1.Characteristics of boningmycin induced cellular senescence of human tumor cells

Cellular senescence is one of the important steps against tumor. This study was to observe the characteristics of boningmycin induced senescence of human tumor cells. MIT method and clone formation assay were used to detect the growth-inhibitory effect. Cellular senescence was detected with senescence-associated beta-galactosidase staining. Cell cycle distribution and accumulation of intracellular reactive oxygen species (ROS) were analyzed with flow cytometry. Protein expression was detected by Western blotting. The results showed that the growth-inhibitory effect of boningmycin was obviously stronger on human oral epithelial carcinoma KB cells than that on non-small cell lung cancer A549 cells. Comparison to the similar action of doxorubicin, that boningmycin induced the features of cellular senescence in both cell lines, its due to the arrest at G2/M phase and an increase of ROS level. The molecular senescence marker P21 increased significantly after boningmycin treatment at a dosage of 0.1 micromol x L(-1), whereas a higher concentration of it induced apoptosis. The results indicated that cellular senescence induced by boningmycin was one of its mechanisms in tumor suppression.

Objective To observe the changes of mRNA expression related to kidney yang deficiency and the ul-trastructure of hypothalamus in Lewis rats, and further study the constitution of kidney yang deficiency in Lewis rats.Meth-ods Ten 7-8-week old SPF male Lewis rats and twenty 7-8-week old SPF male Wistar rats were used in this study.The rats received subcutaneous injection of hydrocortisone to establish kidney yang deficiency model.The expressions of TNF-α, IFN-γ, IL-10, CRH, MR and GR mRNA in the hypothalamus of the two groups were detected and the ultrastructural changes of hypothalamus and adrenal gland were observed to compare the differences between the two groups, and to ex-plore the mechanism of kidney yang deficiency constitution in the Lewis rats.Results Compared with the normal Wistar rats, the expressions of GR and IL-2 mRNA in the hypothalamus were significantly increased ( P<0.01 ) , while the ex-pressions of CRH, TNF-α, IFN-γand IL-10 mRNA were significantly decreased in the Lewis rats (P<0.01).The zona fasciculata of the adrenal cortex was slightly thinner, the mitochondria were slightly swollen and the amount of mitochondria was decreased.The nuclei of hypothalamic neurons were larger, and the mitochondria, neuron synapses and secretory vesi-cles in the presynaptic neurons were decreased.Conclusions There are abnormal expression of immune-related cytokines and CRH mRNA, and ultrastructural changes in the hypothalamus and adrenal gland of Lewis rats with kidney yang defi-ciency.Such changes may be related to the functional imbalance of the hypothalamus-pituitury-adrenal axis.
3.Study on dynamic changes and gender difference of heart rate variability and blood pressure variability via implantable telemetry technology in spontaneously hypertensive rats

Objective To observation dynamic changes and gender difference of heart rate variability and blood pressure variability in unbonded spontaneously hypertensive rats using Telemetry technology .Method Taking sixteen spontaneously hypertensive rats of 3-month-old in SPF grade, there were implanted TL11M2-C50-PXT device intraperitoneally.Seven days later, respectively monitoring chainless blood pressure and electrocardiogram for 24 h continuously time using telemetry system when 3, 5, 7 months of age in spontaneously hypertensive rats .Analysing 24 h systolic pressure , mean blood pressure , diastolic blood pressure , heart rate variability and blood pressure variability . Result Systolic pressure , diastolic pressure and mean pressure , blood pressure variability increased with age of the moon increased in spontaneously hypertensive rats , moreover female SHR rats of blood pressure variability was significantly lower than that of the male rats .Correlated indexes of heart rate variability had no obvious change Between three to seven month -old in male spontaneously hypertensive rats .But RR interval , SDNN, TP, VLF and HF increased significantly with age of the moon increased in female spontaneously hypertensive rats , moreover the female SHR rats of TP , VLF and HF index of heart rate variability were lower than the male rats .Conclusion With the SHR rats age and blood pressure , blood pressure variability increased , and the female SHR rat variability in blood pressure and heart rate was significantly lower than that of the male .
4.Kidney yang deficiency constitution and hypothalamic-pituitary-adrenal axis dysfunction in Lewis rats

