Acta Universitatis Medicinalis Anhui 2015;(8):1115-1118
Objective To investigate the protective effects of melatonin and possible mechanisms on rats with alco-holic fatty liver (AFL). Methods All rats were randomly divided into 4 groups: normal group (n = 10), model group (n = 12) and melatonin groups (10 mg / kg, 20 mg / kg; n = 10, respectively). The model of rats’ alcoholic fatty liver was induced by intragastric influsion of ethanol for 8 weeks. The melatonin groups’ rats received melato-nin by intraperitoneal injection after intragastric infusion of ethanol. Histopathological changes were evaluated by hematoxylin and eosin staining. The expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected by immunohistochemical methods. The detection of aspartate aminotransferase (AST) levels and alanine transarninase (ALT) levels and the total bilirubin ( TBIL) levels in serum were provided by routine laboratory methods using an autoanalyzer. The levels of malondialdehyde ( MDA) and activities of glutathione peroxidase ( GPx) were measured by spectrophotometry. Results Compared with the normal group, the liver cells of the mod-el group showed obvious steatosis and significant swelling. However, less degree and less extensive of steatosis and swelling were observed in the melatonin groups. Compared with the normal group, the levels of ALT, AST and TBIL in serum and the levels of MDA in liver homogenates were significantly increased in the model group (P <0. 01), and the activities of GPx were distinctly decreased in the model group(P < 0. 01). But in the melatonin groups, the levels of ALT, AST and TBIL in serum and the levels of MDA in liver homogenates were decreased (P< 0. 01), and the activities of GPx were increased (P < 0. 01). Additionally, melatonin lessened the expression of TNF-α and IL-6 in liver obviously (P < 0. 01). Conclusion Melatonin may inhibit the development of alcoholic fatty liver and its possible mechanism is the ability to resist oxidative stress and lessen the expression of TNF-α and IL-6 and other relevant factors in liver.

Journal of Guangzhou University of Traditional Chinese Medicine 2001;0(03):-
Quality of ulcer healing (QUH) was applied to explore the mechanism of the recurrence of peptic ulcer. The relationship of QUH and epidermal growth factor, prostaglandins, heat shock proteins, immune function and Helico-bacter pylori was assayed. Mechanism of Chinese herbal medicine for the recurrence of peptic ulcer was presented as follows: improving blood circulation and barrier function of gastric mucosa, eliminating Helicobacter pylori and increas ing QUH.

AIM:To study the effect of bcr - abl gene antisense phosphorothioate oligonucleotides(Aspo) on K562 cell line and explore its significance in chronic myelogencous leukemia (CML) gene therapy. METHODS: Cells were exposed to oligomers, observed by inverted microscope. Cells inhibitory rate were determined by 0.4% trypan blue exclusion. CFU - K562 were cultured in 0.8% methylcellulose. P210 was measured by flow cytometry. RESULTS:K562 cells were treated with Aspo, they still grew in clone state and show antisense sequence specific and dose dependent. When the concentration of Aspo was more than 5?mol/L, the growth of cells was inhibited and P210 was down regulated or completely suppressed, and the greatest growth inhibition was at 120h. There was significant inhibition of cell proliferation in a rang of cells number from 1 ? 10-4/mL to 5 ? 10-4/mL after treatment with 10?mol/L Aspo. b2a2 Aspo was also effect on K562 cells which expressing b3a2 mRNA. CONCLUSION: bcr - abl Aspo has a specific growth inhibition effect on K562 cells, and worths further study in CML gene therapy.

A UPLC-ESIMS method was developed for simultaneous analysis of seven major bioactive flavonolignans in silymarin including silychristins A(1) and B(2), silydianin(3), silybins A(4) and B(5), and isosilybins A(6) and B(7). In this study, the seven major active flavonolignans including the diastereomers 1/2, 4/5, and 6/7 were completely separated using UPLC with an ACQUITY UPLC C18 column and MeOH-water (formic acid) mobile phase system. The collision-induced dissociation (CID) MS/MS spectra of these flavonolignans were studied systematically using ESI-MS. The results with the present methodology showed that UPLC-MS/MS can be used for general screening of active natural products from plant extracts and for the specific quality control of silymarin.

Chinese Journal of Radiological Medicine and Protection 2021;41(11):851-855
Objective:To study the feasibility of using pelvic iterative cone beam CT images for dose calculation of radiation therapy planning, so as to provide support for adaptive radiotherapy. Methods:The CIRS 062 M phantom was scanned by Varian Halcyon v2.0 o-ring accelerator, and the average CT number under different scattering conditions was calculated, and then the ICBCT-ED conversion curve was established. CT images of CIRS 002PRA pelvic IMRT phantom and ICBCT images at different positions were collected. Treatment plan using VMAT technique based on CT image was designed and transplanted into ICBCT image with dose recalculated. The differences of gamma passing rate among target volume, organs at risk and 3-dimensional dose were compared. Based on the actual treatment plan of patients, the differences of 3-dimensional dose gamma passing rate in 10 pelvic patients were analyzed retrospectively. Results:There was a large CT value deviation at central position between the isolated no-scattering condition and the full-scattering condition, and the maximum deviation was 144 HU. The CT values of other positions in full-scattering condition were similar to those of the central position, and the maximum deviation was less than 50 HU. Based on the calculated result of ICBCT images at different positions of the pelvic phantom, the dose deviation of the target volume or organs at risk was less than 1 Gy. Compared with the plan based on CT images, the average 3-dimensional dose gamma passing rate under the criteria of 1% dose difference (DD)/1 mm distance-to-agreement (DTA) and 2% DD/2 mm DTA in plan based on ICBCT images were (88.86 ±1.18)% and (98.38±0.89)%, respectively. The ranges of average 3-dimensional dose gamma passing rate under the criteria of 2% DD/2 mm DTA and 3% DD/3 mm DTA in 10 patients with pelvic tumors were 90.03%-95.43% and 93.58%-97.78%, respectively, and the worst result was only 85.90% and 92.90%, respectively. The main reason of the worst result was the dose difference caused by large variation of bladder contour due to over-filling.Conclusions:Under comprehensive scattering conditions, the ICBCT-ED conversion curve is reconstructed and the treatment plan can be designed by using the ICBCT image of Halcyon v2.0 linear accelerator. The accuracy meets the standards of clinical application, which provides assurance for adaptive radiotherapy in the future.
