1.Mediating effect of positive coping style on health literacy and lifestyle among medical students

Objective:
To examine the mediating effect of positive coping style on health literacy and lifestyle among medical students, so as to provide the reference for health education and health promotion among medical students in universities.
Methods:
Medical students of six full-time medical colleges in Guizhou Province were sampled using a multi-stage stratified cluster random sampling method from April to June 2023, and their gender, age and educational level were investigated by questionnaires. The College Students' Healthy Lifestyle Scale, the Health Literacy Questionnaire and the Simplified Coping Style Scale were used to evaluate lifestyle, health literacy and coping style of students. The Process program was used to analyze the mediating effect of positive coping style on health literacy and lifestyle, and Bootstrap method was used to examine the significance of the mediating effect.
Results:
A total of 9 266 peoples were investigated, including 3 177 males (34.29%) and 6 089 females (65.71%), and had a mean age of (21.06±2.61) years. There were 2 775 junior college students (29.95%), 4 864 undergraduates (52.49%) and 1 627 master students (17.56%). The scores of lifestyle, health literacy and positive coping style among medical students were 118.34±19.06, 54.57±10.81 and 2.81±0.68, respectively. Health literacy had a direct positive effect on lifestyle (effect value=0.286, 95%CI: 0.257-0.316) and a indirect positive effect on lifestyle through positive coping style (effect value=0.215, 95%CI: 0.193-0.236), and the mediating effect contributed 42.91% to the total effect.
Conclusion
Health literacy could improve lifestyle indirectly through positive coping style among medical students.
2.Investigation on human Cryptosporidium infection in local area of Anhui Province

Objective To investigate the status of Cryptosporidium infection in the population in the local area of Anhui Prov?ince,and discuss the risk factors of the infection,so as to provide the evidence for the prevention and treatment of cryptosporidi?osis. Methods Qianshan County and Lingbi County of Anhui Province were selected as investigation spots,and the oocysts of Cryptosporidium in the feces of the investigation objects and the specific IgG antibody against Cryptosporidium in the serum were checked by using the pathogenic modified acid fast staining method and ELISA,respectively,so as to determine the status of Cryptosporidium infection in these investigation objects. At the same time,the questionnaire surveys were conducted in the inves?tigation objects so as to know about the risk factors of Cryptosporidium infection. Results A total of 668 people were investigat?ed in the two counties,635 people received etiological examinations,and 15 people were positive with the positive rate of 2.36%;642 people received serological examinations,and 140 people were positive with the positive rate of 21.81%;628 peo?ple received pathogenic and serological examinations at the same time,and the examination results of the both methods showed that 12 people were positive(there were 4 people in Qianshan County and 8 people in Lingbi County),and the positive rate was 1.94%. The rates of Cryptosporidium infection in the population of Qianshan County and Lingbi County were 1.24%(4/322)and 2.71%(8/295)respectively,and the difference had no statistical significance(P>0.05). The single factor analysis found that the rate of Cryptosporidium infection was higher in the children and diarrhea patients;the multivariate logistics regression analy?sis indicated that the rate of Cryptosporidium infection was higher in the people who bred poultry and the diarrhea patients. Con?clusions The positive rate of serum antibody of Cryptosporidium in the population of the local area of Anhui Province is higher, which indicates that the previous infection is serious,and the rate of Cryptosporidium infection in human is relative to the age, diarrhea and whether there are poultries to be bred in the family,which is worthy of attention in the future prevention and treat?ment.
3.Changes of neuron specific enolase in serum of patients with silicosis

Objective:To explore the changes of neuron specific enolase (NSE) concentration in serum samples of patients with silicosis.Methods:In January 2020, 455 cases of silicosis diagnosed in Yantaishan Hospital from January 2018 to December 2019 were collected, and 60 healthy cases and 120 cases of lung cancer were selected as the healthy group and the lung cancer group. The serum levels of NSE were detected by chemical immunofluorescence assay and compared.Results:The serum NSE level of silicosis patients was [(22.88±7.86) ng/ml], higher than that of healthy group [(17.96±4.42) ng/ml] ( P<0.05) . Serum NSE levels in the first, second and third stage silicosis groups were higher than those in the healthy group ( P<0.05) , but there was no statistically significant difference between the silicosis groups at different periods ( P>0.05) . The serum NSE level of silicosis patients was lower than that of the small cell lung cancer group ( P<0.05) , but there was no statistically significant difference between silicosis group and non-small cell lung cancer group ( P>0.05) . The area under curve of silicosis NSE was 0.718 ( P<0.01) when the cut-off was 17.49 ng/ml, the sensitivity and specificity of NSE was 78% and 57% respectively. Conclusion:The serum NSE level of patients with silicosis is significantly increased, which can be used as an important reference index for the diagnosis and the differential diagnosis of silicosis.