ObjectiveToinvestigatethekidneyyangdeficiencyconstitutionandhypothalamus-pituitary-adrenal ( HPA) axis dysfunction in Lewis rats .Method Two kidney yang deficiency models were established by subcutaneous injection of hydrocortisone and by adrenalectomy to induce hypothalamic -pituitary-adrenal ( HPA) axis dysfunction in Lewis rats.Wistar rats were used as control and compared with the two types of Lewis rat models of kidney yang deficiency .The degree of kidney yang deficiency of the Lewis rats was evaluated by examination and detection of the general signs , behavior , and neuroendocrine function , and so on .Result Compared with the normal Wistar rats , the body weight of Lewis rats was significantly higher than that of Wistar rats at the same age .The body temperature , urine volume and grip strength were significantly lower than those of Wistar rats ( P<0.01 for all ) .The memory ability of the Lewis rats was slightly decreased.The liver, kidney and adrenal indexes were significantly decreased in the Lewis rats (P<0.01).The levels of serum ACTH, CRH, cGMP, Cort and urine 17OHCS were significantly decreased (P<0.01).Conclusion A very slight deficiency of adrenal cortex function of Lewis rats is caused by genetic inheritance , and Lewis rats have the constitution of congenital kidney yang deficiency ( criticality or prophase of kidney yang deficiency ) .
5.Study on influencing factors of seroma after laparoscopic preperitoneal tension-free inguinal hernia repair

Objective:To explore the influencing factors of seroma after transabdominal preperitoneal prosthetic (TAPP) inguinal hernia by laparoscopy.Methods:A retrospective cohort study was used to analyze the clinical data of 320 patients with inguinal hernia who received TAPP in Fuyang Fifth People′s Hospital from December 2019 to December 2022, including 226 males and 94 females, with an average age of (61.46±10.22) years (range: 23-76 years). Patients were divided into seroma group ( n=18) and non-seroma group ( n=302) according to whether seroma occurred after surgery. Multivariate Logistic analysis was used to screen the influencing factors of seroma after TAPP tension-free repair, and based on Softmax strategy, the artificial neural network model was constructed with binary classification variables survival 0 (no outcome event occurred) and 1 (outcome event occurred) as outcome variables. receiver operating characteristics (ROC) and cumulative gain graph were used to analyze the model differentiation and application value. The measurement data of normal distribution were expressed as mean±standard deviation ( ± s), and independent sample t-test was used for comparison between groups. Chi-square test was used for comparison between count data groups. Results:The incidence of postoperative seroma in 320 patients was 5.63% (18/320), including 7 cases of type Ⅰ, 4 cases of type Ⅱ, and 7 cases of type Ⅲ, all of which were improved after symptomatic treatment. Combined with underlying diseases, anticoagulant drugs, duration of disease, operation time, intraoperative blood loss, hernia sac diameter, mesh fixation method, neutrophil to lymphocyte ratio (NLR) were factors influencing seroma ofter TAPP in inguinal hernia patients ( OR=1.732, 2.414, 2.346, 1.480, 2.159, 1.725, 1.248, 2.179; 95% CI: 1.385-2.942, 1.764-3.176, 1.280-3.209, 1.263-2.275, 1.331-2.861, 1.308-2.239, 1.005-1.764, 1.644-2.982; P<0.05). The ROC curve and cumulative gain graph showed that the artificial neural network model could well predict the probability of postoperative seroma. Conclusions:The occurrence of seroma after TAPP tension-free repair in inguinal hernia patients is related to underlying diseases, taking anticoagulant drugs, course of disease, operation time, intraoperative blood loss, diameter of hernia sac, patch fixation method, NLR and many other factors. Clinical attention should be paid to these problems to reduce the incidence of postoperative seroma.
6.Preparation and Biocompatibility Study of Contrast-Enhanced Hernia Mesh Material

BACKGROUND:
Meshes play a crucial role in hernia repair. However, the displacement of mesh inevitably leads to various associated complications. This process is difficult to be traced by conventional imaging means. The purpose of this study is to create a contrast-enhanced material with high-density property that can be detected by computed tomography (CT).
METHODS:
The contrast-enhanced monofilament was manufactured from barium sulfate nanoparticles and medical polypropylene (PP/Ba). To characterize the composite, stress tensile tests and scanning electron microscopy (SEM) was performed. Toxicity and biocompatibility of PP/Ba materials was verified by in vitro cellular assays. Meanwhile, the inflammatory response was tested by protein adsorption assay. In addition, an animal model was established to demonstrate the long-term radiographic effect of the composite material in vivo. Subsequent pathological tests confirmed its in vivo compatibility.
RESULTS:
The SEM revealed that the main component of the monofilament is carbon. In vitro cell experiments demonstrated that novel material does not affect cell activity and proliferation. Protein adsorption assays indicated that the contrast-enhanced material does not cause additional inflammatory responses. In addition, in vivo experiments illustrated that PP/Ba mesh can be detected by CT and has good in vivo compatibility.
CONCLUSION
These results highlight the excellent biocompatibility of the contrast-enhanced material, which is suitable for human abdominal wall tissue engineering.
