 2 groups. The dogs in group A were maintained intravenous glucose saline transfusion and breathed air freely for 24 h after injury. Those in group B received a massive bronchoscopic lavage with 250 ml of saline to injured lung at 1 hour after injury and then were administrated similarly to those in group A. Gas exchange function of bilateral lungs in the process were observed and pathologic and phyiopathologic changes of the lung specimen were examined after the process. Results While the total lavaging volume was kept to 250 ml, the liquid volume remained in left lung after each operating was from 90 ml to 140 ml. All animals had a significant decline in pulmonary function after smoke inhalation injury. PaO2 in group B decreased significantly soon after the lavage and then increased gradually to exceeding that in group A. The left lung showed serious edema similarly in two groups but in right lung the water content was less, dynamic and static compliance was more in group B than in group A. Conclusion Bronchoalveolar massive lavage at early post-injury stage to injured lung after unilateral lung smoke inhalation injury can mitigate secondary damage of the non-injured lung. Lung lavage can decrease the intensity of secondary systemic inflammatory reaction and show a therapeutic validity to smoke inhalation injury.
4.Analyses on 959 cases of inhalation injury

Objective To review the experience in the management of burned patients inflicted by inhalation injury in our institute in the past 42 years. Methods Patients with inhalation injury were analyzed chronically in three different periods as 1958 to 1980, 1981 to 1990, and 1991 to 2000. Results The mortality rates in general and in terms of different degrees decreased obviously with the elapse of time. In other words, the rates were 48.93%, 26.60% and 8.53 for the above three periods. Conclusion The mortality of inhalation injury was decreased dramatically during the last decade in our institute. Five main measures consisting of inhalation of high concentration or pure oxygen, preventive tracheotomy, frequent tracheal lavage with small amount of lavage fluid at early stage of postburn, mechanical ventilation with PEEP and humidification as early as possible, and application of exogenous pulmonary surfactant contribute greatly to the decrement of death rate in the patients with inhalation injury.
5.Association of MICA-STR with leukemia in Hunan Han population

Objective: To evaluate the effect of partial liquid ventilation with perfluorocarbon on the static lung compliance in rat with smoke inhalation injury. Method: Partial liquid ventilation (PLV)and mechanical ventilation (MV)were set up on rat's model,total static lung compliance (C_(2.94))and low volume static lung compliance (C_(0.49))were measured with hydraulic pressometer,the expansion index(EI)of lung was calculated. Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buyang Huanwu Decoction (BHD). Methods Capillary zone electrophoresis was applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electrolyte buffer was composed of 25 mmol/L borax-methanol (85∶15). Capillary electrophoresis was performed using a 52 cm (30 cm to detector)?50 ?m fused-silica capillary tube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30 ℃. Results Ferulic acid was successfully separated within 6 min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respectively. Conclusion This method can supply reference for the determination of ferulic acid in serum samples and BHD.
8.Apply Value of LHD and RDW in Diagnosis and Treatment of Children with Iron Deficiency Anemia

Objective To investigate the low hemoglobin density (low haemoglobin density,LHD) and red blood cell volume distribution width (red blood cell volume distribution width,RDW) in iron deficiency anemia (iron-deficiency anemia,IDA)in children with the value of diagnosis and treatment.Methods From February 2016 to May 2017,in the Second People's Hospital of Longgang District in Shenzhen City,86 cases of children with iron deficiency anemia for IDA group,and 120 cases of healthy children (as the control group) at the same time were confirmed.Blood routine of children with IDA were detected before and after treatment hemoglobin concentration and the results were analyzed statistically.Results 120 cases of healthy children in the peripheral blood LHD value was 2.74 % ± 0.90 % and the boy was 3.07 % ± 0.81%,higher than that of 2.26 % ± 0.69 % of the girls.Between them,there was statistically significant difference (t=3.815,P<0.05).The value of RDW was 12.37 % ± 2.84 %,12.09 % ± 2.80 % for the boys and 12.56 % ± 2.87 % for the girls,there was no statistically significant difference between the them (t=0.517,P>0.517).Before treatment,86 cases of children with IDA LHD and RDW values in peripheral blood were 30.67 % ± 20.12 % and 16.62 % ± 3.27 %,significantly higher than the control group,the difference was statistically significant between (t=4.025 ~ 4.025,P<0.05 ~ 0.01),and there was no statistically significant difference between male and female children (t =0.761 ~ 0.917,P> 0.05).After treatment L HD and RDW values were 10.65 % ± 8.92 % and 14.02 % ± 2.93 %,significantlylower than before the treatment,between them was statistically significant difference (t=2.912~9.675,P<0.05).Conclusion Children with iron deficiency anemia treatment before the LHD and RDW values significantly elevated in the blood,significantly decreased after treatment,the LHD and RDW values for diagnosis and treatment of children with iron deficiency anemia dynamic monitoring has certain application value,worthy of popularization and application.
9.HPCE fingerprinting of Gegen Qinlian Decoction

Object To study the HPCE fingerprinting of Gegen Qinlian Decoction (GQD). Methods A buffer was composed of 30 mmol/L sodium phosphate and 40 mmol/L borate solution. Capillary electrophoresis was performed using a 65 cm (43 cm to detector) ?50 ?m fused-silica capillary tube. Separation voltage was 22 kV, sampling time was 1 s, detected wavelength was 254 nm, and the temperature was maintained at 30 ℃. Results The 27 components in GQD were successfully separated. The observation of methodology was in keeping with quantitatine determination and qualitative study. Conclusion This method can be used for the quality control of the preparation of GQD with good precision.
10.Study on the cytocompatibility of biodegradable poly(epsilon-caprolactone) microspheres in vitro.

Poly(epsilon-caprolactone) microspheres were fabricated with an average particle size of 5.08 +/- 0.23 microns. The effect of poly(epsilon-caprolactone) microspheres on apoptosis and cell cycle of fibroblast was studied with flow cytometry. The data obtained clearly indicated that poly(epsilon-caprolactone) microspheres purified in different ways showed different cytocompatibility. The well purified microspheres have good cytocompatibility.
