1.Study on relationship between serum cystatin C and AOPP with early preeclampsia pregnancy outcome

Objective:To discuss the relationship between serum advanced oxidation protein products (AOPP)and cystatin C with the early preeclampsia pregnancy outcome.Methods:Clinical data of 75 patients received treatment and labor at our hospital from 2011 to 2013 was retrospectively analyzed.The control group included 60 normal pregnant women who received prenatal examination in our hospital at the same time.The difference in cystatin C and AOPP level of the patients with preeclampsia and the normal group was compared.The clinical data and the adverse pregnancy outcome of the patients with preeclampsia in different severity and different outcome of pregnancy were compared.Results:A total of 135 patients were retrospective analyzed ,including 75 in observe group and 60 in control group.The SBP level in the patients with preeclampsia is obviously higher than in the normal group .The level of SB,UA,Cr and AOPP in the patients of mild group is obviously lower than in the severe group.The incidence of oligohydramnios , placental abruption,fetal growth restriction and HELLP syndrome in the patients of mild group was obviously lower than in the severe group .The differences were statistically significant.The level of cystatin C,UA,Cr and AOPP in the patients of adverse pregnancy outcome were obviously higher than in the group of good outcome of pregnancy.Conclusion:The cystatin C and AOPP level of the early preeclampsia patients is obviously increased and it has significant correlation with the pregnancy outcome in patients .

3.Therapeutic value of endoscopic resection of tumors originated from gastric fundus muscularis propria

Objective To investigate the safety and effectiveness of endoscopic resection of tumors originated from gastric fundus muscularis propria.Methods Data of 53 patients with tumors originated from gastric fundus muscularis propria detected by endoscopic ultrasonograpy,treated by endoscopic resection and followed up at our hospital between January 2012 and June 2014 were reviewed.The postoperative pathology and complications were retrospectively analyzed to evaluate the therapeutic effect and safety.Results The procedure was successfully performed on all patients and all lesions were removed in one procedure.The lesion size ranged from 0.5 to 4.5 cm and the operation time was 25-155 min[mean(46.7 ±18.2)min].Mild bleeding (5 ~150 ml)occurred in all cases,which was successfully managed by argon plasma coagulation,hot biopsy probe or endoclip.Perforation occurred in 8 patients(8 /53),seven of whom were closed with titanium clips and titanium clips combined with nylon cord.Laparoscopic intervention was applied to 1 case because of severe perforation.Gastrointestinal decompression,acid suppression with proton pump inhibitors and antibiotics were performed on all cases.No severe hemorrhage occurred.The average length of hospitalization was (5.3 ± 1.4)days(3-14 d).Pathology confirmed 46 cases of gastrointestinal stromal tumors and 7 cases of leiomyoma. The patients were followed up for 3 to 27 months,and no tumor residue or recurrence was observed. Conclusion Endoscopic resection is a method not only to get the accurate pathologic diagnosis but also to meet principle of the local resection for stomach.It is safe,effective and worthy of recommendation.
4.Establishment of cisplatin-resistant human endometrial cancer cell line and the study of its resistant mechanisms