5.Association of insulin sensitivity and β-cell function with body fat in patients with PCOS

A total of 187 patients with polycystic ovary syndrome (PCOS) in our hospital from January 2016 to December 2018 were enrolled, and 190 healthy people served as control group. The levels of homeostasis model assessment of insulin resistance index ( HOMA-IR), β-cell function index ( HOMA-β), total cholesterol ( TC), triglyceride (TG), low density lipoprotein-cholesterol ( LDL-C), high density lipoprotein-cholesterol ( HDL-C), body fat content ( BF), and miR-93 were compared between the two groups. The results showed that HOMA-IR, HOMA-β, TG, TC, LDL-C, BF, and miR-93 in PCOS group were significantly higher while HDL-C was significantly lower than those in control group (all P<0.05). HOMA-IR, HOMA-β, TG, TC, and LDL-C levels in patients with Fat≥35% of PCOS group were significantly higher compared with those in patients with BF<35% ( P<0.05) while HDL-C was significantly lower (P<0.05). There were no significant differences in TC and miR-93 between patients with BF≥35% and Fat<35% in PCOS group (P>0.05). HOMA-IR and HOMA-β were positively correlated with BF level (r=0.427 and 0.224, P<0.05), while miR-93 was not correlated with BF level (P>0.05).
6.Surveillance on pinworm infection among rural children in Anhui Province from 2017 to 2021

Objective:
To understand the status of pinworm infection in rural children aged 3-9 years in Anhui Province, and to provide scientific basis for the prevention and control strategy of pinworm disease.
Methods:
According to the National Surveillance Program of Liver Fluke Disease and Soil Transmitted Nematodiasis(Trial), no less than 10% counties(cities and districts) in Anhui Province were selected as mobile surveillance sites every year. Each surveillance site was divided into 5 areas on the basis of geographical location(east, west, south, north and middle), from each of the areas, one administrative village was selected from one township(town, community) for conducting surveillance. Children at age 3-9 years from each site were examined for pinworm infection with the modified Kato-Katz thick smear method and the adhesive cellophane tape perianal swab method. Chi square test was used to compare the infection rate.
Results:
From 2017 to 2021, the 5 year average infection rate of pinworm in rural Anhui was 1.34%(128/9 557), and there was no significant difference in the infection rate over the years( P >0.05). The detection rates of the modified Kato-Katz thick smear method and the adhesive cellophane tape perianal swab method were 0.28% and 1.23%, respectively, the difference was statistically significant( χ 2=72.97, P <0.01). In different regions, the 5 year average infection rate of Fuyang City was the highest(4.27%), and the rate of each city was positively correlated with the number of local resident population( r =0.54, P <0.05). There was no significant sex difference in the 5 year average infection rates( P >0.05). The 5 year average infection rate of children aged 3 to 9 years in rural areas were 0.62%, 1.10%, 1.44%, 1.57%, 0.94%, 2.09% and 1.57%, respectively, showed an increasing trend with the increase of age( χ 2=14.41, χ 2 trend =6.70, P <0.05). There was no significant difference in the average infection rate between scattered children and collectively living children( P >0.05).
Conclusion
From 2017 to 2021, the infection rate of pinworm among children in rural Anhui province remains at a low level. In the future, health education and monitoring should be strengthened.
7.Cerebral cavernous hemangioma in a family with mutation p.L436fs in KRIT1 gene

Objective:To investigate the clinical manifestations and pathogenic gene mutation sites of familial cavernous hemangioma by a pedigree study of this disease.Methods:A family of cerebral cavernous hemangioma who was admitted to the Department of Neurology of the First Affiliated Hospital of Henan University of Science and Technology in April 2019 was diagnosed as cerebral cavernous hemangioma type 1 based on clinical manifestations and head magnetic resonance imaging (MRI), diffusion weighted imaging and susceptibility weighted imaging screening. According to Zabramski classification criteria, the family′s clinical data were collected and genes were sequenced.Results:A 58-year-old female proband had dizziness and headache as the main symptoms, her daughter and son had no clinical symptoms, and her granddaughter had clinical manifestations of cerebral hemorrhage and seizures. The proband and her family members showed multiple cavernous hemangioma on cranial MRI,and the p.L436fs mutation in the KRIT1 gene of familial cerebral cavernous malformation type 1 was confirmed through genetic examination, which was consistent with the Zabramski typing results based on head MRI. The mutation site of the familial spongiform malformation type 1 pathogenic gene was found to be p.L436fs in KRIT1 gene, which has not been reported in familial cerebral cavernous hemangioma type 1 until now.Conclusion:A new p.L436fs mutation of KRIT1 gene was found in familial cerebral cavernous malformation type 1, which expands understanding of the clinical manifestations and pathogenic gene mutation sites of familial cavernous hemangioma.
8.Small interfering RNA delivery to the neurons near the amyloid plaques for improved treatment of Alzheimer׳s disease.

Gene therapy represents a promising treatment for the Alzheimer׳s disease (AD). However, gene delivery specific to brain lesions through systemic administration remains big challenge. In our previous work, we have developed an siRNA nanocomplex able to be specifically delivered to the amyloid plaques through surface modification with both CGN peptide for the blood-brain barrier (BBB) penetration and QSH peptide for -amyloid binding. But, whether the as-designed nanocomplex could indeed improve the gene accumulation in the impaired neuron cells and ameliorate AD-associated symptoms remains further study. Herein, we prepared the nanocomplexes with an siRNA against -site amyloid precursor protein-cleaving enzyme 1 (BACE1), the rate-limiting enzyme of A production, as the therapeutic siRNA of AD. The nanocomplexes exhibited high distribution in the A deposits-enriched hippocampus, especially in the neurons near the amyloid plaques after intravenous administration. In APP/PS1 transgenic mice, the nanocomplexes down-regulated BACE1 in both mRNA and protein levels, as well as A and amyloid plaques to the level of wild-type mice. Moreover, the nanocomplexes significantly increased the level of synaptophysin and rescued memory loss of the AD transgenic mice without hematological or histological toxicity. Taken together, this work presented direct evidences that the design of precise gene delivery to the AD lesions markedly improves the therapeutic outcome.
