1.In vitro study of bone marrow mesenchymal stem cells differentiated into dopaminergic neuron-like calls

Objective Bone marrow mesenchymal stem cells (BMSCs) were induced to differentiate to the special histological types of neurons in vitro.The morphological change of cells and positive expression of specific antigen on membrane were studied,and the function of connection between the induced BMSCs was also detected.The feasibility of BMSCs differentiate to the special histological types of neurons was investigated.Methods BMSCs were divided into group Ⅰ (induced with bFGF+GDNF),group Ⅱ (induced with bFGF+GDNF+WHI-P131 +Shh),and control group (no revulsive).The morphologic change of cells was observed,and the positive rate of neuron specific surface antigen and the content of dopamine were detected.Formation of mature synaptic structure was detected by immunohistochemical assay of postsynaptic density protein 95 (PSD-95) expression,and synaptic loop was shown by FM1-43 stain synaptic vesicles.Results By immunohistochemical staining,the positive rates of dopamine transporter (DAT) and tyrosine hydroxylase (TH) in group Ⅱ were significantly higher than those in group Ⅰ,and dopamine can been detected in cell culture supematant of group Ⅱ.After BMSCs was induced into dopamine neuron-like cells,number and length of cell protrusions,positive rate of PSD-95 and fluorescence intensity of FM1-43 in group Ⅱ were significantly higher than those of group Ⅰ.Conclusions There were no significant change in positive rate of neuron-specific surface markers,rate of cell survival and differentiation rate after BMSCs differentiated to dopaminergic neuron-like cells.The number and length of cell protrusions,content of dopamine in cell culture supematant,positive rate of dopaminergic neuron-specific surface antigen (DAT and TH),synaptic function index (positive rate of PSD-95 and fluorescence intensity of synaptic loop) of group Ⅱ were all significantly higher than that of group Ⅰ.
2.The clinical significance of ultrasound guided biopsy of peritoneum in diagnosis of unexplained ascites

Objective To investigate the clinical significance of ultrasound guided biopsy of peritoneum in diagnosis of unex-plained ascites .Methods A total of 95 patients with unexplained ascites after conventional examination were collected ,then under-went ultrasound inspection which showed thickened peritoneum and the relations to surrounding tissues around the planned perito-neum puncture point ,and conducted the peritoneal biopsy .Results A total of 95 were successfully biopsied with 2 -4 peritoneal tissues .Pathological examination showed that 37 cases (38 .9% ) were diagnosed as peritoneal metastatic carcinoma ,41 cases (43 . 1% ) with peritoneal tuberculosis ,5 cases (5 .2% ) with peritoneal mesothelioma ,1 case (1 .1% ) with eosinophilic granuloma ,4 ca-ses (4 .2% ) with fibrous hyperplasia ,and 7 cases (7 .4% ) with chronic inflammatory cell infiltration .The positive rate of ultra-sound guided peritoneal biopsy was 88 .4% .Conclusion The ultrasound guided biopsy of peritoneum is of clinically great signifi-cance in diagnosis of unexplained ascites .
3.Comparison of the effects of meal replacements and a standard food-based low calorie diet on weight loss and glycolipid metabolism

Objective To investigate the effects of a standard food-based low calorie diet and meal replacements on compliance,weight loss,and glycolipid metabolism in obese patients,and to provide an optimal nutritional weight reduction method for obese individuals in China.Methods Height,weight,waist circumference,body mass index (BMI),body fat,visceral fat in low calorie diet group (n =23) and meal replacements group (n =17) were assessed at baseline,4 and 8 weeks after the intervention.Fasting plasma glucose (FPG),triglyceride (TG),total cholesterol (TC),high density lipoprotein-cholesterol (HDL-C),low density lipoprotein-cholesterol (LDL-C),aspartate aminotransferase (AST),alanine transaminase (ALT),gamma-glutamyl transpeptidase (GGT),alkaline phosphatase (ALP),fasting insulin (FINS),and homeostasis model assessment for insulin resistance (HOMA-IR) were assessed at both baseline and 8 weeks after the intervention.Results Both low calorie diet and meal replacements led to significant decreases in body weight,waist circumference,BMI,body fat,and visceral fat(all P<0.01) after 4 and 8 weeks.Body fat (P< 0.01) and total body water (P< 0.05) in low calorie diet group were decreased more obviously compared with those in meal replacements group at 4 weeks.However,there were no difference in all these parameters between two groups at 8 weeks (P>0.05).Both groups experienced improvements in biochemical outcomes and metabolic indicators.TG,AST,ALT,GGT,ALP,FINS,and HOMA-IR levels were all decreased (P<0.05 or P<0.01) while HDL-C level was increased (P<0.01) 8 weeks after standard food-based low calorie diet.FPG,TG,TC,LDL-C,AST,ALT,ALP,FINS,and HOMA-IR levels were declined 8 weeks after meal replacements(P<0.05 or P<0.01).TC,LDL-C,and ALT levels were more evidently to reduce in meal replacements group compared with low calorie diet group.Conclusions Present data suggest that meal replacements and low calorie diet may have a similar effect on weight loss,while meal replacements seems to obtain a better metabolic results.
4. Combined treatment of perianal condyloma acuminatum inHIV seropositive men who have sex with men with microwave and photo-dynamic therapy

Objective:
To analyze the clinical features of perianal condyloma acuminatum in HIV infected men who have sex with men (MSM) and assess the effects of combined treatment of microwave and photodynamic therapy.
Methods:
Clinical and laboratory data of 260 male patients with perianal condyloma acuminatum were collected and microwave combined with photodynamic therapy was applied on them.
Results:
The majority of the patients with perianal condyloma acuminatum were young people (243/260). Warts were mainly located in the anus or from the outside to inside of the anus (245/260). Among them, the proportion of warts position of extraanal in HIV-infected group (8/220) was lower than that of non-HIV-infected group (7/40). HPV types 11 (70/118), 6 (44/118) and 16 (40/118) were the most common types of HPV infection. People with HIV infection were more likely to develop high-risk, mixed types HPV infection and multiple HPV infection than those without HIV infection. More than half of the patients (146/260) needed only 1-3 sessions of photodynamic treatments. There was no significant difference in the times of photodynamic treatments and the numbers of photosensitizers required between the HIV-infected and non-HIV-infected groups. The recurrence rate of HIV-infected group (13.6%, 30/220) was also similar to that of non-HIV-infected group (15.0%, 6/40), but far lower than those who underwent microwave therapy alone (45%, 18/40).
Conclusions
The combination therapy significantly reduced the recurrence rate of perianal condyloma acuminatum in HIV infected MSM.