Objective To establish the cisplatin(DDP)-resistant cell line from human endometrial cancer cell line Ishikawa and to investigate its resistant mechanism to DDP. Methods A resistant endometrial cancer cell line ISH/DDP was established by gradually increasing dose of cisplatin and high-dose stimulation. The resistant index was estimated by 3-(4,5-dimethylthiazol-zyl)-5-(3-carboxymethoxyphenyl-2-(4-sulfophenyl)-2H-tetrazolium,inner salt (MTS) assay. Cell growth curve, doubling time and cell cycle phase distribution were measured; drug-resistant protein of breast cancer resistance protein (BCRP),P-glycoprotein (P-gp) and glutathione-S-transferase-π (GST-π) were examined by immuocytochemistry. Results The DDP-resistant cell line ISH/DDP was established with the resistant index of 3. 77. The proliferation of ISH/DDP got slow, doubling time prolonged, which were 40. 1 hours, while it was 34. 1 hours in Ishikawa (P<0.05); and the cell number of G0/G1 phase [(44. 3 ±5. 7)% and (39. 0 ± 10. 7)% ,P >0. 05] and G2/M phase [(11.9 ±0.7)% and (5. 7 ±2. 4)% ,P <0. 05] decreased, while S-phase [(44. 2 ±6.1)% and (55. 3 ± 8. 4) % , P < 0. 05] increased compared with parent cells. The comprehensive score of the expression of BCRP in ISH/DDP was 16. 3 ± 2. 0, while it was 13.4 ± 1. 5 in Ishikawa (P < 0. 05). The score of the expression of P-gp in ISH/DDP and Ishikawa were 15. 5 ± 1. 2 and 16. 1 ± 1. 0 (P > 0. 05) ,respectively. The score of the expression of GST-π in ISH/DDP and Ishikawa were 15. 2 ± 1. 9 and 14. 9 ± 1.1 (P > 0. 05) . Conclusion ISH/DDP cell line showed a typical resistant phenotype and biological characteristics, which may be accounted for high BCRP expression.
5.Repair of UVB damage in mouse skin by adipose derived stem cells

Objective:To explore the therapeutic effect of adipose-derived stem cells (ADSC) on long-wave UV damage in mouse skin in order to provide ideas for the treatment of skin photodamage.Methods:The inguinal and perirenal adipose tissues of C57BL/6 mice were extracted and processed to obtain mouse ADSCs, and the surface markers, adipogenic and osteogenic differentiation capabilities were identified. The mouse photoaging model was irradiated with the SS-03AB UV illuminator, the total UVB dose was 9.45 J/cm 2, and the total UVA dose was 94.5 J/cm 2. Experimental mice (72 in total) were divided into normal group, model group, DMEM (medium) group and ADSC group, each with 18 mice. In the normal group and model group, the materials were taken two weeks after the end of irradiation. After irradiation, the ADSC group was given a subcutaneous injection of 200 μl ADSC suspension, and the DMEM group was given 200 μl of serum-free medium for treatment, and the materials were taken for pathological staining after 2 weeks. The experimental data was processed by analysis of variance. This study was carried out from August 2018 to July 2019 in the First Affiliated Hospital of Zhengzhou University. Results:The extracted cells were identified as adipose-derived stem cells. HE staining showed that the inflammatory cell infiltration in the ADSC group was significantly reduced compared with the DMEM group ( t=20.649, P<0.001) and the normal group ( t=16.147, P<0.001), and the thickness of the dermis layer was significantly increased. Masson staining showed collagen fibers were arranged neatly and the density increased significantly after ADSC treatment. Conclusions:Subcutaneous injection of ADSC can reduce inflammation, promote collagen tissue proliferation, increase the thickness of the dermis, effectively resist inflammatory damage and collagen breakdown caused by UVB.

Parkinson's disease (PD) is a common neurodegenerative disease characterized by bradykinesia,resting tremor,muscle rigidity,and abnormal gait posture.Ocular motor function test plays an important part in neurological examinations.It has been widely accepted that specific ocular motor patterns contribute to diagnosis of Parkinsonian syndrome including progressive supranuclear palsy and multiple system atrophy.However,recent studies have shown that patients of PD may also exhibit specific eye movement disorders,which will be helpful in the early diagnosis,evaluation and differential diagnosis of PD.In 2015,the Movement Disorder Society clinical diagnostic criteria for Parkinson's disease suggested that sustained staring evoked nystagmus could be used as an exclusive criterion for PD.The clinical features,detection methods and clinical significance of oculomotor dysfunction in PD are reviewed in this article.
7.Study on the clinical features and related factors of cognitive impairment in Parkinson's disease

Objective To explore the correlated factors and clinical features of cognitive impariment in parkinson's disease (PD).Methods A total of 419 patients with PD were collected from Xiangya Hospital of Centre-South University during Mar 1st,2017 to Nov 30th,2017.The cognitive functions of patients were assessed with the Mini-Mental State Examination (MMSE),and the basic information and the motor symptoms of 419 PD patients were selected at the same time.The PD patients were classified into three groups according to the MMSE score:PD with no cognitive impairment (PD-NC),mild cognitive impairment in PD (PD-MCI),and Dementia in PD (PD-D).The data were analyzed by SPSS 20.0.Results There were 156 patients with PD-MCI (37.2%) and 64 patients with PD-D (15.3%).The difference of sex and disease duration among three groups were not statistically significant (P > 0.05).The significant difference was found among PD-D,PD-MCI,and PD-NC groups in age of onset,age,educational attainment,Unified Parkinson's disease Rating Scale (UPDRS)-Ⅱ score,UPDRS-Ⅲ score and Hoehn-Yahr stage (P < 0.05).There were significant differences among three groups in MMSE score and its items (P < 0.01).Logistics regression analysis found that the age of onset,educational attainment,and Hoehn-Yahr stage were the risk factors of cognitive impairment in PD patients (P < 0.05).Conclusions Cognitive impairment is common in PD patients,and it is relevant to the age of onset,educational attainment and the severity of illness of PD patients.
8.Tube-nesting EP:A new faster method to get more mouse bone marrow cells

Objective:To explore a method to obtain more mouse bone marrow cells more quickly.Methods:Take both lower limbs of the same mouse,the bone marrow cells were obtained by either the tube-nesting EP method or the traditional syringe irrigation method,recorded the time and quantity of bone marrow cells,to analyze the proportion of viable cells by flow cytometry,and to detect the proliferation of these bone marrow cells used CCK-8 method.Results:Compared with the traditional syringe irrigation method,the time of obtaining mouse bone marrow cells by tube-nesting EP method was shorter(P<0.000 1).Compared with the traditional syringe irrigation method,the number of bone marrow cells obtained by tube-nesting EP method was more(P<0.05).The proportion of live cells and proliferation ability of bone marrow cells obtained by the two methods were identical.Conclusion:Tube-nesting EP method is a new faster method to get more mouse bone marrow cells.
9.NOTCH3 gene mutations in two Chinese families featuring cerebral autosomal dominant arteriopathy with subcortical infarct and leucoencephalopathy.

OBJECTIVETo analyze potential mutations of the NOTCH3 gene in two Chinese families featuring cerebral autosomal dominant arteriopathy with subcortical infarct and leucoencephalopathy (CADASIL).
METHODSThe two probands and related family members and 100 healthy controls were recruited. Potential mutations of the NOTCH3 gene were screened by PCR and direct sequencing. PolyPhen-2 and SIFT software were used to predict the protein function.
RESULTSThe conditions of both probands were adult-onset, with main clinical features including recurrent transient ischemic attacks and/or strokes, cognitive impairment. MRI findings suggested multiple cerebral infarcts and severe leukoencephalopathy. A heterozygous mutation c.328C>T (p.Arg110Cys), which was located in exon 3 of the NOTCH3 gene and known as a causative mutation, was identified in proband 1. A novel heterozygous mutation c.1013 G>C (p.Cys338Ser) located in exon 6 of the NOTCH3 gene was identified in the proband 2, which was not reported previously. The same mutations were not detected among the 100 unrelated healthy controls. Function analysis suggested that heterozygous mutation c.1013G>C can severely affect the functions of NOTCH3 protein.
CONCLUSIONTwo heterozygous missense mutations in the NOTCH3 gene have been identified in two families affected with CADASIL. The novel heterozygous Cys338Ser mutation in exon 6 of the NOTCH3 gene probably underlies the CADASIL.

OBJECTIVE:
To investigate the mutation of small sequence changes in microRNA-7 gene in Chinese patients with Parkinson's disease (PD).
METHODS:
We analyzed miR-7 variants in 225 PD patients from Chinese Han group by DNA sequence.
RESULTS:
None of the patients had miR-7 variants.
CONCLUSION
MiR-7 variation is not associated with PD in Chinese patients.
